  • In order to independently examine the effects of long-chain saturated fatty acids and dietary cholesterol levels on plasma and hepatic cholesterol concentrations, six different diets were fed to male Mongolian gerbils (14 gerbils pes grcup) for an 8-week period. Purified diets contained 36% energy as fat (each saturated fatty acid tested comprised about 20% of the total fat energy) and 0.06% (w/w) cholesterol, corresponding to typical human consumption patterns in Western diets. Fat blends were formulated with natural fat sources. To determine the effects of different saturated fatty acids on plasma and liver cholesterol levels, four of the six diets contained constant levels of all nutrients except for the amounts of lauric acid (12 0), myristic acid (14 : 0), palmitic acid (16 0), and stearic acid (18 : 0). Dietary cholesterol effects were tested using 16 : 0-enriched diets containing 0, 0.006, and 0.06% (w/w) cholesterol. None of the plasma lipids were influenced by fatty acid treatment, including triglycerides, plasma total-, VLDL+LDL-, and HDL-cholesterol. However, hepatic esterified cholesterol concentrations were increased in the palmitic and stearic arid diet groups compared to the lauric and myristic acid diet group. The molar ratios of hepatic EC/FC were the highest in the palmitic acid diet (12.2 $\pm$0.6) and the lowest in the myristic acid diet (6.4$\pm$0.2). Dietary cholesterol significantly (p<0.001) increased the plasma total cholesterol which was due to the increase of both HDL-and VLDL+LDL-cholesterol. In the absence of dietary cholesterol and compared to other species, the gerbil exhibited a high molar ratio of hepatic EC/FC, which was funker elevated by dietary cholesterol feeding (0.06%). The results from this study indicate that hepatic cholesterol concentrations are sensitive to both low levels of dietary cholesterol and saturated fatty acid chain length and also, that plasma cholesterol concentrations are sensitive to low levels of dietary cholesterol .

  • The present studies were designed to compare the effects of both dietary fat levels and P / S ratio on lipid components in plasma and tissues. Changes in plasma HDL-cholesterol, cholesterol and TG, and also in tissue cholesterol and TG were determined in young rats fed diets providing total dietary fat as 10%, 25% or 45% of calories and P / S ratio as 0.2 or 4.0. Plasma cholesterol levels were getting higher as dietary fat levels increased at P / S 0.2. Plasma cholesterol was lower in rats fed dietary fat either 25% or 45 %, each with P / S 4.0. But at 10% no change in plasma cholesterol were observed by P / S 4.0 because of a possible insufficiency of the absolute amount of PUFA. HDL-cholesterol was rather less sensitive to the modification of dietary fat level, but was reduced in rats fed diets of P / S 4.0 at either 25% or 45% fat, even though HDL-cholesterol were increased in the group of 10% with P / S 0.2. Total cholesterol per g- liver were significantly increased as dietary fat levels increased. Liver cholesterol levels were higher in rats fed diets of P / S 4.0 at higher fat levels (25% or 45%) which possibly suggested that a reduction of plasma cholesterol by high PUFA diet was not at least from a decreased synthesis of cholesterol in liver. However, in muscle no significant differences were found by feeding high P / S ratio at each levels of fat. At 10% fat level, compared to 25% or 45%, cholesterol level was lower in g-liver but higher in g- muscle. Plasma TG was decreased as more dietary fat were supplied at P / S ratio, but no consistant response obtained at low P / S ratio. TG per g-liver were reduced by feeding P / S 4.0 diet at 10% or 45% fat level but no differences were found in muscle. P / S 4.0 diet was more efficient in lowering plasma cholesterol TG and HDL-cholesterolt levels only if fat level was more than 25% of the total calories And young rats were more resistant to dietary fat modification.

  • Objectives: The aim of the study was to assess the intake of dietary cholesterol and its major food sources in the Korea National Health and Nutrition Examination Survey (KNHANES). Methods: A total of 20,671 nationally representative sample who had 24-hour recall data from the KNHANES VI (2013-2015) was included in this study. Mean cholesterol intake and the prevalence of subjects with cholesterol intake over the Intake Goal of the Dietary Reference Intakes (DRIs) for Koreans were analyzed. Intakes of cholesterol by food groups or each food were calculated to find out the major food sources for cholesterol intake in Koreans. Results: The mean dietary cholesterol intake was 261.3 mg, which was higher in men (303.5 mg) compared to women (219.1 mg). Dietary cholesterol intake and the prevalence of subjects with cholesterol intake over the Intake Goal of DRIs were the highest in the 19-29 year old group. The eggs was the first major food group source for cholesterol intake in all age groups. Major food sources for cholesterol intake among Korean were egg, chicken, pork, squid and beef, which contributed 66.9% to total cholesterol intake. Conclusions: Although the mean dietary cholesterol intake was under 300 mg, the prevalence of subjects with cholesterol intake over the Intake Goal of DRIs was about 30% in adults. Because both the mean intake and the prevalence of subjects with cholesterol intake over the Intake Goal of DRIs were higher in young adult groups, the dietary cholesterol intake was expected to be increased.

