Altered acetylcholine (Ach) homeostasis is associated with loss of viability in flies, developmental defects in mice, and cognitive deficits in human. Here, we assessed the importance of Ach in Caenorhabditis elegans development, focusing on the role of Ach during dauer formation. We found that dauer formation was disturbed in choline acetyltransferase (cha-1) and acetylcholinesterase (ace) mutants defective in Ach biosynthesis and degradation, respectively. When examined the potential role of G-proteins in dauer formation, goa-1 and egl-30 mutant worms, expressing mutated versions of mammalian $G_o$ and $G_q$ homolog, respectively, showed some abnormalities in dauer formation. Using quantitative mass spectrometry, we also found that dauer larvae had lower Ach content than did reproductively grown larvae. In addition, a proteomic analysis of acetylcholinesterase mutant worms, which have excessive levels of Ach, showed differential expression of metabolic genes. Collectively, these results indicate that alterations in Ach release may influence dauer formation in C. elegans.
In order to investigate an endocrinological effect on metamorposis of the midgut in silkworm, Bombyx mori, 20-hydroxyecdysone and rude extract of brain (PTTH) were injected into pupal isolated abdomens, devoid of prothoracic glands and also in the brainless diapausing pupae with the prothoracic glands (so-called dauer pupae). When ecdysone was assayed abdomens, drastic morphological changes occurred in the midgut. The wet weights of midguts were found decrease as like the normal pupae at metamorphosis. Proteineous changes in epithelial midgut were also observed; pupal specific proteins (PSP) vanished while the adult proteins were newly synthesized. Similar results were also found in the changes of protein in the midgut of dauer pupae by injecting ecdysone and PTTH.

Uranium resources of Mongolia are generally confined to sediments deposited during Jurassic to Cretaceous volcanism. Territory of Mongolian uranium deposits is divided into four districts as follows; Mongol-Priargun, Gobi-Tamtsag, Hentii-Dauer, North-Mongolian. Potential uranium deposits were discovered by Airborne Gamma ray Spectrometric Survey(AGSM). One of them, Haraat deposit, which was interested to us, has been under detailed survey for exploitation by one of American companies, Concord company. The Erdes uranium mine is partly operated by about hundred Russian staffs at the open pit, while underground mining facilities such as the main hoist are almost closed. Ore minerals of the Erdes Mine are coffinite and pitchblende. Uranium content in ore ranges from 0.06% to 1%, averaging 0.2%. Ore reserves of uranium ore in the Dornod deposit including the Erdes Mine accounts 29,000 ton. It is reported that Uranium resources of Mongolia are 1,471,000 ton.

Bei Verurteilung zu Freiheitsstrafe wegen eines Wahldelikts ${\ddot{A}}mter$ sich die Dauer des Verlusts der $Amtsf{\ddot{a}}higkeit$ und des aktiven bzw. passiven Walhrechts auf 10 Jahre. We investigate Korean learners' speech-timing of English before and after instruction in comparison with native speech, in an attempt to resolve disagreements in the literature as to whether speech-timing is measurable (Lehiste, 1977; Roach, 1982; Dauer, 1983 vs. Low et al., 2000; Yun 2002; Jian, 2004). We measured the pair-wise variability between the adjacent stressed and unstressed syllables within a foot as well as that among adjacent feet in approximately 555 English sentences, which were read by 29 native speakers and 41 Korean learners in the intermediate proficiency level. The results show that in comparison with native American English, Korean learner speech is before instruction significantly (p<.001) smaller for the pair-wise variability between the adjacent stressed and unstressed syllables within a foot; and significantly (p=.01) bigger for the variability among adjacent feet within the utterance. The learner speech after instruction showed significant (p=.01) improvement in the pair-wise variability of syllable sequence toward native speech values. The variability among adjacent feet was progressively smaller for learner speech before and after instruction and for native speech (p=.03). We thus conclude that the speech timing difference between Korean English and American English is measurable in terms of the duration. of stressed and unstressed syllables and that the latter is stress-timed and the former is syllable-timing interfered.

Targeted degradation of proteins through ubiquitin-mediated proteolysis is an important control mechanism in various cellular processes. The process of ubiquitin conjugation is achieved by three enzyme complexes, among which the ubiquitin ligase complex (E3) is in charge of substrate specificity. The SCF (SKP1-CUL1-F-box) family portrays the largest and the most characterized member of the E3 ligases. For each SCF complex, the ubiquitination target is recognized by the F-box protein subunit, which interacts with the substrate through a unique C-terminal domain. We have characterized a novel F-box protein CFL-1 that represents a single LRR-type F-box (FBXL) in the Caenorhabditis elegans genome. CFL-1 is highly homologous to FBXL20 and FBXL2 of mammals, which are known to regulate synaptic vesicle release and cell cycle, respectively. A green fluorescence protein (GFP)-reporter gene fused to the cfl-1 promoter showed restricted expression around the amphid and the anus. Modulation of CFL-1 activity by RNAi affected the time interval between defecations. RNAi-treated worms also exhibited reduced tendency to form dauer when exposed to daumone. The potential involvement of CFL-1 in the control of defecation and pheromone response adds to the ever expanding list of cellular processes controlled by ubiquitin-mediated proteolysis in C. elegans. We suggest that CFL-1, as a single LRR-type F-box protein in C. elegans, may portray a prototype gene exerting diverse functions that are allocated among multiple FBXLs in higher organisms.
Nematodes were isolated using silkwom trap through the investigation of 100 soil samples from various biotopes in Korea. The 30 nematode strains from soil and dead insects by the pathogenicity aganinst silkworms (Bombyx mori mori) and insect pests of Calliphora vomitoria, Pseufazetia separata, Palomena angulosa, and Melolontha incana. Mortailty of the silkworm larvae and pupae were as high as 100% by nematode infection, those of insect of pests were varied from 20 to 100%. The 30 strains of entemopathogenic nematodes were classified into five groups of Rhabditidae, Diplogatroidae, Heterorhabitidae, Steinernematidae, and Tylenchida by morphological criteria. The genetic relationships among the 30 nematode strains were analyzed by various RAPD bands with twenty primers. The 30 nematode strains were classified into six major subgroups on the basis of the genetic similarity coefficient of 0.853. The grouping by RAPD was agree with those of morphological taxa in discrimination of the higher group, however, was not completely agree in the subgroup. The family Steinernematidae belong to Rhabditida was clarified as closer to the Tylenchida, rather than the other Rhabditida of Heterorhabitidae, Rhabditidae, and Diplogatroidae in genetic distance valule. From the result of the morphological classification and RAPD of the genomic DNA showed that genetic relationship analysis furnish infurmation on phylogenetic classification and relationships of entomopathogenic nematodes. The application of genetic similarity will overcome the limitation of taxonomy and classification of morphologically simple nematode. Several primers were confirmed those utility of identification for individual nematode strains, the methods of molecular genetics secured the simplicity, rapidity and accuracy on the selection of entomopathogenic nematodes.
