Ultrasonic Sludge Disintegration for Improving Anaerobic Digestion and Simulation of ADM1 (혐기성 소화효율 향상을 위한 초음파를 이용한 슬러지 전처리 및 ADM1 모의)

  • The objectives of this study were to demonstrate that enhanced anaerobic digestion could be achieved by adopting ultrasonic cavitation pretreatment on the basis of the biogas production and to compare the simulation results of ADM1 (Anaerobic Digestion Model No. 1) with results of the experimentally operated digester the ultrasonic pretreatment of sewage sludge showed the hindered effect on the dewaterbility and the increase of SCOD production. In this study, four sets of lab-scale anaerobic digester were operated with untreated(control), 30 min, 60 min and 90 min ultrasonic pretreated sludge. TCOD removal efficiencies in digesters of control, 30, 60, 90 min sonicated sludge were 31.9%, 37.9, 38.5% and42.2%, and 75 removal were 15.9%, 20.8%, 21.5%, 24.1% respectively. Also more biogas was produced gradually with the increased sludge loading and the pretreatment time. Overall the simulation results had a correspondence tendency with the experimental efficiencies.

  • Biogas created from animal waste is a precious energy source. A practical and successful utilization of the biogas is not easy, because there lie some difficulties in biogas production and facilities investment. In this study, the requisites for a successful biogas utilization were discussed. The production results obtained in the previous operation of anaerobic digestion plant were used for the simulation. When the slurry heating was designed for constant biogas generation, depreciation costs of the facilities amounted 1,175,000 yen per year, and biogas productions at $24.5^{\circ}C$, $30.0^{\circ}C$ and $35.5^{\circ}C$ were $16.8m^3$, $17.6m^3$ and $25.1m^3$, respectively. Removal ratios of organic matters were not so high. At $35.5^{\circ}C$, energy value of the biogas produced was estimated 125.5 Mcal per day, and the following heat loss (y Mcal/day) was brought about by the temperature difference ($X^{\circ}C$) between the digester and atmosphere; y = 0.769X - 5.375. The costs of biogas production per cow were assumed to decrease according to enlargement of feeding scale, especially on scales of more than 30 cows. On recent levels of costs and prices of energy in Japan, they were nearly equal to 2 to 3 fold of the price of municipal mixed gas when a anaerobic digester was compulsorily heated and kept at $30.0^{\circ}C$ or $35.5^{\circ}C$.

  • Small-scale household digesters have been promoted across Asia as a sustainable way of handling manure. The major advantages are that they produce biogas and reduce odor. However their disadvantages include the low recycling of nutrients, because digestate is dilute and therefore difficult to transport, and the loss of biogas as a result of cracks and the intentional release of excess biogas. In this study, life cycle assessment (LCA) methodology was used to assess the environmental impacts associated with biogas digesters in Vietnam. Handling 1,000 kg of liquid manure and 100 kg of solid manure in a system with a biogas digester reduced the impact potential from 4.4 kg carbon dioxide ($CO_2$) equivalents to 3.2 kg $CO_2$ equivalents compared with traditional manure management. However, this advantage could easily be compromised if digester construction is considered in the LCA or in situations where there is an excess of biogas which is intentionally released. A sensitivity analysis showed that biogas digesters could be a means of reducing global warming if methane emissions can be kept low. In terms of eutrophication, farms with biogas digesters had 3 to 4 times greater impacts. In order to make biogas digesters sustainable, methods for recycling digestates are urgently required.

Evolutionary Optimization of Pulp Digester Process Using D-optimal DOE and RSM

  • Optimization of existing processes becomes more important than the past as environmental problems and concerns about energy savings stand out. When we can model a process mathematically, we can easily optimize it by using the model as constraints. However, modeling is very difficult for most chemical processes as they include numerous units together with their correlation and we can hardly obtain parameters. Therefore, optimization that is based on the process models is, in turn, hard to perform. Especially, f3r unknown processes, such as bioprocess or microelectronics materials process, optimization using mathematical model (first principle model) is nearly impossible, as we cannot understand the inside mechanism. Consequently, we propose a few optimization method using empirical model evolutionarily instead of mathematical model. In this method, firstly, designing experiments is executed fur removing unecessary experiments. D-optimal DOE is the most developed one among DOEs. It calculates design points so as to minimize the parameters variances of empirical model. Experiments must be performed in order to see the causation between input variables and output variables as only correlation structure can be detected in historical data. And then, using data generated by experiments, empirical model, i.e. response surface is built by PLS or MLR. Now, as process model is constructed, it is used as objective function for optimization. As the optimum point is a local one. above procedures are repeated while moving to a new experiment region fur finding the global optimum point. As a result of application to the pulp digester benchmark model, kappa number that is an indication fur impurity contents decreased to very low value, 3.0394 from 29.7091. From the result, we can see that the proposed methodology has sufficient good performance fur optimization, and is also applicable to real processes.

Molecular Cloning and Characterization of a Novel Cold-Adapted Family VIII Esterase from a Biogas Slurry Metagenomic Library

  • A novel esterase gene, est01, was successfully unearthed from a biogas digester microbiota metagenomic library. The 1,194 bp est01 gene encodes a protein of 44,804 Da (designated Est01). The amino acid sequence of Est01 shows only moderate (33%) identity to a lipase/esterase. Phylogenetic analysis and biochemical characterization confirmed that Est01 is a new member of family VIII esterases. The purified Est01 from recombinant Escherichia coli BL21 (DE3) showed high hydrolytic activity against short-chain fatty acid esters, suggesting that it is a typical carboxylesterase rather than a lipase. Furthermore, the Est01 was even active at $10^{\circ}C$ (43% activity remained), with the optimal temperature at $20^{\circ}C$, and had a broad pH range from 5.0 to 10.0, with the optimal pH of 8.0. These properties suggest that Est01 is a cold-adaptive esterase and could have good potential for low-temperature hydrolysis application.

