본 연구에서는 퇴적물에서 일어나는 유기물 분해과정 중 이용된 전자 수용체와 분해과정에서 생산된 환원 물질의 농도 변화를 적절히 파악하고 정량화하기 위한 수치모델을 개발하였다. 퇴적물에 서식하는 미생물이 전자 수용체를 이용하여 유기물을 분해하는 반응들은 전자 수용체의 종류에 따라서 다음과 같이 6가지 형태로 구분 할 수 있다: (1) 호기성 분해(Aerobic Respiration), (2) 탈질(Denitrification), (3) 망간 환원(Manganese Reduction), (4) 철 환원(Iron Reduction), (5) 황산염 환원(Sulfate Reduction), (6) 메탄 환원(Methanogenesis). 이와 같은 6가지 반응은 양론(Stoichiometry)적으로 표현되어지며, 여기에 관여하는 3개의 고형물질(침전성 유기물질, manganese oxides, iron oxides)과 8개의 용존물질 (oxygen, nitrate, sulfate, ammonia, dissolved manganese, dissolved iron, sulfide, methane)의 움직임은 1차원 물질수지 방정식으로 모델에서 재현되어졌다. 퇴적물에서 미생물에 의한 유기물 분해 반응은 Monod 반응식을 이용하여 간단히 표현되어졌다. 퇴적물에 포함된 물질들에 대한 물질수지 방정식들은 Monod 반응식에 포함된 비선형성을 제거하기 위해서 다단계의 반복적인 수치해석법에 의해 안정적인 해를 구할 수 있었다. 모델의 타당성을 검토하기 위하여 Sweert et al.(1991a)이 Netherland의 Veluwe 호수의 퇴적물에서 조사한 자료에 모델결과를 비교하였고, 결과로써 산출된 전자 수용체와 환원 물질의 수직적 분포형태는 관측간과 비교적 잘 부합하였다. 그러나 모델을 통하여 예측된 $NH_4^+$의 농도는 측정된 농도보다 훨씬 낮은 것이 관찰이 되었는데, 이는 모델에서 유기물질을 표현할 때 사용한 Redfield의 유기물식이 본 연구에 적용된 퇴적물에서의 높은 질소 함유율을 적절히 표현하지 못한 결과로 해석되어 질 수 있다. 퇴적물 깊이에 따른 전자 수용체와 환원된 물질의 분포변화는 중금속의 재용출과 생물이용도를 조절하는 주요인이 되기 때문에, 이 연구에서 개발된 수치모델은 퇴적물에서 일어나는 미량 오염 물질의 거동을 파악하기 위해 유용하게 사용되어질 수 있을 것으로 사료된다.
유기물과 질소 함량이 높은 하천수가 대하천에 유입되기전 소하천의 잡초가 자라는 홍수터에 살포하여 사질 토층을 수직 이동하는 동안 잡초의 근권에서 유기물의 분해와 함께 탈질에 의해 질소가 제거되도록 하는 홍수터 여과공법을 개발하였다. 직경 15cm, 길이 150cm의 PVC pipe에 실제 홍수터에서 채취한 사질의 토양을 충진하여 제작된 홍수터 모형에 하천수를 27.2, 40.8, $68.0\;ml/day/m_2$의 유량으로 연속적으로 살포하고 정상상태에 이른 후 토양 깊이별로 토양 용액을 채취하여 유기물과 $NO_3-N$을 비롯한 무기질소의 이동과 제거 현상을 조사하였고 토양 기체를 채취하여 $N_2$와 $N_20$의 발생 현상을 측정하였다. 포화상태에 가까운 수준으로 유량을 조절할 경우 하천수에 포함된 유기물만을 이용하더라도 매우 효과적인 탈질 환경이 5cm깊이 부근의 표층 토양에서부터 형성되었으며 유기물의 제거와 함께 질소도 효과적으로 제거할 수 있는 것으로 나타났다. 90cm깊이의 홍수터 토양을 통과하는 동안 평균적으로 COD는 18.7에서 5mg/l로 무기질소함량은 2.7에서 0.4mg/l로 정화되었다. 탈질 기체는 대부분 $N_2$ 형태로 발생되었으며 온실효과와 오존층 파괴를 유발하는 $N_2O$ 발생량은 매우 적었다. 표층 토양에 잡초의 근권이 형성되어 있는 실제 홍수터에 이와 같은 기법을 적용할 경우 모형 실험에서 나타난 결과보다 더욱 활발한 탈질 현상이 유발될 수 있을 것으로 판단된다. 이러한 홍수터 여과는 부지가 따로 필요하지 않으므로 시설 및 운영비가 경쟁기술에 비해 싸고, 화학약품 처리나 슬러지 발생이 없는 환경친화적인 하천수 처리방법이 될 것으로 기대되며, 하천수 외에도 도시하수나 산업폐수의 3차 처리에도 응용되어 하폐수의 재활용을 통한 수자원의 절약과 하천수량의 증대에도 기여할 수 있을 것이다.
