Effect of Polyphosphates on the Growth of Penicillium griseofulvum and the Production of Patulin (Penicillium griseofulvum 성장과 Patulin 생성에 미치는 인산염의 효과)

  • To extend the shelf lives of rice and corn products, the effects of the polyphosphates[$Na(PO_3)n$, n=11] on the growth of Penicillium griseofulvum and patulin production were investigated. The growth was completely inhibited in the potatoes dextrose agar medium treated with 2% polyphosphate. Moisture content had a considerable influence on the production of patulin. At 30% moisture content, the amounts of patulin produced in rice and corn were $61.40 \mu g/ml$ and $40.74 \mu g/ml$, respectively, but the level of the toxin was significantly decreased to 93~95% by addition of 1% polyphosphates. No patulin was detected in both rice and corn medium added 2% polyphosphate when the incubation time prolonged. The result of scanning electron microscopy was supposed that the biocidal action of polyphosphate on fungi was related to the collapse of cell wall structure.

Mariannaea samuelsii Isolated from a Bark Beetle-Infested Elm Tree in Korea

  • During an investigation of fungi from an elm tree infested with bark beetles in Korea, one isolate, DUCC401, was isolated from elm wood. Based on morphological characteristics and phylogenetic analysis of the internal transcribed spacer and 28S rDNA (large subunit) sequences, the isolate, DUCC401, was identified as Mariannaea samuelsii. Mycelia of the fungus grew faster on malt extract agar than on potato dextrose agar and oatmeal agar media. Temperature and pH for optimal growth of fungal mycelia were 25oC and pH 7.0, respectively. The fungus demonstrated the capacity to degrade cellobiose, starch, and xylan. This is the first report on isolation of Mariannaea samuelsii in Korea.

Dermatophytosis by Trichophyton rubrum and T raubitschekii in dogs (Trichophyton rubrum 및 Trichophyton raubitschekii에 의한 개 피부사상균증에 관한 연구)

  • This study was carried out to determine the causative agent and the clinical features of dermatophytosis occurred in 3 dogs from Tae-gu, Korea between 1996 and 1997. Specimens of hair and scale, collected from skin lesions were inoculated on potato dextrose agar and mycobiotic agar supplemented with thiamine and inositol. The agar plates were incubated at $25^{\circ}C$ for 2 weeks. Growing fungi isolated were identified by the morphological and growth characterization on bromocresol purple-milk solids-glucose medium, Christensen urea broth and trichophyton media, and by hair perforation tests. Skin lesions were observed in legs, abdomen, ears, face and head with alopecia of various sizes accompanied by itching and/or inflammation. The causative agent of dermatophytosis in the 3 dogs was identified as Microsprum gypseum and Trichophyton rubrum (mixed infection, dog 1), T rubrum (dog 2) and T raubitschekii (dog 3). The dermatophytosis by T rubrum and T raubitschekii was first found on animals in Korea.

Characterization of Myrothecium roridum Isolated from Imported Anthurium Plant Culture Medium

  • During an investigation of microorganisms and pests in plant culture media from imported anthurium pots, a fungal isolate (DUCC4002) was detected. Based on its morphological characters including colony shape on potato dextrose agar, the microstructures of spores observed by light and scanning electron microscopy and the results of phylogenetic analysis using an internal transcribed spacer rDNA sequence, the fungal isolate was identified as Myrothecium roridum. Pathogenicity testing on anthurium leaves revealed that the fungus could colonize and produce sporodochia on the inoculated leaves. This is the first report of M. roridum detected in imported plant culture medium in Korea.

Characteristics of Red Mold Isolated from Traditional Meju (전통메주에 증식하는 붉은 곰팡이의 특성)

  • Red mold was isolated from meju prepared by traditional mtehod and characterized. The isolated red mold grew well on potato dextrose agar medium, In microscopic observation, it had a septum in mycelium and ellipsoidal spore. Optimal temperature and pH for growth were 30$\^{C}$ and 6.0, respectively. Enzyme activities such as protease, a-amylase and glucoamulase in ted mold were lower than those in Aspergillus oryzae. A competitive growth between red mold and Asp. oryzae was greatly affecten by cultivation temperature. The growth of isolated red mold on meju was predominant at below 30$\^{C}$ as compared with Asp. oryzae.

