PARK2 Induces Osteoclastogenesis through Activation of the NF-κB Pathway

  • Osteoclast generation from monocyte/macrophage lineage precursor cells needs to be tightly regulated to maintain bone homeostasis and is frequently over-activated in inflammatory conditions. PARK2, a protein associated with Parkinson's disease, plays an important role in mitophagy via its ubiquitin ligase function. In this study, we investigated whether PARK2 is involved in osteoclastogenesis. PARK2 expression was found to be increased during the receptor activator of nuclear factor-κB ligand (RANKL)-induced osteoclast differentiation. PARK2 gene silencing with siRNA significantly reduced osteoclastogenesis induced by RANKL, LPS (lipopolysaccharide), TNFα (tumor necrosis factor α), and IL-1β (interleukin-1β). On the other hand, overexpression of PARK2 promoted osteoclastogenesis. This regulation of osteoclastogenesis by PARK2 was mediated by IKK (inhibitory κB kinase) and NF-κB activation while MAPK (mitogen-activated protein kinases) activation was not involved. Additionally, administration of PARK2 siRNA significantly reduced osteoclastogenesis and bone loss in an in vivo model of inflammatory bone erosion. Taken together, this study establishes a novel role for PARK2 as a positive regulator in osteoclast differentiation and inflammatory bone destruction.

Developmental toxicity and anti-inflammatory effect of the soft coral Dendronephthya gigantea collected from Jeju Island in zebrafish model

  • Recent in vitro studies have demonstrated that extract of soft coral Dendronephthya gigantea (SCDE) had strong anti-inflammatory activities. However, the direct effects of SCDE on anti-inflammatory activities in vivo model remained to be determined. Therefore, the present study was designed to assess in vivo anti-inflammatory effect of SCDE using lipopolysaccharide (LPS)-stimulated zebrafish model. We also investigated whether SCDE has toxic effects in zebrafish model. The survival, heart beat rate, and developmental abnormalities were no significant change in the zebrafish embryos exposed to at a concentration below $100{\mu}g/ml$ of SCDE. However, lethal toxicity was caused after exposure to 200 and $400{\mu}g/ml$ of SCDE. Treating zebrafish model with LPS treatment significantly increased the reactive oxygen species (ROS) and nitric oxide (NO) generation. However, SCDE inhibited this LPS-stimulated ROS and NO generation in a dose-dependent manner. These results show that SCDE alleviated inflammation by inhibiting the ROS and NO generation induced by LPS treatment. In addition, SCDE has a protective effect against the cell damage induced by LPS exposure in zebrafish embryos. This outcome could explain the profound anti-inflammatory effect of SCDE both in vitro as well as in vivo, suggesting that the SCDE might be a strong anti-inflammatory agent.

발달적 독서치료 프로그램 운영의 효과 - 시범학교의 사례연구 - (The Effects of Developmental Bibliotherapy in School : In the Case of the Model School)

  • 본 논문은 대구광역시 교육청이 독서치료 시범학교로 지정한 중학교의 독서치료 프로그램 운영효과를 분석한 것이다. 참여 학생은 77명이며 내재화 문제와 외현화 문제를 개선하기 위한 두 개의 프로그램에 분리 배치되었으며 각각 15회 실시되었다. 두 프로그램은 각 문제 영역에서 매우 긍정적이며 통계적으로 유의한 효과를 얻었으며, 독서치료 목표와 독서자료에 따라 뚜렷한 효과의 차이를 보였다. 본 연구에서 독서치료는 내재화 문제, 여학생에게 더 큰 효과가 있었다. 독서치료 프로그램은 독서태도에 영향을 미치는 것으로 분석되었으므로 발달적 독서치료는 향후 자가 독서치료로 발전할 수 있도록 독서교육과 연계될 필요가 있다.

  • 한국의 전통사상중에 가장 중요하다고 할 수 있는 효(孝)의 개념과 기능을 발달학적 측면에서 고찰하였다. 효(孝)란 부모-자녀의 상호의존적인 관계로서 사랑을 주고 받는 애착의 관계로 볼 수 있으며, 공격성을 적절히 조절하고 oedipus complex를 해결하는 기능이 있고, 자녀는 부모를 존경하는 한편 부모는 자녀에게 긍정적 동일시(positive identification)의 model이 되어 자녀로 하여금 자아를 실현하도록 도와주는, 일생을 통해 인격형성 전반에 걸쳐 영향을 주는 역동의 개념으로 이해할 수 있겠다.

