Aβ42로 유도된 알츠하이머 마우스 모델에서 이중 가공 인삼열매 추출물의 학습 및 기억 손실 개선 효과 (Double-processed ginseng berry extracts enhance learning and memory in an Aβ42-induced Alzheimer's mouse model)

  • AD는 뇌의 신경세포 사멸뿐만 아니라 학습 및 기억능 소실을 초래하는 퇴행성 뇌 질환이며, 기억과 관련된 주요 뇌 구역인 해마(hippocampus)는 콜린성 조절(cholinergic modulation)에 의해 영향을 받는다(Konishi 등, 2015). AD 병인에 대한 다양한 해석과 설명이 있으나 크게는 amyloid cascade hypothesis과 함께 cholinergic hypothesis가 주류를 이루고 있다. 몇몇의 연구에서 ChAT 합성, ACh 분비, nicotin 및 muscarinic 수용체 감소가 AD 뇌의 대뇌피질과 해마에서 관찰되어(Tata 등, 2014) 퇴행성 뇌질환에서의 중요성이 제시되었고, 이를 배경으로 한 acetylcholinesterase inhibitors (AChEI)가 AD 증상 완화의 목적으로 미국 FDA로부터 승인되어 시판되고 있다. 본 연구에서는 인삼의 활성 성분인 진세노사이드가 다량 함유된 PGBC가 $A{\beta}42$로 유도된 치매 모델에서 뇌세포 보호, ACh 분비 증가, 학습력/기억력 증가, ChAT 발현 증가를 확인하여 인삼열매 추출물의 치매 적용 여부를 확인하고자 하였다. 결과에서 언급한 바와 같이 PGBC는 익기 직전의 4년근 인삼 열매에 추출 및 발효 단계를 추가하여 확보된 물질로서, $A{\beta}42$ 섭취, 제거 및 ACh 분비 촉진 활성이 있는 Re, Rd, Rg3 함량이 증가되어(Kim 등, 2014; Jang 등, 2015), PGBC 자체로서 치매 인자에 대한 조절 효능이 예측되었다. 특히, 7증 7포 및 발효과정을 거친 이중가공 인삼 열매 추출물이 비 발효 증포 추출물에 비해 Rg3가 현저히 증가하는 사전 연구결과와 Rg3가 $A{\beta}42$ 제거 활성을 가지는 것으로 확인된 결과를 종합할 때 PGBC 투여가 AD 증상 완화의 조절자 역할을 할 것으로 생각된다(Kim 등, 2013; Jang 등 2015). 본 연구에 따르면, 마우스 치매 모델에 PGBC 처리 시 PAT 및 Morris water-maze test를 통해 대조군 대비 유의한 수준의 인지능력 개선, ACh 합성을 유도하는 ChAT 유전자 발현 증가, ACh 분비량 증가 등이 확인되어 전체적인 인지능 개선에 극적인 영향을 준 것으로 판단된다. 특히 $A{\beta}42$를 뇌 실로 주입(intracranial injection) 하여 나타나는 뇌세포 손상이 PGBC 투여를 통해 보호된 것으로 사료되었으며, 이는 주요 뇌세포 중 하나인 성상세포에서 관찰되는 GFAP 분석을 통해 확인되었다. 뇌 균질액을 이용한 AChE 활성 연구에서도 PGBC가 AChE를 현저하게 저해하는 것으로 확인되어, AD 환자에게 처방이 가능한 2대 의약품 군중 하나인 시냅스 내 신경전달물질 ACh bioavailability 증가 목적의 처방 보조요법 적용이 기대된다(${\check{C}}olovi{\acute{c}}$ 등, 2013). 결론적으로, 본 연구에 사용된 PGBC는 학습 및 기억력을 개선하는 활성물질을 포함하고 있어 1차적인 퇴행성 뇌질환 보조재로서 직간접적인 대증요법 역할과 2차적으로는 뇌 세포 보호를 통한 질병 악화 지연 소재로 개발이 기대되며, 보다 심도 있는 기전연구를 통해 천연물 신소재 개발도 가능할 것으로 사료된다.

