Synergistic effects of dietary $\beta$-1,3 glucan and feed stimulants in juvenile olive flounder (Paralichthys olivaceus)

  • The present study was conducted to investigate the effects of dietary supplementation of $\beta$-1,3 glucan and feed stimulants(BAISM) as a feed additive for juvenile olive flounder (Paralichthys olivaceus). Eight experimental diets supplemented with $\beta$-1,3 glucan and feed stimulants at 0%, $\beta$-1,3 glucan 0.05% + Baism 0.45%, $\beta$-1,3 glucan 0.05% + Baism 0.95%, $\beta$-1,3 glucan 0.10% + Baism 0.90%, $\beta$-1,3 glucan 0.10% + Baism 1.90%, $\beta$-1,3 glucan 0.15% + Baism 1.35%, $\beta$-1,3 glucan 0.15% + Baism 2.85% and $\beta$-1,3 glucan 0.30% + Baism 2.70% of diets as a dry-matter(DM) basis were prepared. Three replicate groups of fish averaging 9.2 $\pm$ 0.1g (Mean $\pm$ SD) were randomly distributed in each aquarium as a group of 15 fish and fed one of eight experimental diets for seven weeks. After the feeding trial, $\beta$-1,3 glucan 0.10% + Baism 0.90%, $\beta$-1,3 glucan 0.10% + Baism 1.90% diets had a higher weight gain (WG), feed efficiency(FE), specific growth rate(SGR) and protein efficiency ratio(PER) than did fish fed 0%, $\beta$-1,3 glucan 0.05% + Baism 0.45%, $\beta$-1,3 glucan 0.05% + Baism 0.95%, $\beta$-1,3 glucan 0.15% + Baism 2.85% and $\beta$-1,3 glucan 0.30% + Baism 2.70% (P<0.05). however, there was no significant difference among fish fed $\beta$-1,3 glucan 0.05% + Baism 0.45%, $\beta$-1,3 glucan 0.05% + Baism 0.95%, $\beta$-1,3 glucan 0.15% + Baism 2.85% and $\beta$-1,3 glucan 0.30% + Baism 2.70%(P>0.05). and $\beta$-1,3 glucan 0.10% + Baism 0.90% diets had a higher peak value of CL(Chemiluminescence) and lysozyme activity, than did fish fed the other diets (P<0.05). These results indicated that dietary sipplementation of $\beta$-1, 3 glucan and Baism affected growth, feed efficiency, specific growth rate, protein efficiency ratio, Peak value of CL and Lysozyme activity, and the optimum dietary supplementation level of $\beta$-1, 3 glucan and Baism as a feed additive could be approximately $\beta$-1, 3 glucan 0.10% + Baism 0.90% of diet in juvenile olive flounder (Paralichthys olivaceus).

Effects of Dietary β-Glucan on Short Chain Fatty Acids Composition and Intestinal Environment in Rats (식이 베타-글루칸이 흰쥐의 장내 단쇄지방산 조성 및 장내환경 개선에 미치는 영향)

  • The effects of dietary ${\beta}$-glucan, obtained from bacterial fermentation, on the intestinal mass, short chain fatty acids, lactate production and pH in Sprague-Dawley (SD) rats were evaluated. SD rats fed with 0% (control group), 1% or 5% ${\beta}$-glucan supplemented diets (w/w) for 3 weeks. The presence of ${\beta}$-glucan in the diets resulted in a significant increase in colonic contents in a dose dependent manner. The amount of short chain fatty acids increased in rats fed ${\beta}$-glucan diets. Rats fed the 5% ${\beta}$-glucan diets had higher levels of acetate, propionate and butyrate by 1.8, 1.7 and 3.0 fold of the control group in the cecum, and 2.2, 2.9 and 3.1 fold of the control group in the colon, respectively. The ${\beta}$-glucan diets also significantly increased the levels of cecal and colonic lactate by 1.4~3.4 fold, when compared to the control diet, indicating that dietary ${\beta}$-glucan stimulated the growth of lactic acid bacteria within the intestine. These results suggest that dietary ${\beta}$-glucan, by providing short chain fatty acids and reducing the cecal and colonic pH, may be beneficial in improving gut health, and provide evidence for the use of ${\beta}$-glucan as a dietary supplement for human consumption.

Changes In Dietary Fiber Content of Barley during Pearling and Cooking (도정 및 가열조리중 보리의 식이섬유 함량변화)

  • Three hull-less barleys and three covered barleys grown in Korea were pearled to give 0% and 60% yield, respectively. Whole barleys and pearled barleys were analyzed for total, insoluble, soluble dietary fiber and ${\beta}-glucan$ contents. Whole hull-less barleys contained average 17.1% total dietary fiber, and whole covered barleys contained average 23.9% total dietary fiber. Pearled hull-less barleys contained 9,2% total dietary fiber and 4.8% solule dietary fiber. Pearled covered barleys contained 11.9% total dietary fiber and 6.0% soluble dietary fiber. Whole barleys contained $3.2{\sim}3.9%$ (${\beta}-glucan$, and pearled barleys contained $3.5{\sim}5.4%$ (${\beta}-glucan$. Soluble dietary fiber and (${\beta}-glucan$ contents of barley were not affected by cooking, while insoluble dietary fiber content was increased by cooking.

