  • 2,3,7,8-Tetrachlorodibenzo-$\rho$-dioxin(TCDD) displays high toxicity in animals and has been implicated in human carcinogenesis. Although TCDD is recognized as potent carcinogens, relatively little is known about their role in the tumor promotion and carcinogenesis. It is known that TCDD can increase of cancer risk from various types of tissue by a mechanism possibly involving the aryl hydrocarbon receptor (AhR) activation. In this study, effects of TCDD on cellular proliferation of normal human skin and lung fibroblasts, Detroit551 and WI38 cells were investigated. In addition, to enhance our understanding of TCDD-mediated carcinogenesis, we have investigated process in which expression of Erk1/2, cyclinD1, oncogene such as Ha-ras and c-myc, and their cognate signaling pathway. TCDD that are potent activators of AhR-mediated activity was found to induce significant increase of cytochrome P4501A1 mRNA expression, suggesting a presence of functional AhR. These results support that CYP1A1 enzyme may be involved in the generation of TCDD-induced toxicity. Moreover mitogen-activated protein kinases (MARKs) phosphorylation and cyclin D1 overexpression are induced by TCDD, which corresponded with the progression of cellular proliferation. However, TCDD did not affected Ha-ras and c-myc mRNA expression. Taken together, it seems that TCDD are could be a part of cellular proliferation in non-tumorigenic normal human cells such as Detroit551 and WI38 cells through the upregulation of MAPKs signaling pathway regulating growth of cell population. Therefore, AhR-activating TCDD could potentially contribute to tumor promotion and Detroit551 and WI38 cells have been used as a detection system of tumorigenic effects of TCDD.

  • Steam generator(SG) tube, as a barrier isolating primary to the secondary coolant system of nuclear power plants(NPP), must maintain the structural integrity far the public safety and its efficient power generation capacity. And SG tubes bearing defects must be timely detected and taken repair measures if needed. For the accomplishment of these objectives, SG tubes have been periodically examined by eddy current testing(ECT) on the basis of administrative notices and intensified SG management program(SGMP). Stress corrosion cracking(SCC) on the SG tubes is not easily detected and even missed since it has lower signal amplitude and other disturbing factors against its detection. However once SCC is developed, that can cause detrimental affects to the SG tubes due to its rapid propagation rate. Accordingly SCC is categorized as prime damage mechanism challenging the soundness of the SG tubes. In this study, reproduced EDM notch specimens are examined for the detectability and quantitative characterization of the axial ODSCC by +PT MRPC probe, containing pancake, +PT and shielded pancake coils apart in a single plane around the circumference. The results of this study are assumed to be applicable fur providing key information of engineering evaluation of SCC and improvement of confidence level of ECT on SG tubes.

  • To understand the molecular structure and pathogenesis mechanism of Korean garlic viruses, we have isolated cDNA clones for garlic viruses. The partial nucleotide sequences of 24 cDNA clones were determined and those of five clones containing poly(A) tail were compared with sequences of other plant viruses. One of these clones, V9, has a primary structure similar to the carlavirus group, suggesting that the clone V9 derived from a part of garlic latent virus (GLV). Northern blot analysis with the clone V9 as a probe demonstrated that GLV genome is 8.5 knt long and has a poly(A) tail. The clone V9 encodes coat protein (CP) of 33 kDa and nucleic acid binding protein of 10 kDa in different reading frame. The hexanucleotide motif, 5'-ACCUAA, which is conserved in the 3' noncoding region arid was proposed to be a cis-acting element involved in the production of negative strand genomic RNA was noticed. Complementary sequence to the hexanucleotide motif, 5'-TTAGGT, is also found in the positive strand of V9 RNA. The putative CP gene was cloned into the pRSET-A expression vector and expressed in E. coli BL21. The expressed recombinant V9CP protein was purified by $Ni^{2+}$ NTA affinity chromatography. The anti-V9CP antibody recognizes 34 kDa polypeptide which could be CP of GLV in infected garlic leaf extract. Immunoblot and Northern blot analysis of various cultivars shows wide occurrence of GLV in Korean garlic plants.

