The Pharmacognostical Studies of Angelica Plants in Korea (II) A Classification Method of the Roots of some Anegelica Genus Plants by Thin Layer Chromatography (한국산 anglica 속식물의 생약학적연구 (II) 수종 Angelica 속식물근의 Thin Layer Chromatography를 이용한 감별법에 대하여)

  • The internal structures of "Chunho", "Kangwhal" and "Dangui" and their original plants had been compared with each other in the prior report. Coumarin derivatives, khellactone from the roots of Angelica purpuraefolia $C_{HUNG}$ and decursin and decursinol from A. gigas $N_{AKAI}$ were also isolated. In this report, we made a simple and convenient identification method for the powdered drugs of several Angelica genus by using a thin layer chromatogrophic technique. The ehter extract of the powdered drugs were applied to a silica gel plate, and was developed with n-hexane and ethyl acetate (1:1) mixture. The characteristic U.V. absorption patterns of each drug were shown.

Studies on the Coumarins of the Root of Angelica gigas $N_{AKAI}$ (I) (참당귀근의 coumarin성분에 관한 연구(I))

  • From the root of Angelica gigas $N_{AKAI}$ (Umbelliferae), known as "Tang Gui" in Korea, substance A (colorless needle crystal, $C_{14}H_{14}O_{4}$, m.p. $178^{\circ}$) and B (white plate crystal, $C_{19}H_{20}O_{5}$, m.p. $111^{\circ}$) were obtained. Acetylation of "A" gave its monoacetate, white needles, $C_{16}H_{16}O_{5}$. Saponification of "B" with 5% sodium hydroxide produced "A" and senecionic acid. Examination using I.R. and N.M.R. spectra shows that "A" is identical with decursinol and that "B" with decursin. The root also appears to contain five coumarin derivatives and two steroidal compounds.

  • The effect of tannic acid on GAs and cyclic-AMP promoted amylase induction in barley aleurone layers was examined. Of a variety of adenine compounds, only cyclic-AMP and ADP showed significant activity, and these activities were promoted by addition of theophylline to the incubation medium. When aleurone layers of barley endosperm tissues were incubated with GAs in the presence of tannic acid, the amylase activity in the incubation medium was reduced. Cyclic-AMP induced amylase activity was also reduced by additiion of tannic acid. The cyclic-AMP response promoted was more sensitive to tannin inhibition than GAs response. The inhibitory effect of tannic acid shwoed reversibility by addition of higher concentration of GAs or cyclic-AMP. The tannic acid effect on GAs response was also recovered by addition of a higher concentration of cyclic-AMP. Experiment with polyacrylamide disc electrophoresis showed different isozyme patterns according to the additions in the incubation medium. Inhibitory effects of decursinol and coumarin was compared with that of tannic acid. They showed different zymogram patterns.

  • Peucedanum terebinthaceum Fischer (Umbelliferae) known as "Gi Reum Na Mool" in Korea and used for the treatment of cough, phlegm, headache and common cold etc. The substances were isolated from the fruits of Peucedanum terebinthaceum and identified by UV, GLC, IR, NMR, Mass spectra and physico-chemical tests. The steroidal compounds from the fruits were identified as $\beta$-sitosterol (tR 33.01 min.) and stigmasterol (tR 28.87 min.) by GLC. Substance I (colorless prismatic crystal, $C_{19}H_{20}O_5$, mp $110~111^{\circ}$) was identified as decursin which is one of the pyranocoumarin. Hydrolysis of substance I with 5%-potassium hydroxide produced decursinol. Acetylation of decursinot gave its monoacetate, white needles, $C_{16}H_{16}O_5$. Substance II(light yellowish white needle crystal, $C_9H_6O_3$, mp $227~228^{\circ}$) was proved to be umbelliferone. Substance I, decursin, was isolated for the first time from the fruits of Peucedanum spp. (Umbelliferae).

  • During the preparation of Ssang Wha tea it was observed that the highest yield was obtained when it was extracted with 50% ethanol, of which guantity was added 10 times more then that of Sample. The most effective extraction temperature and time was $90^{\circ}C$ and 8hours on the water bath, which was resulted in saving of production cost. From the HPLC Analysis, it was also observed that product extracted with 50% ethanol give higher yield than that of exfrac ted with water.

  • Angelica gigas Nakai (AGN) is a traditional medicinal herb especially in Korea. It contains pyranocoumarins, which are major active components including decursin (D) and decursinol angelate (DA). This study was carried out to determine the change in active component content and antioxidant activity depending on the root diameter of AGN. Several processing steps are involved to use AGN roots as medicine. The dried AGN roots are divided into body (B), thick root (TkR), medium root (MR) and thin root (TnR) according to their diameter before cutting into medicine. The recovery rates of each root parts per 100 kg were measured as $32.3{\pm}2.5$, $9.0{\pm}1.0$, $39.3{\pm}2.1$ and $15.0{\pm}1.0%$, respectively and the mean diameters were measured as $51.95{\pm}4.55$, $7.05{\pm}0.89$, $2.88{\pm}0.49$ and $1.57{\pm}0.32mm$. Two index components, D and DA, were analyzed. The change of both D and DA content showed a similar tendency. Both D and DA content were increased as the root diameter decreased (higher in TnR). In addition, antioxidant activity was higher in B and TnR, and lower in TkR and MR than control. This study showed that the thinner the root diameter, the higher the D and DA content in AGN roots and that TnR has excellent antioxidant activity compared to other root parts, suggesting that the thinner root part of AGN could be used as a potential material.