  • Dietary pattern has paramount importance in shaping the gut microbiota and its associated host health. Herein this study, long term (12 weeks) impact of mushroom derived dietary fiber, ${\beta}-glucan$, is investigated for its effect on low fiber diet consumption. Inclusion of dietary fiber into the low fiber diet (LFD) increased the abundance of genera Lactobacillus and Anaerostipes, the microbes responsible for butyrate (major 'fuel source' of colonocytes) production. Mice fed LFD with ${\beta}-glucan$ showed significant increase in the length of small intestine compared to that of the LFD group without ${\beta}-glucan$. Further, dietary fiber consumption enhanced goblet cell density along with mucosal layer thickness. These results indicate promising effects of ${\beta}-glucan$ towards maintenance of healthy gut and gut microbiota.

Effect of Dietary β-1,3/1,6-glucan Supplementation on Growth Performance, Immune Response and Plasma Prostaglandin E2, Growth Hormone and Ghrelin in Weanling Piglets

  • The experiment was conducted to evaluate the effect of ${\beta}$-1,3/1,6-glucan on growth performance, immunity and endocrine responses of weanling piglets. One hundred and eighty weanling piglets (Landrace$\times$Large White, $7.20{\pm}0.25kg$ BW and $28{\pm}2$ d of age) were randomly fed 1 of 5 treatment diets containing dietary ${\beta}$-1,3/1,6-glucan supplemented at 0, 25, 50, 100 and 200 mg/kg for 4 wks. Each treatment was replicated in 6 pens containing 6 pigs per pen. On d 14 and 28, body weight gain, feed consumption and feed efficiency were recorded as measures of growth performance. Peripheral blood lymphocyte proliferation and serum immunoglobulin G (IgG) were measured to study the effect of dietary ${\beta}$-1,3/1,6-glucan supplementation on immune function. Plasma prostaglandin E2 (PGE2), growth hormone (GH) and ghrelin were measured to investigate endocrine response to ${\beta}$-1,3/1,6-glucan supplementation. Our results suggest that average daily gain (ADG) and feed efficiency had a quadratic increase trend with dietary ${\beta}$-1,3/1,6-glucan supplementation from d 14 to 28, whereas it had no significant effect on average daily feed intake (ADFI). The treatment group fed with 50 mg/kg dietary ${\beta}$-1,3/1,6-glucan supplementation showed a numerical increase in ghrelin, a similar change trend with ADG and no significant effect on GH. Lymphocyte proliferation indices, serum IgG and plasma PGE2 concentrations varied linearly with dietary supplementation levels of ${\beta}$-1,3/1,6-glucan on d 14. Higher levels of ${\beta}$-1,3/1,6-glucan may have a transient immuno-enhancing effect on the cellular and humoral immune function of weanling piglets via decreased PGE2. Taking into account both immune response and growth performance, the most suitable dietary supplementation level of ${\beta}$-1,3/1,6-glucan is 50 mg/kg for weanling piglets.

Effect of Dietary Beta-Glucan on the Performance of Broilers and the Quality of Broiler Breast Meat

  • A total of 400, one day-old commercial broiler chicks were divided into five diet groups (negative control, positive control group with 55 ppm Zn-bacitracin, 15 ppm ${\beta}$-glucan, 30 ppm ${\beta}$-glucan, and 60 ppm ${\beta}$-glucan) and fed for six weeks. Ten broilers were allotted to each of 40 floor pens. Eight floor pens were randomly assigned to one of the 5 diets. Each diet was fed to the broilers for 6 weeks with free access to water and diet. The survival rate, growth rate, feed efficiency, and feed conversion rate of the broilers were calculated. At the end of the feeding trial, the birds were slaughtered, breast muscles deboned, and quality parameters of the breast meat during storage were determined. The high level of dietary ${\beta}$-glucan (60 ppm) showed better feed conversion ratio and survival rate than the negative control. The survival rate of 60 ppm ${\beta}$-glucan-treated group was the same as that of the antibiotic-treated group, which showed the highest survival rate among the treatments. There was no significant difference in carcass yield, water holding capacity, pH, color, and 2-thiobarbituric acid reactive substances values of chicken breast meat among the 5 treatment groups. Supplementation of 60 ppm ${\beta}$-glucan to broiler diet improved the survival rate and feed conversion rate of broilers to the same level as 55 ppm Zn-bacitracin group. The result indicated that use of ${\beta}$-glucan (60 ppm) can be a potential alternative to antibiotics to improve the survival and performance of broilers. However, dietary ${\beta}$-glucan showed no effects on the quality parameters of chicken breast meat.