  • According to the increase of various AV media devices and contents in the digital home, the DLNA becomes to play an important role as the interoperability standard between then Since this guideline only focuses on the interoperability among home networked devices, media players, and media contents existing inside of the homenetwork, there is no retrieval and transmission method for sharing multimedia contents located over several homes via Internet. Additionally, this guideline lets device-detection and notification messages to be transmitted using W multicast methods, and current Internet environment cannot guarantee consistent IP multicast services, it has the limitation that it cannot retrieve and control DLNA devices in other digital homes remotely via the Internet. Therefore, in this paper, we propose the IHM(Inter-Home Media) proxy system and its operating mechanism to provide a way of sharing media contents distributed over multiple DLNA-based homes, through analyzing these limitations and building up a sharing method for A/V media contents distributed over the DLNA homes based on the dynamic or private IP networks. Our method removes the limitation on the user locations through sharing distributed media contents, and also makes cost-downs for storing media contents, from the view point of individual residents.

  • Climate change is believed to increase the amount of dissolved organic matter in surface water, as a result of the release of bulk organic matter, which make difficult to achieve a high quality of drinking water via conventional water treatment techniques. Therefore, the natural water treatment techniques, such as managed aquifer recharge (MAR), can be proposed as a alternative method to improve water quality greatly. Removal of bulk organic matter using managed aquifer recharge system is mainly achieved by biodegradation. Biodegradable dissolved organic carbon (BDOC) and assimilable organic carbon (AOC) can be used as water quality indicators for biological stability of drinking water. In this study, we compared the change of BDOC and AOC with respect to pretreatment methods (i.e., ozone or peroxone). The oxidative pretreatment can transform the recalcitrant organic matter into readily biodegradable one (i.e., BDOC and AOC). We also investigated the differences of organic matter characteristics between BDOC and AOC. We observed the decreases in dissolved organic carbon (DOC) and the tryptophan-like fluorescence intensities. Liquid chromatographic - organic carbon detection (LC-OCD) analysis also showed the reduction of the low molecular weight (LMW) fraction (15% removed, less than 500 Da), which is known to be easily biodegradable, and the biopolymers, high molecular weight fractions (66%). Therefore, BDOC consists of a broad range of organic matter characteristics with respect to molecular weight. In AOC, low molecular weight organic matter and biopolymers fraction was reduced by 11 and 6%, respectively. It confirmed that biodegradation by microorganisms as the main removal mechanism in AOC, while BDOC has biodegradation by microorganism as well as the sorption effects from the sand. $O_3$ and $O_3+H_2O_2$ were compared with respect to biological stability and dissolved organic matter characteristics. BDOC and AOC were determined to be about 1.9 times for $O_3$ and about 1.4 times for $O_3+H_2O_2$. It was confirmed that $O_3$ enhanced the biodegradability by increasing LMW dissolved organic matter.

