Chlorpromazine 의 용혈작용(溶血作用)에 관(關)한 연구(硏究) (Studies on the hemolytic action of chlorpromazine)

  • The author studied the hemolytic action of chlorpromazine on rabbit erythrocytes and obtained the following results: 1. Chlorpromazine caused hemolysis in vitro. The hemolytic action of chlorpromazine was milder than that of saponin(Colleman and Bell Co ) 2. Cholesterol had no marked effect on chlorpromazine hemolysis. 3. Dextrose, albumin and blood plasma protected erythrocytes against chlorpromazine hemolysis. 4. The mechanism of chlorpromazine hemolysis seemed to be different from that of saponin hemolysis. 5. Chlorpromazine increased tile osmotic fragility of rabbit erythrocytes. 6. The intravenous injection of chlorpromazine caused hemolysis in vivo.

인삼제품에서 분리한 부패성 효모의 특성 (Characterization of the Spoilage Yeast Isolated from Ginseng Product)

  • A yeast was isolated from the spoiled ginseng product. The isolate was ellipsoidal shaped yeast measured around 2.0 to 2.5 w in diameter. The strain formed pseudomycelium on potato-dextrose agar medium. The isolated yeast used glucose as fermentable sugar, and showed assimilation activity for glucose, sorbitol and mannitol. The strain was also able to grow in the presence of 1% acetic acid and 50% (w/v) glucose-yeast extract agar. The isolated osmophilic yeast was identified as a strain of Zygosauharomyce sp.

순창전통고추장의 물리화학적 특성에 관한 연구 (Studies on the Physicochemical Characteristics of Sunchang Traditional Kochujang)

  • This study was conducted to investigate the physicochemical characteristics of Sunchang traditional kochujang(fermented hot pepper-soy bean paste) for quality standardization. The kochujang samples, which have been fermented for 8 months in 1999, were collected from 20 firms at folk village in Sunchang area and analyzed their chemical compositions and color reference. The mean value of moisture, amino-type nitrogen, pH, acidity and salt content of the samples collected were $44.62{\pm}1.79%$, $132.66{\pm}21.67\;mg%$, $4.52{\pm}0.08$, $15.77{\pm}1.62$ and $8.76{\pm}1.55%$, respectively. The moisture, pH, acidity and salt content of each sample did not show much differences among samples. The Hunter values(L, a and b) of Sunchang traditional kochujang were $25.72{\pm}1.58$, $23.26{\pm}1.71$ and $9.86{\pm}0.94$, respectively. The mean content of amino-type nitrogen of Sunchang traditional kochujang was $132.66{\pm}21.67\;mg%$, and there were a little difference between the minimum(100.33 mg%) and the maximum(164.56 mg%). The main free sugars of Sunchang traditional kochujang were fructose($1.86{\pm}1.01%$), dextrose($4.29{\pm}2.06%$), sucrose($0.54{\pm}1.21%$), and maltose($1.48{\pm}0.77%$). The contents of fructose, dextrose, and maltose had little difference among samples. The fatty acids in Sunchang traditional kochujang were composed of palmitic, stearic, oleic, linoleic, linolenic, octadecatetraenoic, arachidonic and behenic acid. The linoleic acid(18:2) showed the highest, occuping 59.37% of the total fatty acids.

송이균 접종으로 외생균근을 형성한 소나무 묘목의 PDMP 및 Tween 용액 처리에 의한 순화 (Acclimation of Ectomycorrhizal Pine (Pinus densiflora) Seedlings Inoculated with Tricholoma matsutake by the Treatment of PDMP and Tween Solutions)