지속 가능하고 친환경적인 농업 관행을 추구하기 위해 우리는 40년이 넘는 장기간 동안 화학 비료를 사용하지 않은 토양에 서식하는 근권 박테리아에 대한 광범위한 연구를 수행하였다. 이번 조사를 통해 식물생장촉진 근권박테리아 총 80종을 분리하고 이들의 식물생장 증진 가능성을 평가했다. 이러한 분리균중에서 Burkholderia cepacia CD2는 가장 우수한 식물 성장촉진 활성과 생장능을 나타내어 추가 분석을 위한 최적의 후보균주로 선정되었다. Burkholderia cepacia CD2는 인 가용화 능력, 사이드로포어 생산, 탈질화 능력, 아질산 이온 활용능력 및 요소분해효소 활성을 포함하여 식물 성장에 도움이 되는 다양한 유익한 특성을 나타내었다. 이러한 특성은 식물의 성장과 발달에 긍정적인 영향을 미치는 것으로 잘 알려져 있다. 균주의 분류학적 분류를 검증하기 위해 16S rRNA 유전자 서열분석을 통해 Burkholderia 속 내 위치를 확인하여 계통발생 관계에 대한 추가 통찰력을 제공하였다. 식물 생장 촉진 특성의 기본 메커니즘을 더 깊이 조사하기 위해 우리는 CD2에서 식물 생장촉진과 관련된 특정 유전자의 존재를 확인하려고 하였다. 이를 달성하기 위해 옥스포드 나노포어를 활용하여 전장 유전체 시퀀싱을 수행하였다. CD2 게놈에 대한 전장유전체 분석을 통해 식물 생장 개선에 중추적 요인으로 생각되는 하위 시스템 기능을 확인하였다. 이러한 발견을 바탕으로 Burkholderia cepacia CD2는 미생물 비료로 작용하여 화학 비료에 대한 지속 가능한 대안을 제공할 수 있는 잠재력을 가지고 있다는 결론을 내릴수 있다.
Cold water paddy field soils are relatively unproductive, but can be ameliorated by supplementing with inorganic fertilizer from animal waste-based composts. The yield of two rice cultivars was significantly raised by providing either chicken manure or cow dung-based compost. The application of these composts raised the soil pH as well as both the total nitrogen and ammonium nitrogen content, which improved the soil's fertility and raised its nitrification potential. The composts had a measurable effect on the abundance of nitrogencycling-related soil microbes, as measured by estimating the copy number of various bacterial and archaeal genes using quantitative real-time PCR. The abundance of ammonia oxidizing archaea and bacteria was markedly encouraged by the application of chicken manure-based compost. Supplementation with the composts helped promote the availability of soil nitrogen in the cold water paddy field, thereby improving the soil's productivity and increasing the yield of the rice crop.
Nitrate ($NO_3^-$), a common surface water and groundwater pollutant, poses a serious environmental problem in regions with intensive agricultural activities and dense population. It is thus important to identify the source of nitrate contamination to better manage water quality. Due to the distinct isotope compositions of nitrate among different origins, the dual isotope analysis (${\delta}^{15}N$ and ${\delta}^{18}O$) of nitrate has been widely applied to track contamination sources. This paper provided the underlying backgrounds in the isotope analysis of nitrate, which included typical ranges of ${\delta}^{15}N$ and ${\delta}^{18}O$ from various nitrate sources, isotope fractionation, the analytical methods used to concentrate nitrate from samples, and the potential limitations of the dual isotope analysis along with the resolutions. To enhance the applicability of the dual isotope analysis as well as increase the ability to interpret field data, this paper also introduced several case studies. Furthermore, other environmental tracers including ${\delta}^{11}B$ and $Cl^-/Br^-$ ratios were discussed to accompany the dual isotope analysis for better assignment of contamination sources even when microbial transformation of nitrate and/or mixing between contaminant plumes occur.

Severe loss or hydrogen occurred in most anaerobic hydrogen fermentation reactors. Several selected methods were applied to suppress the consumption of hydrogen and increase the potential of production. As the first trial, pH shock was applied. The pH of reactor was dropped nearly to 3.0 by stopping alkalinity supply and on]y feeding glucose (5 g/L-d). As the pH was increase to $4.8{\pm}0.2,$ the degradation pathway was derived to solventogenesis resulting in disappearance of hydrogen in the headspace. In the aspect of bacterial community, methanogens weren't detected after 22 and 35 day, respectively. Even though, however, there was no methanogenic bacterium detected with fluorescence in-situ hybridization (FISH) method, hydrogen loss still occurred in the reactor showing a continuous increase of acetate when the pH was increased to $5.5{\pm}0.2$. This result was suggesting the possibility of the survival of spore fanning acetogenic bacteria enduring the severely acidic pH. As an alternative and additive method, nitrate was added in a batch experiment. It resulted in the increase of maximum hydrogen fraction from 29 (blank) to 61 % $(500\;mg\;NO_3/L)$. However, unfortunately, the loss of hydrogen occurred right after the depletion of nitrate by denitrification. In order to prevent the loss entangled with acetate formation, $CO_2$ scavenging in the headspace was applied to the hydrogen fermentation with heat-treated sludge since it was the primer of acetogenesis. As the $CO_2$ scavenging was applied, the maximum fraction of hydrogen was enhanced from 68 % to 87 %. And the loss of hydrogen could be protected effectively.
