Monitoring of VHS and RSIVD in cultured Paralichthys olivaceus of Jeju in 2014 (2014년 제주도 양식 넙치, Paralichthys olivaceus에 대한 VHS 및 RSIVD 모니터링)

  • The outbreak of viral diseases caused by viral haemorrhagic septicaemia virus (VHSV) and red seabream iridovirus (RSIV) have been reported in cultured olive flounder, Paralichthys olivaceus. VHSV has been a serious viral disease that infects the olive flounders in South Korea. Clinical signs of VHSV infection are skin darkening, abdominal distension and haemorrhages. Outbreaks of fish iridoviral disease was first reported from red seabream, Pagrus major farms in Japan. Recently, iridovirus infection have occurred frequently from olive flounder farms in South Korea. In this study, disease surveillance was performed to monitor the prevalence of VHSV and RSIV in olive flounder in 2014. The samples were collected from 60 different olive flounder farms in Jeju from April, May, September, November and December in 2014. RT-PCR (VHSV) or PCR (RSIV) results showed that VHSV were detected in 5 farms, but RSIV has not been detected in any farms. The migration of olive flounder was restricted for the quarantine in 5 farms of VHS outbreak. The nucleocapsid protein (N) gene and glycoprotein (G) gene sequences of the 5 Korean VHSV isolates were successfully amplified and sequenced. Phylogenetic analysis was performed using the VHSV sequences reported here together comparison with the nucleotide sequences available from the GenBank database. Phylogenetic analysis indicated that most of Korea VHSV belong to the genotype IVa and closely related to the strains from Japan and China.

Analysis on deterioration behaviors of the beewax-treated Annals of Joseon Dynasty (「조선왕조실록」 밀납본의 열화특성 분석)

  • Paper cultural asset show various deterioration characteristics and behaviors according to the environmental conditions during the storage. The understanding of the paper itself and its conservation knowledge is thus prerequisite to preserve paper cultural assets and pass them to descendants in a good condition. The Annals of Joseon Dynasty is one of the most important our written cultural assets (National treasure No.151) and it is registered as a UNESCO Memory of the World. The Annals of Joseon Dynasty is composed of 1707 volumes and 1,187 books that were written on the official history of 472 years(AD 1392-1863) from King Taejo to King Chuljong (25 generations). Among them, beewax-treated Annals show severe damages due to the deterioration of wax materials such as breaking, hardening, darkening, discoloring and whitening. Urgent measures should be taken to preserve the Annals and prevent further deterioration. Therefore, a systematic and scientific approach on the conservation and restoration of the Annals of Joseon Dynasty is definitely necessary. So it can be used an example to show how much the preservation science can conserve the paper cultural assets. In this respect, this study focuses on analyzing paper properties and deterioration behaviors of the Annals of King Sejong, the most damaged one, using non-destructive micro analysis techniques.

Phyllite as a New Flame Retardant Synergist for ABS Resin Containing Bromine Flame Retardant (브롬계 난연제를 사용한 ABS 수지에 대한 신규 난연 상승작용제로시의 천매암)

  • Flame retardant synergism of phyllite was studied in ABS resins containing brominated flame retardant(tetrabromobisphenol A(TBBA) or brominated epoxy oligomer(BEO)) and antimony trioxide($Sb_2O_3)$. Talc was used for the comparison purpose. ABS compounds were manufactured by a twin-screw co-rotating extruder and subsequently injection molded into several specimen for mechanical and thermal properties. Flame retardancy of ABS compounds measured by UL 94 vertical test with 1.6 mm thick bar specimen was enhanced by the replacement of antimony trioxide into phyllite or talc in the range of 12.5%(0.5 wt%) to 37.5%(1.5 wt%). Phyllite showed better synergistic effect comparing with talc especially for BEO. Only phyllite enhanced the flowability of ABS compounds. Notched Izod impact strength decreased with the proportion of phyllite or talc content. Phyllite could replace the antimony trioxide up to the content of 25%(1 wt%) to give better flame retardancy and flowability without darkening problem.

Effects of lemon or cinnamon essential oil vapor on physicochemical properties of strawberries during storage

  • Recently, consumers have gained an interest in natural and minimally processed foods, inciting the food industry to consider using of natural products as preservatives. Strawberries are a widely consumed fruit but are also highly perishable. Therefore, in this study, the physicochemical properties of strawberries (Fragaria×ananassa) were evaluated after a 12-h treatment with lemon essential oil (Citrus×limon) or cinnamon essential oil (Cinnamomum cassia) vapor during storage at 22℃ for 4 days in an accelerated shelf-life study and 4℃ for 18 days in a validation study. Weight loss was blunted in fruit treated with oil vapor during the first days of storage (p<0.05). Lemon essential oil delayed fruit darkening (p<0.05) but reduced the firmness of strawberries (p<0.05). Strawberries treated with cinnamon essential oil had a higher concentration of reducing sugars (p<0.05), and a decrease of 16.7% visible decay, although the difference was insignificant. Oil vapor treatment did not alter the pH, organic acid content, or soluble solid content during storage compared to the control. Since lemon and cinnamon essential oils have well-documented antimicrobial properties, they may be suitable for the natural preservation of fruit. This study provides new information on using essential oil vapor treatment to preserve fruits, and potentially decrease fruit loss and waste.

