Objective : This study was designed to investigate effect of immature fruits from Citrus unshiu Markovich to mature them on antioxidative and antiaging-related enzyme activities in vitro. Methods : Fruits from Open field-cultivated C. unshiu Markovich (ripe and unripe fruits of Citri) made a purchase in Jeju island. We measured total polyphenol and flavonoid contents in mature C. unshiu Markovich (MC) and immature C. unshiu Markovich (IMC) 70% ethanol extract. DPPH free radical, ABTS radical, and hydroxyl radical scavenging activities in both extracts were determined. Also, reducing power, SOD-like activities, elastase inhibition, and collagenase inhibition activities of these extract were assayed at 5, 1, 0.5, and 0.1 mg/㎖. Results : The total polyphenol contents in the ethanol extract of IMC were higher than those of MC (26.99 ± 0.89 mg/TAEg). And total flavonoid contents in them were very similar to both extract (MC; 7.25 ± 0.4 mg/RUEg and IMC; 7.44 ± 0.18 mg/RUEg). DPPH free radical scavenging activities, ABTS radical scavenging capacity, reducing power, and SOD-like activities of IMC-treated group showed significant increase compared to those of MC group in all treated concentrations. Particularly, DPPH free radical scavenging and elastase inhibition activities in IMC-treated group at 5 mg/㎖ showed similar with positive control group. Besides, IMC extract at 5 mg/㎖ concentration surpassed positive control (BHA or Vit. C) in SOD-like activities and ABTS radical scavenging capacity. Conclusion : These results suggest that IMC ethanol extract may be used as a natural antioxidant and a antiaging material for development of functional foods and cosmeceuticals.
This study was carried out to investigate the structure of antioxidative constituents and the free radical scavenging effect of the main ingredients from Galla Rhois(Rhus javanica Linne). Antioxidative activities of n-hexane, EtOAc and BuOH extracts of Galla Rhois were similar or even higher than that of natural (tocopherol) or synthetic antioxidant (BHA). It is suggested that major fractions for the antioxidative activity of Galla Rhois were the n-hexane, EtOAc and BuOH extract compartments. In the subsequently experiment, one active compound from n-hexane extract, three active compounds from EtOAc extract and one active compound from BuOH extract were isolated. Their chemical structures were identified as syringic acid, protocatechuic acid, gallic acid methylester, gallic acid and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ on the basis of the speculation of spectral data and chemical reaction. Among the compounds, protocatechuic acid, gallic acid methylester and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ showed most potent radical scavenging effect using DPPH method.
In order to examine oxidative stress of reactive oxygen species and the antioxidant effect of Citri Reticulatae Pericarpium (CRP) extract, human skin melanoma cells were treated with various concentrations of hydrogen peroxide ($H_2O_2$). Antioxidant effect of CRP extract on $H_2O_2$-induced cytotoxicity, cell viability, DPPH-radical scavenging activity and superoxide dismutase (SOD)-like activity. In this study, $H_2O_2$ decreased cell viability of cultured human skin melanoma cells in dose- and time-dependent manners, and then, midcytotoxicity value (MCV) was determined at $60\;{\mu}M$ after human skin melanoma cells were cultured for 5 hours in the media containing $20{\sim}60\;{\mu}M$ of $H_2O_2$, respectively. The $H_2O_2$ was on cultured human skin melanoma cells because MCV of $H_2O_2$ was lower than $100\;{\mu}M$. In the antioxidant effect of CRP extract, CRP extract increased cell viability DPPH-radical scavenging activity and SOD-like activity. From these results, it is suggested that $H_2O_2$ was very toxic on cultured human skin melanoma cells. And also, CRP extract has the antioxidant effect on $H_2O_2$-induced cytotoxicity.

This study was conducted to evaluate the effects of drying and extraction methods on antioxidant activity and total phenol content of Gnaphalium affine D. DON (GA). Hot-air, shade-drying, and freeze-drying were used for drying, after which magnetic stirring and ultrasonification were applied. Extracting solvents were water, 80% ethanol, and 80% methanol. Total phenol content was highest in 80% ethanol extract of freeze-dried and stirred GA. Total flavonoid content was highest in 80% methanol extract of freeze-dried and stirred GA. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity was higher in 80% methanol and 80% ethanol extracts than in water extract. 2,2-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity was highest in 80% ethanol extract of shade-dried and ultrasonicated GA. Reducing power was generally higher in 80% methanol extract than in 80% ethanol and water extracts of GA. Total phenol and total flavonoid contents were highly correlated with DPPH radical scavenging activity and reducing power, respectively. This result implies that the antioxidant activity of GA can be attributed to phenol compounds such as flavonoids. Conclusively, phenol compounds such as flavonoids are responsible for the antioxidant activity of GA, and there was no significant effect of drying and stirring conditions on antioxidant activity of 80% ethanol. Meanwhile, DPPH radical scavenging activity of water extract and reducing power of 80% methanol extract were higher in hot-air and shade-dried GAs than in freeze-dried GA.
A peroxynitrite is formed when superoxide and nitric oxide exist at near eqimolar ratio in biological systems. Although not a free radical by chemical nature, peroxynitrite is a powerful oxidant having a wide array of tissue damaging effects ranging from lipid oxidation and inactivation of enzymes and ion channels through protein oxidation and nitration to inhibition of mitochondrial respiration. During our search for new antioxidizing components from natural resources, twenty salt marsh plants were screened for their ONOO and DPPH radical scavenging activities. Among them, methanol extract of Rosa rugosa, lxeris tamagawaensis, Erigeron annus, Tetragonia tetragonoides, Imperata cylindrica, and Suaeda japonica inhibited more than 85% of peroxynitrite produced by 3-morpholinsydnonimine (SIN-1) at a concentration of 5 $\mu\textrm{g}$/$m\ell$. In addition, Rosa rugosa, Artemisia capillaris, Erigeron annus and Ixeris tamagawaensis showed significant scavenging effect against DPPH (1,1-diphenyl-2-picrylhydrazyl radical).
