Mycelial growth properties of an Amanita javanica strain under various culture conditions (노란달걀버섯 균주의 여러 환경 조건에 따른 균사 생장 특성)

  • The edible ectomycorrhizal mushroom Amanita javanica is a valuable species protected by forest law in Korea. However, basic characterization data on its use as an important forest resource has been limited. This study was performed to determine mycelia growth characteristics of the domestically isolated Amanita javanica strain NIFoS 1267 on potato dextrose agar media under diverse culture conditions. Physical factors temperature, pH, and light, as well as chemical factors salts, heavy metals, and pesticides were examined for their effects on the growth of the mushroom strain. The mycelia of A. javanica strain exhibited optimal growth when cultured in dark at 30℃ in media with a pH of 5-6. Normal levels of growth were observed in media containing up to 2% saline. At a heavy metal ion content of 50 ppm, mycelial growth was not affected by arsenic ion but was affected by cadmium and lead ions. In the tests performed with two pesticides used in Korean forests, the growth of the mushroom strain was not affected by the presence of abamectin, but was inhibited in media containing acetamiprid, emamectin benzoate, or thiacloprid. These results are expected to facilitate artificial cultivation of A. javanica as a new commercial product.

Isolation and Characterization of Cryptococcus sp. CS-2 Secreting Polygalacturonase from Soil (토양으로부터 Cryptococcus sp. CS-2의 분리 및 균주가 분비하는 Polygalacturonase의 특성에 관한 연구)

  • A ploygalacturonase-produchg yeast was isolated from Cheju soil by selective eivichment media. One strain which has the highesl activity of polygalacturonase was selected. The characle~ishcs of the strain CS-2 were as follows: CS-2 utilized xylose. sucrose, maltose, u.ehalose, cellobiose. melibiose, lactose, raffinose, inosiiol, dulicilol, and dextrose, but did not utilized galactose, nitrate. nit~te, and lysine. Growth of CS-2 was inhibited by cyclohexamide, 1% acetic acid, and high concenaation (over 50%) of glucose. It grew at $30^{\circ}C$ but did 'IIOL $35^{\circ}C$. The cell size ofthe strain CS-2 was 2.9 p ~ n in length and 1.3 $\mu$ in diameter. Vegetable reproductmn was multiple budding and ascospre was present I to 4. Pseudomycelia or true myceliua formation were not observed In any of the cullureq. These results suggest that strain CS-2 is most likely a strain related Cryptococcus spp. (Cryptococcu spp. CS-2). When polygalacturonase or ihe yeast was induced by addition of polygalactoronic acid, polygalacturonase activity was detected in culture supernatent. There was a peak of specific activity a1 he mid-stationary phase(3 days culture) of growth. Polygalacturonase specific activity of Crylmcoccus sp. CS-2 was 2.96 unitsling. The molecular weighl ol'polygalacturonase was showed to be 46 KDa by both SDS-PAGE and activity stailling.

Isolation and Characterization of Growth Stimulating Thermophilic Fungi on Oyster Mushroom from Oyster Mushroom (Pleurotus ostreatus) Compost (느타리버섯 배지로부터 느타리버섯 균사의 성장을 돕는 고온성 곰팡이의 분리 및 동정)

  • Some of thermophilic fungi which has growth-promoting effect on Pleurotus ostreatus were isolated from compost during high temperature fermentation process. The temperature optima of 7 isolated thermophilic fungi were $50^{\circ}C$ on PDA media. Isolated strains S-1 and S-2 have the best mycelial growing rate, so these isolates were expected as excellent thermophilic fungi for high temperature composting and mycelial growing of oyster mushroom. In liquid culture, the optimal pH of thermophilic fungi observed variously, pH 7.0-10.0 but most of thermophilic fungi grow well in pH 8.0-pH 9.0 and the final pH of media after cultured was done pH 5.5-6.0. In liquid culture of thermophilic fungi on the optimal condition, S-2 have the best mycelial growing rate. The growing rate of thermophilic fungi S-1, S-2, S-5, and S-10 on lignocellulosic substrates was good but Humicola grisea var. thermoidea, well know thermophilic fungi which has growth-promoting effect on Agaricus bisporus, was poor and which was well grown on PDA at $50^{\circ}C$, pH 7.0. Isolated strain S-1 was identified as Trichophyton sp. and other 6 strains were identified as Sepedonium sp. by morphological characteristics.

