This study was performed to investigate the antioxidant activity of extracts of black Panax ginseng (BGE) and its crude saponin (BGECS). The antioxidant activities of BGE and BGECS were evaluated for free radical scavenging activity against stable free radical (1,1-diphenyl-2-picrylhydrazyl) DPPH, nitrite, hydrogen peroxide and superoxide. In addition, the antioxidant activity of BGE and BGECS against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. As a result, BGE and BGECS were found to have a strong inhibitory activity with >90% against the DPPH radical at $1000{\mu}g/m{\ell}$ concentrations. Also, BGE and BGECS exhibited strong inhibitory activity with >80% against hydrogen peroxide at lower concentration ($125{\mu}g/m{\ell}$). Moreover, specific TOSC values (405 and 473 TOSC/mM) of BGE and BGECS against peroxynitrites were higher than GSH (347 TOSC/mM) used a positive control. These results suggest that BGE and BGECS could be useful to develop functional foods against disease related oxidative stress.
Methanol crude extract of the sea grass Zostera marina L. and organic solvent fractions (n-hexane, chloroform, ethyl acetate, n-butanol, and water) were screened for antioxidant activity (total phenolic contents, DPPH scavenging activity, and reducing power) and antimicrobial activity against three human skin pathogens, two bacteria and a yeast; Staphylococcus aureus, Staphylococcus epidermidis, and Candida albicans. Total phenolic contents and 2, 2- diphenyl-1-picrylhydrazyl (DPPH) scavenging activity were highest in the ethyl acetate fraction with 968.50 $\mu$g gallic acid equivalent per milligram of extract, and ca. 95% scavenging activity on the DPPH radicals at 10 mg $ml^{-1}$. In antimicrobial activity tests, MICs (Minimum Inhibitory Concentration) of each Zostera marina extract partitioned ranged from 1mg to 8 mg $ml^{-1}$ (extract/ 10% DMSO) against all three human skin pathogens. The MICs of the ethyl acetate and n-butanol fractions were the same with 1 mg $ml^{-1}$ against S. aureus and C. albicans. The ethyl acetate fraction of Z. marina does protect against free radicals and may be used to inhibit the growth of human skin pathogens.

Since reactive oxygen radicals play an important role in carcinogenesis and other human diseases including neurodegenerative states, antioxidants present in natural products have received considerable attention for alleviation of these disease states. Therefore, in order io identify antioxidants in plant extracts, fifty-seven methanolic extracts derided from indigenous Korean plants were primarily assessed for potential to scavenge stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals. As a result, nine plant extracts were found to exhibit the DPPH free radical scavenging activity in the criteria of $IC_{50}<40\;{\mu}g/ml$. In particular, the extracts of Melioma oldhami $(IC_{50}=0.1\;{\mu}g/ml)$, Myrica rubra $(IC_{50}=16.2\;{\mu}g/ml)$, Sympolocos paniculata $(IC_{50}=23.0\;{\mu}g/ml)$, Carpinus laxiflora $(IC_{50}=25.1\;{\mu}g/ml)$, and Cleyera japonica $(IC_{50}=26.2\;{\mu}g/ml)$ showed a potent radical scavenging activity. Further study for the identification of active compounds from these lead extracts might be warranted.
Objectives: The purpose of this study was to compare the antioxidant and anti-inflammatory effects of Alum (AL) and Burnt Alum (BAL), which are commonly used as external ointments. Methods: Extracts of AL and BAL were classified into three groups: 20, 50, and $100mg/{\mu}{\ell}$. The cytotoxicity was measured by using MTT assays in human keratinocyte cell line (HaCaT). The anti-oxidant effect was measured by using the DPPH (1, 1-diphenyl-2-picryl-hydrazyl-hydrate) radical scavenger. The anti-inflammatory effect was measured by using the inhibitory efficacy for the amount of nitric-oxide (NO) produced in mouse macrophage cell line (RAW 264.7). Results: BAL showed a higher level of cytotoxicity than AL. The AL groups showed a concentration-dependent scavenging effect on DPPH radicals, but no significant relevance was found. The BAL groups showed a concentration-dependent scavenging effect on DPPH radicals. The scavenging effects of the BAL groups were almost insignificant, but the values for the 20, 50, and $100{\mu}g/m{\ell}$ trials were different. The BAL groups showed significant concentration-dependent inhibitory effects on NO production, but the AL groups did not. Conclusions: AL showed an anti-oxidant effect more efficiently than BAL did, which demonstrated a superior anti-inflammatory effect. Therefore, for external usage, AL must be distinguished from BAL.
This study evaluated the antioxidative activity and quality characteristics of Sulgidduk (traditional rice cakes) prepared with ramie leaf powder, in order to measure the antioxidative activity of ramie leaves, which are a potentially useful antioxidant food. Antioxidant activities were measured by total phenolic contents, the scavenging activities of DPPH radicals and ABTS+ radicals, and the reducing power. The total phenolic content of freeze dried ramie leaves was estimated at $58.70{\pm}11.42mg$ GAE/g. The $IC_{50}$ values for DPPH radicals and the ABTS+ radical scavenging activity of the ramie leaves were $23.76{\pm}1.41ug/mL$ and $174.82{\pm}6.4ug/mL$, respectively. The leaves also showed a high reducing power of $1.1057{\pm}0.03$ (O.D). To increase the utilizability of ramie leaf, we made Sulgidduk with added ramie leaf powder. Re-grading the quality characteristics of the altered Sulgidduk, the 3% and 5% addition levels most satisfied texture profile analysis and sensory evaluation. The antioxidative effect of the Sulgidduk significantly increased as the addition level increased, compared to the original Sulgidduk. In term of DPPH radical scavenging activity, the 5% or more addition level Sulgidduk showed an antioxdative effect increase of more than 50%. At the 3% or higher addition level, there was higher antioxidant activity than in the positive control group, which included vitamin C and BHT additions. We thus concluded that Sulgidduk with added ramie leaf power can be manufactured as a food that can satisfy sensory preferences as well as boosting antioxidant activity, at both the 3% and 5% addition level.

