Nondestructive Examination of Ferromagnetic Tube Using Magnetic Saturation Eddy Current Technique (자기포화 와전류기법에 의한 자성 튜브 비파괴검사)

  • The tubes in heat exchanger are typically made from copper alloy, stainless steel, carbon steel, titanium alloy material. type-439 ferritic stainless steel is ferromagnetic material, and furnish higher heat transfer rates than austenitic stainless steels and higher resistance to corrosion-induced flaws. Ferritic stainless steel can typically be found in low-pressure(LP) feedwater heaters and moisture separator reheaters(MSRs). LP feedwater heaters generally utilize thin wall type-439 stainless steel tubing, whereas MSRs typically employ a heavier wall tubing with integral fins. Service-induced damage can occur on the OD(outside diameter) surface of type-439 ferritic stainless steel tubing which is employed for MSRs tubing, and the most typical damage mechanism is vibration-induced tube-to-TSP(tube support plate) wear and fatigue cracking. The wear has been reported that occurs mainly on the OD surface. Accordingly, in this study, we have evaluated the flaw sizing capability of magnetic saturation eddy current technique using magnetic saturation probe and flawed specimen.

Gamma-ray Irradiation on Radio Sensitivity in Cnidium officinale Makino (천궁 돌연변이 유발을 위한 최적 감마선 조사량)

  • Background: Cnidium officinale Makino have been used in traditional medicine in Northeast Asia. Although gamma-ray mutagenesis has been used to develop breeding resources with novel characteristics, research on the radiation sensitivity of C. officinale Makino is limited. Hence, the optimal gamma-ray dosage for mutation breeding in C. officinale Makino was investigated. Methods and Results: Seedstocks were exposed to doses of gamma rays (5 Gy - 50 Gy), and subsequently planted in a greenhouse. After 30 days of sowing, the survival rates and growth decreased rapidly at doses above 20 Gy, while all individuals died at 50 Gy. The median lethal dose (LD50) was 25.65 Gy, and the median reduction doses (RD50) for plant height, number of stems, and fresh weight were 12.81, 9.32, and 23.26 Gy, respectively. Post-irradiation levels of malondialdehyde (MDA), peroxidase (POD), and chlorophyll in the aerial parts of the plant were quantified using spectrophotometry. Relative to the controls, the levels of MDA and POD increased, while the level of chlorophyll decreased at doses ≥ 10 Gy, indicating cellular damage. Conclusions: A dose of 20 Gy was found to be optimal for mutation breeding in C. officinale Makino.

Optimization of orthodontic microimplant thread design (교정용 마이크로 임플란트의 나사산 디자인 최적화)

  • Objective: The purpose of this study was to optimize the thread pattern of orthodontic microimplants. Methods: In search of an optimal thread for orthodontic microimplants, an objective function stability quotient (SQ) was built and solved which will help increase the stability and torsional strength of microimplants while reducing the bone damage during insertion. Selecting the AbsoAnchor SH1312-7 microimplant (Dentos Inc., Daegu, Korea) as a control, and using the thread height (h) and pitch (p) as design parameters, new thread designs with optimal combination of hand p combination were developed. Design soundness of the new threads were examined through insertion strain analyses using 3D finite element simulation, torque test, and clinical test. Results: Solving the function SQ, four new models with optimized thread designs were developed (h200p6, h225p7, h250p8, and h275p8). Finite element analysis has shown that these new designs may cause less bone damage during insertion. The torsional strength of two models h200p6 and h225p7 were significantly higher than the control. On the other hand, clinical test of models h200p6 and h250p8 had similar success rates when compared to the control. Conclusion: Overall, the new thread designs exhibited better performance than the control which indicated that the optimization methodology may be a useful tool when designing orthodontic microimplant threads.

Scoping for Marine Environmental Impact Assessment of Tidal Power Plant Construction (조력에너지 개발사업의 해양환경영향평가 스코핑 방안)

  • This paper assessed environmental impact of tidal power plant construction in the western sea of Korea, and diagnosed problems of the related assessment. We also proposed key assessment items (scoping) and system improvement for environmental impact assessment (EIA). The establishment of a broad consensus of the appropriateness and a clear purpose for business is an important aspect of the aptness of using the waters, and the aggressive collecting of opinions of the stakeholder and institutions will have to be fulfilled. In addition, we presented the following scoping plan in order to minimize the negative effects of the marine ecosystem and the conflict between the people who work in the fishing industry which result due to the construction of the tidal power plants. (1) the change in seawater exchange rates and the aspect of erosion/sedimentation which result from the change in the velocity of running fluid, (2) the destruction of spawns/habitats of fish due to the damage of tidal flats, (3) fishery resources, impacts of fishing grounds, and the spread of suspended sediments, etc. will have to be applied to the key assessment items(scoping). In addition, every citizen will have work hard for an establishment of an active plan which achieves the harmony and balance of environmental preservation and for the policy to be applied through changing their consciousness of environmental preservation.

