감태(甘苔)에서 추출한 Dieckol 성분이 쥐의 인지능력에 미치는 영향 (Effect of Ecklonia cava Extracts Supplementation on cognitive ability in mice)

  • Objectives : The purpose of this study aimed to investigate that dieckol - isolated from Ecklonia cava - supplementation can improve cognitive ability in mice. Methods : 48-male mice(6 weeks old) were divided into four groups; High-Dieckol group(n=12), Low-Dieckol group(n=12), Placebo group(n=12), Control group(n=12) and they were administered orally 5 days per week for 4 weeks at the same time. We performed Morris water maze test, Passive avoidance test, Blood serotonin analysis. And there was examined on neurogenesis in dentate gyrus of hippocampus using 5-bromo-2'-deoxyuridine (BrdU) to label proliferating cells. Results : The results are as follows; As a Morris water maze results, Trial duration was significantly decreased in high dieckol group comparing to placebo group and control group. Distance to target was significantly decreased in high dieckol group and low dieckol group comparing to placebo group and control group. Mean speed was significantly low in high dieckol group comparing to low dieckol group, placebo group and control group. As a Passive avoidance test results, latency time was significantly long in high dieckol group comparing to low dieckol group, placebo group and control group. BrdU cell count was significantly high in high dieckol group comparing to low dieckol group, placebo group and control group. Conclusions : As a conclusion, it is considered that dieckol supplementation might improve learning and cognitive ability.

Apoptosis and Cell Cycle Arrest in Two Human Breast Cancer Cell Lines by Dieckol Isolated from Ecklonia cava

  • Purpose: Dieckol, a phlorotannin compound isolated from Ecklonia cava, has been reported to have antioxidant, antiviral, anti-inflammatory, and anticancer properties. The purpose of this study was to investigate its anticancer effects on human breast cancer cell lines. Methods: In this study, the viability of two human breast cancer cell lines SK-BR-3 and MCF-7 was investigated after dieckol treatment using a WST-1 assay. Apoptosis and cell cycle distribution were assayed via Annexin V-fluorescein isothiocyanate and propidium iodide staining followed by flow cytometric analysis. Immunoblotting analysis was also performed using Bax/Bcl-2 to determine whether the dieckol-induced apoptosis was mediated by the intrinsic apoptotic pathway. Results: In a dose dependent manner, dieckol reduced the number of viable cells and increased the number of apoptotic cells. The effect of dieckol on the cell cycle distribution was analyzed using flow cytometry. Dieckol treatment significantly increased the percentage of MCF-7 and SK-BR-3 in the G2/M phase. Immunoblot analysis revealed that 24 hours of dieckol exposure increased the Bax/Bcl-2 ratio. Conclusion: Dieckol induced cytotoxicity in MCF-7 and SK-BR-3 human breast cancer cells inducing apoptosis and cell cycle arrest. Therefore, it is suggested that dieckol may be a potential therapeutic agent for breast cancer.

감태추출물의 기능성원료 표준화를 위한 지표성분 dieckol의 분석법 검증 (Validation of an analytical method of dieckol for standardization of Ecklonia cava extract as a functional ingredient)

