제주도 대정-한경 유역 효율적 지하수자원 관리를 위한 자료기반 연구 (Data-driven Analysis for Developing the Effective Groundwater Management System in Daejeong-Hangyeong Watershed in Jeju Island)

  • 본 연구에서는 자료기반 분석 기법을 이용하여 제주 대정-한경 유역의 군집형 지하수 이용 관정의 영향력을 평가하고 지하수 자원을 효율적으로 관리하기 위한 도구를 개발하였다. 분석을 위해 대정-한경 유역 내 총 19개 지하수위 관측공의 지하수위 자료, 총 3개 기상 관측소로부터 측정된 강수량 자료, 및 총 4개의 군집형 지하수 이용 관정(저지, 금악, 서광, 및 영어교육도시)으로부터 획득한 이용량 자료가 이용되었다. 먼저, 각 지하수위 관측공에 대하여 강수량 및 이용량 자료를 입력변수로 하는 자료 기반 지하수위 예측모델을 개발하였다. 이때, 과거의 장기적 변동특성을 효과적으로 학습에 이용하기 위하여 누적 장단기 메모리 모델을 이용하였다. 모든 관측 공에 대하여 지하수위 예측모델을 개발하고, 이용량 입력변수에 대한 섭동 민감도 분석을 수행하여 각 군집형 관정의 공간적 영향력을 분석하였다. 금악 수원은 해당 수원 중심으로 영향이 크고, 서광 수원은 하류 지역을 중심으로 영향이 큰 것으로 나타났으며, 영어교육도시는 수원의 상류 지역 중심, 저지 수원은 수원 상류 및 하류 중심으로 영향이 나타났다. 그리고 유역 내 군집형 수원의 영향력은 대략 5km인 것으로 나타났다. 추가적으로, 학습된 예측모델을 기반으로 군집형 이용 관정의 영향 범위에 포함되는 지하수위 관측공에 대해 강수량 대비 배경 지하수위 회복을 위한 적정 지하수 이용량을 산정하였다. 최근의 강수 패턴을 적용하였을 때, 현재 지하수 이용량을 기존의 80%로 제한할 필요성이 있는 것으로 나타났으며, 강수량이 100mm 증가하였을 때, 대략 1,500 m3에서 1,900 m3의 추가적인 취수가 가능할 것으로 평가되었다. 본 연구를 통해 도출된 대정-한경 유역 지하수 거동특성 평가 결과와 자료기반 분석 도구들은 대정-한경 유역의 지속 가능한 지하수 개발을 위한 유용한 정보로 활용될 수 있을 것으로 판단된다.

주문생산 기업을 위한 기계학습 기반 총생산시간 예측 기법 (A Machine Learning-based Total Production Time Prediction Method for Customized-Manufacturing Companies)

  • 4차 산업혁명 기술의 발전으로 사람이 처리하지 못하는 부분을 기계학습 등 인공지능 기법을 활용하여 개선해 보려는 노력이 확대되고 있다. 주문형 생산 기업에서도 주문에 대한 총생산시간을 예측하여 납기 지연 등의 기업 리스크를 줄이고자 하나 주문마다 총생산시간이 모두 달라 이를 예측하는데, 어려움을 겪고 있다. 주문 처리량 증대, 주문 총비용 절감을 위해 효율성이 가장 낮은 영역을 찾아 그 영역을 강화하는 TOC(Theory of constraints) 이론이 개발되었으나 총생산시간 예측은 제시하지 못하였다. 주문생산은 고객의 다양한 요구로 인해 주문마다 그 특성이 모두 다르므로 개별적인 주문의 총생산시간을 사후에 측정할 수는 있으나 사전 예측을 하기는 어렵다. 기존 주문의 이미 측정된 총생산시간도 모두 달라 표준 시간으로 활용할 수 없는 한계성이 있다. 이에 따라 경험이 많은 관리자는 시스템의 이용보다는 감에 의존하고 있고, 경험이 부족한 관리자는 간단한 관리지표(예, 원재료가 파이프이면 총생산시간 60일, 철판이면 총생산시간 90일 등)를 사용하고 있다. 불완전한 감이나 지표를 기초로 하여 작업 지시를 너무 빨리하면 정체가 발생하여 생산성이 저하되고, 너무 늦게 하면 긴급 처리로 인해 생산비용이 증가하거나 납기를 지키지 못하는 경우가 발생한다. 납기를 지키지 못하면 지체상금을 배상해야 하거나 영업, 수금 등의 부문에 악영향을 미친다. 본 연구에서는 이러한 문제를 해결하기 위하여 주문생산시스템을 운영하는 기업의 신규 주문 총생산시간을 추정하는 기계학습 모델을 찾고자 한다. 기계학습에 활용된 자료는 수주, 생산, 공정 실적을 사용한다. 그리고 총생산시간의 추정에 가장 적합한 알고리즘으로 OLS, GLM Gamma, Extra Trees, Random Forest 알고리즘 등을 비교 분석하고 그 결과를 제시하고자 한다.