  • The purpose of this study was to provide a detailed explanation of the mechanism of bisanthracycline, WP 631 in comparison to doxorubicin (DOX), a first generation anthracycline, currently the most widely used pharmaceutical in clinical oncology. Experiments were performed in SKOV-3 ovarian cancer cells which are otherwise resistant to standard drugs such as cis-platinum and adriamycin. As attention was focused on the ability of WP 631 to induce apoptosis, this was examined using a double staining method with Annexin V and propidium iodide probes, with measurement of the level of intracellular calcium ions and cytosolic cytochrome c. The western blotting technique was performed to confirm PARP cleavage. We also investigated the involvement of caspase activation and DNA degradation (comet assay and immunocytochemical detection of phosphorylated H2AX histones) in the development of apoptotic events. WP 631 demonstrated significantly higher effectiveness as a pro-apoptotic drug than DOX. This was evident in the higher levels of markers of apoptosis, such as the externalization of phosphatidylserine and the elevated level of cytochrome c. An extension of incubation time led to an increase in intracellular calcium levels after treatment with DOX. Lower changes in the calcium content were associated with the influence of WP 631. DOX led to the activation of all tested caspases, 8, 9 and 3, whereas WP 631 only induced an increase in caspase 8 activity after 24h of treatment and consequently led to the cleavage of PARP. The lack of active caspase 3 had no outcome on the single and double-stranded DNA breaks. The obtained results show that WP 631 was considerably more genotoxic towards the investigated cell line than DOX. This effect was especially visible after longer times of incubation. The above detailed studies indicate that WP 631 generates early apoptosis and cell death independent of caspase-3, detected at relatively late time points. The observed differences in the mechanisms of the action of WP631 and DOX suggest that this bisanthracycline can be an effective alternative in ovarian cancer treatment.

  • Purpose: It has recently been reported that de novo HBV infection following liver transplantation is caused by grafts from HBcAb positive donors, and this phenomenon has been observed in one third of the liver transplant patients in our center. Therefore, we investigated the presence of HBV virus DNA in liver tissues obtained from HBcAb positive donors to determine the mechanism by which de novo HBV infection occurs. Methods: This study was conducted on 6 patients that were HBsAg negative, HBsAb positive, and HBcAb positive who were donors for liver transplantation between November 1997 and November 1998 at Asan Medical Center. We isolated DNA from a portion of liver biopsy tissues that were obtained during the operation, and then identified the surface and core region of HBV DNA using nested PCR. In addition, four children who received liver grafts from these donors were monitored to determine if they became afflicted with non-HBV related diseases while receiving prophylaxis consisting of short-term HBIG treatment and long-term treatment with an antiviral agent. Results: The surface antigen region was identified in all 6 donors and the core antigen region was observed in 4 of the 6 donors. However, no episodes of de novo HBV infection with prophylaxis were observed. Conclusion: The results of this study support the results of previous studies, which indicated that HBV infection may be the main cause of de novo HBV infection in patients that receive HBsAb positive and HBcAb positive donor grafts.

