Determination of dietary Ca and P levels and their equivalence values of phytase and vitamin D3 for improved growth performance in weanling pigs

  • In Experiment 1, the impact of using diets sufficient in P on the performance of weaned piglet growth with or without a super dose of phytase was investigated. One hundred and twelve piglets were randomized into four treatments with 7 replicates (4 piglets·cage-1). Bodyweight (BW), daily gain (ADG), daily intake (ADFI) and feed conversion ratios (FCR) were measured over three weeks. Treatments included a positive control (PC) with a Ca : P ratio of 0.80 : 0.50%. and the negative control (NC) had a Ca : P ratio of 0.60 : 0.50%. The PC plus phytase was denoted as PC+, and the NC with the phytase diet was designated as NC+. The PC diet was formulated to meet or exceed the NRC (2012) standards for pigs weighing 7 - 25 kgs. Better results were obtained with the PC diet (p > 0.1) compared to the NC diet. The super dose of phytase in the PC+ diet significantly improved the overall FCR (p < 0.1). For Experiment 2, the impact of different calcium (Ca)/phosphorous (P) ratios with or without supplementation of vitamin D3 was determined. Ninety-six piglets were randomized into four treatments with 6 replicates (4 piglets·cage-1). The PC, NC1 and NC2 diets had ratios of 0.80 : 0.50, 0.80 : 0.45, and 0.60 : 0.45%, respectively. The fourth diet (PCV) was the PC diet fortified with vitamin D3. Improved results were observed with the PC diet (p > 0.1) compared to the NC diets. Supplementing dietary 25-OH-D3 in the PCV diet led to numerically higher ADG and ADFI scores with lower FCR values at the early nursery stage (p = 0.0044), but the vitamin supplement did not significantly influence the growth.

  • 본 연구는 인과 칼슘의 수준이 낮은 산란계 사료내 미생물 phytase의 첨가가 생산성 및 영양소 소화율에 미치는 영향을 평가하기 위하여 실시되었다. 사양시험은 50주령 ISA brown 갈색계 192수를 공시하였으며, 사양시험은 7일간의 적응기간 후, 12주간 실시하였다 시험설계는 옥수수-대두박 위주의 사료에 NRC 요구량에 맞춰 Available P과 Ca을 처리한 구(CON; 기초사료), 기초사료에 Available P과 Ca을 각각 대조구 사료의 $50\%$$90\%$를 함유한 사료에 3종류의 미생물phytase PHYTEX, Agranco. Co. $0.01\%$(P1), Natuphos, BASF $0.06\%$(P2), PHOSMAX GENOFOCUS $0.06\%$(P3)를 첨가한 처리구로 4개 처리를 하였다. 총 12주간의 사양 시험기간동안 산란율은 처리구간에 유의적인 차이를 보이지 않았다 (P>0.05). 시험 종료시 난중은 처리구에서 유의적으로 높게 평가되었다(P<0.05). 난각 두께는 시험 9주째 phytase를 급여한 처리구가 대조구와 비교하여 두꺼워졌다(P<0.05). 시험개시시와 종료시 혈청 내 Ca 함량의 차이는 P1처리구에서 가장 적은 변화를 보였으나, 처리구간 유의적인 차이는 보이지 않았으며, P함량에 있어서도 처리구간 유의성은 보이지 않았다(P>0.05). 경골 내 Ca과 P의 함량은 phytase를 급여한 처리구가 대조구에 비하여 유의적인 차이 없이 높은 경향을 보였다(P>0.05). DM, N과 Ash의 소화율에 있어서는 대조구와 P1 처리구가 P2, P3 처리구와 비교하여 유의적으로 증가하였다(P<0.05). Ca과 P에 있어서는 P2 처리구의 소화율이 가장 높게 나타났으며(P<0.05)), 대조구와 P1 처리구는 서로 통계적인 차이를 보이지 않았다(P>0.05). 모든 처리구들이 대조구보다 유의하게(P<0.01) 낮았고 Ercherichia coli의 수는 유의한 차이가 없었다. Lactobacilli의 수는AM30구가 대조구보다 유의하게(P<0.05) 높았다. 결론적으로 활성 미네랄수 $30\%$흡착 zeolite의 사료 내 첨가는 대조구에 비하여 육계의 증체율을 개선시키는 경향이 있었으며 깔짚에서 암모니아 가스 발생도 유의하게 감소시켰고 장내 Lactobacilli의 수를 유의하게 증가시켰다.결과를 얻을 수 있으리라 생각한다.내산에서는 S.typhimurium $(33\%)$이 분리 되었으나, 미국산 닭날개에서는 Salmonella 속균이 전혀 분리되지 않아 대조적이었다. 하지만, 중국산의 닭부분육(혼합육)에서는 S. enteritidis와 L. monocytogenes도 함께 분리되었다. 그러나 부분육의 경우 국내산 및 수입산 모두에서 Campylobacter 속균과 E. coli O157:H7 균주는 전혀 분리되지 않았다. 서울${\cdot}$경기지역의 재래시장과 백화점에서 시판되고 있는 계란 총 446개에 대해서도 동일한 절차와 방법으로 조사하였던바, 재래시장에서 구입했던 계란의 난각부분(Egg-shell)에서만 가금티푸스(fowl Typhoid)의 병원체인 S. gallinarum이 1주$(0.2\%)$만이 분리되었고, 기타 세균으로서는 대장균군이 역시 난각에서 가장 높은 빈도로 분리되었고, 난황(Yolk)에서는 극히 낮은 수준의 세균오염도를 보였다. 다양한 동물종유래 S. aureus 균주들의 유전학적 분석목적에 가장 신뢰도 높고 감별능력이 뛰어난 분석기법으로 선발되었다.cid로부터 cyclooxygenase를 통해 일단 생성된 endoperoxide에서 각각의 prostaglandin을 생성하는 효소, 특히 $G-Rb_{2}$$TxA_{2}$

