In Vitro Antitumor Properties of an Isolate from Leaves of Cassia alata L

  • Leaf extracts of Cassia alata L (akapulko), traditionally used for treatment of a variety of diseases, were evaluated for their potential antitumor properties in vitro. MTT assays were used to examine the cytotoxic effects of crude extracts on five human cancer cell lines, namely MCF-7, derived from a breast carcinoma, SK-BR-3, another breast carcinoma, T24 a bladder carcinoma, Col 2, a colorectal carcinoma, and A549, a nonsmall cell lung adenocarcinoma. Hexane extracts showed remarkable cytotoxicity against MCF-7, T24, and Col 2 in a dose-dependent manner. This observation was confirmed by morphological investigation using light microscopy. Further bioassay-directed fractionation of the cytotoxic extract led to the isolation of a TLC-pure isolate labeled as f6l. Isolate f6l was further evaluated using MTT assay and morphological and biochemical investigations, which likewise showed selectivity to MCF-7, T24, and Col 2 cells with $IC_{50}$ values of 16, 17, and 17 ${\mu}g/ml$, respectively. Isolate f6l, however, showed no cytotoxicity towards the non-cancer Chinese hamster ovarian cell line (CHO-AA8). Cytochemical investigation using DAPI staining and biochemical investigation using terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL)-a method used to detect DNA fragmentation-together with caspase assay, demonstrated apoptotic cell death. Spectral characterization of isolate f6l revealed that it contained polyunsaturated fatty acid esters. Considering the cytotoxicity profile and its mode of action, f6l might represent a new promising compound with potential for development as an anticancer drug with low or no toxicity to non-cancer cells used in this study.

UNDERWATER NOISE GENERATED BY FISHING GEAR -Stern trawl net- (어구에서 발생되는 소음 -트로올 어구-)

  • The main purpose of the present study is to measure the sound spectra of the underwater noises generated by moving trawl net. An underwater recording system was designed to detect underwater noise generated by moving trawl net. The acoustic analysis was made by a heterodyne analyzer (B & K 2010) and level recorder (B & K 2307). The noises generated by the trawl net are appreciably higher (about 10dB) than the background noise in the presence of the fishing vessel. The frequency distribution of underwater noise was DC-6,300 Hz and predominant frequency range was 100-200 Hz, and maximum sound pressure level was $137\;db(re\;1{\mu}Pa)$. Sound pressure level recorded at the ground rope was higher than that recorded at the head rope. The sound pressure level meosured in the course of hawling was higher than that measured in the course of towing. When tile net is being casted tile sound pressure level showed the lowest value.

  • This paper addresses the results of HAUSAT-2 radiation environment and effect analyses, including TID and SEE analyses. Trapped proton and electron, solar proton, galactic cosmic ray models were considered for HAUSAT-2 TID radiation environment analysis. TID was analyzed through total dose-depth curve and the radiation tolerance of TID for HAUSAT-2 components was verified by using DMBP method and sectoring analysis. HAUSAT-2 LET spectrum for heavy ion and proton were also analyzed for SEE investigation. SEE(SEU, SEL) analyses were accomplished for MPC860T2B microprocessor and K6X8008T2B memory. It was estimated that several SEUs may occur without SEL during the HAUSAT-2 mission life(2 years). Software Hamming Code EDAC has been implemented to detect and correct the SEU. In this study, all radiation analyses were conducted by using SPENVIS software.