Organic Matter Degradation and Nitrification Characteristics in Aerobic Digestion of Sewage Sludge (슬러지 호기성 소화공정에서의 유기물 분해 및 질산화 특성)

  • Continuous operation of aerobic sludge digestion reactor was attempted for 279 days. Anaerobic digester sludge, the target material of the experiment, was pretreated by sodium hydroxide at $40^{\circ}C$ for 120 minutes, and the pretreated sludge was fed to 5 L CSTR (continuous stirred tank reactor). Reactor performance was affected by properties of input sludge and HRT (hydraulic retention time). 6 days of HRT showed best and stable performance, and under this condition, removal rates of $NH_3$-N, SCOD, TKN, TCOD, SS, and VSS were 97.4%, 81.7%, 68.7%, 61.4%, 50.6%, and 47.0%, respectively. 73.9% of SS in anaerobic digester sludge was reduced by pretreatment and aerobic digestion. Effluent sludge had low soluble COD of 350 mg/L. This implied the sludge was stabilized and suitable for use as liquid fertilizer. Nitrification took place when HRT was higher than 4 days. $NO_3$-N concentration was as high as 658 mg/L while $NH_3$-N was as low as 20 mg/L.

  • Molten carbonate fuel cell (MCFC) power plants are one of most attractive electricity generation systems for the use of biogas to generate high-efficiency ultra-clean power. However, MCFCs are considerably more expensive than comparable conventional electricity generation systems. The commercialization of MCFCs has been delayed more than expected. After being effective in the Kyoto protocol and considerably increasing the fossil price, the attention focused on $CO_2$ regression and renewable energy sources has increased dramatically. In particular, the commercialization and application of MCFC systems fed with biogas have been revived because of the characteristics of $CO_2$ collection and fuel variety of MCFCs. Better economic results of MCFC systems fed with biogas are expected because biogas is a relatively inexpensive fuel compared to liquefied natural gas (LNG). However, the pretreatment cost is added when using anaerobic digester gas (ADG), one of the biogases, as a fuel of MCFC systems because it contains high $H_2S$ and other contaminants, which are harmful sources to the MCFC stack in ADG. Thus, an accurate economic analysis and comparison between MCFCs fed with biogas and LNG are very necessary before the installation of an MCFC system fed with biogas in a plant. In this paper, the economic analysis of an MCFC fed with ADG was carried out for various conditions of electricity and fuel price and compared with the case of an MCFC fed with LNG.

  • BNR process is an effective method to remove high strength nitrogen included in wastewater and piggery wastewater. There have been many former studies about the induction of nitritation which have many advantages than full nitrification and the impacting factors on nitritation. Especially, it is reported that organic matter has a relation with nitritation. In this study, laboratory sacle reactor was operated using effluent of anaerobic digester, piggery wastewater and anaerobic digester effluent of piggery wastewater. After analyzing the operating results, the impact of organic matter on nitritation was analyzed by classified COD fractions. It was showed that nitritation is affected by organic matter especially by Ss. In conclusion, organic matter should be managed not just as a single gross parameter but in a classified form.

  • Biogas production from agricultural wastes were summarized as follows: 1. Biogas Generation Characteristics of Various Manures and Residues a. Gas yield from crop residues like rice straw, rice hull, corn stalk and coconut husk can be improved by addition of animal manures. b. Gas yield from coconut husk can be improved through aerobic fermentation for at least one week before loading in the digester. c. Gas yield from fresh rice straw is better than from pre-fermented one, whether alone or in combination with animal manures. d. Initial study has shown that fresh azolla can be substituted for animal manures in manurerice straw combinations and gas yield derived based on unit volatile solids loaded is actually better than for manure-residue combinations. e. Gas production is highly sensitive to substrate pH and becomes almost nil at a pH of below 6. 2. Effect of ambient conditions and other factors on biogas production in a house hold-size digester. a. Results showed that compaction of rice straw in straw-manure combination can reduce gas yield compared with loosely mixed straw. b. The effective gas production period extended to 70 days using freshly threshed rice straw and fresh cattle manure as feed material. c. Underground and above ground digesters with shade have relatively more stable substrate temperature than aboveground exposed digesters. This relative temperature instability may likely be the reason for lower gas yield for the exposed aboveground digester loaded with loose straw-cattle manure substrate, compared with the underground digester with the same substrate. 3. Economic Analysis a. Based on prevailing costs of fuel, materials, and labor in the Philippines, biogas produced from the household size system is cheaper than either LPG or kerosene. b. If other benefits like organic fertilizer, pollution control and convenience are considered, biogas will surely be the best alternative fuel source.

Isolation and Identification of Carbon Monxide Utilzing Anaerobe, Eubacterium limosum KIST612 (일산화탄소를 이용하는 혐기성 세균 Eubacterium limosum KIST612의 분리 및 동정)

  • Carbon monoxide (CO)-utilizing acetogens were enriched and KIST612 isolated from anaerobic digester fluid was selected for its abilities to tolerate high CO and acetate concentration. The isolate KIST612 was identified as Eubacterium limosum based on the morphological and biochemical characteristics, G+C content of DNA and 16S rRNA sequence analysis. E. limosum KIST612 produced acetate and butyrate from CO. The optimum temperature and pH for the growth and acids formations were 37$\cdot $C and 7.0, respectively. The growth rate and acids productivity of E. limosum KIST612 were higher than those of any other known acetogens when CO was used as the sole energy and carbon source.
