Oxidative DNA 손상에 대한 Quercetin 및 그 배당체들의 유전독성억제효과와 작용기전 (Antigenotoxicity and Action Mechanism of Quercetin and its Glycosides against Oxidative DNA Damage)

  • Quercetin and its glycosides showed a strong free radical scavenging effect to DPPH radical generation. However, there were not big differences between quercetin aglycone and glycosides under experimental condition of this study. On the other hand, quercetin had pro-oxidant effect in bleomycin-dependent DNA assay. Quercetin aglycone and its glycosides, quercitrin inhibited $H_2$$O_2$- induced DNA damage in CHL cells. They also have an anticlastogenicity toward DNA breakage agent by radical generation like bleomycin. These results indicate that quercetin aglycone and its glycosides are capable of protecting the free radical generation induced by reactive oxygen species like $H_2$$O_2$. The mechanism of inhibition in hydrogen peroxide-induced genotoxicity may be due to their free radical scavenging properties. Therefore, quercetin aglycone and its glycosides may be useful chemopreventive agents by protecting of free radical generation which are involved in carcinogenesis and aging. However, quercetin and its glycosides must also carefully examined for pro-oxidant properties before being proposed for use in vivo.

Antioxidant Effect of Salvia miltiorrhiza

  • A strong antioxidant activity, which was measured by the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl(DPPH) radical, was detected in the methanol extract of Salvia miltiorrhiza Bunge (Labiatae). By activity-directed fractionation, compounds 1 and 2 were isolated as antioxidant principles of S. miltiorrhiza. Compounds 1 and 2 were identified as dimethyl lithospermate and 3-(3, 4-dihydroxyphenyl)lactamide, respectively, on the basis of spectral data. The radical scavenging effect of compounds 1 and 2 on DPPH radical exceeded that of L-ascorbic acid which is a well known antioxidant. These two compounds also showed prominent inhibitory activity against free radical generation in dichlorofluorescein (DCF) method and cytoprotective effect against t-BHP in cultured liver cell.

Screening of Antioxidant Activity of Domestic Trees

  • This study was carried out to investigate the antioxidant activities of domestic trees grown in Korea. Based on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity method, the methanolic extracts of 23 species were screened in order to search for natural antioxidants. Among these species, Acer ginnala, Cotinus coggygria, Acanthopanax koreanum, Thea sinensis and Pinus densiflora showed stronger antioxidative activity comparing with reference compound, ascorbic acid.

고추냉이 추출액의 항산화 효과 (Anti-oxidant Effect of Wasabia Japonica Extracts)

  • This study was carried out to investigate the anti-oxidant effect of Wasabia japonica(WJ). The anti-oxidant activities of various extracts from WJ were investigated. The water and ethanol extracts of WJ leaf were found to cause significant free radical scavenging effects on DPPH and nitric oxide(NO). Sprague-Dawley male rats were randomly assigned to one normal diet and three high cholesterol diet groups which contained 1%(w/w) cholesterol. After 4weeks cholesterol diet, the groups of high cholesterol diet were classified to control(normal diet only), WJR5(normal diet and 5% WJ root) and WJL5(normal diet and 5% WJ leaf). Liver xanthine oxidase(XO) activity in the rats of high cholesterol diet was decreased up to the levels of normal diet group according to WJ diet. The results indicated that WJ possesses antioxidant effects through free radical scavenging effects on DPPH, NO and the decreasing of XO activity. In view of these results, WJ is expected to be an effective material for the anti-oxidant.

Free Radica1 Scavenging and Hepatoprotective Constituents from the Leaves of Juglans sinensis

  • In the course of searching for hepatoprotective agents from natural products, six compounds were isolated from the MeOH extract of the leaves of Juglans sinensis, as guid ed by their DPPH free radical scavenging activity. The structures were determined as juglanoside B (1), quercetin 3-O-${\alpha}$-L-arabinofuranoside (avicularin, 2), quercetin 3-O-${\alpha}$-L-arabinopyranoside (guaijaverin,3), quercetin 3-O-${\alpha}$-L-rhamnopyranoside (quercitrin,4), (+)-catechin (5) and quercetin 3-O-${\beta}$- D-galactopyranoside (hyperin,6). Compounds 2-6 showed significant DPPH free radical scavenging effects. An evaluation for the hepatoprotective activity of the isolated compounds on drug-induced cytotoxicity was conducted, and compounds 1, 2, and 5 showed protective effects against nitrofurantoin-induced cytotoxicity, and compound 5 also exhibited a moderate protective effect on amiodarone-induced cytotoxicity in Hep G2 cells.

