Anti-Angiogenic Activity of Gecko Aqueous Extracts and its Macromolecular Components in CAM and HUVE-12 Cells

  • Gecko is a kind of traditional Chinese medicine with remarkable antineoplastic activity. However, undefined mechanisms and ambiguity regarding active ingredients limit new drug development from gecko. This study was conducted to assess anti-angiogenic properties of the aqueous extracts of fresh gecko (AG) or macromolecular components separated from AG (M-AG). An enzyme-linked immunosorbent assay (ELISA) approach was applied to detect the vascular endothelial growth factor (VEGF) secretion of the tumor cells treated with AG or M-AG. The effect of AG or M-AG on vascular endothelial cell proliferation and migratory ability was analyzed by tetrazolium dye colorimetric method, transwell and wound-healing assays. Chick embryo chorioallantoic membrane (CAM) assays were used to ensure the anti-angiogenic activity of M-AG in vivo. The results showed that AG or M-AG inhibited the VEGF secretion of tumor cells, the relative inhibition rates of AG and M-AG being 27.2% and 53.2% respectively at a concentration of $20{\mu}L/mL$. AG and M-AG inhibited the vascular endothelial (VE) cell proliferation with IC50 values of $11.5{\pm}0.5{\mu}L/mL$ and $12.9{\pm}0.4{\mu}L/mL$ respectively. The VE cell migration potential was inhibited significantly (p<0.01) by the AG (${\geq}24{\mu}L/mL$) or M-AG (${\geq}12\mu}L/mL$) treatment. In vivo, neovascularization of CAM treated with M-AG was inhibited significantly (p<0.05) at a concentration of ${\geq}0.4{\mu}L/mL$. This study provided evidence that anti-angiogenesis is one of the anti-tumor mechanisms of AG and M-AG, with the latter as a promising active component.

  • In this paper, we implemented for the development of a high output induction lamp system, which lamp design is optimized by gas type, mixing ratio, pressure and discharge tube size, amalgam type and mixing ratio, and characteristics of ferrite core in the lamp. It's the circuit design by improving the power factor and efficiency according to the driving method, which has analyzing the characteristics according to the waveform and frequency. Finally, luminaries design part for applying the optimal lighting system considering the surrounding environment, the characteristics of the lighting circuit for electrodeless lamp has analyzed and the improvement has been proceeded. In conclusion, the driving frequency has optimized at 135kHz with degrading $7{\sim}10^{\circ}C$ based on the results of the optical characteristics of the induction lamp on peak noise FET(Q3,Q4) damage.

Long Cut Straw Provides Stable the Rates of Survival, Pregnancy and Live Birth for Vitrification of Human Blasotcysts

  • Most of the commercial devices for vitrification are directly immersed into the warming solution (WS) for increasing of warming rate. However, the previous modified cut standard straw (MCS) which has reported is difficult to immerse into the WS. The aim of this study was to investigate whether the long cut straw (LCS) could be useful as a stable tool for vitrified-warmed human blastocysts. A total of 138 vitrified-warmed cycles were performed between November 2013 and November 2014 (exclusion criteria: women ${\geq}38$ years old, poor responder, surgical retrieval sperm, and severe male factor). The artificial shrinkage was conducted using 29-gauge needles. Ethylene glycol and dimethyl sulfoxide (7.5% and 15% (v/v)) were used as cryoprotectants. Freezing and warming were conducted using the LCS tool. The cap of LCS was removed using the forceps in the liquid nitrogen ($LN_2$) and then directly immersed into the first WS for 1 min at $37^{\circ}C$ (1 M sucrose). Only re-expanded blastocysts were transferred after it was cultured in sequential media for 18-20 h. A total of 294 blastocysts were warmed, and all were recovered (100%). Two hundred eighty-five embryos were survived (96.9%). The vitrified-warmed blastocysts of all patients were transferred without any cancellation. We were able to achieve a reasonable implantation (24.2%), following by clinical pregnancy (36.2%), which then continued to ongoing pregnancy (36.2%), and live birth (31.2%). Using LCS is achieved the acceptable rates of survival, pregnancy and live birth. Therefore, the LCS could be considered as a stable and simple tool for human embryo vitrificaton.

Determinants of Fund Investment Flows: Asymmetry between Fund Inflows and Fund Outflows (펀드투자 자금흐름의 결정요인: 유입자금과 유출자금은 대칭적인가?)

