Examination of ivermectin residues in raw milk after skin administration (원유중 Ivermectin 구충제의 잔류실태 조사)

  • This study was conducted two kinds of aims: 1) to modify the analytical methods (conditions) by high performance liquid chromatography - fluorescence detector for the detection of residual ivermectin in raw milk, 2) to provide basic information for the evaluation of standard of the residual ivermectin in raw milk. It could be considerable that negative ion spectra can be better method in the LC/MS analysis for the detection of residues, Characteristic daughter ions were observed in negative ion spectra, however, linear line was not formed in positive ion one. Three Holstein cows ($500{\pm}10kg$) were applied to commercial ointment of ivermectin just one time at the first day of test, and residues in raw milk were examined for 20day after administration. The limit of detection (LOD) was 0.65ng (n=5) by HPLC/FLD, and recovery rates were $87.85%{\sim}99.47%$. The peak was observed at the 4th day, and residues lasted to the end. Thus ivermectin was prohibited when lactating.

Theoretical Calculation on Alpha Track Density by Using an Electrostatic Ion Spectrometer (정전기분광분석방법에 의한 알파입자비적밀도의 이론적 계산연구)

  • To develop a technique of theoretical alpha track density calculation for comparison with measured track density, an electrostatic ion spectrometer was specially designed and fabricated. The mobility spectrum of first radon daughter(Po-218) in the range of $0.07{\sim}5.0cm^2/V\;s$ from the radon chamber was measured using-the electrostatic ion spectrometer. Measurement was taken in a radon chamber operated using dry particle free air passed through silica gel, activated charcoal and molecular sieve filters. The mobility of a new-born Po-218 ion measured by the electrostatic ion spectrometer was determined to be $1.92cm^2/V\;s$. A comparison of the theoretical and measured alpha track densities was completed and uncertainties concerning the shape of the spectrum were analyzed. It was found that the discrepancies in track densities are primarily Que to the neglect of wall loss of ions in the theoretical track density calculation.

  • The reaction of ethylenecarbonate (I) with bromine was carried out in the presence of benzolperoxide as radical initiator. The following several different esters being ring opened were obtained; bromoacetyl-bromoformate, (1-hyroxy, 1, 2-dibromo)-ethyl bromoformate, (1-hydroxy, 1, 2, 2'-tribromo)diethylacarbonate, 2-bromoethyl-tribromoacetate, (1-acetoxy, 1'-bromomethyl)-bromomalo nate, 2-bromoethyl-bromoacetoxy-tribromoacetate.

Formation of Ultra fine Particle by the Polonium-218 Ions under Different Humidity Conditions (다른 습도조건하에서 Po-218 이온들의 극소입자형성에 관한 연구)

  • A number of investigators have reported the formation of the radiolytic ultrafine particles produced by the interaction of ionizing radiation with water vapor. Previous studies have suggested that a very high localized concentration of the OH radical produced by the radiolysis of water can react with trace gas like organic vapors and produce lower vapor pressure compounds that can then nucleate. In order to determine water vapor dependence of the active, positively charged, first radon daughter(Po-218), an experiment was conducted using a well-controlled radon chamber. The activity size distribution of the radon daughter in the range of 0.5-100nm was measured using the parallel graded wire screens system. Measurements were taken for different relative humidity. The resultant activity size distributions were analyzed. The addition of water vapor to the radon carrier gases resulted in the formation of ultrafine particles by OH radicals formed by radon radiolysis. It may be due to the neutralization of charged Po-218 ion with water vapor through the radio lysis.

Keratanase II Digestion Accompanied with a Liquid Chromatography/Tandem Mass Spectrometry for Urinary Keratan Sulfate Quantitative Analysis