  • This study was intended to find out the effects of the inorganic nutrients in the soil and root on the available constituents contents in Angelica gigas Nakai. The correlation between the contents of decursin and decursinol in roots of the wild and cultivated and the inorganic nutrients contents in the soils and roots was investigated. The contents of decursin in the dried root of the wild was higher than that of the cultivated. The ammonium nitrogen contents in soil and the decursin contents in root had the significant negative correlation, whereas the significant positive correlation was obtained the available phosphorus contents. The significant negative correlation was obtained between the decursin contents and the nitrogen contents of root, and the significant positive correlation between the decursin contents and the contents of phosphorus or potassium in roots.

Geographical origin discrimination of Korean variety, Angelica gigas Nakai by using TDU (Thermal Desorption Unit)-GC/MS (TDU-GC/MS를 이용한 한국품종 참당귀 국산 판별 기법)

  • The objective of this study was to develop a fast and accurate method of variety discrimination and geographical discrimination origin of Korean angelica (Korean variety, Angelica gigas Nakai) by using TDU-GC/MS. Two peaks of decursin and decursinol, which are coumarin derivatives were identified in the range of Total Ion Chromatogram (TIC) RT 26.9-27.2 of the Korean angelica by GC/MS analysis at the time of condensation in a refrigerated condensation system after thermal desorption of sample extracts. In case of Chinese angelica (Chinese variety), ligustilide peak was detected at the RT 17.2. In order to investigate the difference of volatile components according to the geographical origin of Korean variety, the mass spectra were measured by TDU-GC/MS at the range of m/z 40-400 amu. The TIC of domestic cultivation and Chinese cultivation of the Korean variety, Angelica gigas Nakai showed the same tendency as a whole. However, in partial scans of TIC, two peaks detected at 15.54 and 16.05 of RT showed different peak patterns between Korean angelica (Korean variety) cultivated in Korea and in China. The ratio of Peak A (RT 15.54) and B (RT 16.05) was 0.0-0.2 for domestic cultivation and 0.5-2.8 for Chinese cultivation, confirming the possibility of discriminating origin by comparing the TIC peak pattern of TDU-GC/MS.

Effect of Allium cepa (red) and Angelica gigas Nakai on Hair-growth Promotion in C57BL/6 Mice (C57BL/6 마우스에서 모발성장 촉진에 대한 Allium cepa (red)와 Angelica gigas Nakai의 효과)

  • This study was conducted to evaluate the hair-growth promotion effect in C57BL/6 mice of a new substance mixed with Allium cepa (red)-bioconversion extract and Angelica gigas Nakai. The ethanol extract of Allium cepa (red) was bioconverted through the use of the Bacillus subtilis KJ-3 (BS3) strain, which was named Red-BCQ. The quercetin content of Red-BCQ increased by about 7.4-fold after bioconversion. Angelica gigas Nakai extract (Agnex) contains a large amount of coumarins such as decursin (D) and decursinol angelate (DA). A 1 mg portion of Agnex contained 0.4146 mg of D and 0.3659 mg of DA. Minoxidil has been known to promote hair growth. In this study, the hair-growth promotion effects of Red-BCQ, Agnex, and a mixture of both Red-BCQ and Agnex were compared with 5% minoxidil. Twenty-five mice were divided into five experimental groups including saline (CON), 5% minoxidil (PCON), Red-BCQ (RA), Agnex (AG), and a Red-BCQ-Agnex mixture (RAG)-treated group. Samples were administered orally once a day at a fixed time for 4 weeks. Hair growth was monitored by photograph at 7, 14, 21, and 28 days. We also observed 5α-reductase, alkaline phosphatase (ALP), γ-glutamyl transpeptidase (γ-GT), insulin-like growth factor (IGF)-1, transforming growth factor (TGF)-β1, antioxidant enzyme, and the hair follicles of the skin tissue. In all the results, the RAG-administered group showed greater antioxidative and hair-growth promotion effects than the other groups. These data suggest that RAG has potent stimulating activity on hair growth in C57BL/6 mice.

Effects of Nitrogen Fertilization on Leaf Yield and Pyranocurmarine Accumulation in Angelica gigas Nakai

  • Angelica gigas Nakai is one of the most widely used herbal medicines and is known to have many pharmaceutical effects including an anti-oxidant, anti-cancer etc. This study was carried out to investigate an effect of fertilization on leaf yield, production of dry-matter and accumulation of pyranocurmarine compounds such as decursin (DE) and decursinol angelate (DA) in Angelica gigas Nakai. Effect of fertilization was determined from response surface regression equation composing of 2 by 3 factorial arrangement of urea, sodium dihydrogen phosphate and potassium chloride. Yield of leaf in Angelica gigas Nakai significantly increased until 100 days after transplanting. Production of leaf also tended to increase with increasing nitrogen fertilization. Model of regression equation showed that leaf production depended upon nitrogen ($Pr>{\mid}t{\mid}$ : 0.087, 0.256 and 0.079). Also, statistical results between nitrogen application level and production of dry-matter showed significant relationship (p<0.05) and contents of dry-matter was highest in 10 kg 10a-1 treatment on 24 Sep. Active compound isolated and purified from leaf and root of Angelica gigas Nakai was identified as DE and DA by gas chromatograph-mass spectrophotometry (GC-MS). Concentration of DA as prevalent compound in leaf was highest on 20 Aug. but decreased on 24 Sep. Amount of DE and DA accumulated in Angelica gigas Nakai significantly increased with growth stages and nitrogen level. The result of our investigation imply that nitrogen fertilization is important factor for production of leaf and accumulation of pyranocurmarine in Angelica gigas Nakai as a medicinal/food materials.
