Evaluation of Denitrification Efficiency and Functional Gene Change According to Carbon(Fumarate) Concentration and Addition of Nitrate Contaminated-soil in Batch System (회분식반응조 실험을 통한 탄소원(Fumarate) 주입조건에 따른 지하수 중 탈질율 및 탈질 관련 기능성 유전자 분석)

  • Nitrate is on the most seriou pollutant encountered in shallow groundwater aquifer in agricultural area. There are various remediation technologies such as ion exchange, reverse osmosis, and biological denitrification to recover from nitrate contamination. Biological denitrification by indigenous microorganism of the technologies has been reviewed and applied on nitrate contaminated groundwater. In this work, we selected the site where the annual nitrate (NO3) concentration is over 105 mg/L and evaluated denitrification process with sampled soil and groundwater from 3 monitoring wells (MW4, 5, 6). In the results, the nitrate degradation rate in each well (MW 4, 5, and 6) was 25 NO3 mg/L/day, 6 NO3 mg/L/day, and 3.4 NO3 mg/L/day, respectively. Nitrate degradation rate was higher in batch system treated with 2 times higher fumarate as carbon source than control batch system (0.42M fumrate/1M NO3), comparing with batch system with soil sample. This result indicates that increase of carbon source is more efficient to enhance denitrification rate than addition of soil sample to increase microbial dynamics. In this work, we also confirmed that monitoring method of functional genes (nirK and nosZ) involved in denitrification process can be applied to evaluated denitrifcation process possibility before application of field process such as in-situ denitrification by push-pull test.

Development of Biological Filtration Process for Effective Nitrogen Removal in Tertiary Treatment of Sewage (생물막 여과반응기를 이용한 고도질소 제거법의 개발)

  • The treatment performance and operational parameters of a tertiary wastewater treatment process a biological filtration system were investigated. The biological filtration system consisted of a nitrification filter (Fiter 1) and a polishing filter with anoxic and aerobic parts (Filter 2). SS, T-C-BOD, and T-N in effluent were kept stable at less than 3, 5 mg/L, and 5 mgN/L, respectively, under a HRT in Filter (filter-bed) of 0.37~2.3 h. T-N at the outlet of Filter 2 were about 1~5 mgN/L under the condition of LV of 50~202 m/d. In Filter 2, denitrification was accomplished under LV of 50~168 m/d in a 1 m filter-bed. However, the denitrification capacity reached the maximum when the linear velocity was increased to 202 m/d. Relationship between increase in microorganism and headloss was clearer in Filter 2. As a result, the denitrification rate increased from 1.0~2.3 kgN/($m^3-filter-bed{\cdot}d$) as the headloss increased. The COD removal rate was 6.0~9.6 kgCOD/($m^3-filter-bed{\cdot}d$) when operated with Filters 1 and 2. These results mean that captured bacteria contributed a part of COD consumption and denitrification. The maximum nitrification and denitrification rate was 0.5 and 4 kgN/($m^3-filter-bed{\cdot}d$) in Filter 1 and 2.The ratio of backwashing water to the treated water was about 5~10 %. In Filter 1, wasted sludge in backwashing was only 0.7~5.3 gSS/($m^3$-treated water). In Filter 2, added methanol was converted into sludge and its value was 8.0~24 gSS/($m^3$-treated water). These results proved that this process is both convenient to install as tertiary treatment and cost effective to build and operate.

Effect of Organic Acids Fermented from the Settled Sludge and Animal Organic Wastes on the Denitrification (침전 슬러지와 가축분의 유기산화 및 발효유기산이 탈질반응에 미치는 영향)

  • Fermentation efficiencies of organic wastes from the variety of sources were evaluated based on the production of total volatile acids(TVA) in batch reactor. Mixing and pH were not significant factors in producing TVA from the organic wastes. After a 10-day fermentation, final TVA concentrations in piggery, cattle, poultry, and primary settled sludge of domestic wastewater were 8,900, 2,900, 7,370 and 1,630 mg/L, respectively. The pH of organic wastes was decreased from neutral to 5.7. The ratio of TVA to $NH_4{^+}-N$ produced from the animal waste ranged from 11.5 to 30.1, whereas, that in the primary settled sludge of domestic wastewater, was 5.4. Possibility of fermented organic wastes as the electron donors for denitrification in the activated sludge was investigated. In both acclimated and nonacclimated activated sludge, higher denitrification rates were obtained with fermented piggery sludge added than with either methanol or acetate added. The fermented organic acids derived from the primary settled sludge gave the higher denitrification rate ($4.2mg\;NO_3-N/g\;vss{\cdot}hr$) in the acclimated activated sludge. Denitrification rate was $1.5mg\;NO_3-N/g\;vss{\cdot}hr$ in the nonacclimated sludge with the fermented acids from the primary settled sludge of domestic wastewater added.