온라인 과학 기사 텍스트 마이닝을 통해 분석한 에너지 용어 사용의 맥락 (Analyzing Different Contexts for Energy Terms through Text Mining of Online Science News Articles)

  • 본 연구에서는 일상생활에서 에너지 용어가 사용되는 맥락을 알아보기 위하여 온라인 과학 기사를 수집하여 언어 네트워크, 토픽 모델링 분석 기법을 활용해 에너지 관련 기사에 사용된 용어의 빈도, 용어 네트워크, 기사의 주제를 분석하였다. 분석에 사용된 자료는 2018.3.1.부터 1년간의 온라인 과학 분야의 기사 중 에너지를 검색어로 하여 10개의 국내 중앙지에서 검색 및 선정된 2,171편이다. 이 기사들을 자연어 처리하여 51,224개의 문장과 507,901개의 단어로 데이터를 구성하였다. R 프로그램을 활용하여 용어 빈도수 분석 및 언어 네트워크 분석을 실시하였고, 에너지 용어 사용의 맥락 탐색을 위해 구조적 토픽 모델링 분석을 적용해 기사의 주제를 도출하였다. 기사에 사용된 용어 중 빈도수가 유난히 높은 용어는 기술, 연구, 개발로 새로운 소식을 알리는 기사의 특성을 반영한 것으로 나타났다. 한편, 기사 2편당 한 번 이상의 빈도로 사용되는 용어에는 산업 관련 용어(산업, 제품, 시스템, 생산, 시장)와 '전기', '환경'과 같이 에너지 관련 용어로 충분히 기대되는 용어들이 있었다. 한편, 에너지 관련 과학 수업에서 자주 사용되는 '태양', '열', '온도', '발전'도 빈도수 상위에 속하는 용어로 드러났다. 용어 네트워크 분석에서는 산업 및 기술과 관련된 용어와 기초과학 및 연구 관련 용어들이 약한 강도이지만 서로 군집을 이루는 것을 확인하였다. 한편, 에너지와 쌍을 이루는 용어의 분석에서는 '에너지 효율'을 비롯해 '에너지 절감', '에너지 소비' 등과 같이 에너지의 사용에 관한 용어들이 다수를 이루고 그 사용 빈도가 가장 높았다. 에너지 용어가 사용되는 맥락은 16개의 주제를 분류한 4가지 영역으로 '첨단산업', '산업', '기초과학', '환경 및 건강'으로 나타났다. 에너지 사용 관련 용어가 상당히 많이 사용된다는 결과는 에너지 수업의 시작점으로 에너지 저급화 개념의 도입이 효과적일 수 있음을 시사한다. 또한, 첨단산업이나 환경 및 건강의 맥락을 에너지 학습에 도입할 필요성도 보여준다. 본 연구에서 드러난 16개 주제에서 보이는 다양한 에너지 용어가 사용되는 맥락을 재구성해 에너지 관련 수업에 활용한다면 학생들이 학교에서의 에너지 학습과 일상적 상황을 통합적으로 인식하는 데 도움이 될 것이다.