  • This study was conducted to evaluate the physicochemical characteristics of pound cake made of substituted flour with different amount of $\beta-glucan$ i.e. 3, 6 and $9\%$ respectively. $\beta-glucan$ is a functional food material produced from Agrobacterium sup. R259 KCTC 10197BP. The specific gravity and pH of dough were found to be increased according to $\beta-glucan$ content Rheological property such as adhesiveness of dough has no difference between control and $\beta-glucan$ added groups. The pH of baked pound cake was not significantly different between control and $\beta-glucan$ groups. The volume of baked pound cake wag not significantly different between control and the ones with up to $6\%$ $\beta-glucan$, Lightness (L), redness (a) and yellowness (b) value of Hunter color system of cake crumb were not significantly different among teated groups, except a value of crust in $9\%$ $\beta-glucan$ group. the moisture content of cake increased according to $\beta-glucan$ content. Hardness by texture analyser was decreased with increasing $\beta-glucan$ content although cohesiveness wag not changed in β$\beta-glucan$ groups, compared with the control. Sensory evaluation results showed that the mean scores of over·all acceptability in 3 and $6\%$ $\beta-glucan$ were higher than that of control. Based on these results, the addition of $\beta-glucan$ to pound cake up to $6\%$ was considered to be desirable especially in terms of texture.

  • A soluble dietary fiber, $\beta$-glucan, contained in oat and barley has nutritional benefits such as hypocholesterolemic effects and influences blood glucose regulation. The contents of $\beta$-glucan in both cereals range from 3 to 7% with the exception of a certain barley genotype which contains up to 16% $\beta$-glucan. $\beta$-Glucan is distributed mainly in the cell walls of endosperm and the distal (bran) portion of kernel. Various procedures have been developed for increasing the extraction yield of $\beta$-glucan. Oat gum prepared with weak alkali extractionand alcohol proecipitation following protein removal usually contains 80% $\beta$-glucan.The most commonly used method for $\beta$-glucan quantitiation is an enzymatic procedure combining lichenase plus $\beta$-glucosidase followed by measuring the amount of glucos released by glucose oxidase-peroxidase treatment. The increase in foam-and emulsion-stabilizing capacity of $\beta$-glucan is due to the increase in viscosity of the aqueous phase. Therefore, $\beta$-glucan shows great potentials as a thickener and a stabilizer.

  • mechanisms for the hypocholesterolemic effects of $\beta$-glucan remain unclear. Rats were divided into 3 groups ; normal control group, atherogenic group(oral administration of cholesterol 40 mg/kg/day plus vit. $D_2$ 320,000 IU/kg/day), $\beta$-glucan treatment group(atherogenic treatment plus $\beta$-glucan 0.135 g/kg/day). The $\beta$-glucan treatment group showed moderate increases of serum lipids concentration compared with atherogenic group. In histopathological examination, aortas showed no critical lesions. The total fecal neutral sterols and bile acids excreted for 6 days was increased compared with both normal and atherogenic group. To compare effects of soluble fiber and insoluble fiber extracted from barley on postprandial lipemia, 5 healthy male adults ingested on separate days a low-fiber(total dietary fiber 2.61g) control meal or dietary fiber-enriched(12.61g) meals. Fasting and postprandial blood samples were obtained for 6.5h and serum lipids were analyzed. The serum total lipids, total cholesterols, LDL & VLDL-cholesterol were markedly reduced with soluble fiber-enriched meals, but no decrease with insoluble fiber-enriched meals. These results suggest that mechanisms for the hypocholesterolemic effect of $\beta$-glucan on rats were due to the inhibition of cholesterol absorption in the intestinal lumen and acceleration of cholesterol catabolism in the liver. And the soluble dietary fiber($\beta$-glucan) has the hypocholesterolemic effect by dropping serum LDL & VLDL-cholesterol in the clinical study.

Physiological Function in vitro of ${\beta}-Glucan$ Isolated from Barley (보리가루에서 분리한 ${\beta}-Glucan$의 생리적 기능성)

  • In order to prove physiological function of ${\beta}-Glucan$ isolated from barley flour by enzymatic method, in vitro experiments simulating the passive membrane transport of gastrointestinal tract were carried out using dialysis membrane. The yield of ${\beta}-Glucan$ from barley flour was $6.2{\%}$ and its constituents were determined to give $81.6{\%}$ total dietary fiber, $72.9{\%}$ soluble dietary fiber, $8.7{\%}$ insoluble dietary fiber, $8.5{\%}$ moisture, $2.5{\%}$ protein and $7.4{\%}$ ash. The water holding capacity of the ${\beta}-Glucan$ preparation was 6 g water/g dry material. The glucose retardation index after 30 minute dialysis was $13.5{\%}$ in the presence of $3{\%}$ ${\beta}-Glucan$. As the dialysis period became longer, the retarding effect toward glucose absorption decreased and the effect was close to zero after 2 hour dialysis. The bile acid retardation index after 30 minute dialysis was 3, 12 and $18{\%}$ in the presence of 1, 3 and $5{\%}$ ${\beta}-Glucan$, respectively. The effect was higher than the glucose retardation index and decreased as the dialysis time elapsed.
