Effect of Toluene Application to Skin on the Enhancement of Liver Injury in $CCl_4$-Pretreated Rats

  • To evaluate an effect of toluene application to skin on the enhancement of liver damage in $CCl_4$-pretreated rats, toluene (35 mg/$cm^2$) was sequentially applied for 5 days to the skin of liver damaged rats with $CCl_4$ (6 times every other day: 0.1 ml/100 g body weight-50% $CCl_4$ in olive oil) On the basis of the functional and morphological findings in rat liver, appling toluene to the skin in liver damaged animals led to the more enhanced liver damage. In addition, by applying toluene to skin of liver damaged rats, the hepatic cytochrome P450 content was somewhat more increased, but the hepatic benzylalcohol dehydrogenase activity was significantly decreased (P<0.001), whereas benzaldehyde dehydrogenase activity was not statistically changed. In conclusion, the toluene application to skin in liver-damaged rat led to enhancement of liver injury that may be due to the accumulation of toluene metabolite in liver.

Purification and Production Conditions of Antimicrobial Compound from Methylotrophic Actinomycetes MO-16 (Methanol 자화방선균 MO-16으로부터 항균성 물질의 정제 및 생산조건)

  • A methylotrophic actinomycetes strain MO-16, which produce the antimicrobial compound, was isolated from soil and supposed as Amycolatopsis sp. based on taxonomic studies. The cell-free extract of methanol-grown strain MO-16 showed dehydrogenase activity for methanol and formaldehyde when various electron acceptors were added for oxidation. On the other hand, methanol did not affect the production of antimicrobial compounds, and organic nitrogen sources such as corn steep liquor and peptone were better than inorganic nitrogen sources. These compounds showed broad antimicrobial spectrum to the tested strains such as bacteria and yeast. The antimicrobial comounds were very stable under heat(121$^{\circ}C$), acid(pH2.0), alkali(pH11.0) treatments. These compounds were isolated by ethylacetate extract, silica gel column chromatography and reverse phase HPLC. Two compounds(peak 1 and 2) were detected as antimicrobial compounds through the HPLC analysis. The peak 2 was purified as a single compound and revealed a 98% purity.

  • Effects of dammarane glycosides of Panax ginseng on primary cultured chicken embryonic brain cells were studied by microscopic observation and determination of the activity of pyruvate dehydrogenase complex (PDHC). Brain cells were prepared from the brain of 10-day-old chicken embryo and cultured with either a standard medium consisted of 85% Dulbecco's Modified Eagle Medium (DMEM), 10% horse serum and 5% chicken embryonic extracts or a deficient medium consisted of 90% DMEM and 10% horse serum. It was observed that dammarane glycosides of Panax ginseng seemed to show the tendency to stimulate the neurite outgrowth of brain cells which were cultured with a deficient medium under microscopic observation. The activity of PDHC in brain cells cultured with a deficient medium was increased by dammarane glycosides of Panax ginseng.

  • In the periodic amputated rat incisor pulp, the distribution of five isoenzymes of lactic acid dehydrogenase was evaluated and the total LDH activity was assayed. This study hs established the followings; 1) It demonstrates the existence of five distinct isoenzymes of LDH, with LDH-1 and LDH-2 predominating, in the rat incisor pulp. 2) The total LDH activity in periodically amputated rat incisor pulp is markedly increased as compared to the normal rat incisor pulp. 3) It is possible that the periodic amputation of tooth effects the pattern of LDH isoenzymes in the pulp, especially LDH-1 and LDH-2 region.

  • The microbial degradation of $poly-{\beta}-hydroxybutyrate$ (PHB) films was studied in soil microco는 incubated at a constant temperature of 2, 10, 20, 30 and $40^{\circ}C$ for up to 49 days. The degradation rate measured through loss of weight was enhanced by incubation at a higher temperature. At the soil temperature $40^{\circ}C$, $poly-{\beta}-hydroxybutyrate$ was rapidly degraded at a decay rate of 3.5% weight loss per day. The degradation of $poly-{\beta}-hydroxybutyrate$ did not affected significantly the chemical properties of soils such as pH and electric conductivity. However, microbial activity of soil in terms of dehydrogenase activity was increased by the degradation of $poly-{\beta}-hydroxybutyrate$.