발아 서목태 가수분해물의 근위축 억제 효과 (Germinated Rhynchosia nulubilis Hydrolysate Ameliorates Dexamethasone-induced Muscle Atrophy by Downregulating MAFbx Expression in C2C12 Cells and C57BL/6 Mice)

  • 스테로이드 유발 근위축은 장기간 코르티코스테로이드 요법의 일반적인 부작용이며 근육량과 기능의 현저한 감소로 이어질 수 있다. 발아된 Rhynchosia Nulubilis (GRH)는 항염증 및 항산화 특성이 있는 것으로 밝혀진 전통적인 약용 식물이다. 그러나 근 감소 억제 효능에 대하여는 보고된 바가 없다. 본 연구에서는 덱사메타손에 의해 유도된 근위축 C2C12 근관세포 모델과 마우스 모델에서 근위축 억제 활성에 대한 GRH의 영향을 평가하였다. 또한 골격근에 대한 GRH 효과의 기본이 되는 분자 경로를 확인하였다. May Grunwald-Giemsa 염색을 통한 근관 세포 염색 결과는 GRH 처리군에서 근관의 길이와 면적이 증가함을 보여주었다. 또한, muscle ring-finger protein-1 (MuRF-1)와 muscle atrophy F-box (MAFbx) 발현이 GRH 처리군에서 유의하게 감소하였다. 더욱이 덱사메타손에 의해 유도된 근위축 C57BL/6 마우스에서 트레드밀을 사용한 지구력 측정과 악력측정기를 이용한 악력 측정을 통한 근육 기능을 평가한 결과 GRH를 경구 투여한 마우스의 근육 기능이 향상되었다. 또한 이러한 결과는 근육 조직에서 GRH에 의한 MAFbx의 발현 억제를 통해 일어남을 확인하였다. 결론적으로 GRH는 MAFbx 발현을 억제하여 유비퀴틴-프로테아좀 경로를 차단함으로써 덱사메타손에 의해 유발된 근위축을 개선할 수 있을 것이다.

조식동물 탐지 및 모니터링을 위한 딥러닝 기반 객체 탐지 모델의 강인성 평가 (Evaluation of Robustness of Deep Learning-Based Object Detection Models for Invertebrate Grazers Detection and Monitoring)

  • 최근 조식동물로 인한 갯녹음 현상으로 인해 연안 생태계 및 어장환경의 황폐화가 가속화되고 있다. 이러한 갯녹음 현상을 모니터링하고 방지대책을 세우기 위해서는 광범위한 해역에 대한 원격탐사 기반의 모니터링 기술 도입이 필요하다. 본 연구에서는 수중에서 촬영된 동영상으로부터 조식동물을 탐지하고 모니터링하기 위한 딥러닝 기반 객체 탐지 모델의 강인성(robustness)을 비교 분석하였다. 우리나라 연안의 대표적인 조식동물 7종을 대상으로 이미지 데이터셋을 구축하였으며, 이를 활용하여 딥러닝 기반 객체 탐지 모델인 You Only Look Once (YOLO)v7과 YOLOv8을 훈련시켰다. 총 6개의 YOLO 모델(YOLOv7, YOLOv7x, YOLOv8s, YOLOv8m, YOLOv8l, YOLOv8x)에 대해 탐지 성능과 탐지 속도를 평가하였으며, 수중환경에서 촬영 시 발생할 수 있는 다양한 이미지 왜곡에 대해서 강인성 평가를 실시하였다. 평가결과 YOLOv8 계열 모델이 파라미터(parameter) 수 대비 더 높은 탐지 속도(약 71-141 FPS [frame per second])를 보였다. 탐지 성능에 있어서도 YOLOv8 계열 모델(mean average precision [mAP] 0.848-0.882)이 YOLOv7 계열 모델(mAP 0.847-0.850)에 비해 더 높은 성능을 보이는 것을 확인하였다. 모델의 강인함에 있어서 형태 왜곡에 대해서는 YOLOv7 계열 모델이 YOLOv8 계열 모델에 비해 강인한 것을 확인하였으며, 색상 왜곡에 대해서는 YOLOv8 계열 모델이 상대적으로 강인한 것을 확인 하였다. 따라서 실해역에서 수중 영상 촬영 시, 형태 왜곡은 발생 빈도가 낮으며 색상 왜곡은 연안에서 빈번하게 발생한다는 점을 고려했을 때, 연안해역에서 조식동물 탐지와 모니터링을 위해서는 YOLOv8 계열 모델을 활용하는 것이 타당한 것으로 판단된다.

