Protective Effect of NACA on Periodontal Stem Cell (NACA 처리에 따른 치주줄기세포 사멸 억제 효과)

  • Purpose :Periodontal ligament stem cells maintain tissue homeostasis in periodontal ligament. The purpose of this study was to determine the characteristics of periodontal ligament stem cells isolated from premolar teeth and observe protective effects against oxidative damage caused by Triethylene glycol dimethacrylate (TEGDMA) following treatment with N-acetylsysteine amide (NACA) drug known as enzymatic antioxidants. Methods : Primary periodontal ligament stem cell (PDSC) culture was performed from simply extracted human premolar of orthodontic patients. The characteristics of the primary cultured PDSCs was analyzed using the FACS system. PDSCs was incubated with TEGDMA and NACA. The cell proliferation and survival was determined using WST-1 assay. Collected data were analyzed using SPSS Window 20. Results : Primary cultured PDSCs grow on the floor and develop rapidly in a cluster form from up to 14 days. The morphology of PDSCs showed the spindle-shaped cells and grew directionally. FACS analysis, In addition, positive expression of visible cells were observed in mesenchymal stem cell biomarkers. PDLSCs cell viability was significantly decreased at high concentration in both 3 and 6 hours after TEGDMA treatment. We observed a decrease in the number of cells as well as a morphological change of PDLSCs. Antioxidative effect was notable since the death of PDLSC death was significantly inhibited compared to the control group at 24 and 48 hours after NACA treatment. Conclusion : Therefore, based on the results of this study, further research should be encouraged considering the development of clinical treatment methods using various antioxidants as well as regenerative engineering techniques utilizing periodontal ligament stem cells.

  • Electrolytic water(EW), studied in recent decades in the Japan, Russia and United State of America, have shown promise as a method of disinfection whereby low levels of free chlorine, sodium hypochlorite, or hypochlorous acid may be produced in situ in Nacl-containing solution. These methods have shown promise in destruction of microorganisms in medical, dental environment, and in the agriculture and food industry. A recently EW treatment system was evaluated for reducing scouring agent and other surfactants in the washing and scouring process of textile industry Unfortunately, there is, to my knowledge, no serious studies of the properties of EW for textile industry In order to study the characteristics of EW and confirm the possibility of applications in textile industry processes, the pH, surface activity, penetration force, surface tension, and contact angle of EW was measured under various conditions. In general terms, What all this shows is that there is fundamental difference between the properties of EW and that of distilled water.

Glycothermal Synthesis and Characterization of 3Y-TZP Nanoparticles

  • In this study, 3 mol% yttria-tetragonal zirconia polycrystal (3Y-TZP) nanoparticles were synthesized by the glycothermal method under various reaction temperatures and times. The co-precipitated precursor of 3Y-TZP was prepared by adding $NH_4OH$ to starting solutions, and then the mixtures were placed in an autoclave reactor. Tetragonal yttria-doped zirconia nanoparticles were afforded through a glycothermal reaction at a temperature as low as $220^{\circ}C$, using co-precipitated gels of $ZrCl_4$ and $YCl_3{\cdot}6H_2O$ as precursors and 1,4-butanediol as the solvent. The synthesized 3Y-TZP particles were characterized by X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), transmission electron microscopy (TEM), and Raman spectroscopy. The 3Y-TZP particles have a stable tetragonal phase only at glycothermal temperatures above $200^{\circ}C$. To investigate phase transition, the 3Y-TZP particles were heat treated from 400 to $1400^{\circ}C$ for 2 h. Raman analysis indicated that, after heat treatment, the tetragonal phase of the 3Y-TZP particles remained stable. The results of this study, therefore, suggest that 3Y-TZP powders can be prepared by the glycothermal method.

  • Objectives : To discuss the biocompatibility of stainless steel for using acupuncture needles. Results and Conclusions : It is widely assumed that stainless steel is one of the good corrosion-resistant materials and is safe for human body, Most of metal acupuncture needles are made of STS304 and STS316 in light of human body safety: STS304 in more differences between STS304 and STS316 in light of human body safety: STS304 is more vulnerable to oxidation than STS316. A variety of literature survey showed that although STS316 is more corrosion-resistant than STS304 there is no evidence supporting the hypothesis that STS304 has the potential danger to human body, Further study would be necessary to draw more decisive conclusion.

  • Chitosan is a biodegradable and non-toxic material with a molecular weight of 800-1,500Kd which can be obtained in various forms with extraordinary chemical structures and biological characteristics of which enables it to be used in many fields as a biomaterial. Ion irradiation is a useful tool to modify chemical structures and physical properties of high molecular weight polymers. The basic hypothesis of this study is that when surface properties of chitosan in a sponge form are modified with ion beam-irradiation and cell adhesion properties of chitosan would improve and thereby increase the regenerative ability of the damaged bone. The purpose of this study was to illuminate the changes in the cytocompatibility of chitosan sponges after ion beam-irradiation as a preliminary research. Argon($Ar^+$) ions were irradiated at doses of $5{\times}10^{13}$, $5{\times}10^{15}$ at 35 keV on surfaces of each sponges. Cell adhesion and activity of alkaline phosphatases were studied using rat fetal osteoblasts. The results of this study show hat ion beam-irradiation at optimal doses($5{\times}10^^{13}\;Ar^+\;ion/cm^2$) is a useful method to improve cytocompatibility without sacrificing cell viability and any changing cell phenotypes. These results show that ion beam-irradiated chitosan sponges can be further applied as carriers in tissue engineering and as bone filling materials.