On the Identification of Strains and the Cultural Characteristics of Flammulina velutipes in Korea (한국산 팽나무 버섯의 계통류별과 그 배양적 특성에 관한 연구)

  • In order to investigate morphological and cultural characteristics of strains in species, Flammulina velutipes, the author collected isolates of Flammultina velutipes at 49 locations in Korea and cultivated these isolates on the various kinds of solid media. After investigating the cultural characteristics, appeared on the various media, he obtained the following results : 1. The variation of colors in the fruit bodies is connected with the variation of climatic environments(composite effects between mean temperature in January and number of rain days of 1mm and over precipitation). The author, therfore, can find out the trend that brown type is distributed in the midland climatic region and yellow type in the southern climatic regoin. 2. Two types can be classified into several strains respectively : the strain of abundant or insufficient productivity, and strain of selectivity or non-selectivity of media. 3. According to the results of mutual comparison of soluble mycelial proteins by disc electrophoresis using polyacrylamide gels, each type has special common protein fractions(brown type : band located at 26..5mm position from surface of gel, yellow type : band located at 24.5mm position from surface of gel), and each strain has special protein fractions too. Therefore this phenomenon seems to support the results obtained by the above-stated morphological and cultural studies. 4. In the adaptability of strains to the temperature, every strain has the nature of growing in lower temperature(the optimum temperature of 20.deg.C to 25.deg.C) except that YI-1 strain has the optimum temperatue of $25^{\circ}C$-26^{\circ}C. And mycelial growth of every strain is discontinued at $35^{\circ}C.$ 5. In the adaptability of strains to the H-ion concentration, every strain has wide adaptable range of H-ion concentration, and has optimum range of pH 5.5 to 6.6 in mycelial growth excepting YA01, BI-2 and YI-1. 6. In the utility of carbon sources, the mycelial growth of every strain is very poor on the media containing xylose(average diameter of mycelial growth : 18mm), and most strains utilize favorably sucrose(39mm), maltose(37mm) and dextrose(35mm) in mycelial growth. In the utility of nitrogen sources, every strain utilizes favorably organic nitrogens(36 mm)more than inorganic nitrogens (25 mm), and utilizes fully peptone nad asparagine in organic nitrogens. Especially BA-1, BIK-2 and YA-1 strains grow vigorously on each media containing various carbon and nitrogen sources. 7. The characteristic tests of the number of days required for mycelial growth, the number of days requried for sprout of young bodies, the length of stipe and the number of fruit bodies formation seem to be useful methods in the early selection of the strain of the abundant productivity.

Studies on the Cultural Characteristics of Poria cocos (복령(Poria cocos)의 배양학적(培養學的) 특성(特性)에 관한 연구(硏究))

  • The cultural characteristics and some factors such as nutrient sources and supplements effecting on mycelial growth and density were investigated to study the possibility of an artificial cultivation of P. cocos. The optimum pH for P. cocos was 4.0-4.5. The optimal growth temperature ranged from $25^{\circ}C$ to $29^{\circ}C$. Myceial growth of P. cocos was better in SPD than PD media. Adding the nurient sources such as dextrose, yeast and potato infusion to pine extract media practically stimulated the mycelial growth and density of P. cocos comparing to pine extract media alone. When P. cocos was cultured on sawdust media added 3 different supplements composed of corn meal, rice bran and wheat bran, corn meal was the best and its percentage was 30 (w/w) for mycelial growth. On culturing in sawdust media added by varying the mixture ratio of them, the media mixed corn meal and wheat bran (3:1, w/w) supported more vigours for mycelial growth. In inoculation test to pine stem, the fungal growth was good in under or inside pine bark and xylem, but the sclerotium was not observed in the stem. Mycelial growth was also observed in central part of pine stem by cross section.

