Overexpression, Purification, and Characterization of the Herpes Simplex Virus-1 DNA Polymerase-UL42 Protein Complex

  • The herpes simplex virus type-1 (HSV-1)-encoded DNA polymerase consists of two subunits, the products of the UL30 and UL42 genes. UL30 and UL42 were coexpressed in Sf9 cells infected with recombinant baculoviruses carrying the two genes. The UL30 and UL42 gene products remained tightly associated throughout the purification, which led to a near homogeneous heterodimer composed of the DNA polymerase and UL42 protein. The DNA polymerase-UL42 protein heterodimer, purified from the recombinant baculovirus-infected Sf9 cells, showed the same high degree of processivity of deoxynucleotide polymerization as the enzyme purified from the HSV-1 infected primate cells. Like the latter, it contained a 3'-5' exonuclease activity that specifically hydrolyzes an incorrectly matched nucleotide at the 3' terminus of a primer, thereby contributing to the fidelity of DNA replication.

The Preparation of Crystalline Mannobiose from Brown Copra Meal Using the Enzyme System and Yeast Fermentation

  • ${\beta}-1$, 4-Mannobiose was prepared by the enzymatic hydrolysis of brown copra meal and the subsequent elimination of mono-saccharides from the resultant hydrolysate with a yeast. The enzyme system hydrolyzed brown copra meal and produced monosaccharides and $\beta$-1, 4-mannobiose without other oligomers at the final stage of the reaction. Brown copra meal (30 g) was hydrolyzed at $50^{\circ}^C$ and pH 5 for 48 hr with the crude enzyme solution (300 ml) from Penicillium purpurogenum. By the elimination of monosaccharides from the hydrolysis products with a yeast (Candida parapsilosis var. komabaensis k-75), 5.2 g of crystalline mannobiose was obtained without the use of chromatographic techniques. After 50 hours of yeast cultivation, the total sugar content fell from 3.5% to 2.4%, and the average degree of polymerization rose from 1.8 to 2.2.

  • It has been presumed that the molten sulfur above $159^{\circ}C$ consists of an equilibrium mixture of $S_8$ rings and $S_x$ polymers where the number average degree of polymerization, P, is large. But it is known that admixture of halogens with liquid sulfur greatly reduce the viscosity. Constructing a new equilibrium equation, it is possible to evaluate the viscosity when halogens are added to liquid sulfur. Calculated viscosity is in good agreement with experimental values. Using the proposed model, the thermodynamic properties of liquid sulfur are also calculated over a wide range of temperature which the sulfur exists as the polymer.

  • Henequen fibers were treated by electron beam irradiation and by NaOH to make surface modification for better bonding in the manufacture of biocomposite. Impurity removal and carbonyl group formation were noticed in the previous study by electron beam irradiation, but extensive cellulose degradation were also noticed. To evaluate the effects of electron beam irradiation on cellulosic fibers further, henequen fibers, cotton pulp, cotton fibers, and cellophane were irradiated by electron beam, and their changes of cellulose viscosity, chemical composition, and tensile strength were measured and analyzed.

  • $\beta$-1,4-Mannotriose was prepared b he enzymatic hydrolysis of white copra meal (WCM) and the subsequent elimination of monosaccharides from the resultant hydrolysate with a yeast. The enzyme system from sunflower seed hydrolyzed WCM and produced monosaccharides and $\beta$-1,4-mannotriose without other oligomers at the final stage of the reaction. WCM(50g) was hydrolyzed at 5$0^{\circ}C$ and pH 4.5 for 24 hr with crude enzyme solution (500 mL) from sunflower seed. By the elimination of monosaccharides from the hydrolysis products with a yeast (Candida glaebosa), 8.1 g of crystalline mannotriose was obtained without the use of chromatographic techniques. After 48hr of yeast cultivation, the total sugar content decreased from 4.6% to 3.5%, whereas the average degree of polymerization increased from 2.3 to 3.1.