  • 송이균을 접종하여 균근을 형성시킨 소나무 묘목을 실외로 가지고 나갈 경우 송이 균근이 이탈되는 현상을 극복하기 위한 방법을 모색하였다. 실내에서 형성한 송이균 접종 소나무 배지의 초기 토양구조를 유지하며 수분을 공급하고자 배양병의 아래에 구멍을 뚫어 화분형태로 변형을 시킨 후 8종류의 급수방식을 시도하였다. 증류수만을 공급한 경우에는 송이 균근이 이탈되었지만, 2.5% PDMP(24 g potato dextrose broth, 3 g malt extract, 1 g peptone) 용액이나 0.5% Tween 80 용액을 2주 간격으로 관수한 경우에는 3개월의 순화처리 이후에도 송이 균근이 강한 활성을 띠고 있음을 확인하였다. 순화기간동안 송이 균근 형성 비율이 증가하였으며, 순화 성공률은 70% 수준에 이르렀다. 금후 송이를 접종한 소나무 묘목은 이러한 순화처리 과정을 거쳐서 균근을 안정시킨 후 현지에 이식하는 방법을 사용하기를 추천한다.

Protective Effect of Aminoglycosides and Their Combinations Against 2-Chloroethylethyl Sulfide Exposure

  • Exposure of splenocytes to 2-chloroethylethyl sulfide (CEES) resulted in the cell death, and the cytotoxicity of CEES was prevented by inhibitors of lysosomal hydrolases. Therefore, it has been postulated that the cytotoxicity of CEES may be partially due to the lysosomal labilization. This study, based on this mechanism, was undertaken to determine whether aminoglycoside antibiotics as inhibitors of lysosomal phospholipases and their combinations with other lysosome stabilizers can be useful as a treatment to reduce the CEES toxicity in mice. 2-Chloroethylethyl sulfide (20 mg/kg body weight) was injected ip into female ICR mice, and candidate compounds were administered ip before or after the CEES challenge. Kanamycin (40 mg/kg body weight) as effective as deferoxamine (100 mg/kg body weight) enhanced the survival rate after 5 days of intoxication from 10% of control to 50 - 60%. The most effective was found to be the combination of kanamycin, cycloheximide, deferoxamine and dextrose showing an almost full protection against 2LD50 of CEES. Consistent with the protection of the CEES toxicity, the decrease of body weight in mice intoxicated with CEES was effectively prevented by kanamycin or its combinations. It is suggested that kanamycin or its combination (kanamycin, cycloheximide, deferoxamine and dextrose) would be one of effective antidotes against the CEES poisoning in mice.

동결방지제에 의한 딸기펄프와 붉은생고추 페이스트의 저온저장에 관한 연구 (Studies on the Low-temperature Storage of Strawberry Pulp and Red Pepper Paste by Cryoprotectants)

  • 동결방지제를 첨가하여 붉은생고추 페이스트 및 딸기 펄프의 저온저장 실험을 한 결과, 붉은생고추 페이스트는 줄기만을 제거한 후 NaCl 15%, citric acid 0.5%를 첨가하면 $-9.6^{\circ}C$에서 미동결 상태로 저장할 수 있었고, 딸기펄프는 dextrose 25%, fructose 17%, sorbitol 8%, ascorbic acid 0.2%를 첨가했을 때 $-15^{\circ}C$까지도 미동결 상태로 저장할 수 있었다. 또한 동결방지제를 이용한 시료는 무처리한 시료에 비해 저장소요시간이 약 1/2 정도 걸리며, 소요에너지도 약 $70{\sim}80%$의 절감효과를 얻을 수 있었다. 따라서, 동결방지제를 이용하면 저온저장시 빙결정 생성에 따른 조직손상을 억제할 수 있을 뿐만 아니라 에너지 절감효과도 얻을 수 있는 것으로 사료되며, 이와 같은 결과를 일시에 다량 수확되는 과채류에 적용할시 농산물의 저장법 개선에 상당한 효과가 있을 것이다.

