Obesity has become one of the main public health problems. Saussurea lappa (Asteraceae), syn Aucklandia lappa and Saussurea costus, is a well-known herbal medicine that has been used for treating various ailments, such as inflammatory and gastrointestinal diseases. The present study examined the anti-obesity effect of S. lappa extract (SLE) in 3T3-L1 adipocytes and high fat diet (HFD)-induced obese mouse model. SLE significantly inhibited the differentiation from preadipocytes to adipocytes of cultured 3T3-L1 in dose-dependent manner. In addition, SLE significantly decreased the body weight gain and the food efficiency ratio of mice fed HFD during 9 weeks. Further study must be performed for the pharmacological mechanism and safety of SLE as well as the identification of active compound in SLE. Our results revealed that S. lappa suppresses the adipogenesis in cultured cells and the obesity in rodent models. Therefore, S. lappa may be useful toward the development of new potent anti-obesity drugs.
Objectives : Samhwangsasim-tang (SHSST) is a traditional Korean medication, which has been used in Korea for treatment of hypertension and chest pain. Hyperlipidemia and inflammation could influence hypertension and chest pain. This study investigated whether and how SHSST reduces the hyperlipidemia and inflammation related to high-cholesterol diet-induced obesity in rats. Methods : Mice were divided randomly into four groups: the normal diet group, high-cholesterol diet group, low dose treatment group supplemented with 30% ethanol extract of SHSST (L) and high dose treatment group supplemented with 80% ethanol extract of SHSST (H). L and H groups were orally administered with SHSST at the dose of 50mg/kg a day respectively and others were administered with the same volume of physiological saline. Results : Administration of SHSST resulted in a decrease in serum levels of total cholesterol and low-density lipoprotein. Expression of hepatic genes(SREBP2, LXR, LDLR, and HMG-CoA) related with cholesterol metabolism was also suppressed. In addition, SHSST decreased the expression of inflammation-related gene (TNF-${\alpha}$, IL-6, ICAM-1, VCAM-1, TGF-${\beta}1$ and fibronectin). Histological examinations also showed that the size of the adipocytes was smaller in the SHSST treated group than in the high-colesterol diet group. In an in vitro study, SHSST inhibited the production of nitric oxide in a concentration-dependent manner. Conclusions : This study indicates that SHSST has anti-hyperlipidemia and anti-inflammatory effects. It may also suggest that SHSST may be alternative therapy for treatment of hyperlipidemia and its complications.

The purpose of this study was to investigate anti-obesity and anti-hyperlipidemic effects of extracts from Geranium thunbergii (GT) in high-fat-diet-induced obese rats. Animals were randomly divided into four groups [normal diet, high fat diet, MeOH extract of GT (GTM), and BuOH fraction of GT (GTB)] and GT samples were treated with dose of 100 mg/kg for 8 weeks. It was observed that GTB-treated group significantly reduced body weight gain, food intake, epididymal fat weight, and triglyceride level in serum and liver compared to control group. The rats fed GTB also decreased contents of thiobarbituric acid-reactive substances (TBARS), hydroxyl radical, and xanthin oxidase (XO) increased by high fat diet. Furthermore, the anti-oxidant enzymes such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase were increased by GTB treatment. The experimental results indicate that GTB has anti-obesity and anti-hyperlipidemic effects, as well as radical scavenging activity.
This study was undertaken to evaluate the effect of bacteria-derived $\beta$-glucan fiber on serum lipids, adiposity and uncoupling protein (UCP) expression in rats. In order to induce obesity, Sprague-Dawley weanling male rats were allowed free access to AIN-76A diet until 4 weeks of age, and fed high-fat diet (beef tallow, $40\%$ of calories as fat) for 6 weeks until 10 weeks of age. Rats were then fed with $0\%$ thigh- fat control group), $1\%$, or $5\%$ bacterial ~-glucan supplemented high-fat diets (w/w) for another 6 weeks. For comparison, normal control group was fed with AIN-76 diet $11.7\%$ fat). Supplementation with bacterial $\beta$-glucan resulted in a significant reduction of high-fat-induced white fat (i.e., visceral and peritoneal fat) development, adipocyte hypertrophy, and development of hyperinsulinemia and hyperleptinemia. Serum triglyceride, total cholesterol, and free fatty acid levels were greatly reduced, but, HDL-cholesterol concentrations were increased by bacterial $\beta$-glucan supplementation. Serum leptin level was lower in the $\beta$-glucan groups than in the high-fat group. The expression of UCPs (UCP1, UCP2, and UCP3) in brown adipose tissue (BAT) were significantly increased by $5\%$ bacterial $\beta$-glucan-containing diet. This study suggests that the anti-obesity effect of $5\%$ bacterial $\beta$-glucan is attributed to upregulation of UCPs and inefficient energy utilization.

