나리에서 분리한 잎마름병균의 살균제에 대한 감수성 변화와 포장 방제 (Changes in Sensitivity Levels of Botrytis spp. Population Isolated from Lily to Fungicides and Control under Field Condition)

  • 국내 나리 주산단지에서 분리한 Botrytis elliptica 48개 균주와 Botrytis cinerea 23개 균주를 대상으로 농가포장에서 사용되고 있는 살균제에 대한 약제저항성 검정을 실시하였다. 살균제 benomyl과 mancozeb에 대한 $EC_{50}$ 값은 $500-1,000{\mu}g/ml$로 시험에 사용된 모든 약제 중에서 가장 높게 나타나 이 약제들은 나리재배 포장에서 잎마름병에 대한 방제 효과가 거의 없을 것으로 판단되었다. 또한 나리 재배농가에서 잎마름병 방제약제로 가장 많이 사용되고 있는 dicarboximide계 procymidone 살균제와 iprodione 살균제의 경우, $EC_{50}$$5-50{\mu}g/ml$은 각각 93.7%와 100%이었고, diethofencarb/carbendazim 합제와 fludioxonil 살균제의 $EC_{50}$$0-0.1{\mu}g/ml$ 범위는 각각 98.0%와 93.8%이었다. 한편, 비가림 포장 재배에서의 잎마름병 방제 효과는 발병 직후 iprodione 약제, diethofencarb/carbendazim 합제, fludioxonil 약제를 교호적으로 4회 살포하였을 때 가장 우수하였다.

Responses of Peach Blossom Blight and Brown Rot Fungus Monilinia fructicola to Benzimidazole and Diethofencarb in Korea

  • The population shifts of Monilinia fructicola isolates which were resistant to the fungicide benzimidazoles were investigated in four regions of Korea from 1998 to 2000. The isolation frequency of benzimidazole-resistant isolates ranged from 18.8% to 29.6% in Chongdo and from $22.0\%$ to $26.8\%$ in Gyeongsan during the same period. However, the frequency of benzimidazoleresistant isolates was less than $4.0\%$ in Chochiwon and Youngduk during the same period. Benzimidazoleresistant isolates showed cross-resistance among benzimidazoles. On the other hand, none of the isolates showed cross-resistance to diethofencarb and carbendazim. Regardless of the year, the benzimidazole-resistant isolates of $EC_{50}$ higher than 500 $\mug%$ a.i./ml were isolated more frequently in mid and late season than in early season. In an orchard of Gyeongsan that had not been exposed to any fungicides for several years, the population of benzimidazole-resistant isolate had persisted without much fluctuation for three years. These results suggest that benzimidazole resistance of M. fructicola is becoming a problem in controlling brown rot and blossom blight of peach in regions like Chongdo and Gyeongsan.

Survey of Fungicide Resistance for Chemical Control of Botrytis cinerea on Paprika

  • Four hundred and sixty six isolates of B. cinerea were obtained from infected leaves, stems and fruits of paprika grown in greenhouses or plastic film houses in Gangwon province, Korea, between August and November in 2006 and 2007. These isolates were classified into five representative phenotypes of resistant (R) and sensitive (S) reactions as SSR, SRR, RSS, RRS and RSR according to the responses of isolates against benzimidazole, dicarboximide and N-phenyl-carbamate fungicide in order. The percentage of five phenotypes were 51.3, 2.4, 35.6, 8.1 and 2.6%, respectively. The SSR phenotype (51.3%) was the most common. Among the nineteen fungicide products evaluated to compare their efficacy against gray mold pathogen on the paprika fruit inoculated with fungal mycelia, the mixture of diethofencarb and carbendazim was the most effective followed by iprodione, boscalid, the mixture of iprodione and thiophanate-methyl, fludioxonil, polyoxin-B, fluazinam, the mixture of tebuconazole and tolyfluanid and procymidone; while in the assay methods inoculated with fungal spores, the mixture of tebuconazole and tolyfluanid was the most effective in controlling gray mold followed by boscalid, fludioxonil, the mixture of diethofencarb and carbendazim and the mixture of pyrimethanil and chlorothalonil.

