Regulation of Acid Contents in Kiwifruit Irradiated by Various Wavelength of Light Emitting Diode during Postharvest Storage (다양한 파장의 LED 조사를 통한 참다래 과실의 산 함량 조절)

  • The physiological roles of various wavelength of light emitting diode (LED) on ‘Hayward’ kiwifruit experiencing after-ripening were investigated. Various wavelengths from LED light source were irradiated on kiwifruits kept in plastic bags or under open air at $25^{\circ}C$. During two weeks of storage, firmness of Hayward kiwifruits was decreased by $25^{\circ}C$ treatment than by $4^{\circ}C$ treatment. In the $25^{\circ}C$ storage condition, the firmness of kiwifruits was decreased by the treatment of 380 nm UV and 470nm white LED light source. Sweetness of kiwifruits treated with 380 nm UV LED and dark condition at $25^{\circ}C$ increased higher than $15^{\circ}$Brix. The acidity of kiwifruits under open air was decreased 52% by incubating at $25^{\circ}C$ with 660 nm red LED treatment. The acidity of kiwifruits in plastic bags was decreased 52.6, 55.6, 52.8% by the treatment of 440 nm blue, 470 nm white and 660 nm red LED light source, respectively, compared to that of kiwifruits incubated in darkness at $25^{\circ}C$. Decreased acidity irradiated by 660 nm red LED light source can be applied for regulating periods of the kiwifruit after-ripening process. LED light sources emit very narrow wavelength with a power-saving mode, therefore, the usage of these LED light source for regulating the after-ripening process can be classified as a clean biotechnology producing safe and environment-friendly kiwifruits.

Effect of Water Temperature and Photoperiod on the Oxygen Consumption of Four Different Strains of Red Seabream, Pagrus major

  • We determined the effects of different water temperatures (15, 20, and 25℃) and photoperiod cycles (24L:0D, 12L:12D, and 0L:24D) on the oxygen consumption of the offspring of a cultured Japanese strain (JJ), a selected Korean strain (KK), and intraspecific hybrid strains (JK and KJ) of red seabream, Pagrus major, under starvation conditions. The different fish strains, water temperatures, and photoperiod cycles had effects on the mean oxygen consumption of fish. Oxygen consumption increased with increasing water temperatures for all photoperiod treatments (p<0.001). Fish held in continuous darkness (0L:24D) used consistently less oxygen than fish exposed to continuous light (p<0.05). The oxygen consumption of fish exposed to the light phase in a 12L:12D photoperiod was higher than that of fish in the dark phase of the 12L:12D cycle, and differences were significant in three of the strains: JJ (15℃), KK (15 and 20℃), and KJ (25℃). The oxygen consumption of the inbred (JJ and KK) and intraspecific hybrid (JK and KJ) strains varied with differing water temperatures and photoperiod cycles. The JK strain displayed significantly higher oxygen consumption than the other strains under all experimental conditions except 15℃ with a 0L:24D photoperiod. The JK and KJ strains usually showed the highest and lowest oxygen consumption values, respectively, whereas the inbred strains exhibited intermediate values. Oxygen consumption in the JJ and JK strains was usually higher than that of the KK and KJ strains. We propose that differences in the thermal sensitivity and photosensitization properties of the strains contribute to differences in their ability to adapt to changes in water temperature and photoperiod, thus resulting in differences in the amplitude of their metabolic rates.

Shadows and Evil in Inferno of Divine Comedy (신곡의 지옥편에 나타난 그림자와 악)