  • DA-5018 is a synthetic capsaicin derivative under development as a non-narcotic a analgesic ag$\varepsilon$nt. DA-50 18 showed a potent analgesic activity against acute and chronic pain m model(Tablel, 2.), but it had a narrow margin of safety. DA-5018 did not bind to opioid(${\kappa}, {\delta}, {\mu}$), NKl, CGRP receptors in vitro and its analgesic effect was not antagonized by naloxone, a and it did not develop analgesic tolerance. In addition DA-5018 had no inhibitory effects against c cyclooxygenase and 5-lipooxygenase activities. DA-5018 significantly increased the relcase of substance P from the slices of the rat spinal cord. These results suggest that DA-50 18 is not a narcotic nor aspirin-like analgesic and the release of substance P is one of analgesic mechanism of action of DA-5018. We found that DA-5018 was almost ten times more potent and was at l least IOO-times less irritable compared to capsaicin. Accordingly development of topical formula was adopted. Topical formula was desiged and screened by flux test of DA-5018 using hairless mouse skin and several formulas were selected. With these topical formulas we a assessed the analgesic efficacy and carried out the toxicity, skin irritation and pharmacokinetic studies. In streptozotocin-induced hyperalgesic rat and 50 % galactose-fed hyperalgesic rat as diabetic pain models, DA-5018 cream increased the pain thresh이ds up to 77.0% and 24.4% respectively, while Zostrix-HP(capsaicin cream) incr$\varepsilon$as cd by 65.9% and 21.0%. DA-5018 c cream showed a good analgesic effect as welI in FCA-induced arthritic rat. DA-5018 cream did not show any toxicological signs in acute and chronic toxicity test and had little skin irritation in car swclIing and scratching t$\varepsilon$st. Pharmacokinetics of DA-50 18 were studied after topical application of ${14}^C$-Iabelled or unlabelIed DA-5018 cream. Plasma and skin concentrations c except applied skin wcre below the dctection limit and after 7-day cummulative application, plasma concentrations were also below detection limit DA-50 18 may have an advantag$\varepsilon$ ov$\varepsilon$r c capsaicin and is now being developed as a topical agent for the treatment of pains. DA-50 18 cream was approved for Korean IND and is now under a Phase II clinical study for arthritic pain a after finising Phase I study. • Nutritious and functional foods from crop have received great attention in recent years. Colored-grain wheat contains high phenolic compound and a large number of flavonoid. The anthocyanin and polyphenolic synthesis and accumulation is generally stimulated in response to biotic or abiotic stresses. Here, we analyzed genome wide transcripts in seedling of colored-grain wheat response to ABA and PEG treatment. About 900 and 1500 transcripts (p-value < 0.05) from ABA and PEG treatment were aligned to IWGSC1+popseq DB which is composed of over 110,000 transcripts including 100,934 coding genes. NR protein sequences of Poaceae from NCBI and protein sequence of transcription factors originated from 83 species in plant transcription factor database v3.0 were used for annotation of putative transcripts. Gene ontology analysis were conducted and KEGG mapping was performed to show expression pattern of biosynthesis genes related in flavonoid, isoflavonoid, flavons and anthocyanin biopathway. DroughtDB (http://pgsb.helmholtz-muenchen.de/droughtdb/) was used for detection of DEGs to explain that physiological and molecular drought avoidance by drought tolerance mechanisms. Drought response pathway, such as ABA signaling, water and ion channels, detoxification signaling, enzymes of osmolyte biosynthesis, phospholipid metabolism, signal transduction, and transcription factors related DEGs were selected to explain response mechanism under water deficit condition. Anthocyanin, phenol compound, and DPPH radical scavenging activity were measured and antioxidant activity enzyme assays were conducted to show biochemical adaptation under water deficit condition. Several MYB and bHLH transcription factors were up-regulated in both ABA and PEG treated condition, which means highly expressed MYB and bHLH transcription factors enhanced the expression of genes related in the biosynthesis pathways of flavonoids, such as anthocyanin and dihydroflavonols in colored wheat seedlings. Subsequently, the accumulation of total anthocyanin and phenol contents were observed in colored wheat seedlings, and antioxidant capacity was promoted by upregulation of genes involved in maintaining redox state and activation of antioxidant scavengers, such as CAT, APX, POD, and SOD in colored wheat seedlings under water deficit condition. This work may provide valuable and basic information for further investigation of the molecular responses of colored-grain wheat to water deficit stress and for further gene-based studies.

  • In this paper we present a Distributed Object oriented Virtual computing Environment, called DOVE which consists of autonomous distributed objects interacting with one another via method invocations based on a distributed object model. DOVE appears to a user logically as a single virtual computer for a set of heterogeneous hosts connected by a network as if objects in remote site reside in one virtual computer. By supporting efficient parallelism, heterogeneity, group communication, single global name service and fault-tolerance, it provides a transparent and easy-to-use programming environment for parallel applications. Efficient parallelism is supported by diverse remote method invocation, multiple method invocation for object group, multi-threaded architecture and synchronization schemes. Heterogeneity is achieved by automatic data arshalling and unmarshalling, and an easy-to-use and transparent programming environment is provided by stub and skeleton objects generated by DOVE IDL compiler, object life control and naming service of object manager. Autonomy of distributed objects, multi-layered architecture and decentralized approaches in hierarchical naming service and object management make DOVE more extensible and scalable. Also, fault tolerance is provided by fault detection in object using a timeout mechanism, and fault notification using asynchronous exception handling methods