Differential Diagnosis of Hyperphenylalaninemias (고페닐알라닌혈증의 진단 및 평가)

  • All infants should be screened for phenylketonuria (PKU) within the three days of life, in order to allow timely dietary intervention to protect children with PKU from neurologic damage in Korea. A commonly used cut-off level for diagnosis of PKU is $240{\mu}mol/L$ (4 mg/dL). Up to 2% of cases of hyperphenylalaninemias (HPA) detected by the screening test will account for a disorder of $BH_4$ metabolism. Therefore, analysis of blood or urinary pterins is essential, backed up with measurement of DHPR activity, as this allows differentiation of $BH_4$ disorders. A $BH_4$ loading test and measurement of neurotransmitters in CSF provide further important information to the severity of $BH_4$ deficiency and $BH_4$ loading test can detect patients with $BH_4$ deficiency and $BH_4$ responsive PKU. Several protocols for $BH_4$ loading test have been described, involving treatment with $BH_4$ for periods ranging from 1 day to 1 month, and using doses of $BH_4$ of 10-20 mg/kg. There is general agreement that a reduction on blood phenylalanine of at least 30% in response to $BH_4$ loading indicates a clinically significant effect, although in some tests a lower cut-off value may be defined for individual patients, or no specific cut-off value is proposed. The frequency of $BH_4$ responsiveness is highest in patients with mild HPA and mild to moderate PKU resulting from PAH mutations with residual activity.

Rapid and Sensitive Detection of Lettuce Necrotic Yellows Virus and Cucumber Mosaic Virus Infecting Lettuce (Lactuca sativa L.) by Reverse Transcription Loop-Mediated Isothermal Amplification

  • Cucumber mosaic virus (CMV) is damaging to the growth and quality of lettuce crops in Lanzhou, China. Recently, however, for the first time an isolate of lettuce necrotic yellows virus (LNYV) has been detected in lettuce crops in China, and there is concern that this virus may also pose a threat to lettuce production in China. Consequently, there is a need to develop a rapid and efficient detection method to accurately identify LNYV and CMV infections and help limit their spread. Reverse transcription loop-mediated isothermal amplification (RT-LAMP) assays were developed to detect the nucleoprotein (N) and coat protein (CP) genes of LNYV and CMV, respectively. RT-LAMP amplification products were visually assessed in reaction tubes separately using green fluorescence and gel electrophoresis. The assays successfully detected both viruses in infected plants without cross reactivity recorded from either CMV or LNYV or four other related plant viruses. Optimum LAMP reactions were conducted in betaine-free media with 6 mM Mg2+ at 65℃ for LNYV and 60℃ for 60 min for CMV, respectively. The detection limit was 3.5 pg/ml and 20 fg/ml using RT-LAMP for LNYV and CMV plasmids, respectively. Detection sensitivity for both RT-LAMP assays was greater by a factor of 100 compared to the conventional reverse transcription polymerase chain reaction assays. This rapid, specific, and sensitive technique should be more widely applied due to its low cost and minimal equipment requirements.

Clinical Study on the Floating and Sinking Pulse Detection with Piezoresistive Sensors and Contact Pressure Control Robot (압저항 센서와 가압조절 로봇을 이용한 부침맥 검출에 관한 임상연구)

  • The pulse diagnosis is an important and universal method in Oriental medicine. Nevertheless, because of characteristic that depends on subjective sense of Oriental medicine doctor (OMD), it is not recognized by objective basis. The Korean Institute of Oriental Medicine(KIOM) and Daeyo Medi. Co. Ltd. developed the 3-D Mac using arrey piezoresistive sensors and multi-axial robot. 133 healthy subjects participated in this study, 75 males and 58 females, between 20 and 70 years of age. All subjects were relaxed in a supine position on a comfortable chair for twenty minutes before the measurement was taken. The measured position is the radial artery of subject's left wrist and the position is called Chon, Kwan and Chuck in Oriental medicine. To detect floating and sinking pulse, we established coefficient of floating and sinking(CFS). CFS means relative position of maximum pulse pressure in PH curve. The lower CFS value means that the pulse has floating tendency. There was significant diffence between CFS and diagnosis of floating-sinking pulse by OMD(p=0.020). CFS value of over 40's group was significantly larger than those of 20's and 30's(p=0.000). There was no significant difference between male and female(p=0.061).