Characterization of Colletotrichum Isolates Causing Anthracnose of Pepper in Korea

  • A total of 33 isolates of Colletotrichum species obtained from pepper, apple, and strawberry in 2001 and 2002 were identified based on mycological characteristics, responses to fungicides carbendazim and the mixture of carbendazim and diethofencarb, and nucleotide sequence analysis of internal transcribed spacer (ITS) regionMost of the Colletotrichum isolates from pepper could be identified as C. acutatum. The pepper isolates produced grey white mycelia that gradually changed to dark gray. The conidia were variable in size, and almost cylindrical in shape with at least one rounded end. They could grow on PDA amended with carbendazim or with the mixture of carbendazim and diethofencarb at $10{\mu}g/ml$, to which the isolates from apple and strawberry were very sensitive. A part of the ITS regions from the Colletotrichum isolates was amplified with the specific primers designed for C. acutatum (Ca1-1) or C. gloeosporioides (Cg1-3). A primer pair of Ca1-1 and a universal primer (ITS4) amplified a 496-bp DNA fragment from all of the pepper isolates examined and one apple isolate. Taken together, it is conclusive that the Colletotrichum isolates causing the typical lesion of anthracnose on pepper fruits are C. acutatum.

Analyses of the Less Benzimidazole-sensitivity of the Isolates of Colletotrichum spp. Causing the Anthracnose in Pepper and Strawberry

  • The anthracnose disease on pepper fruits in Korea was caused by Colletotrichum acutatum as well as C. gloeosporioides. Since C. acutatum showed less sensitivity to benomyl, it was analyzed whether the less sensitivity was given by the same mechanism for the fungicide resistance of C. gloeosporioides. The isolates of C. acutatum were less sensitive to the three benzimidazole fungicides tested, benomyl, carbendazim, and thiophanate-methyl. However, the of C. acutatum isolates were different from the resistant isolates of C. gloeosporioides in their response to diethofencarb, one of N-phenyl-carbamates; the former was still less sensitive to diethofencarb than the latter. The differences in the resistance mechanisms in two species were conspicuous in sequence analysis of the tub2 genes. The genes from C. acutatum did not show any non-synonymous base substitutions at the regions known to be correlated with the benzimidazole-resistance. All of these data may indicate that the less sensitivity of C. acutatum to benomyl is based on different mechanism(s) from that of C. gloeosporioides.

Species identification and pathogenicity study of Colletotrichum isolates isolated from red-pepper and Chinese matrimony vine

  • This study reports the identification of species of Colletotrichum strains originating from red-pepper and Chinese matrimony vine in Cheongyang. Ninteen isolates of red-pepper and 26 Coiletotrichum isolates of Chinese matrimony vine were compared with 5 isolates of strawberry representing C. gloeosporioides, by use of morphological and cultural criteria. Twenty three isolates among 26 isolates from Chinese matrimony vine were identified as C. acutatum, characterized by the low growth rates and the low sensitivity to carbendazim and diethofencarb. Also, all the isolates of red-pepper were identified as C. acutatum, showing the same characteristics as those of Chinese matrimony vine. Three and five isolates from Chinese matrimony vine and strawberry, respectively, were identified as C. gloeosporioides, characterized by the high growth rates and the high seneitivity to carbendazim and diethofencarb. There were differences in colony color and pathogenicity between Chinese matrimony vine isolates and red-pepper isolates of C. autatum. The isolates of C. acutatum from Chinese matrimony vine producing orange colored colonies with abundant spores showed the strong pathogenicity to Chinese matrimony vine, although they could not infect fruits of red-pepper by the wound inoculation. However, red-pepper isolates of C. acutatum producing gray colonies showed the strong pathogenicity to Chinese matrimony vine as well as red-pepper. Furthermore, comparative study on PCR amplification of ITS regions of rDNA was carried out using a number of Colletotrichum isolates. A species-specific primer could be used for the identification of C. acutatum from red-pepper and Chinese matrimony vine.

