AN EXPERIMENTAL STUDY ON TISSUE REACTIONS OF ALLOGENEIC SCIATIC NERVE GRAFT IN RAT (백서 좌골신경의 동종이식후 조직반응에 관한 실험적 연구)

  • Nerve allografts as a bridge of regeneration is useful in the repair of peripheral nerve defect resulting from trauma, and leprosy. But immunological rejection and complicated scar formation is an unavoidable problem in the application of allogeneic nerves. This article is intended to study of the regeneration of allogeneic nerve grafts in rats with histopathologically, scanning electron microscopically. 24 adult male Sprague-Dawley rats were used as the experimental animals. A 2cm skin incision was made on the lateral aspects of limb, parallel to femur. Segments of sciatic nerve trunk taken from rats, 10mm was resected at the middle of the thigh, nerve graft was inserted between the ends of gaps with perineural and epineural suture method with 10-0 prolene. Obsrevation was made simultaneously at 3 day, 1, 2, 3, 4, 5, 6, 8 weeks after surgery. The results were as follows. 1. In light and electronic microscopic studies, marked degenerative change of the graft nerves were observed at 2 weeks after surgery. 2. After surgery, blood clot fromation was observed at 3 day, granualtion tissue formation was observed at 2 week, and fibrous tissue proliferation was observed at 3 week. 3. In change of nerve fiber, there were Wallerian degeneration at early stage, decrease in degeneration at 4 week but degeneration of myeline was continuded at 8 week. 4. At 4 week, schwann cells proliferate at its cut ends to join with the distal and proximal stump of the damaged nerve. 5. Fibrous scar tissues are formed at 2 weeks and increased progressively in 8 weeks, which was interrupted the regeneration of grafted nerve.

Effect of a 12-week weight management program on the clinical characteristics and dietary intake of the young obese and the contributing factors to the successful weight loss

  • BACKGROUND/OBJECTIVES: The objectives were to investigate the effect of a 12-wk intervention with behavioral modification on clinical characteristics and dietary intakes of young and otherwise healthy obese and to identify factors for successful weight loss. The goal was to lose 0.5 kg per week by reducing 300-500 kcal/day and by increasing physical activities. SUBJECTS AND METHODS: Forty four obese subjects (BMI > 25) and 19 normal weight subjects (BMI 18.5-23) finished the 12-week intervention. Obese subjects participated in 5 group educations and 6 individual counseling sessions. Normal weight subjects attended 6 individual counseling sessions for evaluations of dietary intake and exercise pattern. Anthropometric and clinical characteristics and 3-day dietary records were evaluated at baseline and week12. RESULTS: Weight and serum triglyceride and free fatty acid concentrations in obese group decreased significantly with intervention. Intakes of energy, fat, and cholesterol decreased significantly in the obese. Active participation, realistic weight loss goal setting, and weight gain after high school graduation not during childhood were identified as key factors for successful weight loss. CONCLUSIONS: The 12-week intervention with behavioral modification resulted in reduced energy and fat intakes and led to significant weight loss and improvements of clinical characteristics in the obese. The finding that those who became obese during childhood lost less weight indicates the importance of 'early' intervention.

Bacterial attachment and penetration to Tetracycline-treated resorbable and nonresorbable membranes for GTR (테트라싸이클린 처리된 흡수성 및 비흡수성 조직유도재생술용 막에의 세균부착과 침투양상)

  • The barrier membranes for GTR procedure could be affected bY bacterial contamination after exposure to oral environment. This study was done to evaluate whether the tetracycline impregnated barrier membranes could inhibit bacterial attachment and penetration into membranes. The resorbable membrane(polylactic and polyglycolide copolymer, $Resolute^{(R)}$, W.L Gore and Associates, Inc..USA) and the non-resorbable membrane(e-PTFE; Gore-TexTM, W.L. Gore & Associates, Inc.,USA) were cut into 4mm discs and trated with 5% tridodecylmethylammonium chloride solution in ethanol and dried in air. The membranes were immersed in tetracycline(TC) solution (100mg/ml, pH 8.0) and dried. To the maxillary canine-premolar region in six periodontally healthy volunteers, removable acrylic devices were inserted, on which 8 cylindrical chambers were glued with TC impregnated and non-impregnated discs, the membrane discs were examined for bacterial attachment and penetration, and structural changes under SEM and LM. From the 1st day to the 7th day, membranes showed bacterial plaque formation composed of cocci and rods. Thereafter, filamentous bacteria appeared and the plaque thickness increased. The TC impregnated e-PTFE membranes showed less bacterial attachment and delayed in bacterial plaque maturation than non-treated membranes. As for bacterial penetration, the TC impregnated e-PTFE membranes showed superficial invasion and infrequent presence of bacteria in unexposed inner surface at the 4th week. while the non-treated e-PTFE membranes showed deep bacterial invasion at the 2nd week and frequent presence of internal bacteria at the 4th week. The resorbable membranes started to be resorbed at the 2nd week and were perforated at the 4th week, regardless of TC treatment. In conclusion, bacterial plaque formation and penetration was efficiently delayed in TC impregnated e-PTFE membranes, whereas resorbable membranes were similar in bacterial invasion due to membrane degradation and perforation, regardless of TC treatment.