Developmental Abnormalities in Zebrafish Angiogenesis with Chronic Exposure to Crude Oil and Dispersant

  • Oil spills have occurred throughout the years of industrialization and represent a global challenge as they affect vast areas of the ocean. The toxicity of crude oil to aquatic organisms has been extensively investigated, but the potential impacts of crude oil on vertebrate development remain largely unknown. Here, we investigated the effects of dispersants used in treating a recent oil spill, as well as that of crude oil, on vertebrates by using the zebrafish (Danio rerio) model species, which has been widely used in empirical studies of both early embryonic development and adult physiology. Chronic exposure to crude oil resulted in marked developmental abnormalities, including pericardial edema, abnormal trunk vessel development, retardation of axonal branching, and abnormal jaw development. Embryonic development was affected more severely by exposure to the oil-dispersant combination than to the oil alone. Thus, the zebrafish in vivo model system suggests that dispersant treatment can have detrimental developmental effects on vertebrates and its potential impact on marine life, as well as humans, should be carefully considered in clean-up efforts at the site of an oil spill.

  • The dog frontal sinus may represent an alternative model dental implant research; its topographical resemblance to the maxillary sinus renders it a potentially favorable experimental environment. The aim of this study was thus to elucidate the anatomical configuration of the canine frontal sinus and histological characteristics, and to determine whether it could be a new canine experimental model for dental implant research. Twenty-four sides of canine frontal bones were harvested. The distance from the nasion to the emerging point of the lateral aspect of the canine frontal sinus was measured with the aid of Lucion software. The thicknesses of the canine frontal sinus wall were measured, and the two specimens stained with hematoxylin and eosin. The mean distance from the nasion to the emerging point of the lateral aspect of the canine frontal sinus was 16.0 mm. The mean thicknesses of the canine frontal bone at 3, 6, 9, 12, and 15 mm lateral to the midsagittal plane were 2.3, 2.7, 3.2, 3.8, and 3.7 mm, respectively. The canine frontal sinus was lined with pseudostratified ciliated columnar epithelium. These data suggest that the canine frontal sinus is a suitable alternative to the canine maxillary sinus as a model for studying various sinus augmentation protocols.

  • MicroRNAs control gene expression by inhibiting translation or promoting degradation of their target mRNAs. Since the discovery of the first microRNAs, lin-4 and let-7, in C. elegans, hundreds of microRNAs have been identified as key regulators of cell fate determination, lifespan, and cancer in species ranging from plants to humans. However, while microRNAs have been shown to be particularly abundant in the brain, their role in the development and activity of the nervous system is still largely unknown. In this review, we describe recent advances in our understanding of microRNA function at synapses, the specialized structures required for communication between neurons and their targets. We also propose how these advances might inform the molecular model of memory.

Follistatins have potential functional role in Porcine Embryogenesis

  • In animal reproduction, the quality of oocytes and embryos has been evaluated by the expression of specific molecules. Follistatin (FST), which was isolated from follicular fluid, binds and bio-neutralizes the TGF-${\beta}$ superfamily members. Previous studies using the bovine model showed FST could be an important molecular determinant of embryo developmental competence. However, the effect of FST treatment on porcine embryo developmental competence has not been established. In this study, the effect of exogenous FST on porcine embryo developmental competence was investigated during in vitro culture. FST (10 ng/ml) treatment induced a significant decrease in the rate of cell arrest at the 4-cell stage. The expression levels of DNA-methyltransferase 1 (DNMT1), histone deacetylase 1 (HDAC1), and histone deacetylase 2 (HDAC2) were decreased in 4-cell stage embryos. FST treatment also resulted in significant improvements in developmental competence of embryos in terms of blastocyst formation rate and OCT-4 mRNA levels, the latter being related to pluripotency. In conclusion, during in vitro culture, FST treatment significantly ameliorated 4-cell block during embryonic development and improved embryo developmental competence. Therefore, FST treatment may potentially have a functional role in porcine embryogenesis that is broadly applicable to enhance in vitro embryo development.

멘토링 기능과 멘토 신뢰가 멘토링 프로그램 재참여의도에 미치는 영향: 패션관련학과 대학생을 중심으로 (The Influence of the Mentoring Function and Mentor Trust on the Intention to Reparticipate in the Mentoring Program: Focused on University Students in Fashion Related Departments)

  • The purpose of this study is to investigate the influence of the mentoring function and mentor trust on the intention to reparticipate in the mentoring program, focusing on university students in fashion related departments. To achieve the purpose, research was conducted through a survey questionnaire. The questionnaire data from 223 university students in fashion related departments who experienced mentoring programs were analyzed. SPSS 18.0 and AMOS 7.0 statistical packages were used to investigate a structural model and test research hypotheses. The results derived from the analysis of the questionnaire survey were summarized as follows. First, the result showed that the mentoring function was composed of three factors: a sensitivity developmental function, psychosocial function, and fashion career developmental function. Second, the sensitivity developmental function, psychosocial function, and fashion career developmental function positively affected mentor trust. Third, the sensitivity developmental function positively affected the intention to reparticipate in the mentoring program, while the psychosocial function and fashion career developmental function did not influence the intention to reparticipate in the mentoring program. Fourth, mentor trust positively influenced the intention to reparticipate in the mentoring program. The results of the current study would be useful for the professor and university administrator who would like to establish a mentoring program in a fashion related department, by exploring the relation of mentoring function, mentor trust and the intention to reparticipate in the mentoring program.
