To elucidate the effect of antioxidant complex containing $\beta-carotene$, vitamin E, vitamin C, Ginkgo Biloba leaf extract and selenium on oxygen :tree radical production and detoxification system, rats were fed normal diet and normal diet with antioxidant complex 0.1%, 0.3% and 0.5% for 3 weeks. Feed efficiency ratio, changes in body weight, weight gain and amounts of feces of rat are similar in four groups. Liver weight per body weight and hepatic lipid peroxide weight increased in 0.5% group. However, hepatic glutathione contents in all antioxidant complex added groups were significantly increased compare with normal control group. On the other hand, the activity of xanthine oxidase was a little increased due to the amounts of antioxidant complex. Superoxide dismutase and gutathione peroxidase activity of 0.1% antioxidant complex added group were increased about $10{\sim}20%$ in comparison to normal control group. These results suggest that the supplementation of antioxidant complex 0.1% to basal diet may reduce the hepatic damage caused by free radicals.
This study was conducted to investigate the effects of Forsythia viridissima Lindl. (FVL) on antioxidative defense system and lipid peroxidation of liver in rats fed high-cholesterol diet. Sprague-Dawley male rats weighing 100 $\pm$ 10 g were randomly assigned into five experimental groups fed 0.5% cholesterol ; HC group which was not supplemented FVL extract, 0.05% methanol extract diet group (MSI group), 0.1% methanol extract diet group (MS2 group), 0.025% ethylacetate-souble fraction diet group (ES1 group) and 0.05% ethylacetate-souble fraction diet group (ES2 group). Experimental diets were fed ad libitum to the rats for 3 weeks. The hepatic xanthine oxidase (XOD) activity in the MS2 group was decreased to 20% as compared to HC group. The activities of hepatic superoxide dismutase (SOD) and catalase (CAT) were not significantly different among all the high cholesterol diet groups. The hepatic glutathione peroxidase (GSHpx) activity in MS2, ES2 groups were significantly increased as compared to HC group. The hepatic glutathione S-transferase (GST) activity in the MS2 group was increased to 20% as compared to HC group. The levels of hepatic TBARS in the MS1, MS2, ES1 and ES2 groups were reduced by 13%, 21%, 13% and 21%, respectively, as compared with HC group. The contents of lipofuscin in liver tissue was not significantly different among all the experimental groups. The results indicate that FVL extract may reduce oxidative damage by activating antioxidative defense system of liver in rats fed high-cholesterol diets.
Recently, epidemiological evidence has raised concerns that moderate elevation in body iron stores may increase oxidative stress and the risk of cardiovascular disease and cancer. Onion flesh or peel contains antioxidant such as flavonoids and alk(en)ylcysteine sulphoxides. This study was conducted to examine the effect of onion flesh or peel feeding on antioxidative capacity in aged rats supplemented with high dietary iron. Thirty-two Sprague-Dawley male 16-month-old rats weighing $618{\pm}6g$ were acclimated for 10 days with medium-iron diet (35ppm in diet), and blocked into 4 groups according to their body weights and raised for 3 months on either control diets (adequate iron-35ppm or high iron-350ppm) or experimental diets containing onion flesh/peel (5% w/w in diet) with high iron (350ppm). Rats fed high iron-onion peel diet had significantly high quercetin and isorhamnetin levels in plasma whereas rats fed high iron-onion flesh diet did not show. Plasma TBARS level was lowered by onion flesh or peel diet with high iron supplementation. However, there was no significant difference in cellular DNA damage in brain and kidney tissue among all experimental groups. We concluded that high iron diet (10 times higher than requirement) tend to increase oxidative stress and it is plausible that onion flesh or peel feeding enhances antioxidative capacity in the elderly even with iron supplementation.

Monocyte chemotactic protein-3 (MCP-3), a chemokine that is in a superfamily of structurally related small chemotactic cytokines involved in leukocyte trafficking, is regarded as a key factor in atherogenesis. In this study, we examined the changes in atherogenic parameters including hepatic lipid accumulation and oxidative balance in MCP- 3-overexpressing transgenic mice (MCP-3 mice) under atherogenic conditions. To induce an extreme atherogenic condition, mice were fed a high-fat, high-cholesterol (HFHC) diet for 12 weeks. The body weight and food intake were not changed by MCP-3 overexpression in the aorta. On a HFHC diet, the MCP-3 mice had higher plasma levels of total cholesterol and a higher atherogenic index compared with wild-type mice, although there were no differences in the plasma HDL-cholesterol and triglyceride levels. Furthermore, an increase in lipid accumulation was observed in the aortas as well as the livers of the HFHC diet-fed MCP-3 mice compared with wild-type mice. The activities of antioxidant enzymes increased in the livers of the HFHC diet-fed MCP-3 mice, whereas supplementation with antioxidants, naringin and hesperidin, reversed the activities of the hepatic antioxidant enzymes in HFHC diet-fed MCP-3 mice, indicating that there might be more oxidative damage to the tissues in the HFHC diet-fed MCP-3 mice leading to progression towards atherosclerosis and hepatic steatosis. Microarray analyses of the aorta revealed atherosclerosis-, PPARs-, lipoprotein receptor, and apolipoprotein-related genes that were affected by the HFHC diet in MCP-3 mice. These findings suggest that aortic MCP-3 overexpression may contribute to the development of atherosclerosis and hepatic steatosis under atherogenic conditions.