Population Dynamics and Fitness Comparison of Sensitive and Resistant Phenotypes of Botrytis cinerea to Benzimidazole, Dicarboximide, and N-phenylar-bamate Fungicides

  • A total of 2109 isolates of Botrytis cinerea were collected from infected plants fo strawberry, tomato, and cucumber in Korea from 1994 to 1996. Based on in virtotests for mycelial growth on potato-dextrose agar containing fungicides, the esolates were classified into six phenotypic groups : SSR, SRR, RSS, RRS, RSR, and RRR, representing sensitivity (S) or resistance (R) to carbendazim, procymidone, and diethofencarb. In that order the isolation frequencies of the SSR, SRR, RSS, RRS, RSR, and RRR phenotypes were 28.7, 1.1, 28.8, 39.4, 1.0, and 0.9%, respectively. Three isolates from each SSR, SRR, RSS, RRS, and RSR and an isolate of RRR phenotype were selected and evaluated for their fitness-related characteristics such as pathogenic aggressiveness, mycelial growth rate, sporulation, and sclerotial formation. Competitive abilities of the SSR, SRR, RSS, RRS, and RSR phenotypes were also compared by inculating mixtures of conidial suspensions of two phenotypes to cucumber plant, and then determining re-isolation frequencies from lesions. In general, significant differences in fitness-related characteristics, except pathogenic aggressiveness, were found not only between but also within phenotype groups. In the competitiveness tests, carbendazim-sinsitive phenotypes (SSR and SRR) were found to be more competitive than the resistant ones (RSS and RSR), whereas, the procymidone-resistant phenotypes (SRR and RRS) appeared to be more competitive than the sensitive ones (SSR, RSS, and RSR). There was no consistent dominance in competitiveness between the diethofencarb-resistant and sensitive phenotypes. The RSR phenotype was the least competitive among the five phenotypes.

수삼과 홍삼농축액 중 유기인계와 유기염소계 농약의 잔류 (Residue of Organophosphorus and Organochlorine Pesticides in Fresh Ginseng and Red Ginseng Extract)

  • 본 연구에서는 경북의 풍기와 상주에 있는 인삼재배포장에서 직접 시료를 채취하고, 이를 이용한 홍삼농축액제품을 제조하였다. 수삼과 홍삼농축액 중 유기인계와 유기염소계 농약을 GC-NPD와 GC-ECD로 각각 정량분석 하였으며 GC-MSD로 정성분석을 실시하여, 식품의약품안전청이 2007년 1월1일부터 시행한 농약잔류허용기준에 따라 안전성을 평가하였다. 풍기 수삼에서 유기인계 농약은diazinon을 비롯한pyrimethanil, tolclofos-methyl, metalaxyl, diethofencarb, parathion, cyprodinil, tolylfluanid와 kresoxim-methyl이 모두 불검출 되었다. 그러나 상주 수삼에서는 tolclofos- methyl이 평균 $0.054{\pm}0.008\;mg/kg$ 검출되어 수삼의 농약잔류허용기준 0.3mg/kg의 $18{\pm}2%$ 수준이었다. 풍기와 상주 수삼에서 유기염소계 농약은 BHC, DDT의 이성질체와 aldrin, azoxystrobin, captan, cypermethrin, deltamethrin, dieldrin, difenoconazole, endosulfan-sulfate, endrin, fenhexamid, quintozene, ${\alpha}-endosulfan$, ${\beta}$-endosulfan도 검출 되지 않았다. 한편 풍기와 영주의 인삼으로 제조한 홍삼농축액 중에도 유기인계와 유기염소계 농약은 모두 검출 되지 않았다.

인삼 잿빛곰팡이병균의 procymidone에 대한 감수성 변화와 carbendazim/diethofencarb 합제와의 다중 저항성 (Monitoring for the Resistance of Botrytis cinerea Causing Gingseng Gray Mold to Procymidone and Its Multiple resistance with the Mixture of Carbendazim/Diethofencarb)