The Study on Mucin Release by Airway Goblet Cells in Primary Culture

  • Surface epithelial cells isolated from hamster tracheas and grown on a thick collagen gel become a highly enriched population of mucus-secreting cells. Epithelial cells from tracheas of hamsters were collected using enzymatic procedures and cultured under various conditions. The medium used consisted of a 1:1 mixture of medium 199 and Dulbecco's modified Eagle's (DME) medium which was conditioned before use. Insulin, transferrin, hydrocortisone, epidermal growth factor, and extract from bovine hypothalamus were used as supplement. Due to relatively low basal rates of min secretion from in vitro cultures, cultures are generally radiolabeled using $^3$H-glucosamine as a metabolic precursor. The radiolabeled mucinsreleased are quantitated by precipitation with TCA/PTA. Using this cell culture system, we investigated mucin release of goblet cells by altering the media bathing the apical surface of hamster tracheal surface epithelial(HTSE) cells. Acidic media added sulfuric acid caused sigcificant increases in mucin relesse (155${\pm}$20% at pH 4 and 146${\pm}$16% at, pH 5). Ammonium hydroxide also increased mucin release at pH 9.0(156${\pm}$17%) and pH 10(295${\pm}$9%) respectively. This additional mucin release seems to be associated with cell membrane damage as indicated by release of cellular LDH. SP stimulates secretion of mucin in cultured HTSE cells(154${\pm}$16% at 1${\times}$10$\^$-6/M and 165${\pm}$25% at 1${\times}$10$\^$-5/M. PAF at 5${\times}$10$\^$-6/M and 5${\times}$10$\^$-5/M enhanced by HTSE cells in vitro 168${\pm}$34% and 259${\pm}$30% of mucin secretion, respectively. The increase in mucin release by PAF and SP was not secondary to cell damage or necrosis. SP and PAF may be in mediating mucous secretion induced by inflammation irritantion and infection.

Effect of Calcium Chloride (CaCl2) on the Characteristics of Photosynthetic Apparatus, Stomatal Conductance, and Fluorescence Image of the Leaves of Cornus kousa (염화칼슘 처리가 산딸나무 잎의 광합성 기구, 기공전도도 및 형광이미지 특성에 미치는 영향)

  • Deicing salt is used to melt snow and ice on the road for traffic safety during the winter season, which accumulates in the roadside vegetation and induces visible injuries. The damage may accelerate particularly when it coincides with early spring leaf out. In order to better understand the response mechanisms, C. kousa (3-year-old) was irrigated twice prior to leaf bud in a rhizosphere with solutions of 0.5, 1.0, and 3.0% calcium chloride ($CaCl_2$) concentration, that were made by using an industrial $CaCl_2$ reagent practical deicing material in Seoul. Physiological traits of the mature leaves were progressively reduced by $CaCl_2$ treatment, resulting in reductions of total chlorophyll contents, chlorophyll a:b, photosynthetic rate, quantum yield, stomatal conductance, $F_V/F_M$, and NPQ. On the contrary, light compensation point and dark respiration were increased at high $CaCl_2$ concentration. A decrease in intercellular $CO_2$ concentration by stomatal closure first resulted in a reduced photosynthetic rate and then was accompanied by low substance metabolic rates and photochemical damage. Based on the reduction of physiological activities at all treatments ($CaCl_2$ 0.5%, 1.0%, and 3.0%), C. kousa was determined as one of the sensitive species to $CaCl_2$.

Seasonal Occurrence of Insect Pests and Related Yield Loss in Amaranth Crop in South Korea (국내 아마란스(Amaranthus spp.)에 발생하는 주요 해충의 발생소장과 수량 감소율)

  • This study was carried out to investigate insect pests and their damage caused to commercial amaranth in Gangneung, Gangwon-do, Korea, during 2019-2020. A total of seven families and 18 species were identified including three species of aphids, twelve species of stink bugs, and four species of moths. Among them, five species caused significant economic damage due to a large number of occurrences; Aphis fabae, Nysius plebejus, Polymerus cognatus, Spoladea recurvalis and Spodoptera exigua. Aphids started to occur from the end of April, and showed the greatest incidence in early June. Three Nysius species occurred from mid-May and showed the maximum density in late August, and then its population decreased from the beginning of September. Two species of moth caterpillars began to occur from mid-August and damaged amaranth until mid-October. Comparing the yields between amaranth plants with and without a spray of pesticides for each pest, the yield loss of seed by aphids and stink bugs were 51.9% and 69.8%, respectively. Also, the reduction rates of leaf biomass by S. recurvalis and S. exigua were 72.5% and 36.5%, respectively.