길경에서 추출한 polygalacin D가 근원세포 분화 및 근위축에 미치는 영향 (Effects of polygalacin D extracted from Platycodon grandiflorum on myoblast differentiation and muscle atrophy)

  • 본 연구는 근생성 및 근위축 완화효능을 가진 유효소재 발굴의 필요성에 의해 polygalacin D가 근원세포 분화 및 미토콘드리아에 미치는 영향과 항암제 유도 근위축에 대한 완화효과를 각각 세포 및 동물실험을 통해 확인하고자 하였다. 그 결과, polygalacin D는 다핵을 지닌 근관세포의 수와 분화 종결인자인 MHC isoforms의 발현량을 증가시켰고 근육 내 단백질 분해 인자인 MuRF1, Smad2/3의 발현량은 유의적으로 감소시켰다. 또한 미토콘드리아 생합성 조절인자인 Pgc1α의 발현은 증가시키고 미토콘드리아 분열인자인 Drp1과 Fis1의 발현은 감소시켰다. 한편 zebrafish 동물모델을 통해 항암제 유도 근위축에 대한 개선효과를 확인한 결과, polygalacin D는 항암제에 의해 유도된 근위축과 미토콘드리아 손상을 완화시켰다. 이상의 결과들은 polygalacin D가 미토콘드리아 기능 증진을 매개로 근원세포 분화 촉진 및 근육 단백질 분해 저하 효과를 지닐 뿐만 아니라, 미토콘드리아 손상을 개선하여 항암제로 유도된 근위축에 대한 완화 효과를 나타냄을 시사한다. 따라서 본 연구를 통해 polygalacin D가 근생성 및 근위축 예방과 치료를 위한 잠재적인 유효소재로서의 가능성을 제시하였다.

이중확장칼만필터(DEKF)를 기반한 건설장비용 리튬이온전지의 State of Charge(SOC) 및 State of Health(SOH) 추정 (State of Health and State of Charge Estimation of Li-ion Battery for Construction Equipment based on Dual Extended Kalman Filter)