  • The purpose of this study was to evaluate the sealing properties of the temporary filling materials used in endodontic treatment Access cavities were prepared in 135 extracted human molar teeth. Then, cotton pellets were placed in the pulp chamber until the depth of 5 mm for the temporary filling materials; Caviton, zine oxide-eugenol, double sealing (A) (stopping 15 mm + zinc oxide - eugenol, 3.5mm ) double sealing (B) (stopping 3.0 mm + zinc oxide - eugenol 2.0 mm) and gutta percha stopping. After filling the materials, the teeth were immersed in 1 % methylene blue solutions for 3 days, 1 week and 2 weeks. Then thermal cycling was performed at the temperature of $60^{\circ}C$ and $4^{\circ}C$, followed by longitudinal sections on the center of tooth. Finally, staining on the cotton pellet was evaluated. The following results were obtained. 1. Stopping showed lower marginal sealing quality than Caviton, zinc oxide - eugenol and double sealing. 2. In 1 week group, Caviton showed higher marginal sealing quality than zinc oxide-eugenol, double sealing and stopping. 3. Caviton and double sealing (B) showed a great decrease in marginal sealing quality with the increse of time. 4. Caviton had high marginal sealing quality in 3 day group and 1 week group, but in 2 week group, Caviton showed a great decrease. 5. Double sealing (B) showed fairly high marginal sealing quality in 3 day group, but decreased greatly after 1 week on.

Reduction of long-term potentiation at Schaffer collateral-CA1 synapses in the rat hippocampus at the acute stage of vestibular compensation

  • Vestibular compensation is a recovery process from vestibular symptoms over time after unilateral loss of peripheral vestibular end organs. The aim of the present study was to observe time-dependent changes in long-term potentiation (LTP) at Schaffer collateral-CA1 synapses in the CA1 area of the hippocampus during vestibular compensation. The input-output (I/O) relationships of fEPSP amplitudes and LTP induced by theta burst stimulation to Schaffer's collateral commissural fibers were evaluated from the CA1 area of hippocampal slices at 1 day, 1 week, and 1 month after unilateral labyrinthectomy (UL). The I/O relationships of fEPSPs in the CA1 area was significantly reduced within 1 week post-op and then showed a non-significant reduction at 1 month after UL. Compared with sham-operated animals, there was a significant reduction of LTP induction in the hippocampus at 1 day and 1 week after UL. However, LTP induction levels in the CA1 area of the hippocampus also returned to those of sham-operated animals 1 month following UL. These data suggest that unilateral injury of the peripheral vestibular end organs results in a transient deficit in synaptic plasticity in the CA1 hippocampal area at acute stages of vestibular compensation.

Estimate of laying performance of two crossbred Korean native chickens in 20 to 40 weeks

  • This study was conducted to investigate the laying period performance of two crossbred Korean native chicken (KNC) grand parent stock (GPS) layer during 20 to 40 weeks. A total of 351 20-week-old KNC GPS were allocated in a completely randomized design to give 4 birds per each cage. The chickens were fed commercial diets [i.e., week 20 - 32, crude protein (CP) 18.0% and metabolizable energy (ME) 2,850 kcal/kg; week 32 - 40, CP 17.0% and ME 2,800 kcal/kg)] and fresh water during the whole experiment period ad libitum basis. Body weight, age of sexual maturity, egg weight, hen-day egg production (HDP) and feed conversion ratio (FCR) were measured during the experiment period (week 20 - 40). The results show no significant difference in the Body weight, age of sexual maturity, egg weight, and hen-day egg production (HDP) except for egg weight. 1B had a higher egg weight on week 36 only. We expect that the reason why most factors showed no significant difference is the lack of nutrients caused by limited feeding which follows the standard feeding system for the grand-parent stock. These results provide basic information for laying data in 2-way crossbred Korean native chickens.