Effect of a microbial phytase on growth performance, plasma parameters and apparent ileal amino acid digestibility in Youxian Sheldrake fed a low-phosphorus corn-soybean diet

  • Objective: This study investigated the effect of microbial phytase supplementation on growth performance, tibia ash, plasma parameters, apparent ileal digestibility (AID) of amino acid (AA) and apparent digestibility of nutrients in Youxian Sheldrakes fed with low-phosphorus (P) cornsoybean diets. Methods: A total of 350 Youxian Sheldrakes (7d old) were randomly divided into 5 treatment groups: positive control (PC) group has adequate available P diet (0.42% and 0.38%, starter and grower), negative control (NC) group were deficient in available P (0.32% and 0.28%, starter and grower) and NC diet was supplemented with 3 levels of microbial phytase (500, 750, and 1,000 U/kg). Results: Dietary supplementation of phytase in NC diet improved the average daily gain, increased the levels of serum calcium (Ca), tibia Ca and P, AID of AA and apparent digestibility of energy and Ca in starter stage (p<0.05). There was an increased (p<0.001) in the utilization of P from 17.3% to 23.9%. Phytase supplementation (1,000 U/kg) has shown that the AID of His, Thr, Val, indispensable AA, Glu, Pro, and dispensable AA was higher (p<0.05) than that of NC. Moreover, phytase supplementation improved (p<0.05) serum and tibia Ca and P, AID of AA and apparent digestibility of dry matter, crude protein, energy, P and Ca, and reduced (p<0.05) feed to gain ratio (F/G) and the levels of serum alkaline phosphatase in grower stage. Likewise, an increase (p<0.001) in the utilization of P was noticed from 12.6% to 17.2%. Supplement phytase at 750 U/kg improved the AID of His, Thr, Asp, Cys, Pro, and Ser (p<0.05). Conclusion: The microbial phytase supplement could improve growth performance, AID of some AA and apparent utilization of other nutrients in Youxian Sheldrakes, and reduce excreta P load to environment.