Oncoflash를 적용한 전신 뼈 영상 검사의 스캔 속도 결정에 관한 연구 (A Study on the Determination of Scan Speed in Whole Body Bone Scan Applying Oncoflash)

  • 전신 뼈 영상 검사에서 영상의 질을 유지하면서 검사 시간 단축을 위해 oncoflash와 같은 프로그램을 사용하고 있는데, 이를 임상에 적용할 때 영상의 질과 검사 시간 단축의 두 가지 측면을 동시에 고려해야 하며, 적합한 스캔 속도의 결정에 대한 기준 설정이 필요하여 본 연구를 시행하였다. 서울아산병원 핵의학과에서 전신 뼈 영상 검사를 시행한 환자를 대상으로 하여 분당 30 cm의 스캔 속도로 검사한 영상의 총 계수를 각각 800 K 이하, 800 K, 900 K, 1000 K, 1500 K, 2000 K 이상으로 구분하여 영상의 질을 정성적 분석하고, 전신 총 계수가 1000 K 미만인 비율을 분석하였다. 분당 30 cm의 스캔 속도로 검사한 전신 뼈 영상의 전신 총 계수의 기하학적 평균이 1000 K 미만인 비율을 분석한 결과, 전체 329명 중 58명(17.6%)이다. 전신 총 계수 별로 구분한 영상을 정성적으로 분석 결과, 전신 총 계수 1000 K 이하의 경우에 육안적으로 영상의 입자가 거칠고, 노이즈가 증가한 영상으로 평가되었다. 전신 총 계수의 양과 Oncoflash 적용 여부에 따른 해상력의 비교평가를 위해 전신 뼈 영상 검사의 계수율을 고려하여 2~5 mCi의 선량을 투여한 $^{99m}Tc$ Flood와 4-Quadrant bar phantom을 이용하여 영상을 획득 한 후, Oncoflash 적용 전과 후의 FWHM을 분석하였다. PPM 계수가 3.6 K 이하에서는 Oncoflash 적용 후의 FWHM이 전과 비교해서 높게 나왔으나, 3.6~4.0 K 이상에서는 Oncoflash 적용 전보다 적용 후에 낮거나 동등하게 측정되었다. 전신 뼈 영상 검사의 시작 시점인 두부와 흉부의 일부가 포함된 상태에서 감마카메라의 PPM (Patient Positioning Monitor) 계수를 측정하고, 분당 30 cm의 스캔 속도를 적용하여 획득한 전신 총 계수 사이의 상관 관계를 분석 결과는 PPM 계수가 각각 2.5~3.0 K일 때 전신 총 계수의 평균과 표준 편차는 $965{\pm}173\;K$ (17.8%), 3.1~3.5 K에서 $1,084{\pm}154\;K$ (22.4%), 3.6~4.0 K에서 $1,242{\pm}186\;K$ (18.4%), 4.1~4.5 K에서 $1,359{\pm}170\;K$ (17.1%), 4.6~5.0 K에서 $1,405{\pm}184$ K (10.5%), 5.1~6.0에서 $1,640{\pm}376\;K$ (10.5%), 6.1~7.0 K에서 $1,771{\pm}324\;K$ (1.3%), 7.1 K 이상에서 $1,972{\pm}385\;K$ (2.0%)으로, 매우 강한 양의 상관관계(상관계수 r=0.775, p<0.01)가 있었다. 전신 뼈 영상 검사 시작 시점의 PPM 계수가 3.6 K 이상일 경우에 분당 30 cm으로 스캔한 후 전신 총 계수가 1000 K 이상인 경우에 Oncoflash를 적용하면 검사 시간을 줄이면서도 기존의 분당 15 cm으로 획득한 영상 수준의 화질을 얻을 수 있었다. 그러나 분당 30 cm으로 스캔한 후 전신 총 계수 기준으로 1000 K 미만인 경우에는 Oncoflash를 적용하여도 영상의 질이 저하되므로, 기존의 분당 15 cm의 속도로 다시 시행하는 것을 고려해야 할 것이다.