Mechanism of Free Phosphate Production by Penicillium sp. GL-101, Phosphate Solubilizing Fungus, in the Submerged Culture (인산가용화균 Penicillium sp. GL-101의 유리인산 생성기작에 관한 연구)

  • We investigated the capability of the phosphate-solubilizing fungus, Penicillium sp. GL-101, to solubilize in vitro some insoluble rock phosphate via possible mechanisms: acidification of the medium, production of chelating metabolites, redox activity, and so on. GL-101 was able to solubilize rock phosphate (mostly calcium phosphate) in a liquid potato dextrose broth(PDB) medium, as determined by spectrophotometric analyses. Acidification was the major mechanism of solubilization since the pH of cultures fell below 4.0 and in cultures containing 1.0%(w/v) loess the pH dropped from 7.0 to 3.2. More than 10 mg/mL concentrations of citric acids were detected by high-performance liquid chromatography(HPLC) in the culture supernatants. Also this fungus showed the phosphatase activity (over 1.3 unit) to contribute partially releasing phosphate from rock phosphate, when supplemented with 1.0% loess in culture broth. The chelating activity of GL-101 in culture supernatants was not present because 2-ketogluconic acid, a chelating agent for the phosphate, was produced only a basal level. Therefore, the solubilization mechanism of rock phosphate by Penicillium sp. GL-101 involves both acidification due to citric acid production and phosphatase activity.

Isolation and Utilization of Antagonistic Pseudomonas fluorescens from Soils for the Protection of Soybean Sprouts Rot (콩나물 부패병 방제를 위해 토양으로부터 분리한 길항균 Pseudomonas fluorescens의 이용)

  • Thirty-three bacterial and fungal strains were isolated from the rotten soybeans and soybean sprouts to isolate pathogenic microorganisms which cause soybean sprouts rot during soybean sprouts cultivation. In pathogenicity tests of the isolates on soybean sprouts, two isolates(K-17 and K-28) caused soybean sprouts rot and were identified as Erwinia carotovora and Fusarium sp., respectively. To isolate antagonists aganist K-17 and K-28 pathogens, bacteria were isolated from various soybean-cultivated soils and screened by the inhibition zone method. A bacterial isolate(J-232) which inhibited growth of both pathogens was identified as Pseudomonas fluorescens and further examined. The culture filtrate of P. fluorescens J-232 (dilution rate of 500 times) inhibited the growth of Erwinia carotovora K-17 and Fusarium sp. K-28 both on potato dextrose agar medium and on soybean sprouts cultivated in vessel. The development of soybean sprouts rots was observed during cultivation by inoculation of soybean seeds with culture filtrate of both pathogens. The combined inoculation of soybean seeds with culture filtrate of antagonistic bacterium and that of pathogens prevented soybean sprouts rot, and the growth of soybean sprouts was similar to that of control. The soybean sprouts inoculated with antagonists culture filtrate alone did not develop soybean sprouts rot, and the growth of the seedlings was shown to be slightly promoted as compared with that of control.

Isolation and Characterization of Colletotrichum Isolates Causing Anthracnose of Japanese Plum Fruit (자두 탄저병균의 분리 및 동정)