Bacterial 및 Fungal Phytase의 단일 및 혼합 급여가 산란계의 생산성과 영양소 이용에 미치는 영향 (Effects of Single or Mixed Supplementation of Bacterial Phytase and Fungal Phytase on Laying Performance and Nutrient Digestibility)

  • 본 시험에서는 bacteria 또는 fungi로부터 유래된 phytase를 단일 및 혼합으로 45주령 Hy-Line Brown 산란계 450수에 12주 동안 급여하였다. 공시계는 시험개시 전 일주일 동안 산란조사를 하여 처리간에 비슷한 산란율이 되도록 하였다. 대조구(Control)사료는 nonphytate phosphorus 함량이 0.3% 되게 하였으며, 처리구는 bacterial phytase(BP)와 fungal phytase (FP) 2종류를 각각 T1(300 DPU, 0 DPU), T2(300 DPU, 300 DPU), T3(300 DPU, 3000 DPU), T4(0 DPU, 3000 DPU)로 하여 급여하였다. 처리구들의 nonphytate phosphorus 함량은 Control의 60%로 하였다. 사료와 물은 전 기간 자유채식토록 하였고, 점등은 17L7D로 하였다. 산란율은 T4가 Control에 비해 유의하게 낮았으며(P<0.05), 두 종류 phytase의 동시 급여 효과는 없었다. 평균 난중과 1일 산란량은 처리구간에 통계적인 차이는 없었으나, 후자의 경우 시험 후반기에 T4에서 Control에 비해 유의하게 낮았다(P<0.05). 사료 섭취량, 사료요구율, 계란의 품질은 처리구간에 차이가 없었다. 건물, 조단백질 및 지방 소화율은 처리구간 유사하였으며, 무기물, 칼슘 및 인 이용율은 뚜렷한 경향이 없었다. 본 실험 결과에서 산란계 사료에 BP 300 DPU 수준일 때 NPP 첨가량을 NRC (1994) 권장량 대비 약 40%가 저감되었으며, 인 이용율 역시 개선되었으나 FP와의 혼합 급여로 인한 개선 효과는 나타나지 않았다.

탄수화물 분해 복합효소제와 미생물 파이테이즈의 첨가가 육성돈의 생산성 및 영양소 소화율에 미치는 영향 (Effects of Dietary Carbohydrase Enzyme Complex and Microbial Phytase Supplementation on Productivity and Nutrient Digestibility in Growing Pigs)

