Zebrafish (Danio rerio)의 체색 변이에 관한 유전 분석 (Inheritance of Golden Coloration in the Zebrafish, Danio rerio)

  • The study has been conducted in order to understand the inheritance of body color in the wild type zebrafish (zebra danio), Danio rerio, and its golden mutant (golden danio). The body color was also studied to determine the effect of golden coloration on the survival rate of zebrafish eggs and larvae up to 15 days after fertilization. Reciprocal monohybrid crosses between the wild and the golden type of zebrafish indicated that golden coloration was controlled by a single gene which had two alleles. Transmission of these alleles from parents to their progenies followed the principles of dominance and segregation based on Mendelian inheritance. Similar results from the reciprocal crosses implied that a locus for golden coloration was located on an autosomal chromosome. On the other hand, average survival rates from four different types of mating between, and within, zebra and golden danio suggested that golden coloration seemed to be associated with the survival rate of zebrafish, especially in its early embryonic stage. This indicated that homozygous recessive golden mutation was likely to weaken the golden danio's chance of survival.

Comparative Ultrastructures of the Fertilized Egg Envelopes in Danio rerio and Danio rerio var. frankei, Cyprinidae, Teleostei

  • The leopard danio, Danio rerio var. frankei is a spotted color morph of the zebrafish, Danio rerio caused by a pigment mutation. The structural differences of fertilized egg and egg envelope are poorly documented. To clarify this, we compared the fertilized egg morphology and ultrastructures of surface structures, the micropyle and the cross section of fertilized egg envelopes of zebrafish and leopard danio, variation species of zebrafish using a light and electron microscopes. Although the fertilized egg sizes were different, the external shapes of the fertilized eggs of two species couldn't be differentiated under the light microscope. The characteristics of fertilized eggs, such as a spherical shape, a non-adhesive quality and a large perivitelline space, were shown to be related to spawning habit. In ultrastructure of fertilized egg envelope, there is no morphological difference of micropyle between two species. By contrast, the ultrastructure and the numbers of knob-like structures and semihemisphere-like structures per unit area on the outer surface, and the number of lamellae of inner layer on the fertilized egg envelope section displayed definite species specificity. Collectively, our data indicate that the ultrastructure of fertilized egg envelope in the zebrafish could be differentiated by species variation.

생물정보학을 이용한 Zebrafish Microsomal Epoxide Hydrolase 클로닝 및 특성연구 (Cloning and Characterization of Zebrafish Microsomal Epoxide Hydrolase Based on Bioinformatics)

  • Zebrafish (Danio rerio)의 microsomal epoxide hydrolase(mEH)로 추정되는 유전자를 클로닝하고 그 특성을 연구하였다. D. rerio의 mEH 추정단백질은 포유동물의 mEH 및 세균의 EH들과 아미노산서열 상동성을 보였으므로 결정분자구조(1qo7 및 1ehy)를 template로 하여 homology modelling을 행하였다. 클로된 단백질은 $Asp^{233}$, $Glu^{413}$$His^{440}$으로 구성된 catalytic triad와 2개의 tyrosine 잔기 및 oxyanion hole이 보존되어 있었다. 생물정보학적인 분석 및 EH 활성시험은 추정단백질이 D. rerio의 mEH라는 것을 보여주었다. Racemic styrene oxide를 기질로 하여 활성시험을 행한 결과, 재조합 D. rerio mEH는 (R)-styrene oxide을 입체선택적으로 가수분해하였으며 45분 반응시간에 99%ee의 광학순도를 가진 (S)-styrene oxide를 33.5% 얻을 수 있었다.

Acute toxicity assessment of camphor in bio-pesticides by using Daphnia magna and Danio rerio

  • Objectives An ecofriendly alternative to chemical pesticides is bio-pesticides, which are derived from natural sources. The interest in bio-pesticides is based on the disadvantages associated with chemical pesticides. Methods We conducted acute toxicity assessments of camphor, a major component of bio-pesticides, by using Daphnia magna (D. magna) as well as assessed the morphological abnormalities that occurred in Danio rerio (D. rerio) embryos. Results The median effective concentration of camphor on D. magna after 48 hours was $395.0{\mu}M$, and the median lethal concentration on D. rerio embryos after 96 hours was $838.6{\mu}M$. The no observed effect concentration and predicted no effect concentration of camphor on D. magna, which was more sensitive than D. rerio, were calculated as $55.2{\mu}M$ and $3.95{\mu}M$, respectively. Morphological abnormalities in D. rerio embryos exposed to camphor increased over time. Coagulation, delayed hatching, yolk sac edema, pericardial edema, and pigmentation of embryos mainly appeared between 24 and 48 hours. Further, symptoms of scoliosis and head edema occurred after 72 hours. In addition, bent tails, ocular defects and collapsed symptoms of fertilized embryonic tissue were observed after 96 hours. Conclusions The camphor toxicity results suggest that continuous observations on the ecosystem are necessary to monitor toxicity in areas where biological pesticides containing camphor are sprayed.