  • Beta-1,4-mannotriose was prepared by the enzymatic hydrolysis of poonac and the subsequent elimination with yeast of monosaccharides and disaccharide from the resultant hydrolysate. The enzyme system hydrolyzed poonac and produced monosaccharides, disaccharide and ${\beta}$-1,4-mannotriose without other oligomers at the final reaction stage. Poonac (50 g) was hydrolyzed at $50^{\circ}C$ and pH 6 for 48 hr with the crude enzyme solution (500 mL) from Trichoderma harzianum. The elimination of monosaccharides and disaccharide from the hydrolysis products with a yeast (Candida guilliermondii) produced 10.5 g of crystalline [${\beta}$-1,4-mannotriose without the use of chromatographic techniques. After 48 hr of yeast cultivation, the total sugar content fell from 4.8% to 3.4%, and the average degree of polymerization (D.P) rose from 2.5 to 3.2. The preparation method presented was confirmed to be suitable for the preparation of mannotriose from poonac.

Well-Defined Thermoresponsive Copolymers with Tunable LCST and UCST in Water

  • A thermoresponsive polymer, poly(2-(dimethylamino)ethyl methacrylate) (PDMAEMA), was successfully synthesized by atom transfer radical polymerization (ATRP). Different amounts of 1,3-propanesultone were used as quaternization agent to transit the PDMAEMA into partially modified poly(zwitterions), resulting in p[DMAEMA-co-3-dimethyl(methacryloyloxyethyl)ammonium propanesulfonate] (PDMAEMA-co-PDMAPS). Molecular weight, molecular weight distribution, and degree of quarternization were determined by gel permeation chromatography (GPC) and 1H NMR spectroscopy. The transmission spectra of the 1.0 wt % aqueous solutions of the resulting polymers at 650 nm were measured as a function of temperature. Results showed that the lower critical solution temperature (LCST) and the upper critical solution temperature (UCST) could be easily controlled by the different composition of dimethylamino and zwitterion groups. The effect of partial quaternization on thermoresponsive properties was also studied by dynamic light scattering (DLS) with the same aqueous concentration (1.0 wt %) as employed for turbidimetry studies. The LCST and UCST values measured by DLS correlated well with those determined by turbidimetry.

Characterisation of Ramie Yarn Treated with Sodium Hydroxide and Crosslinked by 1,2,3,4-Butanetetracarboxylic Acid

  • The microstructure and tensile properties of ramie treated with NaOH and crosslinked with 1, 2, 3, 4-butanetetracarboxylic acid were characterised. X-ray diffraction and FTIR were used to study the crystalline structure o f the resultant ramie yarns. Results showed that the maximum change in the structure of the modified ramie took place at 16% NaOH, which would completely transform Cellulose I to Cellulose II. Also, the crystallinity index and fibre orientation decreased to the minimum value while the absorption properties were enhanced. The average degree of polymerization of the treated ramie yams slightly decreased after NaOH treatment.

  • A bacterial strain No. 43 was isolated from soil samples as a levan-assimiating microorganism producing an extracellular levanase and hydrolying levan to levanbiose. According to the taxonomic characteristics of its morphological and physiological properties, the strain was idenified as Pseudomonas sp. No. 43. The optimum culura medium was composed of 10g levan, 5g(NH4)2SO4, 3g NH4Cl, 3g polypepton, 1g K2HPO4, 0.5gMgSO4.7H2O, and 0.2g MnCl2.4H2O per liter. The cultivation for levanase was carried out in pH 7.0 at 4$0^{\circ}C$ for 28hr. The reaction product was a kind of oligosaccharide and it was purified by chilled ethanol precipitation and gel filtration for evalluation of degree of polymerization (DP). The purified product was determined as levanbiose of MW 342 and DP2 by HPLC and FAB-MS.

  • Hydrogels of semi-interpenetrating polymer networks (semi-IPNs) were prepared by two step reactions. Dimethylaminoethyl methacrylate (DMAM) and poly(ethylene glycol)-dimethacrylate (PEGDM) were copolymerized to yield hydrogels, and then acrylic acid (AA) monomer were adsorbed in the hydrogels followed by polymerization of AA to produce semi-IPNs. The swelling behavior of semi-IPNs depends largely on pH of medium, showing that the degree of swelling of the semi-IPNs exhibits a minimum at pH 6.0. It is observed that the elastic modulus of semi-IPNs is closely related to its swelling behavior.