  • This study is to illuminate the problem of shadows and evil appearing in contemporary events in the midst of a period of upheaval through Inferno of Divine Comedy. First, the concept of shadow and evil were briefly summarized in analytical psychology and discussed the importance of considering the concepts with the empirical aspect of relativity and ethics in the field of psychotherapy. The 14th century, the age of Dante, was the embryonic period of the Renaissance beyond the end of the Middle Ages. It was when Dante, who was writing Divine Comedy, had to take off his persona forcibly and live in exile. In a nutshell, it was a transition period for both the individual and the collective. The dark forest is a nigredo, darkness and chaos we face in this transition, but it can be a place of transformation and rebirth. The three beasts (leopard, lion, and she-wolf) encountered in the forest can be considered as the instinctual images that Dante ignored and alienated, which the medieval Christian world had suppressed and eliminated. Especially at the collective level, as destructive instincts, ferocious beasts roam throughout society when a crisis breaks dominant laws of values. The three beasts of Inferno appear as phenomena of shadows and evil. The aspect of leopard was explored Cerberus and Chiaco(pig) as a symbol of greed, and the lion, Farinata as the form of violent passion. The aspect of the she-wolf was examined as Geryon, a deceitful monster with a beast nature hidden behind goodness, the giants in the frozen lake of Cocytus, and Branca D'Oria, who betrayed and murdered the country and family. Inferno reveals the "state" of being trapped when one yields to the evil hidden within oneself and falls into prey.

Optimal concentrations of plant growth regulators and AgNO3 for the improvement of regeneration efficiency in Chrysanthemum morifolium 'Ohblang' (국화 '오블랑'의 재생 효율 증진을 위한 식물생장조절제와 AgNO3 적정 농도 선별)

  • A plant regeneration system was developed through shoot organogenesis from in vitro leaf explants of Chrysanthemum morifolium 'Ohblang'. The effects of different concentrations of plant growth regulators and AgNO3 on efficient shoot regeneration and inhibition of browning were evaluated in chrysanthemum. The explants were cultured on MS shoot induction medium supplemented with 12 combination treatments of 6-benzyladenine (BA) 0.5, 1.0 and 2.0 mg/L, and α-naphthaleneacetic acid (NAA) 0.2, 0.5, 1.0 and 2.0 mg/L in darkness for 6 weeks and cultured under a 16/8 h photoperiod for 6 weeks. The highest shoot regeneration was obtained from the explants cultured on the medium with 1.0 mg/L BA and 1.0 mg/L NAA. Based on this result, AgNO3 was added to a shoot induction medium containing MS salts, vitamins, 1.0 mg/L BA, 1.0 mg/L NAA, 30 g/L sucrose, and 6 g/L agar to reduce browning of chrysanthemum leaf explants. In the control treatment without AgNO3, leaf explants turned brown at the cut edge; however, browning was not observed in AgNO3 treatments. Shoot organogenesis was higher at low concentrations of AgNO3 and decreased with an increase in AgNO3 concentration. The explants cultured on shoot induction medium (MS salts, vitamins, 1.0 mg/L BA, 1.0 mg/L NAA) with 1 mg/L of AgNO3 produced the highest shoot regeneration with 2.6 shoots per explants and a browning index of 0.7. When the regenerated shoots were detached from the explants and cultured on MS medium, the shoots were elongated and rooted successfully.

In a Time of Change: Reflections on Humanities Research and Methodologies (변화의 시대, 인문학적 변화 연구와 방법에 대한 고찰)

  • This study begins with a question about research methods in humanities. It is grounded in the humanities, focusing on the changes that have brought light and darkness to the humanities, and focusing on discourse regarding research methods that explore those changes. If the role of the humanities is to prevent the proverbial "gray rhino," unlike the sciences, and if the humanities have a role to play in moderating the uncontrollable development of the sciences, what kind of research methods should humanities pursue. Furthermore, what kind of research methods should be pursued in the humanities, in line with the development of the sciences and the changing environment? This study discusses research methods in the humanities as follows: first, in Section 2, I advocate for the collaboration between humanities and scientific methods, utilizing accumulated assets produced by humanities and continuously introducing scientific methods. Prediction of change is highly precise and far-reaching in engineering and the natural sciences. However, it is difficult to approach change in these fields in a macro or integrated manner. Because they are not precise, they are not welcome in disciplines that deal with the real world. This is primarily the responsibility of humanities. Where science focuses on precision, humanities focuses on questions of essence. This is because while the ends of change have varied throughout history, the nature of change has not varied that much. Section 3 then discusses the changing environment, proposals for changes to humanistic research methods, reviews and proposals inductive change research methods, and makes some suggestions for humanistic change research. The data produced by the field of humanities accumulated by humankind in the past is abundant and has a wide range of applications. In the future, we should not only actively accept the results of scientific advances but also actively seek systematic humanistic approaches and utilize them across disciplinary boundaries to find solutions at the intersection of scientific methods and humanistic assets.