The antioxidant icariin protects porcine oocytes from age-related damage in vitro

  • Objective: If fertilization does not occur within a specific period, the quality of unfertilized oocytes in the oviduct (in vivo aging) or in culture (in vitro aging) will deteriorate over time. Icariin (ICA), found in all species of Epimedium herbs, has strong antioxidant activity, and is thought to exert anti-aging effects in vitro. We asked whether ICA protects oocytes against age-related changes in vitro. Methods: We analyzed the reactive oxygen species (ROS) levels and expression of antioxidant, maternal, and estrogen receptor genes, and along with spindle morphology, and the developmental competence and quality of embryos in the presence and absence of ICA. Results: Treatment with 5 μM ICA (ICA-5) led to a significant reduction in ROS activity, but increased mRNA expression of glutathione and antioxidant genes (superoxide dismutase 1 [SOD1], SOD2, peroxiredoxin 5, and nuclear factor erythroid 2-like 2), during aging in vitro. In addition, ICA-5 prevented defects in spindle formation and chromosomal alignment, and increased mRNA expression of cytoplasmic maturation factor genes (bone morphogenetic protein 15, cyclin B1, MOS proto-oncogene, serine/threonine kinase, and growth differentiation factor-9). It also prevented apoptosis, increased mRNA expression of antiapoptotic genes (BCL2-like 1 and baculoviral IAP repeat-containing 5), and reduced mRNA expression of pro-apoptotic genes (BCL2 antagonist/killer 1 and activation of caspase-3). Although the maturation and cleavage rates were similar in all groups, the total cell number per blastocyst and the percentage of apoptotic cells at the blastocyst stage were higher and lower, respectively, in the control and ICA-5 groups than in the aging group. Conclusion: ICA protects oocytes against damage during aging in vitro; therefore, it can be used to improve assisted reproductive technologies.

Effect of Aceton Extract from Styela Clava on Oxidative DNA Damage and Anticancer Activity (미더덕 아세톤 추출물이 산화적 DNA 손상억제 및 암세포 독성에 미치는 영향)

  • Styela clava (also called as rough sea squirt or leathery tunicate) is regarded as native to the northwest Pacific region including Korea and widely distributed in parts of northwestern Europe, North America and Australia. To evaluate Styela clava as a potential bioactive agent, the antioxidant activity of aceton extracts from Styela clava (whole, substance and tunic) was tested by measuring inhibitory effect of $H_2O_2$ induced DNA damage using comet assay. Also, anticancer activity on human colon cancer cell (HT-29) was investigated by MTT reduction assay. The $200\;{\mu}M$ $H_2O_2$ induced DNA damage was inhibited with Styela clava aceton extract in dose dependent manner in human leukocytes. The maximum inhibition was by 62.8, 62.1 and 78.3% at the concentration of $50\;{\mu}g/ml$ of whole, substance and tunic extracts, respectively. The aceton extracts from S. clava were also found to inhibit the growth of human colon cancer cell. The cell proliferation rates decreased to 26.9, 30.6 and 12.0% at the concentration of $500\;{\mu}g/ml$ of whole, substance and tunic extracts, respectively. These results support that aceton extracts from S. clava may be a potential candidate as a possible antimutagenic and chemotherapeutic agent.

Preliminary Results of a Phase I/II Study of Simultaneous Boost Irradiation Radiotherapy for Locally Advanced Nasopharyngeal Carcinoma

  • Background: The purpose of this article is to present preliminary results of simultaneous boost irradiation radiotherapy for locally advanced nasopharyngeal carcinoma (NPC). Methods: Fifty-eight patients who underwent simultaneous boost irradiation radiotherapy for NPC in Cancer Center of Sun Yat-sen University between September 2004 and December 2009 were eligible. Acute and late toxicities were scored weekly according to the Radiation Therapy Oncology Group (RTOG) acute and late radiation morbidity scoring schemes. An especial focus was on evidence of post-radiation brain injury. Also quality of life was analysed according to the EORTC (European Organisation for Research and Treatment of Cancer) recommendations. Discrete variables were compared by ${\chi}^2$ test. The Kaplan-Meier method was used to calculate the survival rates and generate survival curves. Results: A total of 58 patients with a mean follow-up time of 36 months completed clinical trials.Fifty-seven patients (98.3) achieved complete remission in the primary sites and cervical lymph nodes, with only one patient (1.7%) showing partial remission.The most frequently observed acute toxicities during the concurrent chemoradiotherapy were mucositis and leucopenia. Four patients (6.9%) had RTOG grade 3 mucositis, whereas four patients (6.9%) had grade 3 leucopenia. No patient had grade 4 acute toxicity. Three (5.17%) of the patients exhibited injury to the brain on routine MRI examination, with a median observation of 32 months (range, 25-42months). All of them were RTOG grade 0. The 3-year overall, regional-free and distant metastasis-free survival rates were 85%, 94% and 91%, respectively. Conclusion: Simultaneous boost irradiation radiotherapy is feasible in patients with locally advanced nasopharyngeal carcinoma. The results showed excellent local control and overall survival, with no significant increase the incidence of radiation brain injury or the extent of damage. A larger population of patients and a longer follow-up period are needed to evaluate ultimate tumor control and late toxicity.