  • Purpose: Mucopolysaccharidosis IV (MPS IV) is a disease characterized by deficient activity of N-acetylgalactosamine-6-sulfatase (GALNS) causing excessive lysosomal storage of keratan sulfate (KS). The identification of the relevant disaccharide units of KS after keratanase II digestion followed by liquid chromatography/tandem mass spectrometry detection (LC-MS/MS) is validated and applicable for the preliminary diagnosis of MPS IV. Methods: A total of 67 urine samples were collected and analyzed from 11 MPS IV patients comprising 10 MPS IVA and one MPS IVB patients, and 56 normal controls. Urinary glycosaminoglycan was first precipitated by the Alcian blue method followed by a digestion of keratanase II. The protonated species of the digested disaccharide products were detected by using multiple reaction monitoring experiment. Results: One particular disaccharide of KS was selected. The transition mass-to-charge (m/z) of the parent ion and its daughter ion after collision was $462.0{\rightarrow}97.0$, whereas the chondrosine used as an internal standard in this assay was m/z $353.9{\rightarrow}73.0$. The results corresponded well with the two-dimensional electrophoresis method. The quantities of urinary KS were significantly raised in confirmed MPS IV patients when comparing with those of normal controls ($170.2{\pm}81.1$ vs. $4.06{\pm}1.92{\mu}g/mL$). Conclusion: The LC-MS/MS method for MPS IVA determination is specific, sensitive, validated, and applicable for urinary KS quantification. This method can be used not only as a first-line biochemistry examination of MPS IVA, but also as an outcome survey after enzyme replacement therapy.

Synthesis and Fragmentation Behavior Study of n-alkyl/benzyl Isatin Derivatives Present in Small/Complex Molecules: Precursor for the Preparation of Biological Active Heterocycles

  • N-Alkyl/benzyl substituted isatin derivatives are intermediates and synthetic precursors for the preparation of biological active heterocycles. N-alkyl/benzyl isatins have showed various biological activities, such as cytotoxicity, antiviral, caspase inhibition, cannabinoid receptor 2 agonists for the treatment of neuropathic pain, etc. In this study, N-alkyl/benzyl isatin derivatives were synthesized from isatin and alkyl/benzyl halides in presence of K2CO3 in DMF and excellent to quantitative yields (~95%) were obtained. Isatins and benzyl-isatins were condensed with fluorescein hydrazide to form fluorescein hydrazone. All the compounds were subjected to their fragmentation behavior study using LC/MSn. N-Alkyl substituted isatin derivatives fragmented at nitrogen-carbon (N-C) bond, hence gave daughter ion as [RN+H]+. Whereas, N-benzyl substituted isatin derivatives fragmented at carbon-carbon (C-C) bond of alkyl chain which linked with nitrogen molecules, therefore gave N-methyl fragments [RNCH2]+. This study demonstrated that, isatin moiety present in a small/large molecule or in a matrix of reaction mixture with/without N-alkyl/benzyl substituents can be identified by mass spectroscopic fragmentation behavior study.

Mineral Absorption by Cymbidium Jungfrau in the Solution Culture (서양 심비디움 양액재배에서의 무기물 흡수)

  • $N(^{15}N)$ and $P(^{32}P)$ absorption by 2 year-old Cymbidium Jungfrau in solution culture were investigated. Growth, photosynthesis rate, chlorophyll content and mineral composition of Cymbidium in the solution culture with bark or granular rockwool were compared with these parameters in the conventional pot culture. Nitrogen absorption by Cymbidium was higher in full sunlight than in 60% of sunlight while P absorption was higher in 60% of sunlight. Sixty seven % of N absorbed in plant was redistributed to the bulb(39%) and leaves(28%) while 46% of P absorbed was fund in the bulb (36.2%) and leaves (10.2%). Accumulation of P in leaves was 3-fold lower than that of N. N and P absorption in 0.5 or 1 year- old daughter plant Bowing vigorously were greater than in immature daughter or mother plant. The absorption rate of phosphorus in Cymbidium was 350-fold lower than that of barley. Greater shoot length and bulb diameter, and higher fresh weight, photosynthesis rate and chloroployll content were observed in the solution culture than in the conventional pot culture. Solution culture had-also more content of N, P, K and Mg in leaves, bulb and root than conventional pot culture but did not that of Ca. A large part of the nutrient absorption was occurred during vegetative growth. Also, There was no difference between bark and rockwool in the solution culture due to the improvement of poor dispersion of nutrient solution in bark.