  • 본 연구는 해양수산물 유래 기능성 소재인 감태의 지표성분 분석법 검증 및 표준화를 위하여 HPLC를 이용하여 dieckol의 분석법을 확립하고 분석법에 대한 유효성 검증(validation)을 실시하고자 하였다. Dieckol 분석을 위하여 0.1% trifluoroacetic acid가 첨가된 water와 acetonitrile을 이동상으로 Capcell pak $C_{18}$ ($250{\times}4.6mm$, $5.0{\mu}m$) 컬럼을 사용하여 HPLC 방법을 통해 분석한 결과, 표준용액과 감태추출물의 dieckol RT이 일치하고 3.37의 높은 분리도(Rs)를 나타내었으며, UV spectrum이 일치하는 특이성을 확인하였다. 또한 검량선의 상관계수($r^2$)는 0.9994로 높은 직선성을 나타냈으며, 검출한계(LOD)와 정량한계(LOQ)는 0.38과 $1.16{\mu}g/mL$로 각각 확인되었다. 분석법의 재현성을 검증하기 위한 intra-day 및 inter-day 분석에서 RSD는 각각 1.7%와 1.00-1.25%로 나타났으며, 회수율은 99.42-101.32%이였으며 RSD는 2% 이하로 산출되어 정밀성이 있음을 확인하였다. 이상의 결과로부터 HPLC를 이용한 dieckol의 분석법은 감태추출물의 기능성원료 표준화를 위한 지표성분 분석법으로 적합함을 확인하였다. 본 시험법에 따라 분석한 감태추출물 내의 dieckol의 평균 함량은 약 0.11 g/g (10.54%)로 상대표준편차는 0.5% 이하였다. 따라서 이상의 분석결과를 통해 확립된 dieckol 분석법이 향후 감태추출물의 건강기능성식품 기능성 원료 개발과 표준화를 위한 기초자료로 크게 활용될 것으로 기대된다.

Anti-hyperlipidemic Effect of Polyphenol Extract (SeapolynolTM) and Dieckol Isolated from Ecklonia cava in in vivo and in vitro Models

  • The inhibitory effect of polyphenol extracts (Seapolynol$^{TM}$, SPN) of the marine brown algae Ecklonia cava and dieckol, a major component of SPN, on hyperlipidemia was investigated in ICR mice fed a high-fat diet (HFD) for five weeks. For analysis of the anti-hyperlipidemic effects of SPN and dieckol, these two agents were given orally on a daily basis to HFD-fed mice for four weeks, starting one week after the beginning of HFD feeding. Groups administered with SPN as well as dieckol showed lower body weight gains than the HFD only group. Administration of SPN and dieckol also resulted in a significant reduction of the level of total cholesterol (TCHO), triglyceride (TG), and low-density lipoprotein (LDL) cholesterol in the serum of HFD-fed mice. In Oil Red O staining using 3T3-L1 preadipocytes, it was shown that both SPN and dieckol markedly inhibited lipid accumulation of 3T3-L1 cells. Furthermore, SPN and dieckol (50 ${\mu}g$/mL) significantly inhibited 3-hydroxyl-methyl glutaryl coenzyme A (HMGCoA) reductase activity in vitro. Taken together, these results suggest that polyphenols of Ecklonia cava (SPN) and dieckol reduce body weight gain and fat accumulation in HFD-induced obese mice, and that their hypolipidemic effect is related to the inhibition of adipogenesis of adipocytes and HMGCoA reductase activity.

Improvement of Memory by Dieckol and Phlorofucofuroeckol in Ethanol-Treated Mice: Possible Involvement of the Inhibition of Acetylcholinesterase

  • Phlorotannins, the polyphonic compounds found in brown Eisenia and Ecklonia algae, have several pharmacologically beneficial effects such as anti-inflammation. In addition, our recent data show that these compounds may improve the cognitive functions of aged humans suggesting the potential ability to enhance memory in several neurodegenerative disorders. To examine the experimental hypothesis that two effective components of Ecklonia cava, dieckol and phlorofucofuroeckol (PFF), have memory-enhancing abilities, both were administered orally to mice before a passive avoidance test. The repeated administration of either dieckol or PFF dose-dependently reduced the inhibition of latency by the administration of ethanol. To investigate the mode of memory-enhancing actions, the levels of major central neurotransmitters in three different regions (striatum, hippocampus, and frontal cortex) of the mouse brain were measured. The levels of some of the neurotransmitters were significantly changed by ethanol. Both dieckol and PFF altered the levels of some neurotransmitters modified by the ethanol treatment. It is noteworthy that both dieckol and PFF increased the level of acetylcho-line, and they exerted anticholinesterase activities. Overall, the memory-enhancing abilities of dieckol and PFF may result from, at least in part, the increment of the brain level of acetylcho-line by inhibiting acetylcholinesterase.

