  • This study was conducted to improve the postharvest storage techniques of managing and storing seeds, to test qualities and viabilities of the seeds and to examine the germination rate and the protein expression of Achyranthes japonica Nakai. The seeds collected from different areas of Je-Cheon and Gwang-Ju were stored with different temperatures and durations. Two plant growth regulators and two seed priming were treated to investigate their effect on the germination rates and the days required for germination. The weight of one hundred seed collected in Gwang-Ju was heavier than those in Je-Cheon. Seed length collected in Gwang-Ju was also longer about 5.12 mm than those in Je-Cheon about 4.90 mm and seed width was longer in Gwang-Ju than those in Je-Cheon. The rates of seed germination in two different collection areas were higher about 2.9 to 13.0% in Gwang-Ju compared to those in Je-Cheon. Comparing its rates with the storing temperatures and durations, they were not clearly different in between $4^{\circ}C$ and $25^{\circ}C$ and they also were gradually decreased with getting longer storing durations. The germination rates treated by plant growth regulators were higher with $GA_3$ than those with Kinetin. The highest seed germination rate was appeared at 50 ppm of $GA_3$. Comparing its rates with different seed priming, they were relatively higher with $KNO_3$ than those with PEG6000. In protein expression patterns between before the germinating and after the germinating of seeds, more and clear bands were appeared in the seed after the germination compared to those before the germination of seeds, especially 10 ~ 20 kDa. These results showing more and clear bands were more clearly appeared in Gwang-Ju compared to Je-Cheon. Comparing the protein expression with plant growth regulators and seed primings, $GA_3$ was better expression than those with Kinetin and $KNO_3$ was better than those with PEG6000. More and clear bands were closely related to the germination rates of seeds and more detailed studies would be required.

  • Expression of 43 flowering-related genes was examined in two inbred lines of Chinese cabbage, Chiifu and Kenshin, under different photoperiod, vernalization and flower development stages. The floral genes cloned by RT-PCR with degenerated primers showed high homology with Arabidopsis counterparts. Genes in two inbred lines, TOC, CRY1, CO, RGAL and GAl, were highly expressed under all tested conditions. However, expression of three genes was regulated by particular experimental conditions. The expression of LHY gene was predominant in Chiifu under the short-day conditions, whereas the expression of RGAL gene was influenced by vernalization in both inbred lines. Besides, the expression of NAP gene was induced by vernalization only in Chiifu. Most of the flower identity-related genes were expressed during flower development. The transcript level for several genes was not detected in this experiment.

  • From the present study, following conclusions can be drawn: 1. lide in other species, axolotl FGF-8 is proposed to play a similar role in the early phase of limb development. However, the mechanism of its expression might be somewhat different from amniotes considering its characteristic mesenchymal expression. 2. In the regenerating axolotl limbs, Fgf-8 expression profile suggests that it is involved in wound gealing, dedifferentiation, and blastema formation. 3. Exoggenously supplied FGF-8 can accelerate blastema formation and concomitantly increase the Msx-1 expression level at the early stage of limb regeneration. Furthermore, it can partially substitute for nerve factor(s) as has been indicated by the induction of blastema formation in the denervated regenerates after FGF-8 application. 4. The unique expression feature of Fgf-8 in hte mesenechymal tissue of the regenerating axolotl limb might be casually related to its remarkable regeneration capacity of urodele.

  • In the present study, immune gene expression of rainbow trout, Oncorhynchus mykiss, against bacterial endotoxin (LPS) was studied in vivo. The expression of prointflammatory cytokine genes (IL-1$\beta$ and TNF-$\alpha$) and IFN-related genes (IRF-I, Mx-3) at gill was assessed by RT-PCR at different time point of day1 and day 3 post-injection. It was shown that the proinflammatory cytokine gene expression at gill was induced 1 day after LPS injection but the expression was not sustained until day 3. Meanwhile upregulated expression of IFN-related genes was found to be only at day 3 post injection, indicating indirect effect of LPS on these genes.