Assessment of the Cardioprotection Offered by Fisetin in H2O2-induced Zebrafish (Danio rerio)-Tg (cmlc2: egfp)

  • The aim of this study was to evaluate the protective function of fisetin, a natural flavonoid in zebrafish heart for the treatment of myocardial infarction in coronary and ischemic heart disease. For this purpose, we induced oxidative stress zebrafish (Danio rerio)-Tg (cmlc2: egfp) by $H_2O_2$ and then administered fisetin, the protective effect of fisetin was determined by measuring the heart rate following fisetin administration. After testing the toxicity of fisetin, we found that the heartt increased in a concentration-dependent manner, however there was no difference between the heart rates of embryos and adults. The improved heart rate demonstrated the cardioprotective effect of fisetin. The result showed that fisetin, at concentration of 3and $5{\mu}M$, significantly increased heart rate compared with the heart with $H_2O_2$ alone. This indicates that fisetin plays an important role in the prevention of heart damage and treatment of cardiovascular diseases caused by oxidative stress due to ischemia / reperfusion.

  • We investigated the relationship between the $TiO_2$ photocatalytic decomposition of bisphenol A in water and biological toxicity to zebrafish (Danio rerio) during $1\sim28$ weeks post development stage. The bisphenol A in water was completely degraded by the $TiO_2$ photocatalysis in 50 hours. After the photocatalysis, no toxic effects on the morphogenesis of the zebrafish were observed during the development, growth, and maturate stages. Catalase activity of control group was not different from $1\sim5$ week post fertilized group. However, toxic effect on the catalase activity of adult stage(28 weeks) decreased 50% than control group.

  • Historically, research in toxicology has utilized non-human mammalian species, particularly rats and mice, to study in vivo the effects of toxic exposure on physiology and behavior. However, ethical considerations and the overwhelming increase in the number of chemicals to be screened has led to a shift away from in vivo work. The decline in in vivo experimentation has been accompanied by an increase in alternative methods for detecting and predicting detrimental effects: in vitro experimentation and in silico modeling. Yet, these new methodologies can not replace the need for in vivo work on animal physiology and behavior. The development of new, non-mammalian model systems shows great promise in restoring our ability to use behavioral endpoints in toxicological testing. Of these systems, the zebrafish, Danio rerio, is the model organism for which we are accumulating enough knowledge in vivo, in vitro, and in silico to enable us to develop a comprehensive, high-throughput toxicology screening system.

Protective Effects of Plathymenia reticulata and Connarus favosus Aqueous Extracts against Cadmium- and Mercury-Induced Toxicities

  • The extracts of Plathymenia reticulata and Connarus favosus are widely used in the folk medicine. The potential protective effects of these extracts have been evaluated against cadmium in the yeast Saccharomyces cerevisiae, and against mercurial contamination in zebrafish Danio rerio. In yeast, both extracts efficiently protected the ${\Delta}ycf1$ mutant strain exposed to cadmium chloride restoring the growth, the expression of stress-response genes and decreasing the level of oxidative stress. In zebrafish, the supplementation of methylmercury-contaminated diet with both plant extracts similarly protected fish through the suppression of the methylmercury-induced lipid peroxidation, decrease of acetylcholinesterase activity, and restoring the expression levels of stress-response genes. This study particularly demonstrates the protective potential of both aqueous extracts against methylmercury, and could represent an interesting alternative for the Amazonian fish-eating communities to cope with the impact of chronic exposure to contaminated diets.

Long arm octopus (Octopus minor) extract prevents eye injury caused by particulate matter exposure in zebrafish (Danio rerio) embryos

  • Particulate matter (PM) is a mixture of microscopic solid inhalable particles including airborne liquid droplets, and it is implicated with several diseases. The eye does not have a protective barrier among the human organs, consequently it get directly exposed to environmental substances such as PM. The scarcity of treatments for damage to the eyesight and the vision and eye structure being closely related to the structure and function of the central nervous system highlights the cruciality of novel therapeutics. In this study was conducted using in vivo zebrafish vertebrate model which is a useful model due to the conserved genes between human. We found that protective effect of Octopus minor extract against particulate matter-induced adverse effects on eye development in zebrafish (Danio rerio) embryos by regulating antioxidant and anti-inflammatory mRNA expression.
