Humanized Mice for the Evaluation of Francisella tularensis Vaccine Candidates

  • Francisella tularensis (FT), a highly infectious pathogen, is considered to be a potential biological weapon owing to the current lack of a human vaccine against it. Tul4 and FopA, both outer membrane proteins of FT, play an important role in the bacterium's immunogenicity. In the present study, we evaluated the immune response of mice - humanized with human CD34+ cells (hu-mice) - to a cocktail of recombinant Tul4 and FopA (rTul4 and rFopA), which were codon-optimized and expressed in Escherichia coli. Not only did the cocktail-immunized hu-mice produce a significant human immunoglobulin response, they also exhibited prolonged survival against an attenuated live vaccine strain as well as human T cells in the spleen. These results suggest that the cocktail of rTul4 and rFopA had successfully induced an immune response in the hu-mice, demonstrating the potential of this mouse model for use in the evaluation of FT vaccine candidates.

  • Combat vehicles need an air-conditioning unit, although new combat systems tend to use an integrated system for heating, cooling, and ventilating. The specifications of an air-conditioning unit depend on the combat vehicle's purpose. It is difficult to send cooling air from the air-conditioning unit to a gun turret through the drum basket because the gun turret rotates and consists of a closed anti-aircraft shell magazine. In this study, we considered an air-conditioning unit for armored combat vehicle based on the special requirements and military specifications. We evaluated the performance of the air-conditioning unit despite the rotating gun turret through analysis and tests in terms of flow improvement compared to the previous model.

  • In this paper, an integrated radar resource manager for an airborne multi-function radar and a radar-environment simulator are presented. The radar-environment simulator includes target detection/measurement models, a nonlinear tracking filter for the airborne radar and an effective target generation algorithm. The structures and functions of modules in the radar resource manager are established and validated by the radar-environment simulator.

An Experimental Study of Film Cooling Characteristics at Supersonic Free Stream Conditions (초음속 주유동 환경에서의 막냉각 특성 시험 연구)

  • In this paper, film cooling characteristics at supersonic free stream conditions were examined experimentally by applying an IR-thermography. Film cooling experiments were carried out in a free-jet facility at Mach number of 3.0 and with unit Reynolds number of $42.53{\times}10^6$ and $69.35{\times}10^6$ using wedge shaped film cooling model which has a converging film cooling nozzle. Film cooling efficiency was calculated by measuring the surface temperature of PEEK(Polyether Ether Ketone) and the effects of angle of attack and blowing ratios on the film cooling efficiency were examined. The measured wall temperature was significantly reduced by the film cooling flow compared with the results without the film cooling flow. The usefulness of film cooling was also confirmed by the surface heat flux calculated using the surface temperature history of PEEK. As the blowing ratio increases the protected area of PEEK was also expanded along the direction of free stream and film cooling flow.

Design and Test of an Assembly of Air Intake and Variable Geometry Inertial Separator for a Turboprop Aircraft (터보프롭 항공기용 흡입구 덕트 및 가변형 관성분리기 조립체 설계 및 시험)

  • A turboprop aircraft for this study is required to operate at icing condition in order that it performs its given mission. So an air intake system of the turboprop aircraft should be designed and verified not only to provide the maximum possible total pressure at engine inlet at normal flight condition, but also to include an inertial separator which protects Foreign Object Debris (FOD) like ice or snow at icing condition from entering into the engine inlet screen which can cause or lead an catastrophic engine failure like engine flame-out or severe damage. So an air intake assembly incorporating a variable geometry inertial separator has been designed and then CFD/structural analysis for the assembly was performed to see its design results. Then 35% scaled model of the air intake assembly was manufactured and wind tunnel test was done. This paper describes the detailed design results for the aerodynamic design, analysis and wind tunnel testing during the development process of the air intake assembly.

  • The ability to measure groundwater contaminant flux is increasingly being recognized as crucial in order to prioritize contaminated site cleanups, estimate the efficiency of remediation technologies, measure rates of natural attenuation, and apply proper source terms to model groundwater contaminant transport. Recently, a number of methods have been developed and subsequently applied to measure contaminant mass flux in groundwater in the field. Flux measurement methods can be categorized as either point methods or integral methods. As the name suggests, point methods measure flux at a specific point or points in the subsurface. To increase confidence in the accuracy of the measurement, it is necessary to increase the number of points (and therefore, the cost) of the sampling network. Integral methods avoid this disadvantage by using pumping wells to interrogate large volumes of the subsurface. Unfortunately, integral methods are expensive because they require that large volumes of contaminated water be extracted and managed. Recent work has investigated the development of an integral method that does not require extraction of contaminated water from the subsurface. We begin with a review of the significance and importance of measuring groundwater contaminant mass flux. We then review groundwater contaminant flux measurement methods that are either currently in use or under development. Finally, we conclude with a qualitative comparison of the various flux measurement methods.