  • Attempts were made to see if feeding of ginseng ethanol extract could affect proper- ties of rat brain lactate dehydrogenase such as specific activity, heat stability, Km for substrate, inactivation by 3-bromopyruvate and trypsin, and immune response. The following results were obtained. Specific activity of LDH was observed to reach maximum in 5 month after birth and then decrease steadily. However, that of LDH from rat fed with ginseng ethanol extract was found in rat fed with ginseng ethanol extract. 3-bromopyruvate was shown to inactivate LDH-5 from old rat fed. Inactivation of LDH-5 by trypsin was remarkable in old rat fed. Km value for pyruvate in old rat fed was remarkably decreased. Cumulative results suggest that ginseng ethanol extract could affect conformational change of LDH responsible for altered properties through unknown mechanism.

Antioxidant Enzymes in Postpartum Anoestrus Buffaloes Supplemented with Vitamin E and Selenium

  • The 15 buffaloes were divided into three groups, viz. group 1: normal cyclic buffaloes; group 2: postpartum anoestrus buffaloes and group 3: post partum anoestrus buffaloes supplemented with intramuscular injections of Vit. E.-care Se containing 500 mg $\alpha$-tocopheryl acetate and 15 mg selenium at weekly intervals for two months. The postpartum anoestrus buffaloes had significantly higher levels of erythrocytic lipid peroxidation, superoxide dismutase and glucose-6 phosphate dehydrogenase activities but lower glutathione peroxidase activity as compared to normal cyclic buffaloes. The supplementation of vitamin E and selenium lowered the level of erythrocytic lipid peroxidation, superoxide dismutase and glucose-6 phosphate dehydrogenase activities but it had no effect on whole blood selenium and erythrocytic gluathione peroxidase activity. All the animals in group 3 became cyclic and showed 60% conception rate.

Peroxidase Isozyme in Root Differentiation from Cultured Ginseng Root Explants (인삼 근절편 배양시 Peroxidase Isozyme에 관한 연구)

  • In order to pursue some physiological studies on organogenesis in ginseng tissue culture, ginseng root explants were cultured on a modified MS medium containing NAA and kinetin. The activities of peroxidase and some enzymes were investigated and their isoenzyme patterns were also observed. The activity of peroxidase decreased by 20% in one week's culture and increased thereafter by 80% in culturing for 7 weeks compared with the control group. Glucose-6-phosphate dehydrogenase activity increased by 400% after culturing for 5 weeks and increased during the days preceeding root formation. The activities of glutamate dehydrogenase and acid phosphatase also increased during the culture. After 3 weeks' culture, new peroxidase isozyme (pH 7.6) appeared and 7 weeks' culture, another new peroxidase isozyme (pH unidentified) appeared. These patterns were also identified by using FPLC. After 7 weeks' culture, a new esterase isozyme of pH 8.5 appeared and isozyme patterns of acid phosphatase were quite changed compared with the isozyme patterns of tissue cultured for 5 weeks. In so far as these new isoenzymes appear distinctively after 7 weeks' culture, root differentiation is supposed to be induced after 7 weeks' culture.

  • Succinate dehydrogenase was activated by ethyl acetate extract from the methanol extract of white ginseng previously treated with petroleum ether and ethyl ether to remove all highly nonpolar components, and the residual aqueous solution from the ethyl acetate extraction. Also, all of the extracted fractions of red ginseng except the ether extract gave positive results. On the contrary to some suggestions by other workers that alkaloids of ginseng may enhance the succinate degydrogenase activity, our results show that the alkaloids may have exhibited some inhibitory eject on this enzyme.

  • The purpose of this study was to probe the influences of NaF oral administration on a dose-effect relationship between fluoride levels of serum enzyme activity such as alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) in rats fed experimental diets for 5 weeks. All groups increased the activity of serum ALP, AST, ALT, and LDH levels with increasing NaF. In addition the fluoride levels of serum and organ tissues (liver, brain, heart, lung, kidney) in oral NaF groups (NF3~NF50) were significantly increased by adding sodium fluoride in comparison with normal diet group (ND) (p<0.05). These results, a high concentration of sodium fluoride was determined that the toxicity to various organ tissues.
