Localizations of substance P, CGRP and calcium binding proteins in Korean native goat duodenum (한국재래산양 십이지장의 장관신경계통에 분포하는 Substance P, CGRP 및 칼슘결합단백질 반응세포에 대한 면역조직화학적 연구)

  • The localization of substance P(SP), calcitonin gene-related peptide(CGRP) and three calcium binding proteins, calbindin D-28k(CB), calretinin(CR) and parvalbumin(PA) was immunohistochemically examined in the myenteric and submucous plexuses of Korean native goat duodenum. In the neurons of myenteric and submucous plexuses of duodenum, immunoreactivities of SP, CGRP and CB were confirmed in both nerve cell bodies and fibers. In contrast, CR immunoreactivity was found only in nerve fibers of myenteric plexuses, while PA immunoreactivity was found only in nerve cell bodies of submucous plexuses. In the inner circular muscle layer, dense SP-like immunoreactive fibers were prominent but only a few CGRP-like immunoreactivities were observed. Most of SP- and CGRP-like immunoreactive neurons of both plexuses colocalized with CB. This result showed that SP and CGRP may have a important role for the movement of small intestine. The colocalizations of CB with SP or CGRP in myenteric and submucous plexuses suggest that CB may serve neuromodulatory role for SP- and CGRP-immunoreacted neurons on the movement of intestinal wall.

  • Al2O3/Al composites were produced by immersing the sintered silica preform in molten aluminum which contained magnesium as impurity. Three distinct regions existed in the penetration behavior of molten me-tal with changing the reaction temperature. These regions are denoted as low temperature regime(75$0^{\circ}C$-85$0^{\circ}C$) intermediate regime(90$0^{\circ}C$-95$0^{\circ}C$) and high temperature regime(100$0^{\circ}C$$\leq$) In the low temperature regime the penetration speed of molten aluminum increased with increasing reaction temperature whereas it decreased in the intermediate regime due to the phase transition of alumina formed by displacement reac-tion. In the high temperature regime the penetration speed of molten aluminum was the highest at 100$0^{\circ}C$ which was 3.6 mm/hr But above 105$0^{\circ}C$ molten aluminum did not penetrate into the silica preform because of the formation of a dense spinel layer at the preform surface by magnesium in molten Al.

Study of Enzyme Immobilization on Composite of CTA and PCL Membrane for Biosensor (바이오센서용 CTA와 PCL 혼합막에의 효소고정화 기법의 개발)

  • The disposable glucose bio-sensor using composite of CTA and PCL membrane was developed for measurement of glucose. The most effective membrane was composed of CTA/PCL(80/20, w/w) and glutaraldehyde one-step immobilization method ($10{\mu}m$ thickness) for glucose sensor gave the best result among various methods, considering oxygen permeability and electronic sensitivity. A scanning electron micrograph of the cross-section of a typical asymmetric CTA/PCL composite membrane showed that the membrane fused with a dense layer covered with a GOD-glutaraldehyde. Glucose oxidase immoblilized on the membrane showed the linearity between difference of absolute amperometric values and glucose concentrations within 7mM when the GOD immobilized electrode was used. About 35% of activity was remained after 8 days when the tyrosinase was immobilized on CTA/PCL (80/20) membrane.

Lyotropic Behaviors of a Phospholipid-based Lamella Liquid Crystalline Phase Hydrated by Propylene Glycol as a Polar Solvent: Correlation of DSPC vs PG Concentration

  • The lyotropic behaviors to form the structure of distearoylphosphatidylcholine (DSPC)-based liquid crystal (LC) hydrated by only propylene glycol (PG) without water were examined by differential scanning calorimetry (DSC), X-ray diffractions (XRD), polarized microscope (PM) and transmission electron microscope (TEM). By increasing the amount of PG instead of water, it showed the phase transition to be gradually changed from anisotropic structures to other structures more close to isotropic ones and their appearance to be changed from solid-like states to liquid-like ones with more fluidity. Below 50% w/w PG, the mixtures of DSPC and PG resulted in no direct observation of LC structure through PM because they were very close to solid-states. From 55% w/w to 90% w/w of PG, the dense lamella crystalline structures were observed through PM, and their thickness and area decreased as the content of PG increased. Measured by DSC with heating process, the main phase transition from α -lamella phase to isotropic phase appeared from 52.89 °C to 47.41 °C to show linearly decreasing behaviors because PG affects the hydrophobic region of DSPC-based lamella phase. The repeating distance of the lamella phase and the interlayer distance between bilayers were calculated with XRDs and the average number of bilayers related to the thickness in LC structure was approximately estimated by combining with TEM results. The WAXS and DSC measurements showed that all of PG molecules contributed to swelling both the lipid layer in the edge region of lamella phase close to phosphate groups and the interlayer between bilayers below 90% w/w of PG. The phase and thermal behaviors were found to depend on the amount of PG used by means of dissolving DSPC as a phospholipid and rearranging its structure. Instead of water, the inducement of PG as a polar solvent in solid-lamella phase is discussed in terms of the swelling effect of PG for DSPC-based lamella membrane.

  • The paper describes a Large Eddy Simulation (LES) study of turbulent flow around a cavity. A series of three-dimensional cavities placed in a turbulent boundary layer are simulated at a Reynolds number of $1.0{\times}10^5$ by considering U and h, which represent the velocity at the top and the depth of the cavity, respectively. In order to obtain the appropriate solution for the filtered Navier-Stokes equation for incompressible flow, the computational mesh forms dense close to the wall of the cavity but relatively coarse away from the wall; this helps reduce computation cost and ensure rapid convergence. The Boussinesq hypothesis is employed in the subgrid-scale turbulence model. In order to determine the subgrid-scale turbulent viscosity, the Smagorinsky-Lilly SGS model is applied and the CFL number for time marching is set as 1.0. The results show the flow variations inside cavities of different sizes and shapes.