Objectives: The objective of this research is to develop new animal-experimental model for Sasang Constitutional Medicine, especially for partial Taeyangin(one of four constitution which has good pulmonary function and poor hepatic function) by AAP intraperitoneal injection, and to estimate from the viewpoint of obesity and lipid metabolism. Methods: The C57bl/6J mice was divided into 4 groups ; Normal group, AAP group, High-Fat-Diet(HFD) group, and HFD+AAP group. 200mg AAP was injected intraperitoneally to the AAP group twice a week for six weeks, and HFD group was fed with 60%-High-fat Diet for six weeks. HFD+AAP group got both AAP injection and 60%-High-fat Diet at the same time for the same period. In this period, We measured the weight and Food Efficiency Ratio(FER, %) once a week. After six weeks, We conducted the blood chemical test from the groups, and extracted the fat tissue to measure weight. Results & conclusion: In the liver function test, two AAP groups had higher AST and ALP, and normal LDH. The blood level of creatinine from all groups were normal. The rate in weight was lesser by 7.8% in HFD+AAP group, and had lesser FER than HFD group. Also They had lesser Total cholesterol and LDL cholesterol, and had more HDL cholesterol than HFD group. HFD+AAP group hadmore glucose in serum and lesser Insulin-like Growth Factor 1(IGF-1) than HFD group.
This study was performed to investigate the effect of lysine-limited diets containing different levels of L-carnitine on body weight and lipid metabolism in obesity-induced adult rats. Eight-month-old male Sprague-Dawley rats (n = 90) were raised for one month with high fat diet (40% fat as calorie) to induce obesity. After induction of obesity, rats weighing 739.5 g were randomly blocked into three groups according to the body weight and raised for eight weeks with control diet (Co), 50% lysine-limited diet (-L), 50% lysine limitation with 0.3% pivalate diet (-L + P). Each of three groups was allotted to 0.0% L-carnitine (0.0% CT), 0.5% L-carnitine (0.5% CT) and 2.5% L-carnitine (2.5% CT) groups, respectively. The levels of AST, ALT, total protein and albumin in plasma were within the normal range. Daily food intake and calorie intake tended to be lower in 2.5% CT groups than those of other groups regardless lysine limitation or pivalate intake. And body weight gain and calorie efficiency ratio (weight gain (g) /calorie intake (100 kcal)) were significantly the lowest in 2.5% CT groups among all experimental groups regardless of lysine limitation or pivalate intake. The weights of perirenal, epididymal fat pads and brown adipose tissue in 2.5% CT groups were significantly lower than 0.0% CT groups. Plasma total lipid, triglyceride, total cholesterol concentrations in all groups were not significant by experimental compound. HDL-cholesterol concentrations in -L + P +2.5% CT group were highest in -L + P groups. Levels of hepatic total lipid, triglyceride and total cholesterol in 2.5% CT groups were tend to be lower those than in 0.0% CT groups regardless of dietary lysine limitation and pivalate intake. Fecal total lipid excretions of 2.5% CT groups were significantly lower than in 0.0% CT groups in all experimental groups. But fecal triglyceride excretions of 2.5% CT groups were significantly higher than 0.0% CT groups regardless of lysine limitation and pivalate. In conclusion, there was no difference on body weight and lipid metabolism by dietary lysine limitation and pivalate intake. And feeding of 2.5% L-carnitine was more effective than feeding of 0.5% L-carnitine and 0.0% L-carnitine in reduction of body weight, body fat and lipid metabolism.
