Objective : In benign prostatic hyperplasia, dihydrotestosterone acts as a potent cellular androgen and promotes prostate growth. Inhibiting enzyme $5{\alpha}$-reductase that is involved in the conversion of testosterone to the active form dihydrotestosterone reduces this excessive prostate growth. The mechanism on benign prostatic hyperplasia is substantiating evidence to support the clinical value in the evaluation of therapeutic efficacy. In this study, we investigated the effects of Lygodium japonicum on cyto-pathological alterations and expression of $5{\alpha}$-reductase in the rat model of benign prostatic hyperplasia induced by castration and testosterone treatment. Methods : Sprague-Dawley rats were treated with testosterone after castration for induction of experimental benign prostatic hyperplasia, which is similar to human benign prostatic hyperplasia in histopathological profiles. Lygodium japonicum as an experimental specimen, and finasteride as a positive control, were administered orally. The prostates were evaluated by histopathological changes and testosterone levels. Also, the prostates were observed by hematological alterations of AST, ALT, ${\gamma}$-GTP, BUN and creatinine. Results : The rats treated with Lygodium japonicum showed a diminished range of luminal cell and duct epithelial cell damage. The stromal elements and connective tissue proliferation of Lygodium japonicum treated group as compared to the control group decreased. Conclusions : These findings suggest that Lygodium japonicum may protect the glandular epithelial cells. We concluded that Lygodium japonicum could be a useful remedy agent for treating the benign prostatic hyperplasia.
In order to find the effects of testis function on adrenal gland, immature male rats were administered by 20 mg of testosterone propionate with 10 days interval for 50 days, and their adrenal weights and histological changes were compared with those of normal ones, and the results obtained were as follow: Adrenal weight was increased with the period of treatments, and especially the significant difference was recognized after 30 days. Of the zones in adrenal gland, the portion of zona fasciculata and reticularis increased significantly after 20 days and 30 days, respectively, while that of adrenal medulla was decreased significantly after 20 days. In histological changes, hypertrophy and vacuolization were observed in zona fasciculata and reticularis after 20 days, but no changes were recognized in zona glomerulosa and adrenal medulla.
The frequencies of chromosomal aberrations, unmerical variations at various time intervals and DNA synthetic patterns after the treatment with steroids in synchronized human kidney cells treated with 5-AU were investigated in the present experiment. 1. In 5-AU treated group, the frequency of chromosomal aberrations per cell was 0.131, 3 times of control group. In 5-AU + progesterone and 5-AU + testosterone groups, the frequency of chromosomal aberrations per cell was 0.340 and 0.452 respectively. 2. In 5-AU treated group, the frequency of cells with abnormal chromosome number was 0.8%, which was distributed throughout the time regardless of time interval. In 5-AU + progesterone and 5-AU + testosterone groups, the frequencies of cells with abnormal chromosome number were 2.2% and 4.3% respectively and they increased with the time. In 5-AU + progesterone group, the frequency of chromosomal aberrations exhiited the peaks at 12 and 18 hour stage after the treatment with steroids and, in 5-AU + testosterone group, it decreased with the time and in 5-AU treated group no significant difference was observed 3. The increase of labeled metaphases and labelling intensities in 5-AU treated cells are the result of the accumulation of cells at S stage by 5-AU. The decrease of labeled metaphases, labelling intensities and the delay of DNA synthetic time were observed in steroid groups. DNA synthetic pattern of sex chromosomes differs according to the step of cell cycle and DNA synthetic time is irregular because of double treatments with 5-AU and steroids.
The testicular volume measured by a Prader orchidometer was compared with sperm count in semen, the levels of serum FSH, LH, testosterone, prolactin, estradiol and progesterone in 59 nonobstructive infertile men. 1. The causes of infertility were primary hypogonadisms in 50 patients (35 unknown, 9 Klinefelter syndromes, 6 varicocels) and secondary hypogonadism in 9 (5 isolated FSH deficiencies, 1 hyperprolactinerriia, 3 pituitary hypogonadisms). 2. Decreased levels of serum FSH (less than 4 mIU/ml) did not correlate with testis volume but increased level of serum FSH (more than 20 mIU/ml) were mostly noted in the testis less than 10ml. 3. Decreased level of serum testosterone (less than 3 ng/ml) were distinguishably noted in the atrophied testis less than 5 ml. 4. There was no correlation between the testicular volume and the levels of serum prolactin, estradiol, and progesterone. Coclusively, testicular volume less than 11 ml suggests poor spermatogenesis, but normal testicular volume dose not nessarily rule out poor spermatogenesis. Function of Leydig cell is relatively well preserved in atrophied testis of 5 to 10 ml comparing with that of seminiferous tubule.