A Case Study of Mortality Caused by Viral Encephalopathy and Retinopathy (VER) in Cultured Sevenband Grouper, Epinephelus septemfasciatus during Winter (겨울철 양식 능성어의 바이러스성뇌망막증(VER) 감염사례)

  • In 2015, a nervous necrosis virus (NNV) was isolated from sevenband grouper, Epinephelus septemfasciatus, maintained in land-based aquaculture system at below $12^{\circ}C$ in winter. Mortality was up to 30% in brood fish, over 4 kg of body weight. Moribund fish showed clinical sings typical of viral encephalopathy and retinopathy (VER), also called viral nervous necrosis (VNN), such as uncoordinated, corkscrew-like swimming behavior, belly-up at rest, darkening of body, cloudy eyeball and hyperinflation of the swim bladder. Aetiology of the disease was confirmed by gross observation of clinical signs, histopathology and molecular diagnosis. Histological studies revealed severe vacuolation and necrosis in the brain. Molecular diagnosis by revere transcription-polymerase chain reaction (RT-PCR) specific to batanodavirus yielded a positive result. The nucleotide sequences of the PCR-amplified fragment were 99.48~100% similar to barfin flounder nervous necrosis virus (BFNNV) genotype and most closely aligned with Pacific cod betanodavirus (PCNNV). This is the first report of natural batanodavirus, NNV infection in sevenband grouper reared in low water temperature during winter (below $12^{\circ}C$) in Korea.

Study of Alkaline Peroxide Mechanical Pulp Made from Pinus densiflora (국내산 소나무로 제조된 APMP 특성 연구)

  • Alkaline Peroxide Mechanical Pulping (APMP) of Pinus densiflora harvested from domestic mountains was explored. APMP contributes to various advantages including pulp quality, elimination of the need for a bleaching process, and energy savings. Sequential treatment of impregnation of bleaching chemicals and refining not only overcome the concern of alkaline darkening of wood chips during chemical impregnation, but it also brightens the chips to the desired brightness levels suitable for writing and printing papers. APMP pulping from Pinus densiflora was greatly influenced by the dosage levels of hydrogen peroxide and sodium hydroxide. Alkaline peroxide treatment was carried out by applying one of three levels of hydrogen peroxide (1.5, 3, and 4.5% based on the oven-dried weight of the wood chips) and one of three levels of sodium hydroxide (1.5, 3, and 4.5% based on the oven-dried weight of the wood chips). Other chemicals including a peroxide stabilizers and metal chelation were constantly added for all treatments. Chemical treatment with a liquor-to-wood ration of 9:1 was carried out in a laboratory digestor. Compared to BTMP, APMP pulping displayed outstanding characteristics including the less requirement of refining energy, the better improvement of tensile strength, the more reduction of shives, and the greater increase of pulp brightness. In particular, when 4.5% of hydrogen peroxide with impregnation during 90 minutes was used, the brightness of APMP reached 64.9% ISO. Even though bulk of APMP was decreased with the increase of sodium hydroxide, a better and improved balance could be achieved between optical and strength properties. The spent liquor obtained from the discharge of the impregnation process at the dosage level of 4.5% hydrogen peroxide exhibited an equal level of residual peroxide with BTMP. In conclusion, APMP pulping showed successful results with Pinus densiflora due to its better response to the development of optical and physical properties compared to TMP pulping.

Characterization of Surface Deterioration for Stone Property around the Hyeonleung (Royal Tomb of Joseon Dynasty) in Guri, Korea (구리 현릉 능상석물의 표면 손상특성 평가)

  • The materials of sculptured stone property around the Hyeonleung Royal Tomb in Guri consist mainly of high weathered of granitic rocks with magnetite-series. Deterioration characteristics occurred highly with microorganism, soil inflow and black contaminants at the burial mound zone. As a result of deterioration evaluation, stone surface around the burial mound zone show serious comprehensive damage of soil inflow (50.5%) and lichen coverage (47.6%) which are the major damaging factors, and there are about 8.6% of biological growth noticeably in the north side. Surface contaminants and the discoloration had the complex causes. Gypsum occurred between joints of stones and the major contaminant type, darkening which was analyzed organic bodies caused discoloration. From result of ultrasonic velocity measurements, there are mean value of 2,195 m/s with highly weathered (HW) grade. Most of the stone properties showed 4 to 5 weathered grade. Therefore, it turned out that sculpture stone properties require conservation treatments. To remove soil accelerating damage factors and lichen occupying high percents, the cleaning process is necessary and consolidation, rejoining and filling are needed as well. Also, consideration on removing conservation materials containing Ca and e fflorescence is required and retreatments need to be considered.