Free radicals are known to play important role in pathophysiology of hepatic disorders and antioxidants are employed along with other chemotherapeutic agents in treatment of such diseases. In search of natural antioxidant, successive extracts of Hypericum (H.) hookerianum (Family: Hypericaceae) were evaluated by in vitro and in vivo methods. Extracts of aerial parts of H. hookerianum were subjected for 1,1-diphenyl 2-picryl hydrazyl radical scavenging activity (DPPH assay), nitric oxide radicals scavenging assay and thiobarbituric acid reactive substances (TBARS) assay. Methanolic extract was found to be more active than other extracts in DPPH and in vitro TBARS assay with $IC_{50}$ at 5.82 ${\pm}$ 1.33 ${\mu}g/ml$ and 49.78 ${\pm}$ 3.79 ${\mu}g/ml$ respectively. While petroleum ether extract showed more potentials in scavenging the nitric oxide radicals with $IC_{50}$ 220.97 ${\pm}$ 2.69 ${\mu}g/ml$. The administration of $CCl_{4}$ to the control animals caused decrease in the level of catalase and superoxide dismutase, together with significant increase in the level of TBARS in liver and kidney. Reversal of these changes towards normal group was observed by administration of H. hookerianum methanolic extract at 50 and 100 mg/kg body weight, while other extracts were found to be less active.

Degenerative diseases are commonly associated with excess free radicals. Acer okamotoanum, a plant endemic to Korea, is reported to have anti-oxidant, anti-cancer, and anti-viral activities. We previously isolated flavonoids from the ethyl acetate fraction of A. okamotoanum such as quercitrin (QU), isoquercitrin (IQ), and afzelin (AF). In the present study, the in vitro antioxidant activity of flavonoids such as QU, IQ, and AF isolated from the ethyl acetate fraction of A. okamotoanum were investigated by measuring the free radical scavenging activity including 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), and superoxide anion ($O_2{^-}$). The flavonoids (QU, IQ, and AF) concentration-dependently showed a DPPH radical scavenging activity. In particular, QU and IQ showed a higher DPPH radical scavenging activity than that of AF. In addition, the flavonoids (QU, IQ, and AF) at $10{\mu}g/mL$ showed over an 80% scavenging effect against $^{\cdot}OH$ radical production. Furthermore, the $O_2{^-}$ radical scavenging activity of the flavonoids, QU, IQ, and AF increased in a dose-dependent manner. Particularly, IQ exerted the strongest scavenging activities against $^{\cdot}OH$ and $O_2{^-}$ radicals among the other flavonoids. These results indicate that the flavonoids from A. okamotoanum, in particular IQ, would have a protective activity against oxidative stress induced by free radicals, and potentially be considered as a natural antioxidant agent.

Investigation of antioxidative constituents from the ethanol extract of Quercus gilva branches led to the identification of four compounds; catechin (1), epi-catechin (2), tyrosol (3) and tiliroside (4). The antioxidative activity was examined using DPPH radical and superoxide anion radical scavenging method. The isolated compounds 1, 2, 3 and 4 exhibited 94.2 %, 93.4 %, 33.6 % and 11.2 % scavenging activities respectively against DPPH radicals at the concentration of $100{\mu}L$/mL. As well, the compounds $1{\sim}4$ showed respectively 60.2 %, 35.1 %, 20.6 %, 4.5 % inhibition activities against superoxide anion radicals at $200{\mu}L$/mL. Interestingly, the compounds $1{\sim}4$ were isolated for the first time from Quercus gilva Blume.
Physiological functionality of browned oak mushroom was evaluated with focusing its electron-donating ability to DPPH radicals, scavenging ability to superoxide radicals and hydroxyl radicals, and inhibitory activity on lipid peroxidation. The results showed that overall antioxidative activities of browned oak mushroom were superior to those of raw oak mushroom with marketable quality, implying possible involvement of resultant browning reaction products in an increment of antioxidativity. The increased radical-scavenging ability was suggested to mainly be exerted by direct quenching of both superoxide and hydroxyl radicals, not by inhibition of xanthine oxidase activity and $Fe^{2+}$ chelation, respectively. Collectively, these results indicate a possible use of unmarketable browned mushroom as a material for manufacturing various processed functional foods.

Reactive oxygen species (ROS) are reactive and potentially harmful to cells, causing oxidation of lipids, proteins, and DNA. In humans, the deleterious effects of ROS have been linked with aging, carcinogenesis, and atherosclerosis. In order to investigate an antioxidant activity of wood vinegar by bioconversion, we preferentially analyzed the total acidity, acetic acid, pH, and contents of total polyphenols and flavonoids, respectively. Also, we evaluated the scavenging abilities on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, superoxide anion radicals, hydrogen peroxide radicals, and nitric oxide radicals. The total acidity and amount of acetic acid of wood vinegar after bioconversion were lower than those of wood vinegar before bioconversion, but the pH was higher than that of wood vinegar before bioconversion. The contents of total polyphenols and flavonoids of wood vinegar after bioconversion were 11.17 mg/$m{\ell}$ and 0.42 mg/$m{\ell}$, respectively. The $SC_{50}$ values were in order of superoxide anion radical scavenging activity < DPPH radical scavenging activity < hydrogen peroxide radical scavenging activity < nitric oxide radical scavenging activity. Therefore, these results suggest that wood vinegar by bioconversion can be useful as primary antioxidants for medicines and cosmetics.