The antioxidant activities and total phenolics of four Rosaceae species Pourthiaea villosa (Thunb.) Decne, Sorbus commixta Hedlund, Sorbaria sorbifolia var. stellipila Maxim and Pyrus pyrifolia (Burm.f.) Nakai were determined. Phenolic content (polyphenol and flavonoid), radical scavenging activities [2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis (3-ethylnezthiazoline-6-sulfoic acid) (ABTS)] and ferrous ion chelating effect were evaluated. Total polyphenol and flavonoid contents were highest in Pourthiaea villosa (Thunb.) Decne and lowest in Pyrus pyrifolia (Burm.f.) Nakai. Phenolic contents of Pourthiaea villosa (Thunb.) Decne was $331.45{\pm}7.78$ and $90.4{\pm}3.5mg{\cdot}g^{-1}$. DPPH and ABTS radical scavenging activities were found to be lowest in Sorbus commixta Hedlund whereas Sorbaria sorbifolia var. stellipila Maxim and Pourthiaea villosa (Thunb.) Decne showed relatively good DPPH and ABTS radical scavenging activities. Ferrous ion chelating effect was highest in Pyrus pyrifolia (Burm.f.) Nakai ($1.05{\pm}0.04mg{\cdot}ml^{-1}$) and lowest in Sorbus commixta Hundlund ($4.22{\pm}0.71mg{\cdot}ml^{-1}$).
Xanthium strumarium L. is an annual plant belongs to the family Asteraceae which is is called a 'Cocklebur' that is used for medicinal purposes. Convergent phyto-activity of various extracts of Xanthium strumarium L. (Asteraceae) was examined. We estimated antioxidant activity from ground part and fruit extract of X. strumarium using 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH) and ABTS assay. The extract of X. strumarium was separated each fraction that of ethanol, petroleum ether, and ethyl acetate. It showed potent radical scavenging effect against the DPPH radical and ABTS. The study revealed that X. strumarium could be used as a potential source of natural antioxidant.
The MeOH extract of Sorbus commixta (Rosaceae) exhibited strong DPPH radical scavenging activity, and through activity-guided fractionation two antioxidant compounds were isolated and identified as $catechin-7-O-{\beta}-D-xylopyranoside$ (1) and $catechin-7-O-\;{\beta}-D-apiofuranoside$ (2) by physicochemical and spectrometric methods. To evaluate the antioxidant effect of these compounds, some in vitro tests, such as the DPPH radical scavenging activity test, the superoxide radical scavenging activity test and the lipid peroxidation inhibitory activity test, were performed. Compounds 1 and 2 showed stronger activities than both a-tocopherol and butylated hydroxy anisole (BHA) in each assay.
Pylaiella littoralis was collected in the Chuuk lagoon of the Federated States of Micronesia (FSM). The FSM has a variety of coral reef ecosystems, which provide essential materials, such as minerals, vitamins, essential amino acids, for marine organisms. In this study, the antioxidant activities of ethanol and enzymatic extracts of P. littoralis were evaluated by measuring their scavenging activities on DPPH free radical, Alkyl radical, hydroxyl radical and cell viability. The enzymatic extracts were hydrolyzed to prepare water soluble extracts by using five carbohydrate degrading enzymes (AMG, Celluclast, Termamyl, Ultraflo, and Viscozyme) and five proteases (Alcalase, Flavourzyme, Kojizyme, Neutrase, and Protamex). As a result, the enzymatic extracts prepared by Flavourzyme, Ultraflo, and Kojizyme exhibited the greatest effects in DPPH free radical, alkyl radical scavenging activity and cell viability. Also, these enzymatic extracts had a higher antioxidant effect then commercial antioxidants in DPPH free radical and Alkyl radical scavenging activity. This study suggests that P. littoralis might be a useful source of natural antioxidants for the development of dietary supplements.
Objective: The purpose of this study was to evaluate the antioxidative effects of the extract of Scolopendra subspinipes which has been used mainly for detoxication in the oriental medicine and reported to have sedative action, antiinflammatory effect, antihypertensive property and immunity enhancing activity. Method: Inhibitory activities on oxygen radical generating enzymes (aldehyde oxidase and xanthine oxidase) and increasing activities on oxygen radical scavenging enzymes (superoxide dismutase, glutathione peroxidase, glutathione-S-transferase) were investigated. Furthermore, the content of glutathione in the mouse brain, DPPH radical scavenging activity and also anti-lipid peroxidative effects in vivo and in vitro were estimated. Results: The extract showed weak inhibitory effects on the activities of aldehyde oxidase and xanthine oxidase which are oxygen radical generating enzymes. The extract inhibited lipid peroxidation with 26.1% against control group at 500 mg/kg in vivo and with 11.2% against control group at 10 mg/kg in vitro in a dose-dependent manner, which means this drug may protect radical-induced cell damages. The extract showed dose-dependently the scavenging effect on DPPH radical with 24.8% activity at 10 mg/ml in vitro. The extract enhanced the activities of superoxide dismutase, glutathione peroxidase and glutathione-S-transferase, which are oxygen radical scavenging enzymes, with 28.9%, 22.3% and 23.1%, respectively at 500mg/kg in vivo. Finally, this extract strongly increased the glutathione content in the mouse barin. Conclusion: Above results indicated that Scolopendra subspinipes can be useful for the protection or treatment of some diseases caused by reactive oxygen species.