Purpose: Colorectal cancer, which is one of the most commonly diagnosed cancers in developing and developed countries, is highly associated with obesity. The association is largely attributed to changes to western style diets in those countries containing high-fat and high-energy. Luteolin (LUT) is a known potent inhibitor of inflammation, obesity, and cancer. In this study, we investigated the effects of LUT on chemical-induced colon carcinogenesis in high fat diet (HFD)-fed obese mice. Methods: Five-week-old male C57BL/6 mice received a single intraperitoneal injection of azoxymethane (AOM) at a dose of 12.5 mg/kg body weight. Mice were then divided into four groups (n = 10) that received one of the following diets for 11 weeks after the AOM injection: normal diet (ND); HFD; HFD with 0.0025% LUT (HFD LL); HFD with 0.005% LUT (HFD HL). One week after AOM injection, animals received 1~2% dextran sodium sulfate in their drinking water over three cycles consisting of five consecutive days each that were separated by 16 days. Results: Body weight, ratio of colon weight/length, and tumor multiplicity increased significantly in the HFD group compared to the ND group. Luteolin supplementation of the HFD significantly reduced the ratio of colon weight/length and colon tumors, but not body weight. The levels of plasma $TNF-{\alpha}$ and colonic expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 protein increased in response to HFD, but were suppressed by LUT supplementation. Immunohistochemistry analysis also showed that iNOS expression was decreased by LUT. Conclusion: Consumption of LUT may reduce the risk of obesity-associated colorectal cancer by suppression of colonic inflammation.

Cheonggukjang and chaga mushrooms have numerous health benefits, and have been used in alternative medicine. Therefore, a powder mixture of 98: Cheonggukjang and 2: Chaga extracts was fermented with Lactobacillus acidophilus KCTC3925 (FCC) and its anti-obesity effects in high-fat diet (HFD)-induced obese mice were determined. Five-week-old male ICR mice were fed a normal diet or HFD in the presence or absence of 3% and 5% FCC by weight (n = 10 per group). After 12 weeks, the mice were sacrificed, and the serum and tissue samples were collected for analysis. Body weight and epididymal fat pad weight were significantly lowered in the 3% and 5% FCC groups compared with those in the HFD control group (p < 0.01). FCC supplementation suppressed serum triglyceride and increased serum HDL-C levels (p < 0.01). Serum GOT, GPT, and leptin levels, hepatic COX-2 mRNA expression, and splenic COX-2 and IL-4 mRNA expression were significantly higher in the HFD groups than in the control group (p > 0.05); however, except for splenic IL-4 levels, the increases were significantly attenuated by FCC supplementation. Expression of ICAM-1, an aortic inflammatory marker, was significantly increased in the HFD group; this effect was suppressed in the 3% FCC group (p < 0.01) but not in the 5% FCC group. FCC suppressed the body weight and epididymal fat pad weight gain, as well as inflammatory responses in the liver and spleen of HFD-fed mice. Thus, FCC supplementation will be beneficial for the treatment of obesity-related effects.
In order to determine the possibility that Chrysanthemum indicum L. cultured with Lactococcus lactis (CILL) is a material for obesity suppression food, the body weight, body fat mass, and T cells were determined in C57BL/6 mice induced by a high fat diet. The CILL (25.15±2.44 g) demonstrated weight loss from week 4 onward and maintained a low weight gain from week 1 to week 8 (1.00±0.53 g). The 8-week body weight (30.38±4.17 g) indicated loss of 13.15% when compared to the HFD (60% high fat diet, 34.99±2.09 g). Fat mass decreased to 10.3022±2.8813 g, and the absolute liver weight decreased relative to that in the HFD. CD4+ T cells were 4.84±1.33%, CD8+ T cells 7.02±2.26%, and CD4+CD8+ T cells 1.46±0.81%, which were all higher than those in the HFD. As a result, CILL can be used as a material for preventing obesity as an effective measure toward reducing weight when consumed orally.

The aim of this study was to investigate the effect of Taeeumjowuitanggagam-bang (TJV) on the mRNA expression of Sterol regulatory element binding proteins (SREBPs), Tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and Interlukin-6 (IL-6) that are considered to play an important role in lipid and glucose metabolism. For diet-induced obese studies, we split mice into 2 groups. The low fat diet group (LFD, n=8) were supplied with general diet for 10 weeks and the high fat diet group (HFD, n=18) were supplied with 60 kcal% fat diet for 10 weeks. And then The HFD group, the diet-induced obese group, were divided into 3 groups ; a group supplied with normal saline, a group treated with TJV 200 mg/kg and a group treated with TJV 500 mg/kg. They were treated orally with TJV and measured their body weight every day during 10 weeks. After that, we measured mRNA expressions of TNF-$\alpha$, IL-6 and SREBP-1c in liver, and blood concentrations of glucose, total cholesterol and triglyceride too. The results are as follows. The TJV reduced glucose and total cholesterol of blood concentration. The TJV reduced the mRNA expressions of TNF-$\alpha$ and SREBPs in liver. However, We couldn't find the TJV effects on the mRNA expression of IL-6, triglyceride blood concentration, and body weight among groups. The TJV stained liver tissue less red than control group. These results suggest that TJV may be effective for regulation of lipid and glucose metabolism in liver.