This study was conducted to examine the effects of dietary xylooligosaccharides on hepatic HMG-CoA reductase activity and morphological exchange of liver in rats fed high fat diet. Sprague-Dawley male rats weighing 100 $\pm$ 10 g were randomly divided into four groups, two normal diets and two high fat diets containing 1% cholesterol and 10% lard. Two normal diets were classified into a basal diet (normal group) and 10% xylooligosaccharide diet (NX group). The high fat diet groups were classified into a HF group without xylooligosaccharides diet and HFX group supplemented 10% xylooligosacchride diet. Experimental diets were fed ad libidum to the rats for 4 weeks and then they were sacrificed. The body weight of high fat diet (HF group) was increased more than that of normal group, but it was significantly decreased by xylooligosacchrides supplementation. The food intake was not significantly different among the all groups. The weight of liver, small intestine and cecum of all xylooligosaccharide supplemented groups were significantly heavier than those of normal and HF groups. The activity of hepatic HMG-CoA reductase, a rate limiting enzyme of cholesterol biosynthesis, in xylooligosaccharide supplemented groups was higher than that of HF group. Light micrographs revealed that the structures of hepatocytes in xylooligosaccharide supplemented groups were preserved well, compared to HF group. The xylooligosaccharide supplementation exerted a lipid-lowering action by decreasing cholesterol and triglycerides contents in hepatic tissue. In conclusion, the activity of hepatic HMG-CoA reductase and damage of liver in rats fed high fat diets were improved by dietary xylooligosaccharides.

The purpose of the present study was to effect of red pepper seeds powder on antioxidative defense system and oxidative damage in rats fed high fat high cholesterol diet. Rats were divided into five experimental groups which are composed of normal diet group, high fat high cholesterol diet group, high fat high cholesterol diet with 5% red pepper seeds powder supplemented group (SA group), high fat high cholesterol diet with 10% red pepper seeds powder supplemented group (SB group), and high fat.high cholesterol diet with 15% red pepper seeds powder supplemented group (SC group). Supplementation of red seed pepper groups (SA, SB, and SC groups) resulted in increased activities of hepatic glutathione peroxidase and superoxide dismutase. However, there was no significant difference in the activity of hepatic catalase among all experimental groups. Hepatic superoxide radical contents in microsome and mitochondria were significantly reduced in red pepper seeds powder supplemented groups. Hepatic hydrogen peroxide contents in mitochondria were significantly reduced 15% red pepper seeds powder supplemented group. Hepatic carbonyl values in microsome were significantly reduced in 10% and 15% red pepper seeds powder supplemented groups. Thiobarbituric acid reaction substance (TBARS) values in liver and plasma were reduced in red pepper seeds powder supplemented groups. These result suggest that red pepper seeds powder may reduce oxidative damage by the activation of antioxidative defense system in rats high fat.high cholesterol diets.

This study was carried out to determine the action mechanism of sulfur-grain larvae extract (SGE) on anti-obesity and the reduction of blood lipid level in high-fat diet induced obese model animals. Animals were classified into a normal diet group (NC, normal control), HFD (high-fat diet without SGE), HFD 15 (high-fat diet + oral administration of 15 mg of SGE extract per 100 g body weight) and HFD 30 (high-fat diet + oral administration of 30 mg of SGE extract per 100 g body weight). The body weight gain declined in HFD 15 and HFD 30 groups compared with the HFD group, even though the diet intake increased significantly. The weight of liver and adipose tissue increased significantly in HFD group compared with in the HFD 15 and HFD 30 groups. Triglyceride, total cholesterol, LDL-C and AI decreased in HFD 15 and HFD 30 groups compared with in the HFD group, but the contents of HDL-C increased significantly. Expression of SREPB-$1{\alpha}$, SREPB-2 mRNA in the liver was lower in the high-fat diet group compared with the HFD group, but the expression of LPL mRNA in adipose tissue and $PPAR{\alpha}$ increased significantly. Fat accumulation in the liver tissues and liver damage were greatly reduced in HFD 15 and HFD 30 groups compared with in the HFD group. The size of adipocytes became smaller in the HFD 15 and HFD 30 groups compared with HFD group. In conclusion, this research discovered for the first time that grain maggot has anti-obesity effects, by reducing the abdominal fat of obese model animals and lowering blood lipid level through the down-regulation of PPAR-$1{\alpha}$ and SREPB-2 mRNA and the up-regulation of PPAR-${\alpha}$ mRNA.
