On Identifying Operational Risk Factors and Establishing ALARP-Based Mitigation Measures using the Systems Engineering Process for Parcel Storage Devices Utilizing Active Loading Technology

  • Due to the steady growth of the online shopping market and contact-free consumption, the volume of parcels in South Korea continues to increase. However, there is a lack of manpower for delivery workers to handle the growing parcel volume, leading to frequent accidents related to delivery work. As a result, the government and local authorities strive to enhance last-mile logistics efficiency. As one of these measures, unmanned parcel storage lockers are installed and utilized to handle last-mile deliveries. However, the existing parcel storage involves the inconvenience of couriers having to put each parcel in each locker, and this is somewhat insufficient to relieve the workload of delivery workers. In this study, we propose parcel storage devices that use active loading technology to minimize the workload of delivery workers, extract operation risk factors to apply this system to actual sites, and establish risk reduction methods based on the ALARP concept. Through this study, we have laid the groundwork for improving the safety of the system by identifying and proposing mitigation measures for the risk factors associated with the proposed parcel storage devices utilizing active loading technology. When applied in practical settings in the future, this foundation will contribute to the development of a more efficient and secure system. By applying the ALARP concept, a systems engineering technique used in this research, to the development and maintenance of storage devices leveraging active loading technology, it is thought to make the development process more systematic and structured. Furthermore, through the risk management of the proposed system, it is anticipated that a systematic approach to quality management can be employed to minimize defects and provide a stable system. This is expected to be more useful than the existing unmanned parcel storage devices.

Multivesicular Liposomes for Oral Delivery of Recombinant Human Epidermal Growth Factor

  • The purpose of the present study was to prepare multivesicular liposomes with a high drug loading capacity and to investigate its potential applicability in the oral delivery of a peptide, human epidermal growth factor (rhEGF). The multivesicular liposomes containing rhEGF was prepared by a two-step water-in-oil-in-water double emulsification process. The loading efficiency was increased as rhEGF concentration increased from 1 to 5mg/mL, reaching approximately $60\%$ at 5 mg/mL. Approximately $47\%$ and $35\%$ of rhEGF was released from the multivesicular liposomes within 6 h in simulated intra-gastric fluid (pH 1.2) and intra-intestinal fluid (pH 7.4), respectively. rhEGF-loaded multivesicular liposomes markedly suppressed the enzymatic degradation of the peptide in an incubation with the Caco-2 cell homogenate. However, the transport of rhEGF from the multivesicular liposomes to the basolateral side of Caco­2 cells was two times lower than that of the rhEGF in aqueous solution. The gastric ulcer healing effect of rhEGF-loaded multivesicular liposomes was significantly enhanced compared with that of rhEGF in aqueous solution; the healing effect of the liposomes was comparable to that of the cimetidine in rats. Collectively, these results indicate that rhEGF-loaded multivesicular liposomes may be used as a new strategy for the development of an oral delivery system in the treatment of peptic ulcer diseases.

  • As a special topic of the vehicle routing problems (VRP), VRPSDP extends the vehicle routing problem as considering simultaneous pickup and delivery for goods. The past studies have mainly dealt with a only weight constraint of a loading capacity for heterogeneous products. However. this study suggests VRPSDP considering separate loading area according to characteristics of loading species. The objective is to design a set of minimum distance routes for the vehicle routing assignment with independent capacity for heterogeneous species. And then we present a another HVRPSDP model which is easy to utilizes in a unique circumstance that is a guarantee of executing a task simultaneously from the various areas under restricted time and raising an application of vehicles that returns at the depot for the next mission like the military group. The optimal results of the suggested mathematical models are solved by the ILOG CPLEX software ver. 12.4 that is provided by IBM company.