  • 본 연구는 영양소 수준을 낮춘 옥수수-대두박 기초 사료내 $\alpha$-galactosidase와 galactomannanase를 함유한 탄수화물 분해 복합효소제 (ENDO- POWER$^{\circledR}$)와 미생물 파이테이즈 (Natuphos$^{\circledR}$) 의 개별첨가 혹은 동시첨가가 육성돈의 생산성, 영양소 소화율 및 체중 증체 당 사료비에 어떠한 영향을 미치는 가를 평가하고자 실시 되었다. 개시 체중 29.1$\pm$0.14 kg의 육성돈 48두를 공시하여 28일간 사양시험을 실시하였다. 시험설계는 4처리를 두었으며, 처리당 3반복 반복당 4두의 육성돈이 완전임의 배치 되었다. 시험 처리는 1) CON (대조구), 2) LP+NTPS (대조구 사료에서 유효인 함량을 0.15% 단위 낮춘 사료에 미생물 파이테이즈 (Natuphos$^{\circledR}$) 0.1% 첨가 (500 FTU/kg 사료), 3) LEL+ENP (대조구 사료에서 대사에너지와 라이신 함량을 대조구 사료 대비 각각 3% 낮춘 사료에 탄수화물 복합효소제 (ENDP-POWER$^{\circledR}$) 첨가) 및 4) LPEL+ENZ (대조구 사료에서 대사에너지와 라이신 함량을 대조구 사료 대비 각각 3% 낮추고 유효인 함량을 0.15% 단위 낮춘 사료에 미생물 파이테이즈 0.1%와 탄수화물 복합효소제 0.1%를 각각 첨가)를 두었다. 전체 사양시험 기간동안 (28일), 일당 증체량 (ADG), 일당사료섭취량 (ADFI) 및 사료요구량 (Feed/gain)은 사료처리구에 따라 유의적인 차이를 나타내지 않았다(P>0.05). 그러나 저 영양소 수준 사료에 파이테이즈와 탄수화물 복합효소제의 개별적 혹은 동시 첨가는, 대조구에 비해, 시험 전기간에 걸쳐 사료효율을 다소 개선 시키는 경향을 보여주었다. 건물, 조단백질 및 칼슘 소화율에 있어서는 처리구간에 유의적인 차이를 보이지 않았으나, 총 에너지 소화율에 있어서는 LEL+ENP 처리구와 비교해 볼 때, LP+NTPS와 LPEL+ENZ 처리구가 유의적으로 높게 평가되었다 (p<0.05). 건물 배설량은 LPEL+ENZ 처리구가 가장 낮았으며, 칼슘과 인 배설량은 LP+NTPS구가 가장 낮았다(p<0.05). 사양시험 기간동안 소요된 전체 사료비용은 대조구와 비교하여 볼 때, 효소제 첨가 시 낮았으며, 특히 1 kg 증체 하는데 필요한 사료비용은 대조구와 비교하여 LP+NTPS, LEL+ENP, LPEL+ENZ가 각각 3.5% (609.1 vs. 588.3 원/kg), 13.8% (609.1 vs. 535.3원/kg), 12.4% (609.1 vs. 541.9 원/kg) 낮은 것으로 조사되었다. 결론적으로, 본 실험 결과는 옥수수-대두박 위주 육성돈 사료에 유효인, 에너지 및 라이신 함량을 낮추고 파이테이즈와 탄수화물 분해 복합 효소제를 첨가 시 육성돈의 성장에는 유해한 영향 없이 영양소 배설량을 감소 시킬 수 있으며, 또한 양돈 생산비를 감소 시킬 수 있는 가능성을 제시했다고 판단된다.

Comparative Effects of Phytase Derived from Escherichia coli and Aspergillus niger in Sixty Eight-week-old Laying Hens Fed Corn-soy Diet

  • Two hundred and sixteen 68-week-old Hy-Line brown laying hens were used in a 6-week feeding trial to compare the efficacy of phytases Optiphos (OPT) and Natuphos (NAT), which were isolated from Escherichia coli and Aspergillus niger, respectively. Hens were randomly allotted into six treatments with six replications (six layers in three adjacent cages) per treatment according to their initial BW. The hens were then subjected to one of the following dietary treatments: i) Positive Control (PC; available phosphorus (AP) 0.4%); ii) Negative control (NC; AP 0.2%); iii) NAT1 (NC+250 FTU/kg NAT); iv) NAT2 (NC+500 FTU/kg NAT); v) OPT1 (NC+250 FTU/kg OPT); vi) OPT2 (NC+500 FTU/kg OPT). Feed intake, egg production, egg quality, apparent nutrient digestibility and serum P and Ca concentration were evaluated to compare the effect of the two phytases. Feed intake and eggshell thickness were not affected by the treatments. Superior effects (p<0.05) of OPT were only observed in egg production and egg weight compared with NAT. Characteristics such as eggshell breaking strength, apparent digestibility of N, Ca and P and serum P concentration were equally increased with the supplementation of both phytases (p<0.05), where no significant difference was observed in those characteristics between PC and phytase supplementation at 500 FTU/kg. Equally effective improvements (p<0.05) were also observed in egg production and DM digestibility, where no improvements were observed (p<0.05) between the PC group and the groups with phytase supplementation at 500 FTU/kg. Equal increases in the serum Ca level were observed when the groups with phytase supplementation were compared to the PC group. Overall, the results of this study suggest that NAT and OPT are equally effective at liberating phytate-bound complexes when included in 0.2% available phosphorus diets for 68-week laying hens; either source of phytase can be fed to commercial 68-week laying hens at 500 FTU/kg to correct the negative effects associated with a 0.2% available phosphorus diet. In conclusion, either source of phytase can be fed to commercial first cycle laying hens at 500 FTU/kg to effectively replace inorganic phosphorus when economically justified.

