Cloning and Characterization of the HSP70 Gene, and Its Expression in Response to Diapauses and Thermal Stress in the Onion Maggot, Delia antiqua

  • The cytosolic members of the HSP70 family of proteins play key roles in the molecular chaperone machinery of the cell. In the study we cloned and sequenced the full-length cDNA of Delia antiqua HSP70 gene, which is 2461 bp long and encodes 643 a.a. with a calculated molecular mass of 70,787 Da. We investigated gene copies of cytosolic HSP70 members of 4 insect species with complete genome available, and found that they are quite variable with species. In order to characterize this protein we carried out an alignment and a phylogenetic analysis with 41 complete protein sequences from insects. The analysis divided the cytosolic members of the family into two classes, HSP70 and HSC70, distinguishable on the basis of 15 residues. HSP70 class members were slightly shorter in length and smaller in molecular mass relative to the HSC70 class members, and the conservative and functional regions in these sequences were documented. Mainly, we investigated the expression of Delia antiqua HSP70 gene, in response to diapauses and thermal stresses. Both summer and winter diapauses elevated HSP70 transcript levels. Cold-stress led to increased HSP70 expression levels in summer- and winter-diapausing pupae, but heat-stress elevated the levels only in the winter-diapausing pupae. In all cases, the expression levels, after being elevated, gradually decreased with time. HSP70 expression was low in non-diapausing pupae but was up-regulated following cold- and heat-stresses. Heat-stress gradually increased the mRNA level with time whereas cold-stress gradually decreased levels after an initial increase.

Analyses of Valatile Compounds from Allium sup. and Ovipositional Response of Delia antiqua to Various Volatile Chemicals (Allium속 방향성 성분의 분석과 방향성 성분이 고자리파리(Delia antiqua) 산란에 미치는 영향)

  • The major volatiles from Allium species were found to be sulfide compounds and the ratio of sulfide to volatiles was 66.1% in garlic, 66.1% in scallion, 62.3% in green onion, 39.2% in onion, and 4.2% in chive. Trace of cyclooctasulfur was found to be present among the volatiles. The most oviposition of 17.2% occurred at diallyl sulfide and the least of 0.8% at acetylthiophene whereas the most oviposition of 43.3% occurred at ethyl alcohol if concentration was 100%. Among the organic solvents used for dilution, ethyl alcohol received the highest 52.5% of oviposition and ether the lowest of 5.9%. Furfuryl mercaptan which is also one of the volatiles, received 46.9% of oviposition. For oviposition site, D. antiqua preferred sulfides at near 1%, ethyl alcohol at high, and other volatiles at various concentrations. At 100% concentration, most volatiles except dimethyl disulfide and ethyl alcohol received less oviposition than the control which was watered sand with no volatiles added.

Seasonal Occurrence and Summer Diapause of the Onion Maggot, Delia antiqua (Meigen) (Diptera : Anthomyiidae) (고자리파리의 발생소장과 하면상황)

  • Studies were carried out to examine seasonal occurrence and emergence pattern of the onion fly, Delia antiqua (Meigen), in the field of Chinju city in Gyeongnam province from 1984 to 1987. Trap catch revealed that the onion fly had three generations in a year, the second flight being partial; the peaks of adult occurrence were at mid April for the overwintered generation (1st flight), at early June for the first generation (2nd flight), and at the period from late September to early October for the autumn generation (3rd flight). It was shown that parts of the pupae in the first and almost all of the pupae in the second (or autumn) generation underwent summer diapause. The pupal periods were less than 22.4 days for the non-diapaused pupae, while those of the diapause. The pupal periods were less than 22.4 days for the non-diapaused pupae, while those of the diapaused pupae were form 8.5 to 145 days in the first generation in he filed, being shorter for the pupae pupated later. This resutled in synchronization on the emergence of these diapaused pupae at late September, with the adults from the second generation pupae. The ratio of diapausing pupae increased among the later pupated ones in the first generation ; the pupae which had been pupated in early May included 43.8% of diapausing pupae, while more than 86.7% of the pupae were under diapausing state for the pupae pupated in late May and thereafter. More than 93.3% of the pupae went into diapause, regardless of pupation time for the second generation.

  • The Garlic having antibiosis is added to the most cuisine and used to treat dysentery and diarrhea for So-um-in(少陽人). The fact that the garlic, a Kind of Hun-chae, has warm and thermal characteristics couId be proved by a natural ecoIgical action of a fIy, Delia antiqua Meigen. OriginalIy San(蒜) designated Sosan(小蒜), a native species in Korea. But as Daesan(garlic)(大蒜) was imported, it meant both of Sosan(小蒜) and Daesan(大蒜). Now it means only Daesan(大蒜). This process of change of meaning was reviewed by focusing on the Dong-ui-bo-gam(東醫寶鑑). The San-20-mae(蒜二十枚) appearing in the Dangun mythology(檀君神話) should be interpreted not to 'garlic 20 piece(마늘 20개)' but to 'Dal-lae 20 roots(달래 20뿌리)'. It is possible to explain that the hibernant bear compared with the active tiger overcame the animality and the bestiality with perseverance in the dark cave so that it could be transformed to a human at the 21stday, the culmination number of Yang(陽). The garlic decame an abstaining food at sacrificial rituals because it arouses the sexual desire and excites person,s mind. High dose of raw garlic may cause harmful side effects in So-yang-in(少陽人) and Tae-um-in(太陰人) having a thermal characteristic.

Biological Control of Onion Maggot and Tobacco Cutworm with Insect-parasitic Nematodes, Steinernema feltiae and Heterorhabditis heliothidis (곤충기생성 성충, Steinernema feltiae와 Heterorhabditis heliothidis를 이용한 고자리파리 및 담배거세미나방의 생물적 방제)

  • Laboratory trials were conducted for control of onion maggot and tobacco cutworm with ento¬mogenous nematodes. The onion maggot, Delia antiqua, was exposed to Steinernema feltiae at concentration of 0,30,60, 120 or 240 nematodes per larva, and to Heterorhabditis heliot-hidis at concentration of 0, 10, 20, 40 or 80 nematodes per larva. Mortalities of the maggot ranged from 80 to 100% in S. feltiae and from 63.3 to 100% in H. heliothidis. The tobacco cutworm, SpodoPtera litura was exposed to S. feltiae at concentration of 0,50,100,200 or 400 nematodes per larva and to H. heliothidis at concentration of 0,20,40,80 or 160 nemat¬odes per larva with or without kale in petri dish. The 3rd instar larvae of the tobacco cutworm was more susceptible to both nematode species than the 4th or 5th instar at low concentration. Mortalities of the 3rd instar were 100% in S. feltiae and 67.7-100% in H. heliothidis while those of 4th and 5th instar ranged 76.7-100% and 43.3-100% in S. feltiae, and 36.7-90% and 3.3-90% in H. heliothidis, respectively. Mortalities of the tobacco cutworm larvae decreased when the nematodes were sprayed on the kale leaves in petri dish except 3rd instar.