Hypogonadism is a clinical syndrome that results in hormone deficiency and can be classified as 1) primary caused by the gonadal failure and 2) secondary by the hypothalamus-pituitary gland dysfunction and/or cardiometabolic complications. Recently the presence of thyroid hormone receptors in different testicular cell types was demonstrated, and thus thyroid dysfunctions would be another cause of secondary hypogonadism. Thus, we investigated the effects of perinatal hypothyroidism on hypogonadism in male Sprague-Dawley rats. Perinatal hypothyroidism was induced by daily administration of 0.05% 6-propyl-2-thiouracil (PTU) by tap water from gestation day 15, which were compared with negative control (PTU (-)) group. At postnatal day 28, hypothyroid pups were divided into 2 groups: PTU (+) group - continued PTU treatment and PTU (+/-) group - stopped PTU until postnatal day 49. Body weights, dehydrotesosterone (DHT), and testosterone levels were checked 2 and 3 weeks after grouping. Body weights were significantly decreased in PTU(+) and PTU(+/-) groups compared with PTU (-) group at postnatal day 28. 3 weeks later, PTU (+/-) group significantly gained weight compared with PTU (+) group. DHT and testosterone levels significantly decreased with PTU treatment, but increased 3 weeks after stopping PTU administration. Perinatal PTU-induced hypothyroid hypogonadism was sustained for 2 weeks after stopping PTU administration, but restored gonadal hormone levels 3 weeks after stopping PTU. These results suggest that researchers should design an experiment on hypothyroid hypogonadism based on the estimated period.
Globally, esophageal cancer is the seventh most common cancer, and the male-to-female ratio in esophageal adenocarcinoma (EAC) is significantly imbalanced at 4:1 to 8:1. Obesity, reflux, and smoking are known risk factors for this sex difference; however, fully explaining this remains challenging. Studies have investigated the link between exogenous sex hormones and esophageal cancer occurrence. A meta-analysis revealed a lower risk of EAC in female who had undergone hormone replacement therapy. Androgen-deprivation therapy in patients with prostate cancer was associated with a decreased risk of EAC. Tissue-based studies have reported varied results regarding the relationship between estrogen receptor expression and survival in female patients with esophageal squamous cell carcinoma (ESCC). Circulating hormone studies have suggested that higher testosterone and luteinizing hormone levels decreased EAC risk in men, and free testosterone was inversely correlated in female with ESCC. However, a high androgen-estrogen ratio in male patients with EAC was linked to increased odds of EAC. Sex hormones influence carcinogenesis, affecting cell proliferation, differentiation, metabolism, inflammation, and cell death. The studies were limited by the small sample size and varying hormone measurement methods; thus, future studies with definitive conclusions on the association between esophageal cancer and sex hormones are warranted.
The present study was carried out to establish several spermatotoxicity test methods. For this purpose we investigated following parameters in the fertility study of DA-125, a new anticancer agent, in rats: testicular spermatid counts, epididymal sperm counts, daily sperm production rate, sperm morphology, and serum testosterone concentration. Motility and velocity of sperms were also measured using non-treated rats. At 0.3 mg DA-125/kg, spermatids per 1g testis and daily sperm production rate per 1g testis were significantly decreased, when compared with those of control group. Several types of abnormal sperms, such as no head, pin head, double head, hook at wrong angle, no tail, and small sperm, were found in both treated and control groups at a low frequency. Serum testosterone concentration at 0.3 mg DA-125/kg was close to the control value. Sperm motility and velocity measured with non-treated rats were in a good agreement with the results of other investigators. In our study established spermatotoxicity test methods can be used as a tool not only for the close examination of the cause of drug- or chemical-induced infertility, but also for the effective evaluation of reproductive toxicity.
The hair growth effect of the oriental medicine extract which is composed of Polygoni multiflori Radix, Angelica gigantis Radix and Lycii Fructus was studied biochemically and histologically. The study was conducted dividelly into three groups, control, propecia and oriental medicine extract and three groups were compared each other in skin androgen, testosterone (T) and dihydrotestosterone (DHT), content biochemically and the number of hair follicular unit and hair follicle histologically. The oriental medicine extract and propecia decreased skin DHT conetent and increased skin total (T+DHT) content compared with control. In transverse sections of skin specimens the oriental medicine extract and propecia increased total counts of follicular units and follicles compared with control. On the basis of the result, the oriental medicine extract has same hair growth effect as propecia and it is suggested that the oriental medicne extract is capable of a therapeutic agent of alopecia.
We treated KunYangDan (KYK) in SD rats in order toexamine the protective effect against cell damage induced by diethylhexyl phthalate (DEHP). KYD reduced DEHP toxicity by increasing the function of immune cell numbers weight increase of spleen.red blood cells,HB,and HCT content,sperm number and mobility,resulting in improving reproductive function by judging from the recovery of testosterone content. Interestingly the hormone change of testosterone by KYD significantly recovered the decreased its leve. Taken together,these results suggest that KYD specifically affect the reproductive function induced by DEHP,an endocrine disruptor.

Allium tuberosum Rotter(Liliaceae) is a perennial herb of which leaves are used for food. Leek has been reported to have pharmacological effects including alleviations of abdominal pain, diarrhea, hematemesis, snakebite, and asthma. To investigate the effect of dietary leek supplementation on the drug-metaboizing enzymes, quinone reductase(QR) and arylhydrocarbon hydroxylase(AHH) activities in the liver, stomach, small intestine and lung, and on the plasma testosterone and dihydrosterone hormone levels, mice were fed 2% and 5% leek diets for 8 weeks. Quinone reductase, an anticarcinogenic enzyme, was significantly induced in stomach, small intestine, and lung but slightly lowered in hepatic tissue in the experimental groups compared to control group. Arylhydrocarbon hydroxylase activity, involved in bioactivation of procarcinogens, was significantly decreased in liver and lung. Leek feeding led to the reduction in the plasma level of dihydrotestosterone which is associated with the incidence of prostate cancer. These findings support the potential chemopreventive activity of leek supplementation.
