The American Cockroach Peptide Periplanetasin-2 Blocks Clostridium Difficile Toxin A-Induced Cell Damage and Inflammation in the Gut

  • Clostridium difficile, which causes pseudomembranous colitis, releases toxin A and toxin B. These toxins are considered to be the main causative agents for the disease pathogenesis, and their expression is associated with a marked increase of apoptosis in mucosal epithelial cells. Colonic epithelial cells are believed to form a physical barrier between the lumen and the submucosa, and abnormally increased mucosal epithelial cell apoptosis is considered to be an initial step in gut inflammation responses. Therefore, one approach to treating pseudomembranous colitis would be to develop agents that block the mucosal epithelial cell apoptosis caused by toxin A, thus restoring barrier function and curing inflammatory responses in the gut. We recently isolated an antimicrobial peptide, Periplanetasin-2 (Peri-2, YPCKLNLKLGKVPFH) from the American cockroach, whose extracts have shown great potential for clinical use. Here, we assessed whether Peri-2 could inhibit the cell toxicity and inflammation caused by C. difficile toxin A. Indeed, in human colonocyte HT29 cells, Peri-2 inhibited the toxin A-induced decrease in cell proliferation and ameliorated the cell apoptosis induced by this toxin. Moreover, in the toxin A-induced mouse enteritis model, Peri-2 blocked the mucosal disruption and inflammatory response caused by toxin A. These results suggest that the American cockroach peptide Peri-2 could be a possible drug candidate for addressing the pseudomembranous colitis caused by C. difficile toxin A.

  • 상악 전치부 외상으로 인한 치관-치근 파절의 경우 파절선이 치은연하에 위치할 때 결손된 골조직과 연조직을 재생성하기 위해 교정적 정출술이 제안되었다. 교정적 정출술은 심미적인 결과와 좋은 예후를 나타내는 쉬운 교정 접근법 중 하나이다. 그러나 강한 힘을 사용하거나 매우 빠른 교정력을 가하게 되면 조직손상이나 유착이 발생할 수 있다. 다음 증례는 두 환자에서 외상으로 인한 상악전치의 치관-치근 파절에 대해 교정적 정출술을 이용한 임상치료과정을 서술하였다.

  • Background and Objectives : Traditionally, the excision of submandibular gland (SMG) has been commonly used for treatment of calculi in the proximal duct or gland parenchyma. Over the last 10 years several new minimally invasive techniques including lithotripsy, sialendoscope were introduced in the treatment of sialolithiasis. But these have some limitation on large, infected calculi. The aim of this study is to assess the intraoral treatment of submadibular stones. Subjects and Method : The records of one hundred and seventy-three patients who underwent intraoral removal of submadibular sialolithiasis between June 1, 1989 and July 31, 2006 were retrospectively reviewed. Results : Stone location was distal to the edge of the mylohyoid muscle in 127 patients and proximal to gland in 48 patients (mean size of sialoliths, 7.1mm [range 3.0-25mm]). The complete removal of stones was observed in 170 (97.1%) patients regardless of size and location. Recurrence of lithiasis was found in 8 patients (then treated with intraoral removal in 5 patientsand resection of SMG (submandibular glands) in 3 patients). Acalculous sialadenitis in 9 patients (5.1%) and cyst formation in 2 patients (1.1%) was found. But no evidence of postoperative complications including hemorrhage, fistula, damage to lingual nerve were found. Conclusion : The intraoral removal of submandibular stone is useful in preservation of submandiblar function and effective in palpable stones regardless of location, size.

다중하중에 따른 경사제 피복재의 추계학적 신뢰성 해석 (Stochastic Reliability Analysis of Armor Units of Rubble-Mound Breakwaters Subject to Multiple Loads)

  • 경사제에 불규칙하게 작용하는 임의 크기의 다중하중으로 인해 피복재의 안정성에 대한 성능이 시간에 따라 어떻게 달라지는지를 해석할 수 있는 추계학적 신뢰성 해석 모형이 개발되었다. Hudson의 공식과 Melby 공식을 이용하여 재현기간에 따른 파고의 함수로 경사제 피복재의 초기 저항력 크기와 피해율을 확률적으로 산정할 수 있는 새로운 방법이 제시되었다. 생애기간에 대한 신뢰성 분석을 실시하여 시간에 따른 다중하중의 작용과 사용한계나 극한한계 등 한계상태에 따른 구조물의 성능을 올바로 해석할 수 있었다. 마지막으로 보수보강 목표확률을 시간에 따른 누적파괴확률의 결과와 조합하여 구조물 유지관리에서 가장 중요한 변수인 보수보강 시점을 정량적으로 산정할 수 있는 방법이 제시되었다.