  • Streptococcus spp. of bacterial pathogen of fish were isolated from the cultured flounder (Paralichthys olivaceus) in fish farm of Jeju Island. Clinical signs of the infected flounder which are the most commons symptoms are as follows: erratic swimming, darkening of the body colour, unilateral or bilateral exophthalmia, corneal opacity, hemorrhages in the opercular and the bases of the fins, and the ulceration of the body surface. Biochemical characteristics of pathogenic fish Streptococcus spp, were gram positive, spherical form, catalase negative, oxidase negative and $\beta$-haemolytic, respectively, The viable cells counted from the tissue of the diseased flounder were the largest in the order of the ulcer, the kidney, the blood and the brain, The drugs used were ampicillin, ciprofloxacin, doxycycline, gentamycin, tetracycline, erythromycin, streptomycin and oxytetracycline, Streptococcus spp. were found to be sensitive to ampicillin, ciprofloxacin, doxycycline and gentamycin, but were resistant to tetracycline, erythromycin, streptomycin and oxytetracycline. The pathogenicity of Streptococcus spp. on the cultured flounder with an abdominal cavity injection was high. The haemolytic activity of the toxin against the sheep red blood cells reached the maximum after 30 min incubation at $37^{\circ}C$ or $50^{\circ}C$. The toxin showed highest activity at pH $5.5\sim6.5$.

The study on the causal agent of Streptococcicosis (Lactococcus garvieae), isolated from cultured marine fishes (해산 양식어류로부터 분리된 연쇄구균증의 원인균, Lactococcus garvieae에 대한 연구)

  • The purpose of this study was to investigate the microbiological characteristics and the distributions of the bacteria causing streptococcicosis occurred in marine fish farm, Korea. Many kinds of cultures fishes suffered from the disease accompanied with typical symptoms, including darkening of the skin, exophthalmia, petechiae inside of the opercula and distended abdomen. The isolates from the diseased fishes were compared with Lactococcus garvieae by biochmical, biophysical and serological methods and polymerase chain reaction(PCR) assay. We isolated 35 strains of the geuns Streptococcus from the diseased olive flounder, Paralichthys olivaceus, yellow tail, Seriola quinqueradiata and Korean rockfish, Sebastes schlegeli. 15 strains out of the isolates were identified to L. garvieae and the others were not because of their different biochemical and biophysical charateristics. Seven strains of the isolates were agglutinated by rabbit serum raised against L. garvieae $KG^+$ phenotypic cells(ATCC49156)as a reference strain. Twenty-one strains of the isolates identified to L. garvieae since they were formed the expected band through performing PCR assay using specific primers, pLG-1(5'-CATAACAATGAGATCGC-3') and pLG-2(5'-GCACCCCGCGGTTG-3'). In the present study, it showed that L. garvieae was a dominant strain causing streptococcicosis in the tested area due to occurrence of 21 strains as L. garvieae out of all the isolates, 9 atrains as Streptococcus sp. and 5 strains as Enterococcus sp.

OPTICAL MULTI-CHANNEL INTENSITY INTERFEROMETRY - OR: HOW TO RESOLVE O-STARS IN THE MAGELLANIC CLOUDS

  • Intensity interferometry, based on the Hanbury Brown-Twiss effect, is a simple and inexpensive method for optical interferometry at microarcsecond angular resolutions; its use in astronomy was abandoned in the 1970s because of low sensitivity. Motivated by recent technical developments, we argue that the sensitivity of large modern intensity interferometers can be improved by factors up to approximately 25 000, corresponding to 11 photometric magnitudes, compared to the pioneering Narrabri Stellar Interferometer. This is made possible by (i) using avalanche photodiodes (APD) as light detectors, (ii) distributing the light received from the source over multiple independent spectral channels, and (iii) use of arrays composed of multiple large light collectors. Our approach permits the construction of large (with baselines ranging from few kilometers to intercontinental distances) optical interferometers at the cost of (very) long-baseline radio interferometers. Realistic intensity interferometer designs are able to achieve limiting R-band magnitudes as good as $m_R{\approx}14$, sufficient for spatially resolved observations of main-sequence O-type stars in the Magellanic Clouds. Multi-channel intensity interferometers can address a wide variety of science cases: (i) linear radii, effective temperatures, and luminosities of stars, via direct measurements of stellar angular sizes; (ii) mass-radius relationships of compact stellar remnants, via direct measurements of the angular sizes of white dwarfs; (iii) stellar rotation, via observations of rotation flattening and surface gravity darkening; (iv) stellar convection and the interaction of stellar photospheres and magnetic fields, via observations of dark and bright starspots; (v) the structure and evolution of multiple stars, via mapping of the companion stars and of accretion flows in interacting binaries; (vi) direct measurements of interstellar distances, derived from angular diameters of stars or via the interferometric Baade-Wesselink method; (vii) the physics of gas accretion onto supermassive black holes, via resolved observations of the central engines of luminous active galactic nuclei; and (viii) calibration of amplitude interferometers by providing a sample of calibrator stars.