Effects of $1{\alpha}$-Hydroxycholecalciferol and Phytase on Growth Performance, Tibia Parameter and Meat Quality of 1- to 21-d-old Broilers

  • This experiment was conducted to investigate the effects of interaction between $1{\alpha}$-hydroxycholecalciferol ($1{\alpha}$-OH $D_3$) and phytase on growth performance, parameters of tibia and plasma, and meat quality of 1- to 21-d-old broilers. Two hundred and forty male, 1-d-old Arbor Acres broilers were randomly assigned to 20 cages, with 12 chicks per cage. Five treatments were designed, with four cages each. A 2${\times}$2 factorial experiment was designed to test 0 and 5 ${\mu}g/kg$ of $1{\alpha}$-OH $D_3$ in combination with 0 and 500 U/kg of phytase. A basal diet was formulated to contain 2.9 g/kg of non-phytate phosphorus (NPP), and the control diet was formulated to contain a normal level of NPP (4.5 g/kg). Results showed that $1{\alpha}$-OH $D_3$ alone increased tibia ash, contents of calcium and phosphate, breaking strength, concentrations of plasma calcium and phosphate, and water-holding capacity of breast and thigh meat, while it decreased growth of broilers. Phytase alone improved performance and tibia quality. Although growth of broilers was lower than that of the positive control when the diet was supplemented with $1{\alpha}$-OH $D_3$ and phytase, tibia quality was significantly improved by the addition of $1{\alpha}$-OH $D_3$ and phytase. These data suggest that interaction between $1{\alpha}$-OH $D_3$ and phytase at 2.9 g/kg of dietary NPP could significantly increase bone quality of 1- to 21-d-old broilers, while not improving growth performance.

Effects of Multiple Enzyme (ROVABIO® Max) Containing Carbohydrolases and Phytase on Growth Performance and Intestinal Viscosity in Broiler Chicks Fed Corn-Wheat-Soybean Meal Based Diets

  • This study was conducted to investigate the effects of dietary supplementation with multiple enzymes composed of phytase plus carbohydrolases (ROVABIO$^{(R)}$ Max, RM) on growth performance, nutritional availability and intestinal viscosity in broiler chicks. A total of one thousand, one-day-old male broiler chicks were randomly allotted into treatment groups that received one of five experimental diets for 32 days. Each group consisted of 40 birds and all experiments included five replicates. The dietary treatments included PC (a positive control diet), NC1 (65 kcal/kg, 0.15% and 0.10% less ME, available phosphorus and calcium levels, respectively, than the PC diet), NC2 (85 kcal/kg, 0.20% and 0.10% less ME, available phosphorus and calcium levels, respectively, than the PC diet), NC1+RM (NC1 plus ROVABIO$^{(R)}$ Max) and NC2+RM (NC2 plus ROVABIO$^{(R)}$ Max). The average body weights, daily body weight gains and feed conversion rates of the chicks fed a diet containing RM improved significantly or tended to improve. The treatments also had no effect on the carcass characteristics or blood parameters, but the viscosity of the intestinal contents of the chicks fed the diet containing RM was significantly lower than that of chicks in the NC without RM groups. Additionally, chicks fed the dietary RM showed increased breaking strength and ash content of the tibia when compared to chicks that received the non-RM diets. Taken together, the results of the present study indicated that the addition of multiple enzymes consisting of phytase plus NSP enzymes improved the growth performance and mineral status of the tibia in broiler chickens fed corn-wheat-soybean meal-based diets with reduced levels of nutrients. Further, these findings suggest that the improved animal performance is associated with reduced intestinal viscosity by the dietary enzyme complex.

