Up-regulation of Prothymosin alpha in THP-1 Cells Infected with Mycobacterium tuberculosis (결핵균 감염에 의한 THP-1 세포에서의 Prothymosin alpha 유전자 발현증가)

  • Mycobacterium tuberculosis is capable of growing and survival within macrophage. The purpose of this study was to identify the genes regulated by infection of mycobacteria in human monocytic THP-1 cells. We used the differential display reverse transcriptase polymerase chain reaction (DD RT-PCR) and nothern blot analysis to confirm the differentially expressed genes from THP-1 cells infected with live Mycobacterium tuberculosis H37Rv, heat-killed Mycobacterium tuberculosis H37Rv and live Mycobacterium bovis BCG. Among many up or down-regulated clones, 27 clones were sequenced and compared with known genes on GenBank. Thirteen of over-expressed clones from THP-1 cells infected with live Mycobacterium tuberculosis H37Rv were identical to human prothymosin alpha, eight were novel clones and six clones showed homology with Human ferritin H chain, Esherichia coli bgl, Mouse RNA-dependent EIF-2 alpha kinase, E. coli htrL, Hyaluronan receptor and T cell receptor. Our result suggests that Mycobacterium tuberculosis might regulate prothymosin alpha gene transcription in monocytic THP-1 cell.

  • To understand molecular and cellular mechanisms of many gene products in the female reproductive organs including the ovary and uterine endometrium as well as during embryo development, researchers have developed and utilized many effective methodologies to analyze gene expression in cells, tissues and animals over the last several decades. For example, blotting techniques have helped to understand molecular functions at DNA, RNA and protein levels, and the reverse transcription-polymerase chain reaction (RT-PCR) method has been widely used in gene expression analysis. However, some conventional methods are not sufficient to understand regulation and function of genes expressed in very complex patterns in many organs. Thus, it is required to adopt more high-throughput and reliable techniques. Here, we describe several techniques used widely recently to analyze gene expression, including annealing control based-PCR, differential display-PCR, expressed sequence tag, suppression subtractive hybridization and microarray techniques. Use of these techniques will help to analyze expression pattern of many genes from small scale to large scale and to compare expression patterns of genes in one sample to another. In this review, we described principles of these methodologies and summarized examples of comparative analysis of gene expression in female reproductive organs with help of those methodologies.

Preparation of High Molecular Weight Atactic Poly(vinyl alcohol) Hydrogel by Electron Beam Irradiation Technique (전자빔 조사를 이용한 고분자량 혼성배열 폴리(비닐 알코올) 수화젤의 제조)

  • High molecular weight poly(vinyl alcohol) (PVA) hydrogel to be expected as a candidate material for the wound-dressing was successfully prepared by electron beam (EB) irradiation. To produce PVA hydrogels with various gel fractions, degrees of swelling in water, gel strengths, and elongations, two different number-average degrees of polymerization [($P_n$)s] of PVA were adapted such as 1700 and 4000, and the PVA solution concentration and irradiation dose of EB were controlled to range of 5 $\sim$ 20% and 30 $\sim$ 100 kGy, respectively. The gel fraction and strength of PV A hydrogel were increased with increasing molecular weight of PVA, solution concentration, and irradiation dose of EB. On the contrary, the degree of swelling and elongation of PVA hydrogel were decreased. The thermal property and crystallinity related to degree of crosslinking of PVA hydrogel were examined by the analyses of differential scanning calorimetry and X-ray diffraction.

Isolation and Characterization of a Wound or UV Induced cDNA Fragment from Pleurotus sajor-caju (상처 및 자외선 자극에 반응하는 여름느타리 cDNA 단편의 분리 및 그 발현 특성)

  • A 0.4 kb cDNA fragment was isolated from mRNA of UV or mechanical wound damaged Pleurotus sajor-caju by the differential display method. Expression of the gene corresponding to this cDNA fragment was highly induced by mechanical wound damage or UV treatment. This gene showed only basal level expression in mycelia, stipe, and cap under normal growth conditions. Sequencing analysis showed that this cDNA fragment contains partial open reading frame. Homology search using genbank database revealed that although this gene do not have homology with already reported wound induced genes, it has a significant sequence homology in defined region with the cdc2-related protein kinase gene which is known to be involved in negative regulation of meiotic maturation in Xenopus oocytes.

Design of 60-GHz Back-to-back Differential Patch Antenna on Silicon Substrate

  • This paper presents a novel design of a differential patch antenna for 60-GHz millimeter-wave applications. The design process of the back-to-back (BTB) patch antenna is based on the conventional single-patch antenna. The initial design of the BTB patch antenna (Type-I) has a patch size of 0.66 × 0.98 mm2 and a substrate size of 0.99 × 1.48 mm2. It has a gain of 1.83 dBi and an efficiency of 94.4% with an omni-directional radiation pattern. A 0.4 mm-thick high-resistivity silicon (HRS) is employed for the substrate of the BTB patch antenna. The proposed antenna is further analyzed to investigate the effect of substrate size and resistivity. As the substrate resistivity decreases, the gain and efficiency degrade due to the substrate loss. As the substrate (HRS) size decreases approaching the patch size, the resonant frequency increases with a higher gain and efficiency. The BTB patch antenna has optimal performances when the substrate size matches the patch size on the HRS substrate (Type-II). The antenna is redesigned to have a patch size of 0.81 × 1.18 mm2 on the HRS substrate in the same size. It has an efficiency of 94.9% and a gain of 1.97 dBi at the resonant frequency of 60 GHz with an omni-directional radiation pattern. Compared to the initial design of the BTB patch antenna (Type-I), the optimal BTB patch antenna (Type-II) has a slightly higher efficiency and gain with a considerable reduction in antenna area by 34.8%.