Bacillus licheniformis Isolated from Korean Traditional Food Sources Enhances the Resistance of Caenorhabditis elegans to Infection by Staphylococcus aureus

  • We investigated whether Bacillus spp., newly isolated from Korean traditional food resources, influence the resistance of hosts to foodborne pathogens, by using Caenorhabditis elegans as a surrogate host model. Initially, we selected 20 Bacillus spp. that possess antimicrobial activity against various foodborne pathogens, including Staphylococcus aureus. Among the selected strains, six strains of Bacillus spp. used in preconditioning significantly prolonged the survival of nematodes exposed to S. aureus. Based on 16S rRNA gene sequencing, all six strains were identified as B. licheniformis. Our findings suggest that preconditioning with B. licheniformis may modulate the host defense response against S. aureus.

Design and Performance Analysis of Lateral Type MEMS Inertial Switch (수평 구동형 MEMS 관성 스위치 설계 및 성능해석)

  • A lateral type MEMS inertial switch was designed on the same principle as spring-mass system. The MEMS switch is used for arming mechanism of the arm-fire device by sensing the applied acceleration. We analyzed the switching capability of the MEMS switch under various acceleration conditions via performance model. Simulation results showed that the MEMS switch works very well at 10 g when the applied acceleration slope does not exceed 10 g/msec. On the other hand, the threshold operating acceleration level simulation exceeded the requirement (10±2 g) due to the width and length of the spring by considering 10% tolerance of the design values. Design modification of doubling the width of the spring, which is difficult to reduce less than 10% tolerance in fabrication process, was proposed after confirming the simulation results comply the requirement.

  • For hypersonic aircraft, increase of flight speeds causes heat loads that are from aerodynamic heat and engine heat. The heat loads could lead structural change of aircraft's component and malfunctioning. Endothermic fuels are liquid hydrocarbon fuels which are able to absorb the heat loads by undergoing endothermic reactions, such as thermal and catalytic cracking. In this study, methylcyclohexane was selected as a model endothermic fuel and experiments on endothermic properties were implemented. To improve heat of endothermic reaction, we applied zeolites and confirmed that HZSM-5 was the best catalyst for the catalytic performance. The objective is to investigate catalytic effects for heat sink improvement. The catalyst could be applied to system that use kerosene fuel as endothermic fuel.

Gpx3-dependent Responses Against Oxidative Stress in Saccharomyces cerevisiae

  • The yeast Saccharomyces cerevisiae has defense mechanisms identical to higher eukaryotes. It offers the potential for genome-wide experimental approaches owing to its smaller genome size and the availability of the complete sequence. It therefore represents an ideal eukaryotic model for studying cellular redox control and oxidative stress responses. S. cerevisiae Yap1 is a well-known transcription factor that is required for $H_2O_2$-dependent stress responses. Yap1 is involved in various signaling pathways in an oxidative stress response. The Gpx3 (Orp1/PHGpx3) protein is one of the factors related to these signaling pathways. It plays the role of a transducer that transfers the hydroperoxide signal to Yap1. In this study, using extensive proteomic and bioinformatics analyses, the function of the Gpx3 protein in an adaptive response against oxidative stress was investigated in wild-type, gpx3-deletion mutant, and gpx3-deletion mutant overexpressing Gpx3 protein strains. We identified 30 proteins that are related to the Gpx3-dependent oxidative stress responses and 17 proteins that are changed in a Gpx3-dependent manner regardless of oxidative stress. As expected, $H_2O_2$-responsive Gpx3-dependent proteins include a number of antioxidants related with cell rescue and defense. In addition, they contain a variety of proteins related to energy and carbohydrate metabolism, transcription, and protein fate. Based upon the experimental results, it is suggested that Gpx3-dependent stress adaptive response includes the regulation of genes related to the capacity to detoxify oxidants and repair oxidative stress-induced damages affected by Yap1 as well as metabolism and protein fate independent from Yap1.