정맥주사제 혼주시 5-fluorouracil의 차광유무에 따른 안정성에 관한 연구 (The Effect of Light on the Stability of IV admixture with 5-Fluorouracil)

  • The effect of the light on the stability of 5-fluorouracil admixture was investigated. Four sets of 5-fluorouracil admixture were prepared using 50 mg/ml injection in $5\%$ dextrose solution in PVC bags and glass bottles: (1) 5-fluorouracil 1 mg/ml concentration in glass bottles, (2) 5-fluorouracil 1 mg/ml in PVC bags, (3) 5-fluorouracil 10 mg/ml in glass bottles, and (4) 5-fluorouracil 10 mg/ml in PVC bags. In each set, one group was protected from the light (control group) and the other group was exposed to the fluorescent light (study group). All admixtures were stored at room temperature for 72 hours. Also, 5-fluorouracil injections (50 mg/ml) were prepared in plastic syringes. Half of the samples of 50 mg/ml concentration were protected from the light (control group) and the other half were exposed to the fluorescent light (study group). These were stored at room temperature for 48 hours. After visual inspection, the pHs of each admixture were determined. The 5-fluorouracil concentrations were measured by high-performance liquid chromatography with UV detection, with 5-bromouracil as an internal standard. Over the study period, no visual changes were observed. The pHs of 5-fluorouracil admixtures were in the range of $8.2\~8.5$. The peak area ratios (5-FU/5-BrU) of 5-fluorouracil admixtures protected from the light were compared with those of the admixtures exposed to the light. There was no statistically significant difference between two groups during the study period (p>0.05). In conclusion, 5-fluorouracil admixtures in $5\%$ dextrose solution exposed to the light were stable for 72 hours.

번데기동충하초(Cordyceps militaris)의 균사 생장 (Cultural Characteristics of Mycelial Growth by Cordyceps militaris)

  • 강원대학교 EFCC에서 보관중인 번데기동충하초의 배양적 특성과 인공재배의 조건을 조사하였다. 균사생육에 적합한 배지로는 SDAY 배지와 YMA 배지, SDAY 배지, HMD 배지, PDA 배지로 조사되었으며 균사생육이 빠른 배지로는 MMM 배지로 조사되었다. 균사생장에 적합한 온도로는 $25^{\circ}C$와 pH는 $6.0{\sim}8.0$으로 조사되었다. 균사배양에 적합한 탄소원으로는 dextrose로 조사되었고 fructose와 lactose에서 균사생장이 빠른 편이였다. 균사생장이 우수한 질소원으로는 hemoglobin으로 나타났으며 tryptone에서 포자형성이 우수하였다. 무기 염류에서는 $KH_2PO_4$$K_2HPO_4$에서 균사생장이 우수하였으며 C/N비에서는 100 : 1의 비율에서 균사생장이 양호하였다.

Sectors from Phyricularia grisea Isolates on Edifenphos and Iprobenfos-Amended Media

  • Sectors of rice blast fungus, Pyricularia grisea, frequently appeared on potato dextrose agar amended with edifenphos and iprobenfos. Thus, we assessed the sector-forming frequency of isolates of P. grisea and compared the fungicide sensitivity between wild types and sectors against the fungicides. The 905 isolates of the fungus were obtained from rice-growing locations in Korea from 1997-1998. When the isolates were grown on potato dextrose agar amended with minimal inhibitory concentrations of edifenphos (20 ${\mu}$g a.i./ml)and iprobenfos (55 ${\mu}$g a.i./ml), they produced sectors that overcame the effect of the fungicides. Among the 905 isolates tested, 9.0% produced sectors against edifenphos and 5.6%, against iprobenfos. Different sector-forming frequencies were also observed among the 11 locations of Korea. Sectors obtained from the fungicide-amended media generally grew more than their counterpart wild types grown on the media with either edifenphos or iprobenfos, regardless of their origins. In this study, greater relative growth of sectors over wild types of tested isolates can support the resistant characteristic of the fungus to survive against the fungicides. Therefore, the results indicate that the sectoring in rice blast fungus, P. grisea, may play a role in the occurrence of fungicide resistance.
