Datura Stramonium Intoxication of Which Resulted in Changing Mentality - A Case Report - (흰꽃독말풀 열매 달인 물 복용 후 발생한 의식 변화 환자 1례)

  • Datura stramonium is a wild growing plant, consumption of which can result in severe toxicity. It contains a variety alkaloids include atropine, hyoscamine, and scopolamine that can all cause anticholonergic poisoning. It has been use of herb medicine in Korea. Side effects from ingesting Datura stramonium include dry mouth, blurred vision, photophobia, and may be followed by hyperthermia, confusion, agitation, hallucination, and aggressive behavior. In severe toxicity, it can cause seizure, coma, life-threatening arrhythmia, and death. We experienced one patient who developed stupor mentality caused by Datura stramonium. Severe presentation, especially mental change of Datura stramonium should draw attention. Poisoning associated with Datura stramonium can be prevented through educational public health program.

  • In order to elucidate the optimum medium on the growth and tropane alkaloid production of hairy root, hairy root were induced by inoculating leaf and stem of Datura stramonium var. tatula Torr. with Agrobacterium spp. Both Agrobacterium tumefaciens $\textrm{A}_{4}$ T and A, rhizogenes ATCC 15834 among tested strains were effective on hairy root formation. Among 23 clones selected in SH (Schenk and Hildebrandt, 1972) liquid medium, DTLA9 clone was shown fast growth of hairy root and DTLE6 clone was shown high level production of tropane alkaloids. When both DTLA9 and DTLE6 clones were cultured in the GD (Gresshoff and Doy, 1972) medium, alkaloid production was higher than in 8 tested media. It was elucidated that optimum medium for root proliferation and for tropane alkaloid production is SH, GD medium, respectively.

  • The optimum concentrations of pH, sucrose and vitamins for the growth and tropane alkaloid production of hairy root clone DTLA9 (best growth line) were investigated. The optimum pH in growth and tropane alkaliod production of DTLA9 clone in SH (Schenk and Hildebrandt, 1972) basal medium without growth regulator were pH 6.3 and 6.5, respectively. Also, the optimum sucrose concentration in growth and tropane alkaliod production in the same medium were 3.0 and 2.8%, respectively. The optimum concentrations of ascorbic acid, D-pantothenate, nicotinic acid, pyridoxine, riboflavin, and thiamine on the growth of DTLA9 clone in SH basal medium without vitamins were 0.1 mM, 0.003 mM, 0.07 mM, 0.002 mM, 0.025 mM, and 0.01 mM, respectively. In particular, supplement of 0.1 mM ascorbic acid to SH basal medium without vitamins stimulated the tropane alkaloid production.

  • Tobacco (Nicotiana tabacum) cultivars including NC 82 and KF 114, and Datura stramonium, Physalis floridana, Gomphrena globosa, and Chenopodium spp. were added to the previous host plants tested for the further examination on the biological characteristics of tobacco mosaic virus (TMV) strains isolated from tobacco (TMV-Common), tomato (TMV-Tomato), and pepper (TMV-Pepper), In TMV-Tomato and TMV-Pepper, different symptoms were noted in P. floridana (no symptom development), and NC 82 (local lesion production on the inoculated leaves) from TMV-Common with which systemic mosaic symptoms were developed. Local lesions were developed in KF 114, D. stramonium, G. globosa, and Chenopodium spp by TMV-Common and TVM-Tomato, while no symptom was observed in KF114 and G. globosa. Also the number and size of local lesions were smaller in KF 114 than Xanthi-nc tobacco (local lesion host) infected with TMV-Tomato. Systemic necrosis was induced in Xanthi-nc and KF 114 when infected with TMV-C at high temperature, but not with the other strains.

  • A strain of Cucumber mosaic virus(CMV) was isolated newly from sweet pepper(Capsicum annuum var. angulosum) showing necrosis with large necrotic spots on fruits and vein banding with malformation on leaf at Cheongdo area in Gyeongsangbukdo. The new strain was designated as a CMV-NP and the shape of virus particles was isometric of 26 nm in diameter from the sweet pepper fruit by Dip method. The strain of CMV-NP was identified genetically by VC/RT-PCR. CMV-NP could infect systemically on the 9 indicator plants including Cucumis sativus, but it could infect locally on Chenopodim amaranticolor and C. quinoa. CMV-NP induced the specific symptoms of necrotic rings on the inoculated and the upper leaves of N. rustica and Tetragonia expansa. On Cucumis sativus, the large chlorotic ring and vein chlorosis were produced on the upper leaves. CMV-NP had no virulence on Datura stramonium.

  • Sterol compositions of three solanaceous plant seed oils i.e., tobacco(Nicotiana tabacum L.), datura (Datura stramonium L.) and Chinese lycium (Lycium chinense Mill.) were analyzed by thin layer chromatography, gas liquid chromatography and gas liquid chromatography-mass spectrometry. Cholesterol, cholest-7-enol, campesterol, 24-methylenecholesterol, stigmasterol, sitosterol, and 28-isofucosterot were identified in 4-des-methylsterol fraction. 31-Norlanost-8-enol, $4{\alpha}$-methylcholest-8-enol, lophenol, 31-norlanosterol, obtusifoliol, 31-norcycloartenol, cyloeucalenol, gramisterol (24-methylenelophenol), and citrostadienol in 4-monomethylsterol fraction, and lanost-8-enol, cycloartanol, lanosterol, ${\beta}$-amyrin, 24-methylenelanost-8-enol, cycloartenol, lupeol and 24-methylenecycloartanol in 4,4-dimethylsterol fraction were identified respectively.