Drug Targeting to Lungs by Way of Microspheres

  • In many conventional drug delivery systems in vogue, failure to deliver efficient drug delivery at the target site/organs; is evident as a result, less efficacious pharmacological response is elicited. Microspheres can be derived a remedial measure which can improve site-specific drug delivery to a considerable extent. As an application, Lung-targeting Ofloxacin-loaded gelatin microspheres (GLOME) were prepared by water in oil emulsion method. The Central Composite Design (CCD) was used to optimize the process of preparation, the appearance and size distribution were examined by scanning electron microscopy, the aspects such as in vitro release characteristics, stability, drug loading, loading efficiency, pharmacokinetics and tissue distribution in albino mice were studied. The experimental results showed that the microspheres in the range of $0.32-22\;{\mu}m$. The drug loading and loading efficiency were 61.05 and 91.55% respectively. The in vitro release profile of the microspheres matched the korsmeyer’s peppas release pattern, and release at 1h was 42%, while for the original drug, ofloxacin under the same conditions 90.02% released in the first half an hour. After i.v. administration (15 min), the drug concentration of microspheres group in lung in albino mice was $1048\;{\mu}g/g$, while that of controlled group was $6.77\;{\mu}g/g$. GLOME found to release the drug to a maximum extent in the target tissue, lungs.

Controlled Transdermal Delivery of Loxoprofen from an Ethylene-Vinyl Acetate Matrix

  • Repeated oral administration of loxoprofen can induce many side effects such as gastric disturbances and acidosis. Therefore, we considered alternative routes of administration for loxoprofen to avoid such adverse effects. The aim of this study was to develop an ethylene-vinyl acetate (EVA) matrix system containing a permeation enhancer for enhanced transdermal delivery of loxoprofen. The EVA matrix containing loxoprofen was fabricated and the effects of drug concentration, temperature, enhancer and plasticizer on drug release were studied from the loxoprofen-EVA matrix. The solubility of loxoprofen was highest at 40% (v/v) PEG 400. The release rate of drug from drug-EVA matrix increased with increased loading dose and temperature. The release rate was proportional to the square root of loading dose. The activation energy (Ea), which was measured from the slope of log P versus 1000/T, was 5.67 kcal/mol for a 2.0% loaded drug dose from the EVA matrix. Among the plasticizer used, diethyl phthalate showed the highest release rate of loxoprofen. Among the enhancers used, polyoxyethylene 2-oleyl ether showed the greatest enhancing effect. In conclusion, for the enhanced controlled transdermal delivery of loxoprofen, the application of the EVA matrix containing plasticizer and penetration enhancer could be useful in the development of a controlled drug delivery system.

Study on Delivery of Military Drones and Transport UGVs with Time Constraints Using Hybrid Genetic Algorithms (하이브리드 유전 알고리즘을 이용한 시간제약이 있는 군수 드론 및 수송 UGV 혼합배송 문제 연구)

  • This paper studies the method of delivering munitions using both drones and UGVs that are developing along with the 4th Industrial Revolution. While drones are more mobile than UGVs, their loading capacity is small, and UGVs have relatively less mobility than drones, but their loading capacity is better. Therefore, by simultaneously operating these two delivery means, each other's shortcomings may be compensated. In addition, on actual battlefields, time constraints are an important factor in delivering munitions. Therefore, assuming an actual battlefield environment with a time limit, we establish delivery routes that minimize delivery time by operating both drones and UGVs with different capacities and speeds. If the delivery is not completed within the time limit, penalties are imposed. We devised the hybrid genetic algorithm to find solutions to the proposed model, and as results of the experiment, we showed the algorithm we presented solved the actual size problems in a short time.

