Extractives from the leaves of Thuja orientalis Linnaeus (측백나무(Thuja orientalis Linnaeus) 잎의 추출성분)

  • 2kg of the dried leaves of Thuja orientalis Linnaeus were ground, extracted with acetone-$H_2O$(7:3, v/v), concentrated, and fractionated with a series of hexane, $CH_2C1_2$ EtOAc and water on a separators funnel. Each fraction was freeze dried to give dark-brown powder and a EtOAc soluble portion. of the powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. Spectrometric analyses such as NMR and FAB-MS including TLC were performed to characterize the structures of isolated compounds. The leave of Thuja orientalis Linnaeus contained a large amount of flavononol derivatives such as quercetin-3-O-$\alpha$-L-rhamnopyranoside and myricetin-3-O-$\alpha$-L-rhamnopyranoside in addition to a small amount of flavan compounds such as (+)-catechin and (+)-gallocatechin. The antioxidative activities of each fractions and isolated compounds were done by DPPH radical scavenging test, and all of them were indicated strong antioxidative activities.

Antioxidant and antimicrobial activity of solvent fractions from black bamboo leaves (오죽 잎 용매분획물의 항산화 및 항균 활성)

  • To investigate the natural antioxidant and antimicrobial phytochemicals from black bamboo (Phyllostachys nigra MUNRO) leaves, the solvent fractions from crude methanol extract were made with n-hexane, ethyl acetate, and butanol, and their antioxidant and antimicrobial activities were determined. The antioxidant activities were examined by 1,1-diphenyl-1-picrylhydrazyl (DPPH) method and ferric ion reducing antioxidant power (FRAP) method, and the antimicrobial activities against Staphylococcus aureus were tested by paper disc agar diffusion method. Total phenolic contents and total flavonoid contents of the solvent fractions were also determined. The ethyl acetate fraction with the highest total phenolic contents among all fractions showed the strong antioxidant activities by DPPH method and FRAP method, and antimicrobial activities against S. aureus at all test concentrations. Caffeic acid, ferulic acid, quercetin, and kaempferol were analyzed by HPLC in the ethyl acetate fraction from black bamboo leaves by the comparison with the standard chemicals. It is supposed that the ethyl acetate fraction from black bamboo leaves could be used as natural preservatives in the food industry.

Screening of Antioxidants and Antimicrobial activity in native plants (국내 자생 식물의 항산화 및 항미생물 활성 탐색)

  • This study was carried out to screen antioxidative by in vitro bioassay method from 89 Korean natural sources extracted by 100% MeOH. Antioxidant activity test was used the DPPH method. MeOH extracts from Castanea crenata and Ulmus davidiana var. japonica showed high antioxidant activity by $5.8{\mu}g\;(RC_{50})$ and $12.2{\mu}g\;(RC_{50})$, respectively, among 13 plants exhibiting the activity. The extracts from Platycarya strobilacea, Lindera erythrocarpa, Chrysanthemum boreale, Rumex crispus and Viburnum awabuki also showed over 90% antimicrobial activity, according to in vivo bioassay method.

Biological Activity and Cosmetic Preservative Effects of Rosa multiflora Ethanol Extracts (찔레꽃 에탄올추출물의 생리활성과 화장품 방부효과)

  • Background: The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity and cosmetic preservative efficacy have not yet been studied. Thus, this species is yet to be defined as a functional cosmetic material. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. Methods and Results: The antioxidant activity was assessed through free radical scavenging assays with 2,2-diphenyl-1-picrylhydrazyl (DPPH). Additionally, the contents of total phenols and flavonoids were analyzed. The phenolic compounds were detected using HPLC. The antimicrobial activity against Staphylococcus aureus, Escherichia coli, and Candida albicans was assessed using the disc diffusion assay. The preservative effect (challenge test) on a formulation of soothing gel was performed for 28days. The DPPH radical scavenging ability, denoted by the $SC_{50}$ (half maximal inhibitory concentration for DPPH radical scavenging) value was found to be $131.63{\mu}g/m{\ell}$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In additon, astragalin and gallic acid were identified in the extract. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 5 - 0.5%, and no significant activity was noted against C. albicans. The ethanol extracts (5% and 10%) met the preservation standards of the Cosmetics, Toiletry, and Fragrance Association (CTFA). Conclusions: Thus the ethanol extract of R. multiflora can be used in cosmetics as a natural preservative and antioxidant.

4-Hydroxy-2'-Nitrodiphenyl Ether Analogues as Novel Tyrosinase Inhibitors

  • Tyrosinase ubiquitously existing from microorganisms to animals and plants is known to be the most critical and rate limiting enzyme during melanin biosynthesis. In order to develop new tyrosinase inhibitor we have synthesized 14 diphenyl ether compounds possessing hydroxyl, bromo, and nitro groups in the structure. Among the compounds prepared, 18 and 19 have shown much stronger inhibition of tyrosinase monophenolase function than arbutin used as a positive control. Both compounds 18 and 19 possess para-hydroxyphenyl moiety in their structure, which might reinforce the importance of p-hydroxyphenyl group in the tyrosinase inhibitory process. In the DPPH radical scavenging activity test, none of the compounds even 18 and 19 showed significant antioxidant activity. The results suggest that elaborate adjustment of diphenyl ether analogues with proper substituents have potential to be developed as new skin whitening agents working on the tyrosinase function.