  • 전기자동차와 신재생에너지에 관한 관심이 높아지면서 건설장비 산업분야에서도 리튬이온 배터리를 접목하려는 요구가 높아지고 있다. 건설중장비는 건설 현장의 다양한 작업으로 인해 전류 용량의 감소가 급속히 진행되기 때문에 SOC(State of Charge) 및 SOH(State of Health) 같은 배터리의 상태를 더욱 정확하게 추정할 필요가 있다. 본 논문에서는 SOC와 SOH를 동시에 추정이 가능한 적응제어 기법 기반 이중확장칼만필터(Dual Extended Kalman Filter, DEKF) 알고리즘을 이용하여 실제 측정데이터와의 오차를 비교하였다. 배터리 충전 상태 예측을 위해 배터리 셀을 완전 충전 후 0.2C-rate조건에서 SOC 5% 간격으로 OCV를 측정하였고, 배터리의 열화를 판단할 수 있는 건전성 지표 확보를 위해 다양한 C-rate(0.2, 0.3, 0.5, 1.0, 1.5C rate) 조건에서 50 Cycle 동안 노화 실험을 수행하였다. DEKF를 이용한 SOC 및 SOH 추정 오차는 C-rate이 커질수록 커지는 경향을 보였으며 특히 SOC 추정결과, 0.2, 0.5 및 1C-rate에서 6%이하로 나타남을 확인하였다. 또한 SOH 추정 결과는 0.2 와 0.3C-rate에서 각각 최대오차 1.0% 및 1.3% 이내로 좋은 성능을 보이는 것으로 확인하였다. 다만, C-rate가 0.5C-rate에서 1.5C-rate으로 증가함에 따라 추정오차도 1.5%에서 2%로 다소 증가하는 것을 확인할 수 있었으나, 모든 C-rate 조건에서 DEKF를 사용한 SOH의 추정 성능은 약 2% 이내인 것으로 나타났다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. To synthesize microspheres for biomedical application, "two stage shot growth method" was developed. The uniformity ratio of synthesized microspheres was always smaller than 1.05. The system based on PC-486DX was designed to acquire medical images produced by medical imaging instruments and related audio information, that is, doctors' reporting results.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo). Dependence of Fo on Nk and Bi has been observed by experiment, and the results have been compared with the numerical results obtained by solving a 2-dimensional conduction equation. Fo and explosion depth (from the surface to the point of maximun temperature) are increased when Nk and Bi are increased.To find out the minimum laser power for explosive evaporative removal process, steady state analysis has been also made. Optimal stimulus parameters, which can minimize muscle fatigue and can achieve effective muscle contraction were determined in slow and fast muscles in Sprague-Dawley rats. Stimulus patterns of our stimulator were designed to simulate electromyographic activity monitored during locomotion of normal subjects. Muscle types of the lower extremity were classified according to their mechanical property of contraction, which are slow muscle (msoleus m.) and fast muscle (medial gastrocneminus m., rectus femoris m., vastus lateralis m.). Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. 보스웰리아 추출물의 골관절염 억제 효과 연구 (Effect of Boswellia serrata Extracts on Degenerative Osteoarthritis in vitro and in vivo Models)

