Construction and validation of a synthetic phage-displayed nanobody library

  • Nanobodies derived from camelids and sharks offer unique advantages in therapeutic applications due to their ability to bind to epitopes that were previously inaccessible. Traditional methods of nanobody development face challenges such as ethical concerns and antigen toxicity. Our study presents a synthetic, phage-displayed nanobody library using trinucleotide-directed mutagenesis technology, which allows precise amino acid composition in complementarity-determining regions (CDRs), with a focus on CDR3 diversity. This approach avoids common problems such as frameshift mutations and stop codon insertions associated with other synthetic antibody library construction methods. By analyzing FDA-approved nanobodies and Protein Data Bank sequences, we designed sub-libraries with different CDR3 lengths and introduced amino acid substitutions to improve solubility. The validation of our library through the successful isolation of nanobodies against targets such as PD-1, ATXN1 and STAT3 demonstrates a versatile and ethical platform for the development of high specificity and affinity nanobodies and represents a significant advance in biotechnology.

Development of Optimum Management System for Irrigation Facilities (수리시설물 최적관리 시스템 개발)

  • For the efficient operation and maintenance( 0 & M), irrigation facilities were graded on the basis of their indices related to the 0 & M condition and the characteristics of reservoirs managed by Farmland Improvement Association(FIA) were examined. The results obtained are summarized as follows : 1. Characteristics of reservoirs include effective storage capacity, irrigation area, basin area, height and length of bank. Each characteristic is scored by the principle component analysis method. Variables which compose one index are categorized on the basis of their unit and each variable is scored so that the score of each unit sums to 100 for each index. 2. The Optimum MAnagement System of Irrigation Facilities(OMASIF) for Pyoungtaek area was developed by connecting general data of the irrigation facilities to image data. The database system is divided into three tables; LookUp Table, Facility Table, and Image Table. Image Table is again divided into five sub-tables, Image Table, Construction Cost Table, Acreage Table, 0 & M Cost Table, and Specification Table. 3. The evaluating criteria for the 0 & M of irrigation facilities can be established using the OMASIF. Irrigation facilities evaluated as poor state by the criteria should be repaired.

  • The effects of bank crest width, slope steepness, soil properties, and soil compactness on the characteristics of levee breach due to overlfow were investigated through a series of experiments. Generally, the major factors influencing the breach phenomenon are compactness, soil properties. crest width, and slope steepness, in that order. Using proper soil, and ensuring enough compactness in bank construction are very essential for extending breach duration and reducing peak overflow ranges for the values of breach duration, breach width, width-to-depth ratio, and side slope of the breach section, proposed by Singh, MacDonald and Fread based on the field data for earth dam breaks were reasonable. We found that those criteria could also be applied to the phenomenon of levee breaching.