고려엉겅퀴 ( 곤드레 ) 의 종자발아 및 차광재배 효과 구명 ( Improvement of Germination Rate by Low temperature and Development of Effectve Shading Cultivation of Cirsium setidens under Rain Shelter in Highland )

  • For improvement germination rate of Cirsium setidens, wet seeds were storaged for 20 to 80 days at $4^{\circ}C$ and germinated at $18^{\circ}C$ in the dark or light condition. To assess the effects of shading and plant age on the growth, fresh yield and seed yield under rain shelter in highland at altitude of 800m above sea level, seeds were sown in an open field in late september,1990. In early May the next the seedlings were transplanted for vegetatable and seed production in the plastic houses for rain shelter. They were grown from 1991 to 1993 in the houses covered with the shading nets ranging from 0 to 70% for vegetable production and 0 to 50% for seed production. Germination rate inceased as the storage period lengthened, and the rate came to $60\sim80%$ at 60days after germination. It was more effective in lighting than in darkness. For vegetable production, the shading net treatments showed more values in almost the characters checked at the third year than the non-shading treatment. No. of stem, no, of leaves, stem length and degree of softening were no different between shading net treatments, but the other characters were significantly different. Ratio of survival, density of chlorophyll and ratio of dry matter of plants were highest in the plots of 50%, 70%, and 30% of shading, respectively. The highest fresh yield was recorded from 2-year plants, 70% of shading from them showed the greatest yield. The highest seed yields were produced 2-year-old plants of non-shading plot. For seed production, the non-shading treatment is best, and the highest seed yields were obtained in both the second and third years.

Development of an Efficient In Vitro Screening Method for Selection of Resistant Lily Cultivars Against Fusarium oxysporum f. sp. lilii (백합 구근부패병 저항성 검정을 위한 기내 간편 검정법 개발)

  • The soilborne fungus Fusarium oxysporum f. sp. lilii (Fol) is a serious threat to all lily cultivars, especially infecting bulbs and flowers. It has become increasingly important to develop varieties resistant against the bulb rot disease. Genetic diversity of cultivars and reliable screening methods are required for this purpose. Here, an efficient in vitro screening system for evaluating resistance to Fol in 38 in vitro-grown lily plants was investigated. Various factors including culture conditions of Fol, inoculum density, appropriate plant materials, inoculation method and duration, and incubation period of plant materials after inoculation were combined to optimize the screening method. As a result, we suggest optimal conditions for an in vitro screening system for the selection of Fol-resistant lily cultivars as follows. Fol was grown on potato dextrose agar (PDA) medium for 6 days at $25^{\circ}C$ in darkness and used as working inoculation. Spore suspensions were prepared (inoculum density: $1.0{\times}10^4$ $spores{\cdot}mL^{-1}$), and then leaf segments $1.5{\times}2.0cm^2$ were inoculated by dipping for 22 hours at $25^{\circ}C$ in dark. Later, leaves were cultured on 0.6% agar plates at $25^{\circ}C$ and 50% humidity with a photoperiod of 16 hours light/8 hours dark (fluence rate of $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) to examine the progress of bulb rot. After 7 days, disease levels were classified into indices 1 (no symptom) to 6 (serious bulb rot). Soil inoculation of Fol carried out with resistant or susceptible lily cultivars that had been selected through in vitro screening confirmed the reproducibility of results. Therefore, the in vitro screening method established in this study is efficient and reliable for selection of lily cultivars resistant against bulb rot disease.

Effect of Heat-Moisture Treatment of Domestic Rice Flours Containing Different Amylose Contents on Rice Noodle Quality (아밀로오스 함량이 다른 국내산 쌀가루의 수분-열처리가 쌀국수 품질에 미치는 영향)

