Effects of Volatile Substances on Rat Lactate Dehydrogenase and Cholinesterase (흡입물질이 흰쥐 Lactate Dehydrogenase와 Cholinesterase 활성변화에 미치는 영향)

  • The effects of volatile substances inhalation on lactate dehydrogenase and cholinesterase in rats were investigated. Male Sprague-Dawley rats were exposed to marketed odorant, ethyl acetate and ethyl ether for 15 days. Enzyme activities were measured in serum and several tissues such as liver, lung, brain, heart, kidney and muscle to find differences of effects according to the organ. Cholinesterase activity in serum and most of tissues revealed time-dependent decrease in the case of marketed odorant inhalation. Especially in heart and kidney significant decrease was observed. Ethyl acetate exposure to rats revealed also decrease in serum and all tissues by 40% to 60%. Ethyl ether inhalation showed significant decrease by 30% to 50%. Lactate dehydrogenase activity was markedly increased in serum and similarly in heart, brain and kidney by exposure to marketed odorant. No changes were observed in liver. Ethyl acetate exposure to rats revealed increase in serum by about 200%, compared to normal group and in other tissues by 40% to 70% except in liver and muscle. Ethyl ether inhalation showed significant increase in serum by about 100%. There was no change in 'liver and slight increase in muscle.

The Effect of Ginseng Saponin on Yeast Alcohol Dehydrogenase (인삼 사포닌이 효모 Alcohol Dehydrogenase에 미치는 영향)

  • Yeast alcohol dehydrogenates and ginseng saponin interaction has been investigated to understand the non-specific enzyme stimulating effect of the saponin of Panax ginseng C.A. Meyer. It was confirmed that several amphiphiles such as sodium dodecyl sulfate(SDS), Triton X-100, sodium taurodeoxycholate (Na-TDC) as well as ginseng saponin mixture and purified ginseng glycosides lowered Km values of yeast alcohol dehydrogenase (ADH) for ethanol and NAD in the presence of the above amphiphiles suggesting that the surface activity of the amphiphiles might play a significant role in the ADH catalyzed reactions. Conformational change of yeast alcohol dehydrogenase in the presence of the above amphiphiles at their optimal concentration for the maximum activity was studied. Circular dichroism (C.D) spectrum of yeast ADH showed that the conformational change of the enzyme occurred in the presence of above amphiphiles. Fluorescence data also showed that the hydrophobic area increased in the presence of above amphiphiles. Examination of the interaction between ADH and ginseng saponin using radioactive saponin showed that there might be a very weak interaction between them. From the above results, it was concluded that the non-specific enzyme stimulating effect of the saponin might be due to the change of polarity of the enzyme solution in the presence of the saponin.

  • Polyacrylamide gel electrophoresis was used to investigate the patterns of LDH and MDH isozymes in the embryo and adult of amphibia; Rana nigromaculata, Rana plancyi chosenica and Hynobius leechii. Rana nigromaculata is considered to be heterozygous for the gene specifying the "B" subunit of LDH, and Hynobius leechii to be heterozygous for the gene specifying the "A" subunit of LDH. The LDH isozyme paatern of embryos of the above three species is characterized by a gradual increase in the activity of LDH-5 (muscular form)during development. Two or three molecular forms of MDH is present steadily from early embryos and in adult. Of the MDH isozymes, the more cathodic one (MDH-m) appears weakly in early developing stages, but increases slowly in the activity as the embryo develops.the embryo develops.

Purification and Characterization of a Cyclohexanol Dehydrogenase from Rhodococcus sp. TK6

  • Activity staining on the native polyacrylamide gel electrophoresis (PAGE) of a cell-free extract of Rhodococcus sp. TK6, grown in media containing alcohols as the carbon source, revealed at least seven isozyme bands, which were identified as alcohol dehydrogenases that oxidize cyclohexanol to cyclohexanone. Among the alcohol dehydrogenases, cyclohexanol dehydrogenase II (CDH II), which is the major enzyme involved in the oxidation of cyclohexanol, was purified to homogeneity. The molecular mass of the CDH II was determined to be 60 kDa by gel filtration, while the molecular mass of each subunit was estimated to be 28 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The CDH II was unstable in acidic and basic pHs, and rapidly inactivated at temperatures above $40^{\circ}C$ . The CDH II activity was enhanced by the addition of divalent metal ions, like $Ba^2+\;and\;Mg^{2+}$. The purified enzyme catalyzed the oxidation of a broad range of alcohols, including cyclohexanol, trans-cyclohexane-1,2-diol, trans-cyclopentane-l,2-diol, cyclopentanol, and hexane-1,2-diol. The $K_m$ values of the CDH II for cyclohexanol, trans-cyclohexane-l,2-diol, cyclopentanol, trans-cyclopentane-l,2-diol, and hexane-l,2-diol were 1.7, 2.8, 14.2, 13.7, and 13.5 mM, respectively. The CDH II would appear to be a major alcohol dehydrogenase for the oxidation of cyclohexanol. The N-terminal sequence of the CDH II was determined to be TVAHVTGAARGIGRA. Furthermore, based on a comparison of the determined sequence with other short chain alcohol dehydrogenases, the purified CDH II was suggested to be a new enzyme.