  • The present studies were designed to compare the effects of dietary fat sources and levels on plasma and tissue cholesterol. Changes in plasma total cholesterol and HDL-cholesterol, cholesterol concentration in aortas, liver and intestines were determined in young rats fed diets providing dietary fat as corn-oil or butter and levels as 10%, 20% or 30% of calories respectively 1) Plasma total cholesterol and HDL-cholesterol levels were little sensitive to the modification of dietary fat sources and levels. 2) Dietary cholesterol increased the levels of plasma total cholesterol and this effect was accentuated by feeding butter. But dietary cholesterol did not increase the levels of HDL-cholesterol in the butter-fed groups, but decreased in the corn-oil-fed groups. 3) Liver cholesterol concentrations were significantly higher in rats fed diets of corn-oil than those fed diets of butter. On the contrary, cholesterol concentrations of intestines were significantly higher in the butter-fed groups than the corn-oil-fed groups. However, in aortas, no significant differences were found.

  • In this study, we investigated the effects of lowering the fat and cholesterol in the diets of 26 Korean hypercholesteolemic men($\geq$240mg/dl). They consumed 2378kcal/day with 20.9% of the energy deriving from fat, and a cholesterol intake of 282mg(118mg/1000kcal). The experimental diet consisted of 2400kcal, the same as their usual diet, but the fat content was restricted to 15%, and the cholesterol level to below 100mg/1000kcal. The subjects kept to this diet for four weeks and were asked to maintain their usual life activities during the experimental period. The dietary intake and levels of plasma lipid, lipoprotein-cholesterol and apoprotein of the subjects were analyzed before, two weeks into, and after four weeks dietary intervention. After two of the dietary intervention, there were no significant changes of plasma total cholesterol or triglyceride levels but there was some changes of phospholipid level. However, after four weeks, the levels of plasma total cholesterol, triglyceride, and phospholipid had decreased significantly: 18.2%, 32.9%, and 11.9%, respectively. And the LDL-cholesterol and VLDL-cholesterol levels also showed a marked reduction of 18.1%, and 33.0% respectively without change of HDL-cholesterol level. There were no changes in the levels of Apo-A 1, Apo-B, or Lp(a). The changes of the plasma lipid levels were significantly associated with the changes in dietary fat intake but not the cholesterol intake. In conclusion, although the responses to the dietary intervention varied among the individual subjects, the lowering of dietary fat component from 21% to 15% of energy intake seems to be an effective way to reduce plasma cholesterol and triglyceride levels without decreasing HDL-cholesterol level. It was also found that the restriction of dietary cholesterol to below 100mg/1000kcal not seem to be effective for the hypercholesterolemic patient who already consuming below 300mg/day of cholesterol.

  • The present studies were designed to observe the effects of both dietary fat levels and P/S ratio on lipid components in plasma and tissues of young rats when cholesterol was supplemented at 1%(w/w) to four dietary groups providing total fat as 10%( LF ) or 45% ( HF ) of calories and P/S ratio as 0.2 or 4.0, Low Fat-0.2 : Low Fat-4.0 : High Fat-0.2 : High Fat-4.0. Plasma total cholesterol was increased almost to the same level in all dietary groups after the cholesterol supplement but the value of HF -0.2 was slightly higher than that of LF -0.2. Plasma TG was also increased in all dietary groups when cholesterol was supplemented but was more significantly increased in LF group than in HF group. HDL-cholesterol was slightly increased by cholesterol supplement but there was no effect by the total dietary fat level of previous diet. However, plasma HDL-cholesterol, total cholesterol and TG levels were slightly reduced in high fat diet of P/S 4.0. Total cholesterol per g -liver was higher in HF group than in LF group before and after cholesterol supplement even though it was increased more in LF group with cholesterol addition. Liver cholesterol was also higher in rats fed diets of P/S 4.0 at high fat level before and after cholesterol supplement. In contrast, total cholesterol per g-muscle was reduced by cholesterol supplement in all groups and it was significantly higher in LF than in HF. There was no significant effect in liver TG by total fat level and P/S ratio when cholesterol was supplemented. Muscle TG was lowered in all groups by cholesterol supplement but no effect by total fat level and P/S ratio.