EFFECT OF WORK CRUSHING SUGARCANE ON PREGNANCY AND LACTATION IN CATTLE AND BUFFALOES

  • The suitability of female cattle ($Brahman{\times}Holstein$) and buffaloes (Murrah / Jafarabadi) for work on a sugarcane crusher was tested. The animals were fed chopped sugarcane tops and elephant grass supplemented with rice bran and urea-molasses blocks. In experiment 1, six cattle and six buffaloes, (with three animals seven to eight months pregnant at the start of the experiment in each group), worked individually, alternate weeks for 90 minutes a day for 6 d/week. Within the 90 min, cattle did more work than buffaloes (p < 0.05), and non-pregnant animals did more than pregnant animals, but cattle crushed significantly (p < 0.05) more cane per day than buffaloes. During work animals lost weight, but gained the weight back during resting weeks, when fodder intakes were higher. There were no significant differences in live weight change and fodder intakes between pregnant and non-pregnant animals during the six weeks. In experiment 2, four pairs of lactating cows and buffaloes crushed sugarcane for 3 h/d, 6 d/week for three weeks. Work done was higher in cattle than buffaloes, but cane crushed was marginally lower. All animals lost weight during work, particularly in the first week, but gained weight during the week after work. Daily milk yield was lower during the working weeks ($-0.55{\pm}0.15kg$) than in the week before work. Butter fat yield showed no significant change. One buffalo showed ovarian activity and oestrus behaviour during the work. Two cows showed ovarian activity within one month after the work. It was concluded that pregnant and lactating cows and buffaloes can effectively operate a cane crusher, however a temporary reduction in milk yield can be expected in the working period, and ovarian activity may be suppressed.

The Effect of Virtual Reality-based Sitting Balance Training Program on Ability of Sitting Balance and Activities of Daily Living in Hemiplegic Patients (앉기자세에서 실시한 가상현실 균형훈련이 뇌졸중 환자의 앉기 균형 및 일상생활활동 능력에 미치는 영향)

  • Purpose : This study examined the effects of balance training using virtual reality program on sitting balance ability and activities of daily living (ADL) in stroke patients. Method : In the study, 20 patients with hemiplegia were divided into two groups: experiment group (EG) of 10 patients and control group(CG) of 10 patients. The two groups received general occupational therapy for 30 minutes, per day, at a rate of 5 times per week for 6 weeks. The EG was additionally conducted which was performed virtual reality balance training and the CG was conducted general occupational therapy balance training for 30-minutes, once a day, 3 times a week for 6 weeks. Result : The evaluations of this study included: limit of stability(LOS), modified Functional Reach Test(mFRT), and modified Barthel Index(MBI). The patients were evaluated before and after their six week training programs. Significant differences in the LOS, mFRT, MBI were found between pretest and posttest scores in both the EG and CG groups(p<.05). Also, LOS, mFRT, MBI were significant different between the groups at post-test(p<.05). Conclusion : The study findings suggest that virtual reality balance training can improve sitting balance and ADL ability in stroke patients.

Immunity of the Buffalo Mammary Gland during Different Physiological Stages

  • To study the immunity of the buffalo mammary gland during involution and around parturition and compare it with the mastitic mammary gland, milk samples were collected from 9 Murrah buffaloes during the above critical periods. SCC of buffalo milk increased significantly (p<0.01) by day 21 of involution and one week prepartum. SCC was significantly higher around parturition but became normal at 14 days postpartum. Phagocytic activity (PA) and phagocytic index (PI) of the buffalo milk neutrophils decreased as the duration of the dry period increased. Elevated levels of immunoglobulins at calving improved the PA and PI, but the lowest PA of 18.8% and PI of 1.75 were recorded at 7 days postpartum. Buffaloes suffering from clinical mastitis had PA of 12.3% and PI of 1.46 that increased significantly (p<0.01) on the third day of treatment. Distance of teat from ground level was found to be minimum at one week before parturition. The investigation showed that in vitro phagocytic activity of buffalo neutrophils was weakest at one week postpartum.

The Different Growth Rate by Predation Risk in Larval Salamander, Hynobius leechii (한국산 도롱뇽의 포식압 유, 무에 따른 성장률의 변화)

  • This study examined the different growth rate of larval salamander Hynobius leechii by the predation risk. We collected salamander's eggs from Mountain Inwang. Eggs were treated by two different conditions: (1) cue - which had a predation risk three times a day; (2) no cue - which had no predation risk. Predation risk was conducted by using chemical cue from Chinese minnows. The chemical cue treatment started from the next day after the collection and ended one week after the hatching. Between the first week and the second week after the hatching we fed salamander larvae with tubifex ad. libitium. After the treatment phase, we measured snout - vent length of the each larva on the first week and the second week after the hatching. The results showed that there was a significant difference in the growth rate by the predation risk. From these results we found that predation risk can cause the different growth rate to the larval salamander and these changes could be affect their mortality.