선량 및 선량률 변화에 따른 말초혈액 임파구의 염색체 이상의 빈도 (The Frequency of Chromosomal Aberrations of Peripheral Lymphocytes according to Radiation Dose and Dose Rate)

  • 목적 : X-선 조사 후 이동원 염색체와 환형 염색체의 빈도가 피폭 선량과 비례하는지를 확인하고, X-선 조사시 선량률의 변화가 생물학적 선량 측정에 영향을 주는지를 찾아서 보정인자로 작용하는지를 알아보았다. 대상 및 방법 : 28세의 건강한 성인남자에서 2회에 걸쳐 각각 16 ml와 18 ml의 전혈을 정맥으로부터 무균채취하여 이미 해파린(heprin) 처리되어 있는 10 ml의 vacutainer (삼화공사, 한국)에 2 ml씩 분주하였다. 분주한 혈액 시료를 1개는 정상대조 군으로 16개는 실험 군으로 총 17개를 실험에 사용하였으며, 방사선 조사는 조사야 10${\times}$10 cm$^{2}$로 분당 240 cGy 의 일정한 선량률로 50, 100, 200, 300, 400, 600, 800 cGy의 선량을 각 1회 조사하였다. 나머지 9개의 실험 군은 동일한 선량(400 cGy)에서 선량률 변화만 주어 20, 40, 60, 80, 100, 160, 240, 320, 400 cGy/min의 각 선량률로 1회 조사하였다. 관찰 시 염색체의 개수가 46${\times}$2개 되는지 여부를 확인하고 또한 염색체의 밀도나 염색체 형태가 염색체 이상 빈도 확인에 적합한지 확인한 후 해당되는 경우에 한해서만 계측하였다. 각 표본에서 최소 50개의 림프구를 관찰하려고 하였다. 통계처리는 SPSS를 이용하여 평균값 비교와 회귀곡선의 기울기 값을 구하였다. 결과 : 방사선 조사된 림프구의 관찰된 총 수에 대한 염색체 이상을 가진 림프구의 비율(yield)은 선량이 50, 100, 200, 300, 400, 600, 800 cGy일 때 각각 0, 9, 20, 27, 55, 88, 100%이었다. 선량변화에 따른 염색체 이상(이동원 및 환형 염색체)의 평균빈도는 50, 100, 200, 300, 400, 500, 800 cGy일 때 각각 0.000, 0.093, 0.200, 0.354, 0.612, 2.040, 2.846이었다. 관찰된 총림프구에서 이동원 염색체와 환형 염색체의 빈도(Ydr)는 선량이 50, 100, 200, 300, 400, 600, m cGy일 때 각각 0.000, 0.093, 0.200, 0.364, 0.613, 2.040, 2.846이었다. 방사선량과 Ydr의 관계는 Ydr=$\alpha$D+$\beta$D$^{2}$에 의한 Liner-quadratic model을 따를 때 Ydr=0.188${\times}$10$^{-2}$ D/Gy+0.422${\times}$10$^{-4}$/Gy$^{2}$${\times}$D$^{2}$로 나타났다. 이동원 염색체와 환형 염색체 이상을 가지는 림프구에서 이동원 염색체와 환형 염색체의 빈도(Qdr)는 50, 100, 200, 300, 400, 600, 800의 선량일 때 각각 0.000, 1.000, 1.000, 1.333, 1.118, 2.318, 2.846 이었으며 이는 앞의 Ydr값으로부터 구한 값과 거의 일치하였다. 선량율을 각각 20, 40, 60, 80, 100, 160, 240, 320, 400 cGy/min으로 달리하여 총 선량을 400 cGy를 각각 조사했을 때 임파구에서 나타난 염색체이상(이동원, 환형, 무동원체단편 염색체)의 빈도의 차는 없었다. 결론 : 방사선의 급성 피폭에 의한 손상은 선량률에 의한 영향은 미미하며 대부분 피폭된 총 선량에 의해 좌우된다. 염색체 이상을 이용한 생물학적 선량측정방법은 피폭 선량이 증가함에 따라 비교적 규칙적으로 증가하므로 피폭 선량 측정을 위한 유용한 방법이라고 생각한다. 그러나 일회 피폭인 급성 피폭 시는 선량률 변화가 보정인자로써 작용하지는 않았다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The frame and leaflet of the polymer valve were made from polyurethane, To evaluate the hemodynamic performance of the polymer valve a comparative study of flow dynamics past a polymer valve and a St. Jude Medical prosthetic valve under physiological pulsatile flow conditions in vitro was made. Comparisons between the valves were made on the transvalvular pressure drop, regurgitation volume and maximum valve opening area. The polymer valve showed smaller regurgitation volume and transvalvular pressure drop compared to the mechanical valve at higher heart rate.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N.