  • With recent increased demand for reverse engineering in dental machining, the 3D laser scanner is widely used for inspection of artificial pontic. In order to overcome the optical drawback of laser scanner, such as irregular scatter, direction of beam, and the influence of surface integrity, it is developed in this study a new 3D measuring system for artificial pontic using spherical coordinate system mechanism by point laser sensor, which keeps the direction of beam normal to surface consistently. The comprehensive integrated system is established to evaluate the improvement of accuracy with data acquisition system. The experimental results for measuring a master ball and pontic models shows the excellent form accuracy and repeatability compared with conventional apparatus. Also, these results shows the possibility to apply this system for the measuring purpose within 0.05mm accuracy of pontic at the sharp edge or margin contour, which was difficult to measure at the conventional systems.

A network-biology approach for identification of key genes and pathways involved in malignant peritoneal mesothelioma

  • Even in the current age of advanced medicine, the prognosis of malignant peritoneal mesothelioma (MPM) remains abysmal. Molecular mechanisms responsible for the initiation and progression of MPM are still largely not understood. Adopting an integrated bioinformatics approach, this study aims to identify the key genes and pathways responsible for MPM. Genes that are differentially expressed in MPM in comparison with the peritoneum of healthy controls have been identified by analyzing a microarray gene expression dataset. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analyses of these differentially expressed genes (DEG) were conducted to gain a better insight. A protein-protein interaction (PPI) network of the proteins encoded by the DEGs was constructed using STRING and hub genes were detected analyzing this network. Next, the transcription factors and miRNAs that have possible regulatory roles on the hub genes were detected. Finally, survival analyses based on the hub genes were conducted using the GEPIA2 web server. Six hundred six genes were found to be differentially expressed in MPM; 133 are upregulated and 473 are downregulated. Analyzing the STRING generated PPI network, six dense modules and 12 hub genes were identified. Fifteen transcription factors and 10 miRNAs were identified to have the most extensive regulatory functions on the DEGs. Through bioinformatics analyses, this work provides an insight into the potential genes and pathways involved in MPM.

Comparative scanning electron microscope analysis of the enamel of permanent human, bovine and porcine teeth

  • Background: Bovine and porcine teeth are often used in in vitro experiments as substitutes of human teeth. Objectives: The aim of the present study was to perform a comparative analysis of enamel morphology of permanent human, bovine and porcine teeth under the scanning electron microscope. Methods: As many as 10 human, 10 bovine, and 10 porcine teeth were studied. All the teeth were sectioned and the halves were randomly divided into 2 groups according to the examined tissue (vestibular enamel at the mid-height of the dental crown and in the cervical area). Human and bovine enamel was etched for 15 sec and porcine enamel for 30 sec. The scanning electron microscope analysis was performed. The length and width of enamel prisms were determined with the "Met-Ilo" 1.1 computer program. Results: All enamel samples revealed the same etching pattern-Silverstone's type 2. Bovine enamel showed a similar porosity and the amount of interprismatic enamel compared to human enamel while the amount and width of interprismatic enamel bands in porcine enamel were evidently greater. The shape of the porcine prisms was visually similar to human prisms, although dimensions were significantly different. However, bovine prisms differed in form and appeared to be distinctly elongated. Conclusions: Reported findings indicate that the results of experimental studies carried out on bovine and porcine enamel should not be compared with the results obtained on human enamel.

Image Segmentation of Fuzzy Deep Learning using Fuzzy Logic (퍼지 논리를 이용한 퍼지 딥러닝 영상 분할)

  • In this paper, we propose a fuzzy U-Net, a fuzzy deep learning model that applies fuzzy logic to improve performance in image segmentation using deep learning. Fuzzy modules using fuzzy logic were combined with U-Net, a deep learning model that showed excellent performance in image segmentation, and various types of fuzzy modules were simulated. The fuzzy module of the proposed deep learning model learns intrinsic and complex rules between feature maps of images and corresponding segmentation results. To this end, the superiority of the proposed method was demonstrated by applying it to dental CBCT data. As a result of the simulation, it can be seen that the performance of the ADD-RELU fuzzy module structure of the model using the addition skip connection in the proposed fuzzy U-Net is 0.7928 for the test dataset and the best.

Effect of Blood Contamination on Vickers Microhardness and Surface Morphology of Mineral Trioxide Aggregate

  • This study aimed to investigate the effects of blood contamination on the Vickers hardness and the surface morphology of premixed MTA and compare them with the effects on conventional MTA. The Vickers microhardness of Endocem MTA Premixed Regular (EP) and ProRoot MTA (PM) was assessed after immersion in fetal bovine serum (FBS) and saline. Stem cells from human exfoliated deciduous teeth (SHED) were seeded on MTA after immersion in FBS, saline, and deionized water (DW). Cell adhesion patterns and surface morphology were visualized via scanning electron microscopy (SEM). The surface microhardness of EP and PM in FBS was lower than in saline. However, short-term exposure of PM to FBS did not reduce the microhardness compared to saline. Angular crystals formed in water, while rounded crystals with more air voids appeared in FBS. Favorable SHED attachment occurred in all groups. Overall, the surface hardness of EP and PM decreased after FBS exposure, although PM was less influenced. We suggest minimizing the amount of bleeding when using MTA clinically; nevertheless, PM remains an option with more expected blood contamination than EP. In summary, exposure to FBS decreased mechanical performance but allowed cell adhesion for both MTAs, with PM being more resistant to these changes.