  • This study investigated whether dietary factors are more influential factor than other health behavior such as drinking, smoking and exercise on abnormal serum cholesterol level inspite of Korean dietary pattern differences compared to Europeans and Americans. A double case control study model has been used for the study design. One model consisted of high blood cholesterol cases and control. the other model consisted of low blood cholesterol cased and controls. 5.398 sedentary male workers who had taken medical examinations at a university hospital were used as the study subjects. Out of the study subjects, 36individuals with high blood cholesterol cases and 30 individuals with low blood cholesterol cases were selected. For the 66 individual control selection, the individual control selection, the individuals matching method was adopted. The food frequency method was used to collect the data for assessment of the dietary factors. A standardized questionnaire was used to investigate other health behavior. logistic regression analysis was employed to measure the relative importance between the factors considered. There were no statistically significant differences observed in nutrients consumption or other health behavior among the low, normal and high blood cholesterol groups, An overmatching effect had been suspected as the cause of those findings. However, the results of logistic regression analysis to identify the factors influencing high serum cholesterol showed that odd ratios of dietary factors such as tocopherol(3.0) and saturated fatty acid(1.6) were higher than I. I of smoking and 1.2 of drinking. Similar results were also observed incases of low serum cholesterol. The above findings imply that although the dietary pattern is quite different from that of Europeans and America, the dietary factor is still a significant factor for abnormal blood cholesterol in Koreans. Therefore, the dietary risk factor identified in high fat consumption populations are still relevant for the relatively healthy Korean as guideline for preventive health practices. • The purpose of this study was to investigate the effects of dietary xylooligosaccharide on lipid levels of serum in rats fed high cholesterol diet. Male Sprage-Dawley rats weiging 100$\pm$10g were randomly assigned to groups of two normal(N, N+10X) and four high cholesterol diets which contained 1%(w/w) cholesterol. High cholesterol diet groups were classified to xylooligosaccharide free diet (C group), 5% xylooligosaccharide diet(C+5X group), 10% xylooligosaccharide diet(C+10X group) and 15% xylooligosaccharide diet(C+15X group) according to the level of dietary xylooligosaccharide supplementation. These experimental diets were fed ad libidum for 4 weeks. The weight gain of high cholesterol diet group were significantly increased more than that of normal group, but those of 10% and 15% dietary xylooligosaccharide groups were significantly decreased more than that of high cholesterol diet(C)group. The higher content of xylooligosaccharide, the more food intake was increased. The food efficiencies of 10%, 15% cholesterol diet groups were lower than that of high cholesterol diet(C)group. The levels of serum triglyceride(TG) and total cholestoral were significantly high in cholesterol diet groups compared with normal diet group but were decreased in groups fed 5% and 10% dietary xylooligosaccharide. Especially, the lowest level showed in group fed high cholesterol diet containing 10% xylooligosacchride. High cholesterol diet group containing 10% xylooligosaccharide increased HDL-cholesterol level and then decreased LDL-cholesterol level and atherogenic index compared with other groups. The level of TBARS(thiobarbituric acid reactive substances) in serum was decreased in rat group fed xylooligosaccharide in high cholesterol diet. The higher content of xylooligosacchride, the more gastrointestinal transit time was decreased. The results indicate that dietary xylooligosaccharide can improve status of TG and total cholesterol and repress lipid peroxidation in serum lipid at hypercholesterolemia induced by high cholesterol diet.

  • This study was conducted to investigate the effects of dietary fibers (cellulose ; polydextrose ; pectin ; ricebran) supplementation on the kidney fucntion of hyperlipidemic-induced rats on a high cholesterol diet (5% cholesterol). Serum lipid concentrations were not influenced by dietary fiber sources, but the pectin group was effective in reducing serum lipid levels. Total serum protein and albumin concentrations tended to higher in the polydextrose group, and serum creatinine and urea-N concentrations were higher in the pectin group than those in the other groups. Even though total urinary protein was high in the pectin group, GFR was reduced (18~30% decrease) as compared to other groups. The urea-N level was elevated in the polydextrose group as compared to that of cellulose group(25% increase). Total lipid, triglyceride and cholesterol concentrations in the liver of pectin group were lower than those of other groups. Total lipid and cholesterol concentrations in the kidneys were reduced in pectin and ricebran groups, respectively. The total cholesterol concentration infecal was significantly high in the polydextrose and pectin groups(p<0.05), and the triglyceride was highest in the pectin group. These results indicate no significant effects of dietary fiber supplementation on the kidney function of hyperlipidemic-induced rats on a high cholesterol diet, but pectin was very effective to improve lipid metabolism and to reduce GFR.

  • The study was conducted to investigated the effect of dietary levels of zinc (Zn) on lipid metabolism in cadmium (Cd) treated rats. Sixty male Sprague-Dawley rats weighing about 110$\pm$10g were divided into 6 groups. The Zn levels were low (0ppm), control (30 ppm) and Cd-treated groups were administrated with Cd chloride (5.0mg/kg of body weight) by oral intubation at the same time once a week. Serum total lipid, triglyceride , total cholesterol and DHL-cholesterol contents decreased significantly in low Zn group. In serum dietary high Zn group, it decreased significantly in the content of triglyceride but total cholesterol and HDL- cholesterol revealed increase slightly . Total lipid and triglyceride contents increased by Cd oral intubation, but triblyceride content of control group decreased significantly in Cd-treated groups. HDL-cholesterol content also decreased by Cd oral intubationl. Cd-treatment increased total cholesterol content in low Zn group. Liver triglyceride content increased as dietary Zn level became more increasing and total lipid was not influenced by dietary Zn levels. Liver cholesterol content was higher in low and high Zn groups than that of control group. Liver phospholipid content decreased in low Zn group . Oral intubation of Cd increased in the contents of triglyceride and total lipid. The content of liver Zn was not influenced by the dietary low Zn but that of serum showed a remarkable increased . Oral Cd intubation increased the Cd contents of liver and serum.