Purpose: Baicalein, a natural flavone found in herbs, exhibits diverse biological activities. Nonalcoholic steatohepatitis (NASH) is an irreversible condition often associated with a poor prognosis. This study aimed to evaluate the effects of baicalein on the development of NASH in mice. Methods: Male C57BL/6J mice were randomly divided into four groups. Three groups were fed a methionine-choline-deficient (MCD) diet to induce NASH and were simultaneously treated with baicalein (at doses of 50 and 100 mg/kg) or vehicle only (sodium carboxymethylcellulose) through oral gavage for 4 weeks. The control group was fed a methionine-choline-sufficient (MCS) diet without the administration of baicalein. Results: The baicalein treatment significantly reduced serum levels of alanine aminotransferase and aspartate aminotransferase, suggestive of reduced liver damage. Histological analysis revealed a marked decrease in nonalcoholic fatty liver activity scores induced by the MCD diet in the mice. Similarly, baicalein treatment at both doses significantly attenuated the degree of hepatic fibrosis, as examined by Sirius red staining, and hepatocellular death, as examined by the terminal deoxynucleotidyl transferase dUTP nick end labeling assay. Baicalein treatment attenuated MCD-diet-induced lipid peroxidation, as evidenced by lower levels of hepatic malondialdehyde and 4-hydroxynonenal, demonstrating a reduction in oxidative stress resulting from lipid peroxidation. Moreover, baicalein treatment suppressed hepatic protein levels of 12-lipoxygenase (12-Lox) induced by the MCD diet. In contrast, baicalein enhanced the activities of antioxidant enzymes such as superoxide dismutase, catalase, and glutathione peroxidase. Additionally, baicalein treatment significantly reduced hepatic non-heme iron concentrations and hepatic ferritin protein levels in mice fed an MCD diet. Conclusion: To summarize, baicalein treatment suppresses hepatic lipid peroxidation, 12-Lox expression, and iron accumulation, all of which are associated with the attenuation of NASH progression.
Cladosiphon okamuranus is edible brown algae cultured commercially and massively at Okinawa coast. Dietary fiber occupying most of C. okamuranus isn't digested and absorbed by digestive enzymes of human. But it is known to prevent and treat constipation as metabolic disease. This study was to investigate the effects of dietary fiber extracted from C. okamuranus on the damage of liver and the blood cholesterol level in the high fat diet-fed rats. The effects were measured by the levels of aspartate aminotransferase (ALT), alanine aminotransferase (AST), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and high density lipoprotein (HDL) in sera. The levels of ALT, AST, TC, TG and LDL were significantly reduced in the C. okamuranus-treated group compared with the high fat diet group. But HDL level was markedly increased. The results showed that the dietary fiber extracts from C. okamuranus have the inhibitive effects of cholesterol biosynthesis in the high fat diet-fed rats.

To evaluate an effect of dietary protein on lipoprotein profile serum of carbon tetrachloride-treated rats, carbon tetrachloride (50% in olive oil) was twice given at 0.1ml/100g body weight at intervals of 24hours to the male rats and then the degree of liver damage in carbon tetrachloride-treated animals fed a low protein diet was compared with that fed a high protein diet. The increasing rate of liver weight/body weight and the serum levels of alanine aminotransferase in carbon tetrachloride-treated rats to the control group were higher in rats fed high protein diet than those fed low protein diet. In the serum levels of lipid (total lipid, total cholesterol and triglyceride) remarkable differences were not found between low protein diet group and high protein diet group. But these serum lipids in carbon tetrachloride-treated rats were decreased and the decreasing rate of serum lipids to control group were higher in carbon tetrachloride-treated rats fed high protein diet than those fed low protein diet. Under the animal model as identified by the present data herein, serum pre $\beta$-lipoprotein and $\alpha$-lipoprotein fractions were decreased in carbon tetrachloride-treated rats, but the serum levels of $\beta$-lipoprotein were rather increased in the both group by the injection of carbon tetrachloride. Especially, the decreasing rate of $\alpha$-lipoprotein fraction was higher in $CCl_4$-treated rats fed a high protein diet than those fed a low protein diet to its control group and the increasing rate of serum $\beta$-lipoprotein fraction was also higher in $CCl_4$-treated rats fed high protein diet than those fed low protein diet.