  • BACKGROUND: Although the filamentous fungal pathogen Colletotrichum species causing anthracnose disease on various fruits including peach, apple, persimmon and grape, there is no report on Japanese plum in Korea. METHODS AND RESULTS: In 2016, diseased fruits showing typical anthracnose symptoms of Japanese plum were collected in market and ochards. Diseased tissue was cut off and disinfected subsequently with 70% ethanol for 1 min, and in 1% sodium hypochloride solution for 1 min, followed by three washes with sterile distilled water. The disinfected tissues were placed onto potato dextrose agar (PDA), and incubated at $25^{\circ}C$ in the dark for 5 to 7 days. For single-spore isolation, conidia were scraped off the plate using a loop, and suspended with 10 mL sterile distilled water. One hundred microliter of the conidial suspension was spread on PDA plates and incubated at $25^{\circ}C$. Finally, one germinated conidium was transferred onto PDA plates. Morphological and cultural characteries of colonies and spores of isolated Colletotrichum were observed after 7 to 10 days incubation on PDA. Molecular identification of isolates were analyzed by comparing rDNA-ITS gene sequences with NCBI GeneBank. CONCLUSION: Of eleven isolates of Colletotrichum isolated from anthracnose diseased Japanese plum fruits, six were identified as C. acutatum, and five as C. gloeosporioides based on diagnostic characteristics such as colony growth rate, shape and size of conidia, and rDNA-ITS sequences. This is the first report of Colletotrichum causing the anthracnose on Japanese plum in Korea.

Effects of Enzyme Treatments on Quality and Yields of Barley Tea (보리의 효소처리가 보리차의 품질 및 수율에 미치는 영향)

  • In order to improve the quality and yields of barley tea(water extracts), enzymatic hydrolysis was carried out with using three kinds of mired enzymes of ${\alpha}-amylase,\;{\beta}-amylase$ and protease. The barley treated with enzymes were the crushed , soaked and slightly roasted(light brown) Youngsanbori(hulless) and the physical, chemical and sensory characteristics were investigated. The results showed that enzymatic hydrolysis caused a signigicant increase in solid yields (80%) of barley tea, particularly with the mixed enzyme 5,000 which has higher activity of ${\alpha}-amylase$ than the other enzymes. The intrinsic viscosity of barley tea was reduced by enzymatic hydrolysis as the value of dextrose equivalent increased The Hunter L, a, b values was lower for the enzyme treated barley tea but showed not significant difference with reaction time. The organoleptic evaluation clearly showed that both intensity and acceptability of odor and taste was markedly Increased. The roasted nutty, and sweety odor and taste were particular in increase by mixed enzyme 5,000.

Effects of Soil Solarization for Control of Cucumber Wilt -Suppression of Fusarium oxysporum f. sp. cucumerium and Promotin of Cucumber Growth- (태양열 소독에 의한 오이덩굴쪼김병 방제 -병원균 생장억제 및 오이생육촉진에 미치는 비닐 피복효과-)

  • The effects of solarization on the suppression of soilborne plant pathogen and the growth promotion of cucumber plants were examined in artificially infested soil by vinyl mulching and not mulching from July 25 to August 25, 1983. During the solarization period, the highest temperatures were $58^{\circ}C,\;45^{\circ}C,\;and\;42^{\circ}C$, at 5cm, 15cm, and 25cm of soil depth respectively. The inoculum of cucumber wilt pathogen, Fusarium oxysporum f. sp. cucumerinum, was mixed with soil 30cm deep and saturated with water. The pathogen was completely killed after 30dys of solarization in 5cm soil depth and 98 percent of inoculum was eliminated in 15cm soil depth. But the survival rate of the fungi in 25cm soil depth of solarized plot did not show significant differences compared with those in nontreated plot in 5cm and 15cm depth. Although some of the pathogenic fungi might survive from solarized soil in 15cm and 25cm depth, the ability of microconidia production was reduced significantly The number of microconidia grown on Komada's medium in isolates the primary colonies from solarized soil was less than that in isolates from nontreated soil approximately by one fourth. The first subcultured solates from the solarized soil grown on potato dextrose agar also produced a small amount of microc. onidia compare with that of subcultured isolates from nontreated soil. Cucumber seedlings planted in the soil collected from solarized plot grew much better than that in the soil from nontreated plot at any of soil loved, especially in 5cm of soil depth. And the fruits harvested from cucumber plants grown in the solarized plot were more in number and leavier in weight than that from nontreated plot. Besides the typical symptom development, significant growth suppression wvas recognized with increase of inoculum density of F. oxysporum f. sp. cucumerinum at early stage of cucumber seedlings in steam sterilized soil.