The Nutrient Removal of Mixed Wastewater composed of Sewage and Stable Wastewater using SBR (SBR을 이용한 하수와 우사폐수로 구성된 혼합폐수의 영양소 제거)

  • This study was carried out to obtain the optimal operating parameter on organic matters and nutrient removal of mixed wastewater which was composed of sewage and stable wastewater using SBR. A laboratory scale SBR was operated with An/Ae(Anaerobic/Aerobic) ratio of 3/3, 2/4 and 4/2(3.5/2.5) at organic loading rate of 0.14 to 0.27 kgBOD/$m^3$/d. TCOD/SCOD ratio of mixed wastewater was 3, so the important operating factor depended upon the resolving the particulate parts of wastewater. Conclusions of this study were as follows: 1) For mixed wastewater, BOD and COD removal efficiencies were 93-96% and 85-89%, respectively. It was not related to each organic loading rate, whereas depended on An/Ae ratio. During Anarobic period, the amount of SCOD consumption was very little, because ICOD in influent was converted to SCOD by hydrolysis of insoluble matter. 2) T-N removal efficiencies of mixed wastewater were 55-62% for Exp. 1, 66-76% for Exp. 2, and 67-81% for Exp. 3, respectively. It was found that nitrification rate was increased according to organic concentration in influent increased. Therefore, the nitrification rate seemed to be achieved by heterotrophs. During anoxic period, denitrification rate depended on SCOD concentration in aerobic period and thus, was not resulted by endogenous denitrification. However, the amount of denitrification during anaerobic period were 3.5-14.1 mg/cycle, and that of BOD consumed were 10-40 mg/cycle. 3) For P removal of mixed wastewater, EBPR appeared only Mode 3($3^*$). It was found that the time in which ICOD was converted to VFA should be sufficient. For mode 3 in each Exp., P removal efficiencies were 74, 87, and 81%, respectively. But for 45-48 of COD/TP ratio in influent, P concentration in effluent was over 1 mg/L. It was caused to a large amount of ICOD in influent. However, as P concnetration in influent was increased, the amounts of P release and uptake were increased linearly.

Change of Sludge Denitrification and Nitrification Rate according to the Operating Conditions in Advanced Wastewater Treatment Processes (하수고도처리공법의 유입하수량 변화에 따른 슬러지 질산화/탈질속도 변화)

  • The purpose of this study is to investigate the changes of sludge characteristics according to the changes of influent sewage flowrate in the advanced wastewater treatment processes including MBR, SBR, and $A_2O$. The ratio of the actual sewage flowrate to the design flowrate is decreased from 100% to 70, 40%, and 10%, and the specific denitrification rate and ammonia oxidation (nitrification) rate was measured. The specific nitrification rate of the sludge collected from the aeration tank of each process was measured at a similar value ($0.10gNH_4/gMLVSS/day$) in all three process under the condition of 100% of sewage flowrate. It has tended to decrease significantly as the sewage flowrate decreased from 70% to 40%. The specific denitrification rate was also decreased by ~50% as the sewage flowrate decreased. However, considering the total nitrogen concentration in the influent and the microbial concentration in the reactor, the changes in kinetic parameter did not affect overall nitrogen removal. Therefore, it can be concluded that stable nitrogen removal will be possible under low influent flowrate condition if the MLVSS concentration is kept high.

Isolation and Nitrogen Removal Characteristics of Heterotrophic Nitrification-Aerobic Denitrifying Bacteria, Stenotrophomonas sp. CW-4Y (종속영양 질산화- 호기적탈질 세균 Stenotrophomonas sp. CW-4Y의 분리와 질소제거 특성)

  • CW-4Y was identified as Stenotrophomonas sp. by morphological and physiological characteristics, and phylogenetic analysis of its 16S rDNA gene sequence. Nitrogen removal by CW-4Y was analyzed in relation to the ammonium concentration, presence of organic carbon, carbon source, and carbon-to-nitrogen ratio (C/N). Stenotrophomonas CW-4Y has heterotrophic nitrification and aerobic denitrification abilities. Stenotrophomonas CW-4Y utilized only glucose as carbon sources, and heterotrophic nitrification and aerobic denitrification were observed regardless of the type of nitrogen source. The maximum ammonium removal rate of CW-4Y was 80 $mg-N{\cdot}L^{-1}{\cdot}d^{-1}$ and its denitrification rate of 192 $mg-N{\cdot}L^{-1}{\cdot}d^{-1}$ at $NO_3{^-}-N$ (about 280 ppm) in shake culture experiments at a C/N ratio of about 15 was about 30 times higher than those of other bacteria with the same ability.