  • Flow accelerated corrosion(FAC) of carbon steel pipes in nuclear power plants has been known as one of the major degradation mechanisms. It could have bad influence on the plant reliability and safety. Also detection of FAC is a significant cost to the nuclear power plant because of the need to remove and replace insulation. Recently, the interest of the guided wave testing(GWT) has grown because it allows long range inspection without removing insulation of the pipe except at the probe position. If GWT can be applied to detection of FAC damages, it will can significantly reduce the cost for the inspection of the pipes. The objective of this study was to determine the capability of GWT to identify location of FAC damages. In this paper, three kinds of techniques were used to measure the amplitude ratio between the first and the second welds at the elbow area of mock-ups that contain real FAC damages. As a result, optimal inspection technique and minimum detectability to detect FAC damages drew a conclusion.

Validation of an Enzyme-Immunoassay for Calcitriol in Human Plasma and Evaluation of Its Pharmacokinetics after Single-dose in Korean Volunteers (인체 혈장 중 칼시트리올의 효소면역 분석법 검증 및 단회투여 후 약물동태 연구)

  • An enzyme immunoassay (EIA) was validated for quantitation of cacitriol in human plasma. Calcitriol was immunoextracted with immunocapsules, which contain monoclonal antibodies to calcitriol linked to solid phase particles in suspension with a vitamin D binding protein inhibitor. Calcitrol was eluated and the eluates were evaporated under a gentle stream of nitrogen gas. The absorbance of analytes was determined using a microplate reader (reference wavelength 650 nm; measurement wavelength 450 nm). The method was specific and sensitive enough to detect as low as 6.5 pmol/L of calcitriol. Linear calibration range was 6.5-491 pmol/L with correlation coefficient greater than 0.99. The overall accuracy was in the range of 83.8 to 111.2% and precision C.V. (%) 0.99 to 8.47%. The recovery was approximately 100% and stability was confirmed during storage and sample preparation. The pharmacokinetic parameters were calculated by baseline subtraction because calcitriol is an endogenous material. Following oral dose of calcitriol, the mean AUC$_{24h}$ was 1038${\pm}$539 pmol/Lhr and C$_{max}$ of 128${\pm}$63.1 pmol/L was reached at 3.50${\pm}$1.07 hr. The mean t$_{1/2}$ of calcitriol was 5.13${\pm}$2.10 hr. The present EIA method was successfully applied to study bioavailability after oral administration of 2 ${\mu}$g of calcitriol in healthy Korean subjects.

A Study on UAV DoA Estimation Accuracy Improvement using Monopulse Tracking (모노펄스 추적을 이용한 무인기 DoA 추정정밀도 향상 방안에 관한 연구)

  • Various studies such as INS(: Inertial Navigation System) are conducting to estimate the position of UAV, because the GPS information of UAV is at risk like the GPS jamming. The position estimation using DoA and RTT are used to apply many radar systems, and that process can be applied in datalink of UAV. The general monopulse feed in UAV datalink is Multi-horn, because of the wide BW(: Band Width) and frequency range. And it needs wide SNR range of tracking because of the limited transmit power of airborne unit. The estimation error of position increase at low SNR, and the DoA is valid in only 3dB beam width but high SNR causes false of mainlobe detection because of large sidelobe. In this paper, We propose the method to achieve higher accuracy of DoA estimation on low SNR and review some idea that able to detect mainlobe.

An Electrical Resistivity Survey for Leachate Investigation at a Solid Waste Landfill (폐기물 매립지 침출수 조사를 위한 전기비저항 탐사)

  • The electrical resistivity method is an effective geophysical tool to detect subsurface contamination because the contaminated zones show generally lower electrical resistivity. In this study, the electrical resistivity surveys were applied to a waste landfill site to image the subsurface structure around the landfill and to identify the contaminated zones. First, the dipole-dipole 2D resistivity surveys were conducted along the boundaries of landfill to define the developed contaminated zones. Then the crosshole resistivity tomography was applied to confirm the suspected contaminated zones at depth. The results of drilling and geochemical analysis of ground water supported that the low resistivity zones coincide well with the contaminated zones and the leachate pathways could be delineated effectively from the resistivity survey.