Dieckol Suppresses CoCl2-induced Angiogenesis in Endothelial Cells

  • Dieckol is a polyphenol compound isolated from brown algae that has anti-oxidant, anti-inflammatory, and anti-tumor activity. We examined the anti-angiogenic effects of dieckol in endothelial cells under hypoxic conditions. Treatment with $CoCl_2$, a hypoxic mimetic agent, increased proliferation, adhesion, migration, and tube formation in HUVECs, as well as vessel sprouting in rat aortic rings, which correlated well with increased expression of hypoxia-inducible factor 1-alpha ($HIF1{\alpha}$) and ${\beta}1$-integrin. Dieckol suppressed $CoCl_2$-induced adhesion, migration, and tube formation in HUVECs and vessel sprouting in rat aortic rings. Dieckol treatment decreased $CoCl_2$-induced overexpression of $HIF1{\alpha}$ and its downstream signaling molecules, including ${\beta}1$-integrin/Fak, Akt/eNOS, and p38 MAPK. These results suggest that dieckol is a novel angiogenesis inhibitor and a potential treatment for angiogenesis-dependent diseases in humans, such as malignant tumors.

곰피추출물의 지표성분 Dieckol의 분석법 개선 및 검증 (Modification and Validation of an Analytical Method for Dieckol in Ecklonia Stolonifera Extract)

  • 본 연구에서는 곰피추출물을 이용하여 건강기능식품 개발 시 원료의 표준화를 위하여 HPLC-PDA를 이용하여 지표성분 dieckol의 분석법 개선 및 검증을 실시하였다. 분석법의 유효성 검증은 ICH가이드라인에 근거하여 특이성, 직선성, 정확성, 정밀성, 검출한계 및 정량한계를 통해 신뢰성 및 타당성을 검증하였다. 지표성분 dieckol 분석을 위하여 0.1% formic acid가 첨가된 water와 acetonitrile을 이동상으로 사용하였고 컬럼은 Capcell pak C18을 사용하여 HPLC를 이용한 분석방법에서 표준용액과 곰피추출물의 지표성분 dieckol 피크 머무름 시간 일치하고 다른 물질의 간섭을 받지 않는 것을 확인하였으며, PDA spectrum이 일치하므로 특이성을 확인하였다. Dieckol의 검량선의 상관계수(R2)은 1에 가까운 높은 상관계수 값 0.9997로 높은 직선성을 확인할 수 있었으며 분석에 적합함을 알 수 있었다. 농도를 알고 있는 곰피추출물에 dieckol 표준용액을 최종 농도가 각각 저농도(2.5 ㎍/mL), 중간농도(5 ㎍/mL), 고농도(10 ㎍/mL)가 되도록 첨가하여 정밀성 및 정확성을 계산하였다. 정밀성은 일간, 일내 정밀성으로 확인하였으며, intra-day 및 inter-day 분석에서 RSD는 각각 1.58-4.39%, 1.37-4.64% 수준으로 5% 이하의 우수한 정밀성을 보였다. 정확성 측정결과 intra-day 및 inter-day 분석에서 회수율은 각각 96.91-102.33% 및 98.41-105.71%로 우수한 정확성을 나타내었다. 검출한계(LOD)와 정량한계(LOQ)는 0.18 ㎍/mL, 0.56 ㎍/mL로 각각 확인되였으며 낮은 농도에서도 검출이 가능함을 보였다. 확립된 분석법은 특이성, 직선성, 정밀성, 정확성, 검출한계 및 정량한계에 대한 분석법 검증결과가 모두 우수한 분석방법임을 증명하였다. 또한 검증된 분석법을 이용하여 곰피추출물 중 함유한 dieckol 함량을 분석한 결과 27.39±0.50 mg/dry weight g로 분석되었다. 본 연구는 HPLC-PDA를 이용한 곰피추출물의 지표성분인 dieckol의 분석방법을 개선 및 유효성 검증을 진행하여 과학적으로 신뢰성이 있는 적합한 분석방법임이 검증되었다.