Effect of Dietary Phytase Transgenic Corn on Physiological Characteristics and the Fate of Recombinant Plant DNA in Laying Hens

  • The study aimed to evaluate the potential effects of feeding with phytase transgenic corn (PTC) on organ weight, serum biochemical parameters and nutrient digestibility, and to determine the fate of the transgenic DNA in laying hens. A total of 144 50-week-old laying hens were grouped randomly into 2 treatments, with 8 replicates per treatment and 9 hens per replicate. Each treatment group of hens was fed with diets containing 62.4% non-transgenic conventional corn (CC) or PTC for 16 weeks. The phytase activity for CC was 37 FTU/kg of DM, whereas the phytase activity for PTC was 8,980 FTU/kg of DM. We observed that feeding PTC to laying hens had no adverse effect on organ weight or serum biochemical parameters (p>0.05). A fragment of a poultry-specific ovalbumin gene (ov) was amplified from all tissues of hens showing that the DNA preparations were amenable to PCR amplification. Neither the corn-specific invertase gene (ivr) nor the transgenic phyA2 gene was detected in the breast muscle, leg muscle, ovary, oviduct and eggs. The digestibility data revealed no significant differences between the hens that received the CC- and PTC-based diets in the digestibility of DM, energy, nitrogen and calcium (p>0.05). Phosphorus digestibility of hens fed the PTC-based diet was greater than that of hens fed the CC-based diet (58.03% vs 47.42%, p<0.01). Based on these results, it was concluded that the PTC had no deleterious effects on the organ weight or serum biochemical parameters of the laying hens. No recombinant phyA2 gene was detected in muscle tissues and reproductive organs of laying hens. The novel plant phytase was efficacious in improving the phosphorus digestibility of laying hens.

Effects of Nutrient Specifications and Xylanase Plus Phytase Supplementation of Wheat-based Diets on Growth Performance and Carcass Traits of Broiler Chicks

  • The simultaneous addition of xylanase (5,600 EXU/kg) and phytase (500 FTU/kg) feed enzymes to wheat-based broiler diets was investigated. Starter, grower and finisher diets, with three tiers of nutrient specifications, were fed to 1,440 broiler chicks kept on deep litter from 1-42 days post-hatch, without and with xylanase plus phytase, to determine the effects of diet type and enzyme supplementation on growth performance. The nutrient specifications of type A diets were standard; energy density and protein/amino acid levels were reduced on a least-cost basis to formulate type B diets and further reduced to type C diets. Phosphorus (P) and calcium (Ca) levels were adjusted in supplemented diets. From 1-42 days post-hatch, diet type significantly influenced growth performance. Birds on type C diets had lower growth rates (2,429 vs. 2,631 g/bird; p<0.001), higher feed intakes (4,753 vs. 4,534 g/bird; p<0.005) and less efficient feed conversion (1.96 vs. 1.72; p<0.001) than birds offered type A diets. Enzyme supplementation increased growth rates by 3.2% (2,580 vs. 2,501 g/bird; p<0.005) and improved feed efficiency by 2.7% (1.80 vs. 1.85; p<0.05) over the entire feeding period. There were no interactions between diet type and enzyme supplementation. At 21 days, 5 out of 30 birds per pen were transferred to cages to ascertain treatment effect on apparent metabolisable energy (AME) and nitrogen (N) retention. Xylanase plus phytase enhanced AME (13.48 to 13.91 MJ/kg DM; p<0.001) and N retention (56.3 to 59.7%; p<0.005). Carcass and breast weights of the caged birds were determined following commercial processing. Diet type significantly influenced breast weight, carcass weight and yield. Birds offered Type A diets, in comparison to Type C diets, supported heavier breast (467 vs. 424 g; p<0.001) and carcass weights (1,868 vs. 1,699 g; p<0.001) with superior carcass yields (71.8 vs. 70.6%; p<0.005). Enzyme addition increased carcass weight by 3.9% (1,752 vs. 1,821 g; p<0.005) and breast weight by 5.8% (431 vs. 456 g; p<0.01) without influencing yields. Feed ingredient costs per kg live weight gain and per kg carcass weight indicated that enzyme addition was economically feasible, where supplementation of Type A diets generated the most effective results. Importantly, soluble and total non-starch polysaccharide and phytate contents of the wheat used were typical by local standards. This study confirms the potential of supplementing wheat-based broiler diets with xylanase plus phytase but further investigations are required to define the most appropriate inclusion rates and dietary nutrient specifications in this context.