A continuous feed and cyclic draw SBR process was developed to overcome flow rate fluctuation and to maximize organic matters utilization efficiency for nitrogen and phosphorus removal. The developed SBR process was operated with two parallel reactors. Influent was supplied to one reactor which was not obligately aerated. At the same time, the other reactor was just aerated without supplying influent. In addition this mode was changed periodically. Cycle time was 6hr and aeration time ratio($t_{aer}/t_{total}$) was 0.33, respectively. $COD_{cr}$ and SS removal efficiencies of 95% or higher were achieved. Nitrogen removal was so greatly influenced by influent $COD_{cr}/T-N$ ratio. At influent $COD_{cr}/T-N$ ratio of 5.7, removal efficiencies of ammonia-N, T-N and T-P were 96%, 78% and 55%, respectively. Influent $COD_{cr}/T-N$ of 4 or higher ratio was necessary to achieve 60% or higher nitrogen removal. Organic matters of influent was efficiently utilized in denitrification reaction and consumed COD has a good correlation with removed T-N(about 6.5 mgCOD/mgTN). Continuous feed and cyclic draw SBR process could be one of alternative processes for the removal of nutrients in rural area where $COD_{cr}/T-N$ ratio was low and fluctuation of flow rate was severe.
In order to remove both nitrate and sulfate present in the concentrate of RO(reverse osmosis) process, a combined bio-regeneration and ion-exchange(IX) system was studied. For this purpose, both denitrifying bacteria(DNB) and sulfate reducing bacteria(SRB) were simultaneously cultivated in a bio-reactor under anaerobic conditions. When the IX column containing a nitrate-selective A520E resin was fully exhausted by nitrate and sulfate, the IX column was bio-regenerated by pumping the supernatant of the bio-reactor, which contains MLSS concentration of $125{\pm}25mg/L$, at the flowrate of 360 BV/hr. Even though the nitrate-selective A520E resin was used, the breakthrough curves of ionic species showed that sulfate was exhausted earlier than nitrate. The reason for this result is due to the fact that the concentration of sulfate in RO concentrate was 36 to 48 times higher than nitrate. The bio-reactor was successfully operated at a volumetric loading rate of 0.6 g $COD/l{\cdot}d$, nitrate-N loading rate of 0.13 g $NO_3{^-}-N/l{\cdot}d$, and sulfate loading rate of 0.08 g $SO_4{^{2-}}/l{\cdot}d$. The removal rate of SCOD, nitrate-N, sulfate was 90, 100, and 85%, respectively. When the virgin resin was fully exhausted and consecutively bio-regenerated for 2 days, 81% of nitrate and 93% of sulfate were reduced. When the virgin resin was repeatedly used up to 4 cycles of service and bio-regeneration, the ion-exchange capacity of bio-regenerated resin decreased to 95, 91, 88, and 81% of virgin resin.
This study investigated the effect of a co-culture of Scenedesmus dimorphus and nitrifiers using artificial wastewater on the removal of ammonium, nitrate and phosphate in the advanced treatment. To test the synergistic effect of the co-culture, we compared the co-culture treatment with the cultures using S. dimorphus-only and nitrifiers-only treatment as controls. After 6 days of incubation, nitrate was removed only in the co-culture treatment and total amount of N removal was 1.3 times and 1.6 times higher in the co-culture treatment compared to those in the S. dimorphus- and nitrifiers-only treatments, respectively. In case of total amount of P, co-culture treatment removed 1.2 times and 12 times more P than the S. dimorphus -and nitrifiers-only conditions, respectively. This indicates that the co-culture improved removal rates for ammonium, nitrate, and phosphate. This further implies that there was no need for denitrification of nitrate and luxury uptake of P processes because nitrate and phosphate can be removed from the uptake by S. dimorphus. In addition, co-culture condition maintained high DO above 7 mg/L without artificial aeration, which is enough for nitrification, implying that co-culture has a potential to decrease or remove aeration cost in the wastewater treatment plants.
최근 국내에서 인공습지는 하 폐수처리장 방류수의 후처리, 비점오염원 관리, 생태하천 복원 등 다양한 용도로 적용되고 있다. 그러나 인공습지가 설치되는 지역의 기후, 유입수의 특성, 지역적 특성 등에 따라 매우 영향을 많이 받기 때문에 인공습지 내에서의 오염물질의 제거기작은 확실하게 이해되고 있지는 않다. 그러므로, 이 연구에서는 인공습지 내에서 질소, 인을 중심으로 한 오염물질의 제거기작을 중심으로 살펴보았다. 그 결과, 질소 제거의 주된 기작은 인공습지 내의 근권(rhizosphere)에서의 질산화/탈질에 의한 것으로 판단되며, 인 제거의 주된 기 작은 인공습지내 여재 등의 기층(substrate)에서의 흡착에 의한 것으로 판단된다. 그러나, 인공습지내의 인의 거동은 산화/환원 전위(ORP), 흡착/탈착, 미생물의 작용 등에 따라 다양한 형태를 가질 수 있는 것으로 판단된다.