  • 세계 수자원 시장은 연평균 6.5%씩 증가하고 있으며, 2025년 기준 1,038조원까지 급성장할 것으로 전문가들은 예상하고 있으나, 기후변화의 가속화로 인해 가뭄 및 홍수피해가 증가하여 수자원 관리가 더욱 어려워져지고 있다. 급속도로 변화는 환경에 대비하기 위해 국내에서는 수자원 및 기후변화에 대한 다양한 연구가 진행되고 있으나, 해외사업 진출 시에는 수자원 기초자료 수집에 대한 어려움을 겪고 있다. 이를 해결하기 위해 글로벌 수문기상자료 제공과 함께 GIS정보, 댐 및 저수지 관련 자료, 인문사회 자료, 물관련 통계자료, 물관련 재해자료 등을 웹으로 제공하는 글로벌 수자원 정보 시스템(GWB, Global World Bank)을 개발하고자 한다. 본 연구에서는 시스템의 통합 DB를 구축하고 관리방안을 도출하기 위해 수집된 메타데이터 속성 및 데이터구조를 파악하고, 세부항목별 자료 포맷을 분석 후 GIS기반 관측소 정보와 자료를 매칭하여 최종적으로 시스템 컨텐츠별로 DB를 맵핑하였다. 강수량과 기상자료는 33개국의 관측소 6,531개소의 일/월/연단위 관측자료와 10,977격자의 격자분석자료를 구축하였다. 수문자료는 33개국의 수문관측소 2,242개의 월/연단위 유량관측자료와 10,977격자의 월/연단위 직접유출, 기저유출, 잠재증발산의 격자분석자료를 구축하였다. 그리고, 수집된 강우와 기상자료는 기계 오작동, 자료 전송 오류 등으로 인한 결측치 및 이상치에 대해 자료품질분석을 통해 오자료에 대한 보정을 진행하였다. 해당자료는 MySQL를 활용하여 DB를 구축하였으며, GIS정보는 GeoServer를 활용하여 운영서버에 구축된 정보를 최종적으로 사용자에게 Web Browser로 표출하였다. 해당 시스템은 추후 전지구 수자원관련 정보를 제공하여 해외사업지역의 댐이나 보 등의 구조물 설계, 수자원산업의 해외 진출시 데이터 수집의 한계점 및 시간단축을 해결할 수 있어 수자원 분야에 기여 할 수 있을 것으로 판단된다.

Construction of PANM Database (Protostome DB) for rapid annotation of NGS data in Mollusks

  • A stand-alone BLAST server is available that provides a convenient and amenable platform for the analysis of molluscan sequence information especially the EST sequences generated by traditional sequencing methods. However, it is found that the server has limitations in the annotation of molluscan sequences generated using next-generation sequencing (NGS) platforms due to inconsistencies in molluscan sequence available at NCBI. We constructed a web-based interface for a new stand-alone BLAST, called PANM-DB (Protostome DB) for the analysis of molluscan NGS data. The PANM-DB includes the amino acid sequences from the protostome groups-Arthropoda, Nematoda, and Mollusca downloaded from GenBank with the NCBI taxonomy Browser. The sequences were translated into multi-FASTA format and stored in the database by using the formatdb program at NCBI. PANM-DB contains 6% of NCBInr database sequences (as of 24-06-2015), and for an input of 10,000 RNA-seq sequences the processing speed was 15 times faster by using PANM-DB when compared with NCBInr DB. It was also noted that PANM-DB show two times more significant hits with diverse annotation profiles as compared with Mollusks DB. Hence, the construction of PANM-DB is a significant step in the annotation of molluscan sequence information obtained from NGS platforms. The PANM-DB is freely downloadable from the web-based interface (Malacological Society of Korea, http://malacol.or/kr/blast) as compressed file system and can run on any compatible operating system.

An Analysis for the Economic Impact of Forest Road Investment (임도시설 투자의 경제적 파급효과 분석)

  • Forest road is an essential infrastructure for forest management such as the composition and management of forest resources, timber and forest byproduct production & transportation. It has recently been utilized forest recreation and forest sports as well as also forest pest control, forest fire prevention and evolution. When you build a forest road, the economic function in the forest is activated, so that it can result in the ripple effect of induced employment, value-added creation and production inducement. The purpose of this study is to analyze the impact caused by forest road construction occurring as the overall economy. For analysis it was applied to inter industry analysis method that is a method for analyzing the quantitative cross-correlation. The data were used in the Input-Output Tables In 2014, the Bank of Korea. When you build a forest road, economic effect due to the construction of the forest road is generated and economic effects are also generated due to the increase in the production of forest products after the construction of the forest road. Therefore, we will analyze the economic impact of the two effects. The estimated economic value of forest products, which is the economic effect of forest product cultivation, was calculated through some assumptions and the economic ripple effect was analyzed. The forest road construction sector is defined as land clearing and reclamation, and irrigation project construction and the forestry forest products sector is defined as the sum of raw timber, edible forest products and misc. forest products. In total, 32 sectors were classified, and except for the two sectors defined as forest road construction and forestry forest products, the remaining sectors were integrated according to the classification system of 30 integrated classifications of the Bank of Korea. As a result, the production inducement coefficient for forest construction was analyzed to be 2.767 and the production inducement coefficient for forestry forest products was analyzed to be 1.565. This means that 2,767 times the production of forest road construction investment is induced in the whole industry and the production of 1.562 times the amount of forestry forest products is caused by the whole industry as the production of forestry forest products increases. The value added inducement coefficient for forest road construction was 0.977 and the value added inducement coefficient for forestry forest products was 0.985. Forest road are essential infrastructure for forestry development and should be continuously invested because they are essential elements of timber production and forest byproduct production with functions such as forest management, forest recreation, forest sports, and town connection.