  • The glycosylation pattern of Erythropoietin (EPO), produced by recombinant CHO cells, was studied using the simple and rapid technique of 'Lectin-blotting'. In this experiment we used three different kinds of lectins, MAA(Maackia amurensis agglutinine), RCA(Ricinus communis agglutinine), and DSA(Datura stramonium agglutinine), which bind to the terminal sialic acid, galactose, and the N-acetyllactosamine chain respectively. The lectin-blotting technique was used to analyze the carbohydrate structure of EPO produced in the presence of two physiologically active chemical compounds, ammonium and chloroquine. The effect of the ammonium ion on the glycosylation of EPO was studied because it accumulated in the medium mainly as a by-product of glutamine matabolism. Ammonium chloride significantly inhibited the sialylation of the terminal galactose residue at concentrations of 8mM or more. Chloroquine, a potent inhibitor of glycosylation, inhibited terminal sialylation at concentrations of 100 and 200 $\mu$M, and at a concentration of 300 $\mu$M, also inhibited Nacetyllactosamine chain synthesis.

Studies on Virus Diseases of Orchids in Korea (우리나라 란 바이러스에 관한 연구)

  • Orchids have been propagated vegetatively for a long time without adequate control measures against virus diseases in Korea. As a result, it is presumed that most of the orchid varieties in Korea may have been degenerated. Nevertheless there has been little work on the virus diseases of orchids in Korea. Therefore studies were initiated to isolate an4 characterize the orchid viruses occurring in Korea. The results obtained are summerized as follows. 1. Symptoms of virus diseases on orchid varieties can be grouped 1) mosaic, 2) necrotic streak with mosaic, 3) ring necrosis, 4) chlorotkc ring and 5) necrotic spot. 2. A total of 102 orchid plants representing 4 genera were investigated on the occurrence of Cymbidium mosaic virus and tobacco mosaic virus by serological agar-gel double diffusion test. The test revealed that approximately $45\%$ of the orchids were infected with Cymbidium mosaic virus. None of the plants were found to be infected with tobacco mosaic virus. 3. Local lesions appeared on the inoculated leaves of Chenopodium amaranticolor Cassia occidentalis and Datura stramonium 7-12 days after mechanical inoculation with Cymbidium mosaic virus. 4. Physical properties of the Cymbidium mosaic virus determined by inoculation on Chenopodium amaranticolor were as follows: Thermal inactivation Point; $75-80^{\circ}C$, dilution end Point; $10^{-5}-10^{-6}\%$ aging in vitro; 8 days. 5. Three different buffers at pH 7.0 and pH 9.0 were compared for the efficiency of agar-gel double diffusion test with Cymbidium mosaic virus. Phosphate, imidazol and tris buffer at pH 7.0 gave equally satisfactory results. 6. Electron microscopic examination of the Cymbidium mosaic virus revealed rod shaped particles measuring 460-580mu.

  • Tomato spotted wilt virus (TSWV) was occurred on the three vegetables of egg plant (Solanum melongena), whole radish (Raphanus acanthiformis) and sugar loaf (Cichorium intybus) at Anyang area infested with TSWV. Whole radish was produced the symptoms of necrotic spots on the leaves, and necrosis and malformation on the roots by TSWV. Egg plant was induced the symptoms of typical multiple ring spots on the leaves and necrotic rings on the fruits. Sugar loaf was infected severely with the typical symptoms of ring spots on the leaves and stunt. The three isolates of TSWV could infect locally on the indicator plants of Chenopodium amaranticolor, C. quinoa and Nicotiana debney, and systemically on N. glutinosa, N. benthamiana and Datura stramonium. Two TSWV isolates from egg plant and sugar loaf were very similar in virulence. However, the virulence of TSWV from whole radish was very different as local infection on 5 Nicotiana species including N. tabacum 'Xanthi NC'.

Characterization of an Isometric virus Infecting Paprika (Capsicum annuum var. glosum) in Korea

  • An Isometric virus was isolated from Paprika (Capsicum annuum var. glossum) showing necrosis spot and malformation on the fruit and the leaves, respectively, at yecheon in Korea. The virus could infect locally on Chenopodium amaranticolr, C. quinoa, Petunia x hybrida and Nicotiana glutinosa, but could not infect on Gomphrena globosa and Physalis floridana. The virus could infect systemically on red pepper and Lycopersicon esculentum. Datura stramonium, N. cleuarandii, N. rustim and N. tabacum cvs. were produced necrosis or necrotic ring spot lesions on the inoculated leaves and mosaic, vein necrosis or lethal death on the upper leaves. The virus was not related serologically to cucumber mosaic virus (CMV). In RT-PCR assay, it could not detected with specific primers of CMV and BBWV-II. The virions contain one molecule of genomic RNA, Which was approximately 3.8Kb and the coat protein (CP) of the purified virion migrated as a single band with molecular wight of about 29KDa in SDS-PAGE.