  • A Study on the characteristics of $Ta/Ta_2O_5/MnO_2$ capacitor applied $MnO_2$ by means of pyrolysis of manganese nitrate solution was carried out. Single phase of $MnO_2$ was obtained in the pyrolysis temperature range of 230 to $250^{\circ}C$ by TG/DSC analysis on manganese nitrate solution. Temperature of pyrolysis, concentration of manganese nitrate solution and the number of pyrolysis were selected for the basic parameters of embodying $MnO_2$ solid electrolyte and then the effects of these parameters on the characteristics of capacitor were estimated. The characteristics of capacitor pyrolyzed radiationally was superior to that of capacitor pyrolyzed convectionally on the basis of these optimized parameter conditions. It was verified that radiational pyrolysis formed smaller spherical $MnO_2$ particles than those of convectional one relatively and these facts resulted in forming uniform and dense solid electrolyte layer into the microporous sintered body of capacitor.

  • This study is observed the skin of the parrot fish, Oplegnathus fasciatus that related study of epidermal alternation with environmental and physiological change. It composed of supporting cells, unicellular glands and accessory cells. The supporting cells are classified into superficial cell, intermediated cell and basal cell. Superficial cell of epidermal layer is squamous or cuboidal and contain nucleus of ovoid type. And its free surface has many microridge which covered with glycocalyx. Intermediated cell is ovoid and has a nucleus of round shape. Basal cell is columnar, and nucleus is situated in the upper cytoplasm. Gland cells are classified into mucous cell and club cell. By the histochemical studies of the epidermal secretions the mucous materials react on blue in ABPAS (pH 2.5). Club cell is observed numerous vacuoles and microfilaments in the cytoplasm. The cytoplasm of chloride cells are occupied with numerous mitochondria. Pigment cells are classified into two type. The one contain pigment granules of electron dense, and the other contain reflecting platelets.

Study on Development in vitro of Follicular Oocytes fertilized in vitro in Korean Native Cattle (한우에 있어서 체외수정된 난포란의 배발생에 관한 연구)

  • This study was carried out to investigate the factors affecting development in vitro of follicular oocytes fertilized in vitro in Korean Native Cattle. The bovine ovaries were obtained at a slaughter house and the follicular oocytes were recovered by aspirating the follicular fluid from the visible follciles of 3~6mm. The bovine oocytes were matured in vitro for 20~24 hours in TCM0-199 containing 10% FCS and hormones (0.02AU/ml FSH, 10$\mu\textrm{g}$/ml LH, 1$\mu\textrm{g}$/ml estradiol-17$\beta$). The matured oocytes were fertilized in vitro using Percoll-separated frozen-thawed spermatozoa in BO solution containing caffeine(5mM) and heparin(10$\mu\textrm{g}$/ml). Twenty-four hours after insemination, the oocytes were cultured in vitro and then the effects of cumulus cell layer, co-culture with cumulus cells, bovine oviduct epithelial cells from ampulla or isthmus on development of ova, were studied. The results obtained are summarized as follows : 1. The in vitro development degree of oocytes attached with compact and dense layered cumulus cells was higher than that with 3~4 layered cumulus cells to be 9~16cells(P<0.01). 2. When the in vitro fertilized oocytes were co-cultured with bovine oviduct epithelial cells or cumulus cells, the development rate to be morula was 20.2% and 12.7%, respectively and the rates were higher than that of control, 2.1%(P<0.05). 3. The development rate to be morula was 15.8% and 23.8%, respectively when the in vitro fertilized oocytes were co-cultured with bovine oviduct epithelial cells from ampulla or isthmus, and the rates were higher than that of control, 0%(P<0.05%).

Preparation and Characterization of BaTiO3 Powders and Thin films (티탄산바륨 분말과 박막의 제조 및 특성 연구)

  • The $BaTiO_3$ powders and thin films were prepared by an alkoxide modified sol-gel process (polymerization-complex route) using ethylene glycol. The stable starting (Ba-Ti)-mixed metal organic sol was made by addition of acetylacetone. The $BaTiO_3$ powders, which had a particle size of 40~77 nm, were crystallized from an amorphous to a tetragonal phase on annealing at 700 and $1100^{\circ}C$ for 1 h. From FT-IR, solid-state $^{13}C$ CP/MAS NMR spectroscopy and X-ray diffractometry, the trace of the Ba-Ti-oxycarbonate phase first appeared at $400^{\circ}C$. Hydrolyzed sol was spin coated on a quartz wafer at 3500 rpm for 60 s and pyrolyzed at $1100^{\circ}C$ for 1 h. After heat treatment, the coated layer became dense and smooth.

Ultrastructure of Compound Starch Granules and Protein Bodies of Starchy Endosperm Cell in Rice (쌀 배유세포 전분복합체와 단백질체의 미세구조)

  • The ultrastructure of the compound starch granules and the protein bodies of Odaebyeo rice of early matured variety were examined by light microscope and electron microscope. The endosperm cell appealed rectangular or octangular shape on the cross section. The thickness of cell wall containing of membraneous materials was about $0.5\;{\mu}m$ in diameter. The starch cell was filled compactly with globular or oval shaped compound starch granules with the size of $20{\sim}25\;{\mu}m$ in diameter. The compound starch granules were consisted of central core starch granule and concentrical $2{\sim}3$ layers of starch granules. The average thickness of the starch granules were about $5\;{\mu}m$. Most protein bodies were found in the aleurone layer The globular protin bodies were scattered near the compound starch granules and $2.5{\sim}3\;{\mu}m$ in diameter. The protein bodies composed of central electron dense materials and peripheral electron loose materials in limiting membrane.