  • We investigate determinants of fund investment flows using Korean equity investment funds. Unlike previous studies which analyzed net-flows (inflow minus outflow), we analyze fund investment inflows and outflows separately that should properly reflect investors' fund selection and redemption decision. We find similar effects of past return, fund age on net-flows to existing studies based on US market data. The analysis of determinants of inflows shows that inflows are related to past return, fund age and sales fee as net-flows. In contrast, outflows are found to behave quite differently from inflows. Apparently, asymmetry exists between fund investment inflows and outflows at the Korean fund selection market. Specifically, high past returns increase fund investment inflows while increase, rather than decrease, fund outflows. Moreover, 'convexity' is detected both in inflows and outflows: higher past returns accelerate outflows as well as inflows. Effects of sales fee also differ between inflows and outflows. In the 'affiliated' fund sample, sales fee is negatively related to inflows while positively related to outflows. In the 'unaffiliated' fund sample, sales fee is positively related to inflows, but no significant relationship exists with outflows. Empirical findings of this paper imply that the rational investor's fund selection view cannot provide a consistent explanation of the Korean fund selection market. In particular, the positive and convex relationship between past returns and fund outflows is inconsistent with the rational investor view. The fact that investor's fund investment appears to display 'disposition effect', which has been reported by studies of individual investors' stock investment behaviour suggests that the behavioral finance view should be a part of explanation for the Korean fund selection market. In addition, the strikingly different patterns between the 'affiliated' funds and the 'unaffiliated' funds, imply that brokers' incentive structure is another prevailing factor for fund investment flows.

Development and Validation of a Canopy Photosynthetic Rate Model of Lettuce Using Light Intensity, CO2 Concentration, and Day after Transplanting in a Plant Factory (광도, CO2 농도 및 정식 후 생육시기에 따른 식물공장 재배 상추의 군락 광합성 모델 확립)

  • The photosynthetic rate is an indicator of the growth state and growth rate of crops and is an important factor in constructing efficient production systems. The objective of this study was to develop a canopy photosynthetic rate model of romaine lettuce using the three variables of $CO_2$ concentration, light intensity, and growth stage. The canopy photosynthetic rates of the lettuce were measured at five different $CO_2$ concentrations ($600-2,200{\mu}mol{\cdot}mol^{-1}$), five light intensities ($60-340{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$), and four growth stages (5-20 days after transplanting) in three closed acrylic chambers ($1.0{\times}0.8{\times}0.5m$). A simple multiplication model expressed by multiplying three single-variable models and the modified rectangular hyperbola model including photochemical efficiency, carboxylation conductance, and dark respiration, which vary with growth stage, were also considered. In validation, the $R^2$ value was 0.923 in the simple multiplication model, while it was 0.941 in the modified rectangular hyperbola model. The modified rectangular hyperbola model appeared to be more appropriate than the simple multiplication model in expressing canopy photosynthetic rates. The model developed in this study will contribute to the determination of an optimal $CO_2$ concentration and light intensity with the growth stage of lettuce in plant factories.

Utilization of Processed Pine Needle Powder in Broiler Chicken Diets

  • This study examined the effects of dietary processed pine needle tree powder (PPNP) on growth performance, visceral organ parameters, ileal digestibility, blood metabolites and ileal morphology of broiler chickens for 5-week after hatch. In total, 220 1-day-old male Ross 308 broiler chicks were randomly allocated to 30 pens (seven birds per pen), and each pen was assigned to one of five dietary treatments, a diet without PPNP (control) and diets with 0.5%, 1.0%, 1.5% and 2.0% PPNP. The body weight and feed intake were measured weekly. One bird from each pen was selected randomly and gently euthanized to collect the blood, visceral organs and digesta samples on 2, 3 and 5 week of age. Birds fed 2.0% PPNP showed decreased (P<0.05) overall body weight, average daily gain and average daily feed intake compared to those fed control, 0.5 and 1.0% PPNP. The small intestine length increased (P<0.05) as the level of PPNP in the diet increased. A deeper crypt depth was observed (P<0.05) when birds fed 2.0% PPNP compared to those fed the control diet at week 2. A higher V:C ratio was observed (P<0.05) in birds fed 1.0% PPNP than in those fed other treatments at week 3. Ileal protein digestibility improved (P<0.05) when birds fed 0.5% and 1.0% PPNP compared to that in those fed the control diet at week 3. The feed conversion ratio, ileal dry matter digestibility and blood urea nitrogen and cholesterol, and gizzard and ceca weights were not affected (P>0.05) by supplementation of any levels of PPNP. These results demonstrated that inclusion of PPNP in broiler diets could be a possible factor in improving overall growth performance suggesting early gut development when 1% PPNP is supplemented.

  • Alveolar bone loss and deformation can be a risk factor in removable prosthetic restoration treatment for partially or fully edentulous patients. The use of implants to solve this problem could improve the support, retention and stability of removable restoration. Attachments used in implant overdenture are versatile. The attachment should be selected according to the patients' conditions. Milled bar has been chosen when readymade bar could not be used because of the narrow distance between implants or firm stability and support of supra-structure were needed. Milled bar design is able to provide cross arch stabilization and comfortability to patients. However, it needs skilled laboratory procedures. Recently, the fabrication of milled bar has become simple and its suitability has been improved through the development of CAD/CAM system. In a 67-year-old female Alzheimer's disease patient with 8 implant fixtures on the fully edentulous site of mandible, implant overdenture with using milled bar and magnet attachment was planned. As rapid treatment was required, CAD/CAM system was used to make a simple laboratory procedure instead of a traditional fabrication process. With this system, implant overdenture with milled bar can be fabricated esthetically and functionally.