Antioxidative Activity and Irritation Response of Lespedeza bicolor

  • In the present study, we evaluated the free radical scavenging and xanthine oxidase inhibitory activities exhibited by extracts obtained from the dried stems (and leaves) of Lespedeza bicolor We also assessed its potential irritation activities with regard to cosmetic use. When the DPPH radical scavenging activities of L. bicolor were assessed at six different concentrations (0, 50, 100, 250, 500 and 1000 ${\mu}g/ml$), the concentration of L. bicolor required to inhibit DPPH radical formation by $50\%$ was found to be $164.90{\mu}g/ml$. The effects of L. bicolor on the inhibition of xanthine oxidase were determined at seven different concentrations. The $50\%$ effective concentration was found to be $282.75{\mu}g/ml$. In the skin irritation test, all animals survived for the duration of the study, and all exhibited normal gains in body weight. The control sites exhibited no response to the control procedures. No edema, erythema, or eschar formation was observed in any of the tested rabbits. In the ocular irritation study, all of the rabbit eyes remained normal. In summary, L. bicolor extracts were considered to be non-irritating to the skin and eye.

Diversity and Biological Activities of Endophytic Fungi of $Emblica$ $officinalis$, an Ethnomedicinal Plant of India

  • In the present study, an attempt to evaluate the antimicrobial and antioxidant activity of fungal endophytes inhabiting $Emblica$ $officinalis$ has been made keeping in view the medicinal importance of the selected host plant in Indian traditional practices. A total of four endophytic fungi belonging to Phylum Ascomycetes were isolated from different parts of the plant which were characterized morphologically and by using rDNA-internal transcribed spacer. The most frequently isolated endophyte was $Phomopsis$ sp. The antioxidant activity by 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and reducing power assay, and total phenol were evaluated using ethanolic extract of endophytic fungi. DPPH activities in all the ethanolic extract increased with the increase in concentrations. Endophytes, $Phomopsis$ sp. and $Xylaria$ sp. showed highest antioxidant activity and also had the higher levels of phenolics. Antimicrobial activity of fungal extract were tested against four bacteria namely, $Escherichia$ $coli$ MTCC730, $Enteroccocus$ $faecalis$ MTCC2729, $Salmonella$ $enterica$ ser. $paratyphi$ MTCC735 and $Streptococcus$ $pyogenes$ MTCC1925, and the fungus $Candida$ $albicans$ MTCC183. In general, the fungal extracts inhibited the growth of test organisms except $E.$ $coli$.

Development of functional cosmetic powder using silkgland powder of silkworm (누에실샘 미세분말을 이용한 기능성 색조용 파우더 개발)

  • In this study, silkgland powder of silkworm were investigated to see possibility for cosmetic powder materials. To test possibility as a baby powder cosmetics, total content rate of amino acids, DPPH free radical scavenging assay, MTT assay, and clinical trial were done. According to the result of the analysis of the amino acids of silkgland powder, serin (26.77%) content was the highest and asparatic acid (15.47%), and glycine (9.62%) were followed. DPPH free radical scavenging activity of silkgland powder was lower than vitamin C by 82.3% and 97%, respectively, which is relatively good. And the moisture and elasticity effect were increased in silkgland powder compared to control cosmetics by 210% and 185%, relatively. Thus, these results suggest that silkgland powder of silkworm may have beneficial properties as a material for cosmeceuticals.

A Study on the Development of Cooked Rice according to the Different Coating Ratio of Yacon(Polymia sonchifolia) Root and Its Antioxidant and Sensory Properties (야콘 뿌리 추출액 코팅 쌀 제조 및 항산화력과 관능적 품질 연구)

  • The objective of this study was to develop functional rice coated with yacon roots extracts and to evaluate the antioxidant properties and physiological activity of rice coated with yacon extract. The washed rice was coated by spraying yacon extract at 10, 20, 30, and $40^{\circ}Brix$ and dried at room temperature. The moisture contents, color changes, antioxidant properties as total polyphenolics and DPPH radical scavenging activity and instrumental texture property and sensory evaluation change in the yacon root extracts coated rice and uncoated cooked rice were observed during 3 days of storage. Increasing the coating concentration was associated with lower color values(L value) and moisture contents. Total phenolics and DPPH radical scavenging properties generally increased in the cooked rice coated with yacon root extract more than the control (non-coated rice). However, D-20($20^{\circ}Brix$ of the yacon root concentration) resulted in better textural properties and sensory overall acceptability in cooked rice on 0 and 3 storage days. It was concluded that the optimum coating ration of rice and added functional yacon rice were $20^{\circ}Brix$ and 7% respectively, in proportion to the total weight of raw rice.

Effects of Shiitake Mushroom on Anti-platelet Aggregation and Anti-thrombotic (표고버섯 추출물의 항혈소판 응집 및 항혈전 효과)

  • In in vitro study, the anti-platelet aggregation effect of Shiitake mushroom extract was examined by measuring the collagen induced platelet aggregation and the DPPH radical scavenging. In in vitro study, anti-thrombotic effect of Shiitake mushroom extract was examined using the carotid artery thrombosis rat model. Carotid artery thrombosis rat model was made by 35% $FeCl_3$ treatment. After that, we investigate thrombus weight and blood flow. In platelet aggregation test, the extract significantly inhibited platelet aggregation in a concentration dependent manner(p<.001). Also, extract increased DPPH radical scavenging activity in a concentration dependent manner. Extract significantly inhibited thrombus weight to compare with control group. And blood passage time were shorter in the Shiitake mushroom extract supplemented groups than in control group. These results provide experimental evidence that Shiitake mushroom can be used to prevent platelet aggregation and thrombosis, then could apply the clinical diseases such as cardiovascular disease, and so on.