  • Transaction value of business-to-business(B2B) is composed of various factors for groups and time series. In this paper, we use the panel data for finding various variables and using this we analyse the factors that is major influence to transaction value of business-to-business. For analysis we looked at transaction value of business-to-business of 7 groups such as manufacturing industry, electric, gas and piped water industry, construction industry, retail & wholesale trade, traffic industry, publish, image; broad-casting & telecommunication and information service industry, etc. In our analysis we looked at the transaction value of business-to-business during the period from 2005.01 to 2009.12. We examined the data in relation to the transaction value of cyber shopping mall, company bond, composite stock price index, transaction value of credit card, loaned rate of interest in deposit bank, rate of exchange looking at the factors which determine the transaction value of business-to-business, evidence was produced supporting the hypothesis that there is a significant positive relationship between the transaction value of cyber shopping mall, composite stock price index and loaned rate of interest in deposit bank, rate of exchange. The company bond is negative relationship, transaction value of credit card is positive relationship and they are not significant variables in terms of the transaction value of business-to-business.

Effect of Artificial Changes in Geographical Features on Local Wind (인공적 지형변화가 국지풍에 미치는 영향)

  • The effect of artificial changes in geographical features on local wind was analyzed at the construction site of bridge and fill-up bank in the southern part of Haui-do. Geographic Information System (GIS) data and Computational Fluid Dynamics (CFD) model were used in this study. Three-dimensional numerical topography based on the GIS data for the target area was constructed for the surface boundary input data of the CFD model. The wind observations at an Automatic Weather Station (AWS) located in Haui-do were used to set-up the model inflows. The seasonal simulations were conducted. The differences in surface wind speed between after and before artificial changes in geographical features were analyzed. The surface wind speed decreases 5 to 20% at the south-western part and below 2% of the spatial average for salt field. There was also marked the effect of artificial changes in geographical features on local wind in the westerly wind case for the target area.

  • The present study evaluated the blocking effect of natural and artificial topography on sunshine duration in the southern coastal area of Haui-do. The geospatial data for the target area was constructed by geographic information system(GIS) data. Three-dimensional modeling based on solar azimuth and altitude angles was conducted for the assessment of sunshine environment. The sunshine area was evaluated over 80~90% of the target area in the daytime, especially in summer. The blocking effect of mountainous terrain on sunshine duration was presented at the northern residential area in the late afternoon. There was also the effect of artificial topography by construction of fill-up bank on sunshine environment at the southern residential area early in the morning and the south-western part of salt field in the late afternoon.

2D Flood Simulation for Estimating the Economic Loss in the Building Areas

  • 2D hydraulic models of urban areas are at the forefront of current research of flood inundation mechanisms, but they are constrained by inadequate parameters of topography and insufficient data. In this paper a numerical model based on DEMs is presented to represent overflow waters due to bank break in urban areas. The surface flow in the building areas is assumed to be properly modeled by solving Saint-Venant equation. In order to represent flooding broken out in Samcheok city, 2002, hydraulic model test using tracer has been carried out and validated. These efforts will serve for making flood hazard map and for estimating economic loss due to inundation of personal properties in urban areas.