  • 본 실험에서는 primary culture된 연골세포 in vitro 실험모델과 MIA로 유발한 골관절염 in vivo 실험모델을 이용하여 보스웰리아 추출물의 골관절염 예방 효과를 확인하였다. 먼저 MTT 시험법을 통해 세포 사용 적정농도를 $20{\mu}g/mL$ 이하로 결정하여 연골세포 사멸 억제를 확인하고, 이를 근간으로 골관절염 동물실험 모델에서 골관절염 예방 효과를 확인하였다. $H_2O_2$ 처리에 따른 산화적 독성으로 연골세포 사멸을 유도한 실험에서 보스웰리아 추출물은 유의적으로 세포사멸을 억제하였으며 이러한 효과는 $5{\sim}20{\mu}g/mL$ 사이농도에서 비교적 높게 나타났다. 세포실험에서는 골관절염 발병에 따라 관절연골에 영향을 미치는 염증인자에 대한 기전연구를 진행하였으며, 연골세포에 LPS를 처리하여 염증을 유도한 후 보스웰리아 추출물의 효과를 확인한 실험 결과 면역과 염증반응을 조절하는 nuclear transcription factor ${\kappa}B(NF{\kappa}B)$, 염증관련 cytokine IL-6, TNF-${\alpha}$의 발현이 모두 보스웰리아 추출물 처리 시 유의적으로 감소하는 것으로 나타났으며 특히 $20{\mu}g/mL$ 농도에서 가장 효과가 뚜렷하고 일관되게 확인되었다. 또한 이들 cytokine에 의해 생성되는 COX-2 발현과 COX-2에 자극 받아 생성이 촉진되는 PGE2 생성을 확인한 결과 역시 $20{\mu}g/mL$ 농도에서 가장 효과적으로 생성이 억제되어 염증을 조절하는 것으로 나타났다. 특히 보스웰리아의 기능성분으로 알려진 보스웰릭산(boswellic acids)에 의해 억제되는 5-LO의 경우, 염증반응 유발의 핵심인자를 생성하는 효소로 알려져 있으며 이를 직접적으로 저해하는 것으로 알려진 보스웰리아의 효능을 확인 하고자 실험을 진행하였다. 실험 결과 염증을 유도한 연골세포에서 보스웰리아 추출물의 처리에 따라 5-LO 활성이 감소하였으며, 이는 곧 보스웰리아 추출물이 염증을 유발 핵심효소인 5-LO 활성을 효과적으로 억제함으로써 연골세포 보호 효과를 나타낸 것이다. 세포실험에서의 기전 결과를 바탕으로 골관절염을 유발한 동물모델에서의 보스웰리아 추출물의 섭취에 따른 효과가 나타날 것으로 기대되어 관절염 유발 동물모델에서 보스웰리아 추출물의 효능검증 실험을 진행하였으며, 세포실험 결과 및 기존의 연구내용을 참고하여 동물에서 보스웰리아의 섭취 농도를 50 mg/kg, 100 mg/kg 및 200 mg/kg으로 결정하고 AIN-93G diet에 보스웰리아 추출물 분말을 섞어 보스웰리아 diet를 제작하여 실험기간 동안 제공하였다. 골관절염 유발 동물모델을 만들기 위해 SD rat의 관절강에 MIA를 injection 하였으며, 보스웰리아 추출물 섭취에 따른 관절염 예방 효과를 관찰하기 위해 관절염 유발 2주일 전부터 제작된 식이를 제공하고 유발 후 3주간 지속적으로 식이 제공 및 관찰하였다. 연골의 주요 구성 성분인 collagen 및 aggrecan은 골관절염 발병 시 여러 인자에 의해 분해되는 것으로 알려져 있으며, 골관절염 유발동물모델에서 collagen type I, collagen type II 및 aggrecan 유전자 발현을 실시간 정량 PCR로 측정하여 변화를 살펴보았다. 그 결과 골관절염 유발 sham군에 비해 보스웰리아 추출물 섭취군에서 유의적으로 collagen type I, collagen type II 및 aggrecan 발현이 증가하였으며, 특히 BW200군에서 CLX 약물대조군과 비슷한 수준으로 발현이 증가하여 관절연골의 보호 효과가 가장 좋은 것으로 확인되었다. 교원질 합성을 억제하고 분해를 촉진시키는 MMPs(MMP-3, MMP-9, MMP-13)의 발현을 실시간 정량 PCR로 측정하여 발현 변화를 살펴보았다. 그 결과 앞선 다른 실험 결과와 마찬가지로 보스웰리아 섭취군에서 MMPs 유전자 발현이 유의적으로 낮아졌음을 살펴볼 수 있었다. 특히 BW200군에서 MMPs 발현이 유의적으로 감소하였으며, 관절염에 효과적으로 사용되는 약물인 CLX 투여 양성대조군과 비슷한 수치를 나타내었다. 이상의 결과를 통하여 보스웰리아 추출물은 골관절염에서 관절연골의 보호 효과가 있으며, 이는 골관절염 발생 시 나타나는 염증발현의 기전적인 측면뿐만 아니라 골관절염 유발 동물에서 섭취 효능까지 모두 일관되게 나타나 골관절염에서의 기능성 소재로써 개발가능성이 충분할 것으로 생각된다. 또한 추후 실험을 통해 골관절염이 유발된 동물에서 보스웰리아 추출물 섭취 시 관절연골의 형태학적인 변화 및 골상태의 분석과 더불어 관절염 유발 동물의 관절연골 및 혈액학적 분석을 진행하여 동물에서 기전적 측면을 보완하고 보스웰리아 추출물 효능에 대한 추가적인 검증을 하고자 한다.