Properties of Glass Melting Using Recycled Refused Coal Ore (선탄 경석 재활용 원료를 이용한 유리 용융 특성)

  • In this study, the glass melting properties are evaluated to examine the possibility of using refused coal ore as replacement for ceramic materials. To fabricate the glass, refused coal ore with calcium carbonate and sodium carbonate in it (which are added as supplementary materials) is put into an alumina crucible, melted at $1,200{\sim}1,500^{\circ}C$ for 1 hr, and then annealed at $600^{\circ}C$ for 2 hrs. We fabricate a black colored glass. The properties of the glass are measured by XRD (X-ray diffractometry) and TG-DTA (thermogravimetry-differential thermal analysis). Glass samples manufactured at more than $1,300^{\circ}C$ with more than 60 % of refused coal ore are found by XRD to be non-crystalline in nature. In the case of the glass sample with 40 % of refused coal ore, from the sample melted at $1,200^{\circ}C$, a sodium aluminum phosphate peak, a disodium calcium silicate peak, and an unknown peak are observed. On the other hand, in the sample melted at $1,300^{\circ}C$, only the sodium aluminum phosphate peak and unknown peak are observed. And, peak changes that affect crystallization of the glass according to melting temperature are found. Therefore, it is concluded that glass with refused coal ore has good melting conditions at more than $1,200^{\circ}C$ and so can be applied to the construction field for materials such as glass tile, foamed glass panels, etc.

Multiple LCD System Development of daisy-chain Method using LVDS (LVDS를 이용한 daisy-chain 방식의 다중 LCD 시스템 개발)

  • This thesis explains the development of multiple LCD system with the additional function to maximize the utilization of PC contents. The newly developed system is composed of host LCD and slave LCD. Host LCD decodes and outputs the image and voice of NTSC, PAL, SECAM signals. It also converts the decoded signals into LVDS signals before transmitting them to slave LCD stage. In addition, the installation of CF Memory and USB Memory helps display multi-media data. Unlike the host LCD, since not including the tuner and memory part, the slave LCD can't receive TV signals and play video signals. It only has the function to receive LVDS image signals and display on a LCD panel. This newly developed multi-LCD system has competitiveness in various aspects. With its simple structure, the failure rate, price and display power are relatively low due to its simplification of the control part. It has price and functional competitiveness as the product whose host LCD can control the entire slave LCD in terms of channel, volume, and video output.

  • The effects of chemically modified thermoplastic starch (CMPS) on the morphology, thermal and mechanical properties of the blends of poly (lactic acid)(PLA) and poly(butylene adipate-co-terephthalate)(PBAT) were studied. Blends of PLA/PBAT with the CMPS contents of 10, 20 and 30 wt% on the basis of PLA/PBAT weight were prepared by a twin screw extruder. The morphology, thermal and mechanical properties of the blends were examined by using scanning electron microscope (SEM), differential scanning calorimeter (DSC) and a tensile tester. The DSC study revealed that PLA/PBAT blends are thermodynamically immiscible, while the compatibility was much improved by addition of the CMPS.

Identification of Differentially Expressed Genes by Gabapentin in Cultured Dorsal Root Ganglion in a Rat Neuropathic Pain Model

  • Neuropathic pain is a chronic pain disorder caused by nervous system lesions as a direct consequence of a lesion or by disease of the portions of the nervous system that normally signal pain. The spinal nerve ligation (SNL) model in rats that reflect some components of clinical pain have played a crucial role in the understanding of neuropathic pain. To investigate the direct effects of gabapentin on differential gene expression in cultured dorsal root ganglion (DRG) cells of SNL model rats, we performed a differential display reverse transcription-polymerase chain reaction analysis with random priming approach using annealing control primer. Genes encoding metallothionein 1a, transforming growth factor-${\beta}1$ and palmitoyl-protein thioesterase-2 were up-regulated in gabapentin-treated DRG cells of SNL model rats. The functional roles of these differentially expressed genes were previously suggested as neuroprotective genes. Further study of these genes is expected to reveal potential targets of gabapentin.

Identification of Differentially Expressed Genes in Nickel[li]-Treated Normal Rat Kidney Cells

  • Nickel(II) compounds are carcinogenic metals which induce genotoxicity and oxidative stress through the generation of reactive oxygen species. In search of new molecular pathways toward understanding the molecular mechanism of nickel(II)-induced carcinogensis, we performed mRNA differential display analysis using total RNA extracted from nickel(II) acetate-treated normal rat kidney cells (NRK-52E). Cells were exposed for 3 days to 160 and 240 uM nickel(II) concentrations. cDNAs corresponding to mRNAs for which expression levels were altered by nickel(II) were isolated, sequenced, and followed by a GenBank Blast homology search. Specificity of differential expression of cDNAs was determined by RT-PCR and Western blot analysis. Two of them (SH3BGRL3 and FHIT) were down-regulated and one (metallothionein) was up-regulated by nickel(II) treatment. The expression of these mRNAs were nickel(II) concentration-dependent. The levels of FHIT and metallothionein proteins were also consistent with the results for mRNAs. Overall, although the fundamental questions related to function of these genes in nickel(II)-mediated carcinogenicity are not answered, our study suggests that they can be interesting candidates for studies of molecular mechanisms of nickel(II) carcinogenesis.