Thermostability of a marine polyphenolic antioxidant dieckol, derived from the brown seaweed Ecklonia cava

  • The thermostability of antioxidant activity of dieckol, a phlorotannin isolated from brown seaweed Ecklonia cava was investigated. The thermostable antioxidant properties of dieckol were evaluated at 30, 60, and $90^{\circ}C$ for 7 days using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radical scavenging activities, and comparing its performance to that of ascorbic acid. The intracellular reactive oxygen species (ROS) scavenging activity and apoptotic body formation were investigated using DCF-DA assay and nuclear staining with Hoechst 33342, propidium iodide and flow cytometry. Dieckol treated at different temperatures during 7 days showed stable scavenging activities on towards DPPH and hydroxyl radicals. In addition, dieckol showed a stable protective effect against $H_2O_2$-induced apoptotic body formation in Vero cells. On the other hand, the radical scavenging activities and intracellular ROS scavenging activities of ascorbic acid, used as a positive control, were significantly decreased at $60^{\circ}C$ and $90^{\circ}C$ from on the 4th day and 3rd days, respectively. In conclusion, the results indicated that food grade antioxidant extracts containing dieckol derived from E. cava remain a stable during the temperatures encountered during the processing of food and cosmetics.

감태에서 분리된 플로로탄닌의 염증 및 알러지 억제 효과 (Anti-inflammatory and Anti-allergic Effects of Phlorofucofuroeckol A and Dieckol Isolated from Ecklonia cava)

  • 갈조류인 감태(Ecklonia cava)의 에틸아세테이트 분획으로부터 phloroglucinol, eckol, eckstolonol, triphlorethol-A, phlorofucofuroeckol A, dieckol의 6종의 phlorotannin이 분리되었으며, NMR 및 MS분석으로 구조가 규명되었다. 감태의 주요 활성물질 분석을 위하여, 분리된 6종 물질에 대한 항산화, 항염증 및 항알러지 효과를 평가하였다. 그 중 phlorofucofuroeckol A와 triphlorethol-A가 라디컬(DPPH, ABTS) 소거 활성이 높았으며, 염증 반응에 대한 활성을 분석하기 위하여 LPS를 처리한 대식세포주인 Raw264.7에서 산화질소(NO) 생성 억제활성을 측정한 결과, phlorofucofuroeckol A의 NO 생성 억제 효과가 가장 큰 것으로 나타났다. 또한, 알러지 반응에 대한 억제활성을 분석하기 위하여 IgE-항체로 활성화시킨 비만세포주 RBL-2H3에서 분비되는 ${\beta}-Hexosaminidase$를 측정한 결과, dieckol이 농도의존적으로 가장 높은 억제 효과를 나타냈다. 따라서, 본 연구에서 항염증 및 항알러지 활성을 중심으로 분리된 6종의 플로로탄닌 중 phlorofucofuroeckol A와 dieckol은 감태의 알러지 염증 억제활성을 나타내는 주요 물질로 사료된다.

Dieckol, a Component of Ecklonia cava, Suppresses the Production of MDC/CCL22 via Down-Regulating STAT1 Pathway in Interferon-γ Stimulated HaCaT Human Keratinocytes

  • Macrophage-derived chemokine, C-C motif chemokine 22 (MDC/CCL22), is one of the inflammatory chemokines that controls the movement of monocytes, monocyte-derived dendritic cells, and natural killer cells. Serum and skin MDC/CCL22 levels are elevated in atopic dermatitis, which suggests that the chemokines produced from keratinocytes are responsible for attracting inflammatory lymphocytes to the skin. A major signaling pathway in the interferon-${\gamma}$ (IFN-${\gamma}$)-stimulated inflammation response involves the signal transducers and activators of transcription 1 (STAT1). In the present study, we investigated the anti-inflammatory effect of dieckol and its possible action mechanisms in the category of skin inflammation including atopic dermatitis. Dieckol inhibited MDC/CCL22 production induced by IFN-${\gamma}$ (10 ng/mL) in a dose dependent manner. Dieckol (5 and $10{\mu}M$) suppressed the phosphorylation and the nuclear translocation of STAT1. These results suggest that dieckol exhibits anti-inflammatory effect via the down-regulation of STAT1 activation.