Changes in Soil Physicochemical Properties and Dehydrogenase Activity by the Formation of Fairy Ring of Tricholoma matsutake (송이 균환(菌環)의 발달(發達)에 따른 토양(土壤)의 이화학적(理化學的) 특성(特性)과 탈수소효소(脫水素酵素)의 활성(活性) 변화(變化))

  • The management of pine-mushroom forest means the environmental control for fairy ring of Tricholoma matsutake. Thus, the management demands intensive study for keeping healthy condition of the fairy ring, and that for the measurement of active portion of the soil ecosystem. This study was conducted to investigate the impact of T. matsutake fungus on the soil physicochemical properties and dehydrogenase activity by dividing the fairy ring into 3 regions such as 'zone of decayed mycorrhizae', 'zone of physiologically active mycorrhizae', and 'in front of fairy ring'. The passing of T. matsutake did not result in significant changes in canon contents of soils, but available phosphorus, carbon, and nitrogen contents were different between the soils of active mycorrhizal zone and that in front of fairy ring. The dehydrogenase activity around the fairy ring of T. matsutake was quite lower than that in general forest soils, which indicated that the fairy ring of T. matsutake was built up in the relatively immature soils which contain little organic matter. Thus, the dehydrogenase activity of soil was thought to be used as an index for the management of pine-mushroom forest with considering that the management practically means the environmental control for keeping good conditions for the development of fairy ring of T. matsutake. Especially, the dehydrogenase activity measurement can be recommended as a tool for time-decision of litter removal by floor raking since the activity is a good index of litter decomposition.

On respiration rate and dehydrogenase activity of yeast cells irradiated by gamma ray from cobalt-60. 1. Oxygen uptake rate of the gamma irraiated yeast cells had been measured with Warburg's manometer, and the $O_{2}$-uptake was compared with those of normal cells. The rate of endogetious respiration increases in its $O_2$-uptake at 150, 000 rentgen dose, and at higher rentoen doses it was decreased. Exogenous respiration begin to decrease in its O_2$-uptake at 5, 000r. doses of irradiation, further decrease with increasing of doses unproportionally. 2. It appears that plasma-membrane and nuclear membrane of yeast cells have changed and denatured by gamma-irradiation, as exogenous respiration of glucose had been decreased at a dose of 200, 000r's irradiation. 3. The activity of glucose, alcoholic, lactic, succinic and glutamic deliydrogenase (G.D.H., A.D.H., L.D.H., S.D.11., and GL.D.H.) in the gamma irradaited cells had been assayed by T.T.C.(Triphenyl tetrazolium chloride) method and spectrophotometry, the obtained results were compared with those of normal cells. 4. At a dose of and 10, 000 rentgens' irradiation of gamma ray, the activty of each debydrogenase (G.D.H., A.D.H., L.D.H., ) shows a sharp and highest peak in optical absorbalicy, but each abtivity of S.D.H and Gl.D.H shows its' maximum peak at a dose of 30, 000r. 5. The curve of each dehydrogenase activity was found to be rhythmical according to dose-rate of gamma irradiation. 6. Comparing with activity of debydrogenase each other, the maximum peak in optical absorbency can be arranged according to order as follows; glucose > alcoholoic > lactic > glutamic > succinic, this order is identical to the order of breakdown utility in respiration of normal yeast cells. 7. The activity of dehydrogenase experimented exhibit a resistance against gamma irradiation at lethal dose of cells, and the activity of dehydrogenase are found to be much resistant than those of respiratory system. We may assume that the membrane substrate of mitochondria or cytoplasm had been destructed by gamma-irradiation much more than that of dehydronase system.

  • The relationships among the contents of Pb, Zn, Cd, Cu and microbial biomass and activity of soil were investigated in Kyongbu, Yongdong, and Chungbu highways. The heavy metal concentrations were the highest in Kyongbu highway with high traffic density. The levels of these metals in soil in three highway sites were much higher than Mt. Kwank as control site. The highest concentrations of heavy metals were found in the upper layer of soil adjacent to the roadside. Dehydrogenase activity (DHA) and adenosine tri-phosphate (ATP) contents were generally lowest at distance of 0.5 m from the roadside. Simple regression analysis indicated that DHA and ATP contents were highly negatively correlated with Pb and Zn concentrations.

  • Effects of organic solvent fractions from perilla ethanol extract on alcohol metabolism in rats were examined and the results were as follows: Ethanol souble fraction, after a single oral administration to rats, was found to cause a significant decrease in the serum ethanol concentration as well as enhancement of liver cytosolic alcohol dehydrogenase (ADH) activity. On the other hand, the fraction insouble in ethanol was found to increase ethanol concentration in the blood and inhibit ADH activity.

  • The G-6-PD (glucose-6-phosphate dehydrogenase) activity, the first step in the pentose phosphate shunt, of rat livers during prenatal and pstnatal development in different sexes was studied. The enzyme activity is very high (54.2 units) at 16 days of embryo ad then decreases to a low level (17.6 units) at 13days after birth. There are significant increase between 13 and 15 days of age and continuously increases to the level of 53.2 units at 19days of age. The G-6-PD activity in female rat livers was slightly higher than in males.

  • It has been known that the ginseng extracts activate the lipid metabolism in animal body. The experiments were undertaken to elucidate the effects of total, diol, and trial saponin of ginseng on the activities of acyl Co-A synthetase and hydroxyacyl Co-A dehydrogenase involved in fatty acid metabolism in normal albino rat liver. The acyl Co-A synthetase activity, in vitro, was increased by 20% with treatment of 2.5${\times}$10-3% total saponin, by 14% with 2.5${\times}$10-3% diol saponin, arid 30% with 2.5${\times}$10-4% triol saponin, respectively. And the enzyme activity was increased by 27% at 2 hours after intraperitoneal injection of total saponin. Hydroxyacyl Co-A dehydrogenase activity, in vitro, was increased by 77% with 10-4% total saponin, by 64% with 10-2% diol saponin, and by 72% with 10-3% triol saponin, respectively. Also, the enzyme activity, in vivo, was increased by 15.3% and 33% at 2 hours and 4 hours.