  • Paclitaxel과 vancomycin을 $5\%$ 포도당주사액, $0.9\%$ 염화나트륨주사액 또는 하트만용액과 함께 Y-Site 장치를 써서 환자에게 주입할 때 두 약물의 안정성에 관하며 면구하였다. Paclitaxel 0.3 mg/ml 및 1.2 mg/ml과 vancomycin 1 mg/ml, 5 mg/ml 및 10 mg/ml을 각각 1 : 1로 혼합한 후 0, 1, 2, 4, 12시간 시점에서 두 약물의 농도를 HPLC로 분석하였다, 방해물질에 의한 분석오차를 줄이기 위해 분석법을 여러상태에서 확인하였으며 각 농도에서 3차례씩 실험하였고 각 샘플은 반복하여 HPLC로 분석하였다. 분석전에 각 시료의 투명도, 색의 변화, 침전상태 및 pH를 검사하였다. Paclitaxel 0.3 mg/ml 및 1.2 mg/ml와 vancomycin 1 mg/ml, 5 mg/ml 및 10 mg/mt를 각각 혼합하였을 때 12시간 동안 안정하였으며 주사액의 혼탁이나 색의 변화 및 침전은 나타나지 않았으며 pH도 변하지 않았다.

Induced Change in DNA Methylation of Fusarium oxysporum f. sp. niveum due to Successive Transfer

  • Changes in pathogenicity of old and successively-cultured isolates of Fusarium oxysporum f. sp. niveum have been observed and the concept that such cultures will become attenuated is generally accepted. However, the genetic basis for this phenomenon has not been studied. In an effort to identify a DNA marker closely linked to variations, DNA methylation was investigated both before and after the successive transfers of F. o. f. sp. niveum isolates on artificial media. A sector of mycelium in F. o. f. sp. niveum race 2 isolate (TXXID) which showed variation in pigmentation and colonial morphology occurred after 18 successive weekly transfers on potato dextrose agar (PDA). The sector characteristics were stable and did not change after more successive transfers. It was shown that DNA methylation preexists in ribosomal RNA gene (rDNA) of F. o. f. sp. niveum and that additional changes in DNA methylation occurred during successive culturing.

  • Since 1950 there has been a dramatic progress in rationalization of the production of sweet potato and potato starch in Japan. This enabled dextrose industry by enzymatic process to develop rapidly due to the success of enzymatic liquefaction and saccharification. Isomerization of glucose to fructose has been studied, and the immobilization of isomerases prompted its products on industrial scale in 1970. Another advance is the development of effective methods of producing high purity maltose. A malto-hexaose forming amylase was discovered in 1971 and attempts are being made for its pharmaceutical utilization. Saccharification of cellulose by cellulase has been studied. Conversion of starch to other polysaccharides is another example for the numerous Japanese activities.

Isolation of Antagonostic Fungi to Phytophthora Capsici for Biological Control of Phytophthora Blight of Red-Pepper (고추역병의 생물학적 방제를 위한 길항진균의 분리)

  • For isolation of antagonistic fungi antagonistic to Phytophthora capsici, a total of 157 isolates of fungi were screened from soil. Among the 157 isolates further screened by the dual culture test on potato dextrose agar and V-8 juice agar, 16 isolates were tested to show their antagonistic activity against P. capsici and Fusarium oxysporum. Fungal cul-ture filtrates of screened 16 isolates were shown to inhibit germination of zoospoorangia of P. capsici entirely and conidia of F. oxysporum considerably. Antagonistic fungi were shown to suppress of P. capsici infection of red-pepper plants maintained in the green house. Four isolates. 27 J5, 37 J10, 36 J13 and 31 K10, with the reduced disease incidence 53.3∼60.0% were identified as Fusarium sp. (27 J5). Trichoderma sp. (37 J10, 36 J13) and Penicillium sp. (31 K10).

Crown and Root Rot of Greenhouse Tomato Caused by Fusarium oxysporum f. sp. radicis-lycopersici in Korea

  • Forty(40) isolates of Fusarium oxysporum isolated from wilting tomato plants at Buyeo of Korea in 1997 were inoculated to four tomato cultivars (Ponderosa, Okitsu 3, Walter, and Zuiken) to examine pathogenic reactions. Isolation rates of F. oxysporum f. sp. lycopersici (FOL) races 1 and 2, and F. oxysporum f. sp. radicis-lycopersici(FORL) were 3.5%, 24.5%, and 57.5%, respectively. Mycelial growth on potato-dextrose agar at different temperature for the three pathogens was $26^{\circ}$. In the pathogenicity tests, however, the range of optimum temperature for disease development for FORL was between 15 and $20^{\circ}$, while that for races 1 and 2 of FOL were specifically pathogenic to tomato only. This suggests that host ranges of FORL and FOL differ significantly.