The effect of embryo catheter loading technique on the live birth rate

  • Objective: Embryo loading (EL) is a major step in embryo transfer (ET) and affect on the success of in vitro fertilization (IVF). This study aimed to compare the effect of two different EL techniques on the rates of pregnancy and delivery in IVF/ET cycles. Methods: 207 fresh ET and 194 Frozen-thawed ET (FET) cycles were included in this retrospective study. Two groups (A and B) were defined based on the EL technique used. In group A, the entire catheter was flushed with Ham's F-10 medium. The embryos were then drawn into the catheter using one air bracket. In group B, $70{\mu}L$ of air was aspirated into the syringe and the catheter was flushed using Ham's F10 medium. The medium, air, embryos, air, and finally another layer of medium were then sequentially drawn into the catheter. The main outcome measures were the pregnancy and delivery rates. Results: The groups did not differ with respect to the etiology of infertility, the source of spermatozoa, the quality of the embryos, the type of EL catheter, and the ease of transfer. The pregnancy rate was similar between two groups. In fresh ET cycles, a higher delivery rate was observed in group B than it group A (78.1% vs. 60%, p=0.1). In FET cycles, the rate of delivery was significantly higher in group B than in group A to a nonsignificant extent (88.9% vs. 58.8%, p=0.06). Conclusion: EL techniques did not have a significant impact on the delivery rate in either fresh or FET cycles.

On the Untact ICT based New Concept Storage Device Design by Interworking SysML and CAD Data to Improve the Development Efficiency

  • In these days, innovative functions are required to unmanned parcel delivery lockers. As non-face-to-face transactions become the center due to the recent COVID-19 pandemic, many problem occurs in society such as theft crimes and lack of loading space. Therefore, New concept storage device is developed in korea. It has the functions that minimizes empty spaces by using the internal transport device to enable efficient space loading and refrigerate goods such as foods. In order to systematically approach the system design of the unmanned parcel delivery lockers which is the new function is applied, We applied model-based system engineering to improve the development efficiency and use a system modeling language to express the system. We conducted interworking research of CAD data including system modeling and design data. it is expected that this method will increase the effective development efficiency such as maintenance traceability and reduction of development period and cost.

Preparation and evaluation of GFP-containing microspheres for oral vaccine delivery system (경구용 백신수송체용 GFP 함유 마이크로스피어의 제조 및 평가)

  • In order to design the oral vaccine delivery system, we prepared the alginate micro spheres containing GFP (green fluorescent protein) as a model drug by spray method. To optimize the preparation conditions of microspheres, we investigated the effects of various parameters including nozzle pressure, nozzle opening angle, and concentrations of sodium alginate and calcium chloride. The prepared microspheres were evaluated by measuring their sizes, loading efficiency, and morphology. The particle size of microspheres was affected by the concentration of sodium alginate and calcium chloride, nozzle pressure, and nozzle opening angle. As the concentration of sodium alginate increased, GFP loading efficiency and particles size of microsphere also increased. However, it was observed to be difficult to spray the sodium alginate solution with concentration greater than 1.5% (w/v), due to high viscosity. The pressure over $3\;kgf/cm^2$ didn't affect the size of particles. As a result, the spraying method enabled us to prepare microspheres for oral vaccine delivery system. In this study, microspheres prepared with 1% (w/v) sodium alginate had greater loading efficiency and better spherical shape.

Layer-by-layer assembled polymeric thin films as prospective drug delivery carriers: design and applications

  • Background: The main purpose of drug delivery systems is to deliver the drugs at the appropriate concentration to the precise target site. Recently, the application of a thin film in the field of drug delivery has gained increasing interest because of its ability to safely load drugs and to release the drug in a controlled manner, which improves drug efficacy. Drug loading by the thin film can be done in various ways, depending on type of the drug, the area of exposure, and the purpose of drug delivery. Main text: This review summarizes the various methods used for preparing thin films with drugs via Layer-by-layer (LbL) assembly. Furthermore, additional functionalities of thin films using surface modification in drug delivery are briefly discussed. There are three types of methods for preparing a drug-carrying multilayered film using LbL assembly. First methods include approaches for direct loading of the drug into the pre-fabricated multilayer film. Second methods are preparing thin films using drugs as building blocks. Thirdly, the drugs are incorporated in the cargo so that the cargo itself can be used as the materials of the film. Conclusion: The appropriate designs of the drug-loaded film were produced in consideration of the release amounts and site of the desired drug. Furthermore, additional surface modification using the LbL technique enabled the preparation of effective drug delivery carriers with improved targeting effect. Therefore, the multilayer thin films fabricated by the LbL technique are a promising candidate for an ideal drug delivery system and the development possibilities of this technology are infinite.