  • 중추신경계는 일단 손상이 되면 손상된 세포의 재생, 손상된 수초의 회복, 신경계의 정상적인 연결 등의 제한성 때문에 그 회복이 매우 힘들다. 이러한 중추신경계의 중요한 손상으로는 다발성 경화증, 뇌졸중, 척수손상, 외상, 축삭의 탈수초화 등이 있다. 이전 연구들은 많은 발생빈도를 보이고 있는 척수손상에서 실질적인 척수의 기능적인 회복을 위해 손상된 척수신경의 재생과 축삭의 재수초화가 중요한 요인이라고 전하고 있다. 최근에는 이러한 척수손상에 대한 치료적 접근으로서 세포이식 기술이 하나의 해결책을 열어주고 있다. 따라서 본 논문에서는 척수손상의 특성을 살펴보고, 척수손상에 의한 기능장애에 대해 세포이식이 기능의 회복을 증진시킬 수 있다는 증거를 논의하고자 한다.

  • 원자로 정지동안에도, 잔열제거계통은 그 기능이 계속 유지되어야 하나, 실제로 가압 경수로에서 냉각상실고가 많이 발생되어 있다. 본 논문은 원자로 정지중의 냉각기능상실을 예방하고, 또한 냉각기능상실로 인한 노심손상의 중대성을 완화시키기 위한 대책을 강구하기 위한 시도로서, 전형적인 가압경수로에 대한 사고/고장 수목과 운전원실수 확률을 위한 HCR 모델, 초기 사상의 빈도를 위한 2단계 bayesian 방법 및 고장난 계통의 회복 활률을 위한 계단함수 모델 등을 이용한 원자로 정지 위해도 모델을 개발하여, 잔열제거계통의 신뢰도를 분석하였다. 그 결과는 원자로가 정지 중일 때의 위해도가 운전중일 때 이것에 비해 별로 낮지 않은 것으로 나타났으며, 몇 가지의 설계개선을 통하여 냉각기능상실로 인한 노심 손상확률을 상당히 낮출 수 있는 것으로 나타났다.

Structural modal identification and MCMC-based model updating by a Bayesian approach

  • Finite element analysis is one of the important methods to study the structural performance. Due to the simplification, discretization and error of structural parameters, numerical model errors always exist. Besides, structural characteristics may also change because of material aging, structural damage, etc., making the initial finite element model cannot simulate the operational response of the structure accurately. Based on Bayesian methods, the initial model can be updated to obtain a more accurate numerical model. This paper presents the work on the field test, modal identification and model updating of a Chinese reinforced concrete pagoda. Based on the ambient vibration test, the acceleration response of the structure under operational environment was collected. The first six translational modes of the structure were identified by the enhanced frequency domain decomposition method. The initial finite element model of the pagoda was established, and the elastic modulus of columns, beams and slabs were selected as model parameters to be updated. Assuming the error between the measured mode and the calculated one follows a Gaussian distribution, the posterior probability density function (PDF) of the parameter to be updated is obtained and the uncertainty is quantitatively evaluated based on the Bayesian statistical theory and the Metropolis-Hastings algorithm, and then the optimal values of model parameters can be obtained. The results show that the difference between the calculated frequency of the finite element model and the measured one is reduced, and the modal correlation of the mode shape is improved. The updated numerical model can be used to evaluate the safety of the structure as a benchmark model for structural health monitoring (SHM).

Reverting Gene Expression Pattern of Cancer into Normal-Like Using Cycle-Consistent Adversarial Network