  • The influence of heat-moisture treatment (HMT) and substitution of rice flour containing different amylose contents on the quality characteristics of rice noodles was investigated. HMT was applied to rice flours with 21% moisture content at 100 and 105$^{\circ}C$ for 30 min. Three rice cultivars were used, including high amylose of Goami (GM) and intermediate amylose of Choochung (CC) as domestic rice flours and imported rice of Taeguk (TG) as a control. HMT and substitution of rice flour with different amylose contents affected the cooking and texture quality of rice noodles. When rice noodles were made of intermediate amylose rice flour with HMT, cooking properties improved with decreased cooking loss and cooking water turbidity and thus were closer to those of control. Especially, the hardness, adhesiveness, tensile strength, and darkness of rice noodles notably increased when HMT rice flour was used. Based on the results of quantitative descriptive analysis for selected rice noodles, the noodles made of HMT CC at 105$^{\circ}C$ (CC105) had high scores for resilience and adhesiveness and low scores for hardness compared with imported commercial rice noodles and other experimental noodles such as TG, HMT GM100, TG+CC, and TG+CC105. In conclusion, rice noodles were made of composite flours containing high amylose and intermediate amylose contents or HMT intermediate amylose content rice flour.

Effect of $GA_3$ and Light Quality on Seed Germination in Three Campanulan Plants ($GA_3$ 및 파종(播種) 후(後) 광질(光質) 처리(處理)에 따른 도라지, 더덕, 만삼의 발아율(發芽率))

  • Gibberellin (GA) has been widely used to break seed dormancy for better stand establishment. The experiment was done to clarify the effect of $GA_3$ (concentration; period) and light quality (red; white; dark) after sowing on seed germination and radicle elongation of Campanulaceae (Platycodon grandiflorum; Codonopsis lanceolata; C. pilosula) to get an information on their field emergence. The germination test was carried out with 12 hours irradiation for 9 days after priming treatment. In the darkness, the mean germination rate of all the species was decreased in the order to P. grandiflorum, C. lanceolata, C. pilosula. Their mean germination rates and radicle lengths were increased with increased concentration to 0.1mM of $GA_3$. Earlier germination rate was higher but later one was less in 4-day $GA_3$ treatment than in 1- or 2-day $GA_3$ treatment. Light treatment. especially red light given after $GA_3$ treatment. eliminated the $GA_3$ treatment effect. Red light done after $GA_3$ treatment nearly blocked the germination of P. grandiflorum and C. pilosula but delayed that of C. pilosula compared to the other light quality treatments having the similar rate. In addition. the radicle elongation of all three species affected by light quality treatment showed the same result as the germination rate.

Phytochrome Signal Transduction Regulates Anthocyanin Biosynthesis in Cell Suspension Cultures of Vitis vinifera (포도 세포현탁배양계에서 Phytochrome 신호전달에 의해 조절되는 안토시아닌 생합성)

  • This experiment was carried out to confirm that phytochrome regulates anthocyanin bio-synthesis during cell suspension culture system of grape or not. In suspension culture of grape, maximum accumulation of anthocyanin was observed at the stationary phase under continuous white light condition. From mono-chromatic light interruption for 24h at the 4th or 7th day on the suspension cultured cells, the anthocyanin accumulation was highly enhanced at the light interruption at 7th day than 4th day under all monochromatic light treatment. However, the cell growth patterns were not affected by any light treatment. In the darkness, the anthocyanin synthesis was very low but remarkably increased by blue light or red light irradiation. However, the increase of anthocyanin accumulation by blue or red light was suppressed by far-red light in the suspension cells of grape. This suppression by far-red light on the anthocyanin synthesis also observed on the cells treated red or far-red light alternatively. These results implied that phytochrome regulation system may be involved in the anthocyanin biosynthesis of the suspension grape cells. By RNA expression analysis, chalcone synthase (CHS) gene was expressed highly by blue and red light but low by far-red light. The synergistic increase of CHS gene expression was also observed at the treatment of blue light followed by red for 24h. This result may explain the increase of anthocyanin accumulation in B/R treatment. Although the expression of phytochrome gene (PHYA or PHYB) was not highly increased by all light treatment (blue, red, and far-red light) the expression of both PHYA gene and PHYB gene was increased a little in cells treated red or far-red light. In grape suspension cells, the red light enhanced the anthocyanin synthesis, whereas the far-red light was suppressed. Although it was not confirmed whether or not phytochrome gene is activated in anthocyanin accumulating grape cells, we believed that anthocyanin biosynthesis in grape cells may be regulated under phytochrome signal transduction system.