  • In the trend of building enlargement, high-rise and irregular shaped building, domestic application of BIM in construction phase is increasing in recent years. BIM that is based on a three-dimensional, requires a specific building expression. Model expression level required for each phase such as design, construction, and maintenance of the construction project can be different. BIM model to apply in practice is difficult because it is not clear to set up the criteria for the expression level of construction BIM model. And it has a limit that is lower productivity in construction project. Therefore, this study considers the way of the improvement and analyzes the application for representation level of construction BIM model through the survey. By analyzing of the survey results, this study will be to present a basis for an appropriate expression level setting of construction BIM model.

  • The combination of Simian Virus40 (SV40)'s large T antigen with its replication origin is commonly used in molecular studies to enhance the expression of heterogeneous genes through multiplying the plasmid copy number. There are no reports related to the impact of the SV40 T antigen on plant, multiple fissional, cell-type. This study explores the response of two multiple-fission microalgal cells, Scenedesmus quadricauda and Chlorella vulgaris, to the expression of the T-antigen, with aim of applying SV40 T-antigen to increase the expression efficiency of foreign genes in the two species. Different levels of low-expression have been constructed to control the expression of SV40 T antigen using three heterogenous promoters (NOS, CaMV35S, and CMV). Chlorella cultures showed slowdown in the growth rate for samples harboring the T antigen under the control of CaMV35S and CMV promoters, unlike Scenedesmus cultures which showed no significant difference between samples and could have silenced the expression.

  • Among the various volatile organic compounds (VOCs) emitted by the plant, floral scent plays a key role in attracting pollinators for reproduction and mediates ecological interactions. Floral scent is an important trait and industry drives the competition for flowers with novel scents. Chrysanthemum is one of the well-known ornamental plants and is a popular cut flower across the world. Floral scent and the genes responsible for the floral scent emission are poorly studied in chrysanthemum. In the present study, floral scent and the expression pattern of floral scent genes were analyzed in two chrysanthemum cultivars 'Golden Egg' and 'Gaya Glory'. Initially, intensity of the floral scent in five developing stages of flower including 'budding (B), bud developing (BD), initial blooming (IB), almost open (AO) and open flower (OF)' was analyzed using electronic nose (E-nose) with six metal oxide sensors. Based on the distance analysis, different stages of flower showed different relative intensity of scent according to the sensory evaluation. Although the scent pattern differed by stage, scent intensity was strongest in the OF stage in the completely opened flower in both the cultivars. Further, expression pattern of six genes in the floral scent pathway including FDS, IDI, ISPH, TPS2, TPS5 and TPS6 was observed in all the five stages of the flower in both the cultivars. The expression pattern of all the six genes differed by stage and the terpene synthase genes TPS2, TPS5 and TPS6 showed good expression levels in the $5^{th}$ flower stage compared to other stages. This study provides a preliminary data for understanding the regulation of floral scent in chrysanthemum.

  • Fish shows great difference in growth rate between individuals during larval development and early growth. This difference seriously reduces the production efficiency in fish culture. Growth hormone (GH)/Insulin-like growth factor 1 (IGF1) system is said to play some pivotal roles in fish growth. In this study, we investigated differences of GH, IGF1 and GHR gene expressions in juvenile red spotted grouper (Epinephelus akaara) with different growth performance. Red spotted groupers were reared under the same environmental condition (water temperature $24{\pm}1^{\circ}C$, natural light) for 96 days after hatching. They were divided into 3 groups by size (fast growing, middle growing and slow growing groups: FGG, MGG, and SGG, respectively). RNA was extracted from the brain, liver and muscle tissues from each group, and target gene expression was examined by real-time PCR. In the brain with pituitary gland, expression of GH gene in FGG was significantly higher than the expression in SGG, but the expression of IGF1 and GHR genes in the muscle was highest in SGG. Difference of GHR and IGF1 mRNA in the liver between groups with different growth performance was less clear than that in other tissues, although level of IGF1 mRNA was higher in SGG than in MGG. These results suggest that hormonal governing of growth is not the same in fast growing and slow growing fish, and size grading could cause a shift of hormonal state and growth pattern in this species.