Study of Factors Influenced on denitrification in wastewater treatment (폐수처리 탈질 공정에 미치는 인자 연구)

  • In this study, the effects of several factors such as initial nitrate concentration, C/N ratio, biomass amount and external carbon source on denitrification process were investigated using synthetic wastewater and sludge obtained from wastewater treatment facility. As a result, the condition of lower initial nitrate concentration was increased to the removal rate of nitrate than that of high concentration. The increases of C/N ratio and initial biomass amount were linearly enhanced the removal rate. The use of ethanol as external carbon source was shown the highest removal yield than that of others.

Seasonal Variation of Nitrogen Loads and Nitrogen Cycling at Tidal Flat Sediments in Nakdong River Estuary (낙동강 하구 갯벌 퇴적물에서 강을 통한 질산염 유입에 따른 질소순환의 계절 변화)

  • We investigated seasonal variation of sediment-water oxygen and inorganic nitrogen fluxes, and denitrification at tidal flat sediments located in the Nakdong River Estuary from July 2005 to September 2006. Net oxygen fluxes, measured with sediment incubations at in situ temperature, varied from -37.0 to $0.5mmol\;O_2\;m^{-2}\;d^{-1}$. Oxygen fluxes into the sediments from the overlying water increased due to the increased water temperature. Denitrification rate ($4{\sim}2732{\mu}mol\;N\;m^{-2}\;d^{-1}$) in this study was higher compared to the other Korean coast measured with the same method. Denitrification showed the same seasonal variation as oxygen fluxes. Denitrification rate based on $^{15}N$-nitrate showed a strong correlation with nitrate flux into the sediments from the overlying water. Denitrification via "water column supplied nitrate ($D_w$)", calculated from Isotope pairing technique, also correlated well with nitrate flux into the sediments. Nitrate from water column seems to account for seasonal variation of denitrification in Nakdong River Estuary. To understand general patterns and trends of biogeochemical processes of sediments in the Nakdong River Estuary, we categorized biogeochemical fluxes measured in this study according to direction and sizes of fluxes. Type 1(high oxygen and inorganic nitrogen fluxes into the sediments and high denitrification) occurred in summer, whereas Type 2(low oxygen and inorganic nitrogen fluxes into the sediments and low denitrification) occurred in rest of the season. Intertidal flat sediments seem to react sensitively to influence of freshwater from the Nakdong River.

A study on the denitrification and microbial community characteristics by the change of C/N ratio of molasses and nitrate nitrogen (당밀과 질산성 질소의 C/N ratio 변화에 따른 탈질 및 미생물 군집 특성에 관한 연구)

  • To compare the denitrification efficiency, this study used molasses and methanol were used as external carbon sources. Specific experimental conditions were classified according to C/N ratio conditions. The batch test showed that the denitrification efficiency increased as C/N ratios of molasses and methanol rose. The most suitable C/N ratio of molasses turned out 4:1 considering the concentration of the residue chemical oxygen demand (COD) and the denitrification efficiency, which was 91.4%. Specific denitrification rate (SDNR) drawn as a kinetic factor demonstrated that molasses and methanol showed similar SDNR values as C/N ratios of molasses and methanol increased. Under the condition of C/N ratio 4:1, 0.0292 g $NO_3{^-}-N$ removal/g mixed liquor volatile suspended solid (MLVSS)/day (molasses), 0.0299 g $NO_3{^-}-N$ removal/g MLVSS/day (methanol) were found. Sludge adapted to molasses showed that Bacterium Pseudomonas sp. and Bergeylla sp. dominated through an analysis of microbial community. In addition, some bacteria were high convergences than the variety of microbial community. Accordingly, it was assumed that molasses focus on growing microorganisms specialized in denitrification and applied as a replaceable external carbon source that can enhance denitrification performance.

Acceleration of Biological Denitrification by Using Bioelectrochemical Reactor (생물전기화학반응기를 이용한 생물학적 탈질반응의 촉진)

  • Nitrate contamination of water environments can create serious problems such as eutrophication of rivers. Conventional biological processes for nitrate removal by heterotrophic denitrification often need additional organic substrates as carbon sources and electron donors. We tried to accelerate biological denitrification by using bioelectrochemical reactor (BER) in which electrode works as an electron donor. Denitrification activity of 8 environmental samples from various sediments, soils, groundwaters, and sludges were tested to establish an efficient enrichment culture for BER. The established enrichment culture from a soil sample showed stable denitrification activity without any nitrite accumulation. Microbial community analysis by using PCR-DGGE method revealed that dominant denitrifiers in the enrichment culture were Pantoea sp., Cronobacter sakazakii, and Castellaniella defragrans. Denitrification rate ($0.08kg/m^3{\cdot}day$) of the enrichment culture in BER with electrode poised at -0.5 V (vs Ag/AgCl) was higher than that ($2.1{\times}10^{-2}kg/m^3{\cdot}day$) of BER without any poised potential. This results suggested that biological denitrification would be improved by supplying potential throughout electrode in BER. Further research using BER without any organic substrate addition is needed to apply this system for bioremediation of water and wastewater contaminated by nitrate.