  • A number of investigators have reported formation of radiolytic ultrafine particles produced by the interaction of ionizing radiation with atmospheric trace gases. Previous studies have suggested that a very high localized concentration of the hydroxyl radical produced by the radiolysis of water can react with atmospheric trace gases such as $SO_2$ and produce lower vapor pressure compounds that can subsequently nucleate. To determine the trace gas and water vapor concentration dependence of the active, positively charged, first decayt product of radon (Po-218), a well-controlled radon chamber was used in this research. The mobility spectrum of the decay products in the range of $0.07-5.0cm^2/V\;sec$ from the radon chamber was measured using alpha track detector installed inside a specially-designed electrostatic spectrometer. Measurements were taken for different concentrations (0.5ppm to 5ppm) of $SO_2$ in Purified, Compressed air. A kinetics Study following the clustering of $SO_2$ around the $PoO_x^+$ ion in an excess of $SO_2$ for interpretation of the reaction processes was performed.

Recent Advancement in the Stem Cell Biology (Stem Cell Biology, 최근의 진보)

  • Stem cells are the primordial, initial cells which usually divide asymmetrically giving rise to on the one hand self-renewals and on the other hand progenitor cells with potential for differentiation. Zygote (fertilized egg), with totipotency, deserves the top-ranking stem cell - he totipotent stem cell (TSC). Both the ICM (inner cell mass) taken from the 6 days-old human blastocyst and ESC (embryonic stem cell) derived from the in vitro cultured ICM have slightly less potency for differentiation than the zygote, and are termed pluripotent stem cells. Stem cells in the tissues and organs of fetus, infant, and adult have highly reduced potency and committed to produce only progenitor cells for particular tissues. These tissue-specific stem cells are called multipotent stem cells. These tissue-specific/committed multipotent stem cells, when placed in altered environment other than their original niche, can yield cells characteristic of the altered environment. These findings are certainly of potential interest from the clinical, therapeutic perspective. The controversial terminology 'somatic stem cell plasticity' coined by the stem cell community seems to have been proved true. Followings are some of the recent knowledges related to the stem cell. Just as the tissues of our body have their own multipotent stem cells, cancerous tumor has undifferentiated cells known as cancer stem cell (CSC). Each time CSC cleaves, it makes two daughter cells with different fate. One is endowed with immortality, the remarkable ability to divide indefinitely, while the other progeny cell divides occasionally but lives forever. In the cancer tumor, CSC is minority being as few as 3-5% of the tumor mass but it is the culprit behind the tumor-malignancy, metastasis, and recurrence of cancer. CSC is like a master print. As long as the original exists, copies can be made and the disease can persist. If the CSC is destroyed, cancer tumor can't grow. In the decades-long cancer therapy, efforts were focused on the reducing of the bulk of cancerous growth. How cancer therapy is changing to destroy the origin of tumor, the CSC. The next generation of treatments should be to recognize and target the root cause of cancerous growth, the CSC, rather than the reducing of the bulk of tumor, Now the strategy is to find a way to identify and isolate the stem cells. The surfaces of normal as well as the cancer stem cells are studded with proteins. In leukaemia stem cell, for example, protein CD 34 is identified. In the new treatment of cancer disease it is needed to look for protein unique to the CSC. Blocking the stem cell's source of nutrients might be another effective strategy. The mystery of sternness of stem cells has begun to be deciphered. ESC can replicate indefinitely and yet retains the potential to turn into any kind of differentiated cells. Polycomb group protein such as Suz 12 repress most of the regulatory genes which, activated, are turned to be developmental genes. These protein molecules keep the ESC in an undifferentiated state. Many of the regulator genes silenced by polycomb proteins are also occupied by such ESC transcription factors as Oct 4, Sox 2, and Nanog. Both polycomb and transcription factor proteins seem to cooperate to keep the ESC in an undifferentiated state, pluripotent, and self-renewable. A normal prion protein (PrP) is found throughout the body from blood to the brain. Prion diseases such as mad cow disease (bovine spongiform encephalopathy) are caused when a normal prion protein misfolds to give rise to PrP$^{SC}$ and assault brain tissue. Why has human body kept such a deadly and enigmatic protein? Although our body has preserved the prion protein, prion diseases are of rare occurrence. Deadly prion diseases have been intensively studied, but normal prion problems are not. Very few facts on the benefit of prion proteins have been known so far. It was found that PrP was hugely expressed on the stem cell surface of bone marrow and on the cells of neural progenitor, PrP seems to have some function in cell maturation and facilitate the division of stem cells and their self-renewal. PrP also might help guide the decision of neural progenitor cell to become a neuron.