A Comparative Analysis of Negative Air Ions according to the Planting Structure at School Forests (학교 숲 식재구조에 따른 음이온 발생량 비교 분석)

  • In this study, negative air ions, which can increase comfort and positively influence human health, was set as an evaluative factor for school forests. The characteristics, location and microclimate of school forests were examined to determine an effective school forest model for maximum negative air ion generation. It also aimed to provide basic data for the development of urban green fields. The negative air ion concentrations were different according to the green types. The order from the highest to the lowest was: single-layer structure($934ea/cm^3$) > multi-layer structure($794ea/cm^3$) > grass($553ea/cm^3$) > bare ground($529ea/cm^3$). As for the correlation with microclimate, negative air ion concentration was negatively correlated with temperature. The negative air ion concentration was significantly different according to planting type. The negative air ion concentration was higher at single-layer structure sites than at multi-layer structure sites, which appears to indicate that the temperature is higher when the vegetation structure was single-layer compared to multi-layer. Wind speed was higher at the single-layer planted site than at the multi-layer planted site. The vegetation wind shielding effect was lower at single-layer planted sites than at multi-layer planted sites. The single-layer planted site provided more favorable conditions for the generation and dispersion of negative air ions.

Quantitative Evaluation of Criticality According to the Major Influence of Applied with Burnup Credit on Dual-purpose Metal Cask (국내 금속겸용용기의 연소도 이득효과 적용 시 주요영향인자에 따른 정량적 핵임계 평가)

  • In general, conventional criticality analysis for spent fuel transport/storage systems have been performed based on the assumption of fresh fuel concerning the potential uncertainties from number density calculations of actinide nuclides and fission products in spent fuel. However, these evaluation methods cause financial losses due to an excessive criticality margin. In order to overcome this disadvantage, many studies have recently been conducted to design and commercialize a transportation and storage cask applied to the Burnup Credit (BUC). This study conducted an assessment to ensure criticality safety for reactor operating parameters, axial burn-up profiles and misload accident conditions, which are the factors that are likely to affect criticality safety when the BUC is applied to the dual-purpose cask under development at the KOrea RADioactive waste agency (KORAD). As a result, it was found that criticality resulting from specific power, changed substantially and relied on conditions of low enrichment and high burn-up. Considering the end effect in the case of high burn-up produced a positive-definite result. In particular, the increment of maximum effective multiplication factors due to misloading was 0.18467, confirming that misload is a factor that must be taken into account when applying the BUC. The results of this study may therefore be utilized as references in developing technologies to apply the BUC to domestic models and operational procedures or preventing any misload accidents during the process of spent fuel loading.

Case Study on Critical Success Factors and Unexpected Consequences of Structured OJT (S-OJT 성공요인과 예기치 않은 성과에 관한 사례연구)

  • Purpose - Recently on-the-job training (OJT) has become the most preferred training and development method with the emergence of the concept that workplace is the best place where learning take place. But many researchers argue that OJT is not helpful for the performance of organization because OJT is not systematic and mostly depend on quality of trainer. Since Jacobs & McGriffin introduced S-OJT (structured OJT), there has been plenty of researches. But most of the researches have focused mainly on employee's attitude and organizational performance caused by S-OJT and neglected a holistic approach of S-OJT as a system. S-OJT need to be analyzed comprehensively to understand training performance because S-OJT is operated as a system consist of input, process, and organizational context. Although S-OJT may create unintended consequences, there were few researches to explore them. Thus, the purpose of this study is to identify the critical success factors for S-OJT and to find unintended consequences of it. Research design, data and methodology - We conducted a case study on M business unit of A company which developed and has been implementing S-OJT program for years. We designed and prepared the process, collected and analyzed data for the study. We set the theoretical framework to analyze the case after reviewing theories and previous studies on S-OJT. We collected and analyzed internal reports and interview results of the employees of the M business unit. We tried to collect as many information as possible to secure the validity of the research results. Results - The critical success factors identified in the study are as follow. First, it is important to select and train proper trainers for S-OJT. Second, it is needed to develop structured training module. Third, organization have to use effective communication system like on-line community. Forth, trainer should have proper skills for training such as facilitating skill, coaching skill, and delivering skill etc. Fifth, proper learning place is needed. Sixth, organizational support is important especially, immediate supervisor support and concern is critical. Eleventh, it is needed to consider situational contexts. Among them, overload to the trainer will affect the effectiveness of S-OJT. In this study, we found an additional unintended consequence. "To teach is the best way to learn." Experience as a trainer give employee an opportunity to organize one's knowledge and skill and to attain facilitation skill, coaching skill, and relation skill. Thus, organization may use S-OJT to train the potential talent. Conclusions - Many organizations introduced S-OJT to train the newcomers because S-OJT drew attention as an important tool to develop employees. Following this trend, there has been increasing number of researches to find the results of S-OJT and identify the determinants of S-OJT success. However, most of the researches concentrated on finding effects of some factors neglecting holistic approach. This study tried to identify critical success factors affecting effectiveness of S-OJT by using case study and find additional unintended consequence. The results of the study will be useful for organizations which have a plan to adopt S-OJT.