  • Cancer show distinct pattern of gene expression when it is compared to normal. This difference results malignant characteristic of cancer. Many cancer drugs are targeting this difference so that it can selectively kill cancer cells. One of the recent demand for personalized treating cancer is retrieving normal tissue from a patient so that the gene expression difference between cancer and normal be assessed. However, in most clinical situation it is hard to retrieve normal tissue from a patient. This is because biopsy of normal tissues may cause damage to the organ function or a risk of infection or side effect what a patient to take. Thus, there is a challenge to estimate normal cell's gene expression where cancers are originated from without taking additional biopsy. In this paper, we propose in-silico based prediction of normal cell's gene expression from gene expression data of a tumor sample. We call this challenge as reverting the cancer into normal. We divided this challenge into two parts. The first part is making a generator that is able to fool a pretrained discriminator. Pretrained discriminator is from the training of public data (9,601 cancers, 7,240 normals) which shows 0.997 of accuracy to discriminate if a given gene expression pattern is cancer or normal. Deceiving this pretrained discriminator means our method is capable of generating very normal-like gene expression data. The second part of the challenge is to address whether generated normal is similar to true reverse form of the input cancer data. We used, cycle-consistent adversarial networks to approach our challenges, since this network is capable of translating one domain to the other while maintaining original domain's feature and at the same time adding the new domain's feature. We evaluated that, if we put cancer data into a cycle-consistent adversarial network, it could retain most of the information from the input (cancer) and at the same time change the data into normal. We also evaluated if this generated gene expression of normal tissue would be the biological reverse form of the gene expression of cancer used as an input.

Effect of multiple-failure events on accident management strategy for CANDU-6 reactors

  • Lessons learned from the Fukushima Daiichi nuclear power plant accident directed that multiple failures should be considered more seriously rather than single failure in the licensing bases and safety cases because attempts to take accident management measures could be unsuccessful under the high radiation environment aggravated by multiple failures, such as complete loss of electric power, uncontrollable loss of coolant inventory, failure of essential safety function recovery. In the case of the complete loss of electric power called station blackout (SBO), if there is no mitigation action for recovering safety functions, the reactor core would be overheated, and severe fuel damage could be anticipated due to the failure of the active heat sink. In such a transient condition at CANDU-6 plants, the seal failure of the primary heat transport (PHT) pumps can facilitate a consequent increase in the fuel sheath temperature and eventually lead to degradation of the fuel integrity. Therefore, it is necessary to specify the regulatory guidelines for multiple failures on a licensing basis so that licensees should prepare the accident management measures to prevent or mitigate accident conditions. In order to explore the efficiency of implementing accident management strategies for CANDU-6 plants, this study proposed a realistic accident analysis approach on the SBO transient with multiple-failure sequences such as seal failure of PHT pumps without operator's recovery actions. In this regard, a comparative study for two PHT pump seal failure modes with and without coolant seal leakage was conducted using a best-estimate code to precisely investigate the behaviors of thermal-hydraulic parameters during transient conditions. Moreover, a sensitivity analysis for different PHT pump seal leakage rates was also carried out to examine the effect of leakage rate on the system responses. This study is expected to provide the technical bases to the accident management strategy for unmitigated transient conditions with multiple failures.

Hot Water Extract of Triticum aestivum L. (Common Wheat) Ameliorates Renal Injury by Inhibiting Apoptosis in a Rat Model of Ischemia/Reperfusion

  • Objectives : Interruption and subsequent restoration of blood flow into the kidney result in renal injury. As an approach to preventing the renal injury, we determined the optimal conditions and the underlying mechanisms by which supernatant of hot water extract of ground Triticum aestivum L. (extract) attenuated ischemia/reperfusion (I/R) injury. Methods : One hour after administration of the extract (400 mg/kg) by intraperitoneal injection, renal I/R injury was generated by clamping the left renal artery in rats after surgical removal of the right kidney, followed by reperfusion. The maximal difference between the vehicle-treated and the extract-treated group under ketamine/xylazine or enflurane anesthetization was assessed at varying periods of ischemia (30-45 min) and reperfusion (3-48 hr), based on the renal function assessed with serum creatinine levels, tissue injury with hematoxylin/eosin staining, and apoptosis with terminal deoxynucleotidyltransferase-mediated dUTP nick-end labeling staining. Results : Enflurane anesthetization with 40 min of ischemia and 24 hr of reperfusion was identified to be the optimal condition, under which condition serum creatinine levels and tubular damage in the extract-treated group were significantly reduced compared with those in the vehicle-treated group ($1.3{\pm}0.2$ versus $2.7{\pm}0.3$ mg/dL, P < 0.01, and average score $1.8{\pm}0.1$ versus $3.5{\pm}0.3$, P < 0.01, respectively). These beneficial effects were mediated by inhibition of apoptotic cascades through attenuation of renal tissue malondialdehyde levels, Bax/Bcl-2 ratio and caspase-3 levels. Conclusions : The extract conferred renal protection against ischemia/reperfusion injury in rats by scavenging reactive oxygen species and consequently blocking apoptotic cascades, plausibly augmented by enflurane protection.