  • 인삼의 잎에서 발생한 잿빛곰팡이병의 병반으로부터 병원균인 Botrytis cinerea의 224개 균주를 단포자 분리하여 실험에 사용하였다. 분리한 B. cinerea는 한천희석법을 사용하여 procymidone, carbendazim, carbendazim과 diethofencarb 합제의 균사생장 억제 효과를 $EC_{50}$(균사생장을 50% 억제하는 농도)과 MIC(균사생장을 100% 억제하는 최소농도) 값을 조사하였다. Procymidone에 대한 MIC값을 가지고서 병원균을 분류하면 0.8과 $4.0{\mu}g/ml$ 사이에 속하는 그룹과 $20{\mu}g/ml$ 이상인 그룹으로 크게 분류할 수 있었다. 2005년에 분리한 균주들은 지역별로 procymidone에 대한 감수성의 변화가 매우 다양하게 나타났다. 실험에 사용한 procymidone의 각각의 농도에 대한 균사 생장 억제 양상을 가지고 병원균을 각각 감수성($EC_{50}$ 값이 $2.0{\mu}g/ml$ 이하), 중도저항성($EC_{50}$ 값이 2.0과 $5.0{\mu}g/ml$ 사이), 저항성($EC_{50}$ 값이 $5.0{\mu}g/ml$ 이상) 등의 3개의 그룹으로 분류할 수 있었다. 이 기준으로 분류하였을 때, procymidone에 대해서 저항성인 B. cinerea의 비율은 2005년에 19.3%가 2006년에는 27.5%로 상승하였으며, 저항성 그룹의 평균 $EC_{50}$ 값도 $10.0{\mu}g/ml$에서 $237.3{\mu}g/ml$으로 상승하였다. Procymidone과 carbendazim의 경우에 다중저항성을 보이는 균주는 90개로 전체의 40.2%이었으며, 합제와의 다중저항성을 보이는 균주는 9개 밖에 없었다.

다양한 작물로부터 분리한 탄저병균(Colletotrichum spp.)의 살균제에 대한 반응 (Response to Fungicides of Colletotrichum Species Isolated from Infected Tissues of Several Crops)

  • 다양한 작물로부터 분리한 4종의 탄저병균, C. gloeosporioided, C. acutatum, C. coccodes 및 C. dematium 등의 균주들에 대하여 여러 가지 살균제, 즉 carbendazim, carbendazim과 diethofencarb와의 혼합제, 4개의 보호용 살균제 및 3개의 ergosterol 생합성 저해(EBI) 살균제 등에 대한 약제 반응을 조사하였다. Benzimidazole계에 속하는 carbendazim에 대해서 4종의 Colletotrichum의 반응은, C. acutatum은 사용한 6개의 균주 모두 $EC_{50}$값은 $0.001{\sim}3.04{\mu}g/ml$ 사이에 있었으며, 모든 균주의 MIC 값은 $100{\mu}g/ml$ 이상이었다 고추와 사과, 딸기에서 분리한 C. gloeosporioides는 carbendazim에 대해서 저항성과 감수성 반응이 뚜렷하게 구별되었는데, 고추의 두 균주는 $EC_{50}$값은 0.24와 $0.01{\mu}g/ml$이었으며 MIC 값 역시 $1.0{\mu}g/ml$ 이었다. 이러한 반응은 사과와 딸기에서 분리한 균주 중에서 감수성 반응을 보이는 균주인 B13과 138, 그리고 CGF89와 94, 252에서도 비슷한 경향을 보였다. 그러나 저항성 균주로 판명된 B90, 92, 147과 CGF50, 54, 222는 $EC_{50}$값과 MIC 값 모두가 $100{\mu}g/ml$를 상회하였다. C. gloeosporioides는 carbendazim과 역상관 교차 저항성을 보이는 diethofencarb와 carbendazim의 혼합제에 대해서는 모두 $1{\sim}100{\mu}g/ml$의 MIC값을 보였지만, C. acutatum은 $100{\mu}g/ml$이상의 MIC값을 보였다. Carbandazim에 대하여 C. coccodes는 C. acutatum과 그리고 C. dematium는 C. gloeosporioides의 감수성 균주들의 반응과 유사한 반응을 보였다. 4종의 보호용 살균제에 대해서는 C. acutatum은 C. gloeosporioides보다도 높은 저항성인 반응을 보였다. 그러나 ergosterol 생합성 저해 살균제에 대해서는 4종의 탄저병균이 일정한 경향을 보였다.