In vitro Free Radical Scavenging and Hepatoprotective Compound from Sanguisorbae Radix

  • In the course of searching for hepatoprotective agents from natural products, four compounds were isolated from the MeOH extract of Sanguisorbae Radix, as guided by their DPPH free radical scavenging activity. The structures were determined as 4,5-dimethoxy-3-hydroxybenzoic acid methyl ester (1), (+)-gallocatechin (2), methyl $6-O-galloyl-{\beta}-D-glucopyranoside$ (3), and pomolic acid $3-O-[{\alpha}-L-arabinopyranoside]-28-O-[{\beta}-D-glucopyranosyl]$ ester (ziyu-glycoside I) (4). Compounds 2 and 3 showed significant DPPH free radical scavenging effects, exhibiting $IC_{50}$ values of 11.4 and $13.0\;{\mu}M$, respectively. L-Ascorbic acid was used as a positive control and exhibited the $IC_{50}$ value of $50.3\;{\mu}M$. In evaluation of the hepatoprotective activity of the isolated compounds on drug-induced cytotoxicity, compound 2 showed the significant hepatoprotective effect with the $EC_{50}$ value of $91.84\;{\pm}\;11.0\;{\mu}M$ on tacrine-induced cytotoxicity in Hep G2 cells, while silybin, a positive control, exhibited $EC_{50}$ value of $122.4\;{\pm}\;12.5\;{\mu}M$.

Antioxidative Activity of the Extract from the Inner Shell of Chestnut

  • The ethanolic extract of chestnut (Castanea crenata S. et Z., Fagaceae) inner shell (CISE) and one of its components, ellagic acid (EA), were evaluated for their protective effects against 1, 1-diphenyl-2-picryl hydrazine (DPPH) free radical generation and hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line. CISE and EA were shown to possess the free radical scavenging effect against DPPH radical generation, significantly. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from Chinese hamster lung (CHL) cell, assessed by single cell gel electrophoresis assay and 8-hydroxy -2'-deoxy guanosine (8-OH-2'dG) assay. Furthermore, topical application of CISE [$12.5\%$(w/w) cream] and ellagic acid [$1.0\%$(w/w) cream] for 14 days potently inhibited malondialdehyde (MDA) formation of mouse dorsal skin (a marker of lipid peroxidation) induced by ultraviolet B exposure. Therefore, CISE and its component, ellagic acid, may be the useful natural antioxidants by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage when topically applied.

Effect of Kaempferol on the Cytotoxicity Induced Oxygen Free Radicals in Skin Fibroblast Derived from Human In Vitro

  • In order to evaluate on the effect of kaempferol on the cytotoxicity of oxygen tree radicals, XTT assay was performed to determine the cell viability after skin fibroblasts derived from human (Detroit 51) that were treated with various concentrations of hydrogen peroxide $(H_2O_2)$. And also, the effect of kaempferol on the cytotoxicity induced by H202 that was examined by cell viability, lactate dehydrogenase (LDH) activity and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in these cultures. $H_2O_2$ decreased cell viability in dose-dependent manner in these cultures and the $XTT_{90}\;and\;XTT_{50}$ values were determined at concentration of $35{\mu}M\;and\;90{\mu}M$ of $H_2O_2$ after skin fibroblasts derived from human were treated with $15{\sim}90{\mu}M$ of $H_2O_2$ for 6 hours, respectively. $H_2O_2$ was highly toxic on cultured skin fibroblasts derived from human by toxic criteria of Brenfreund and Puerner (1984). In the protective effect of kaempferol on $H_2O_2$-induced cytotoxicity, kaempferol increased DPPH radical scavenging activity and significantly decreased LDH activity. From these results, it is suggested that oxygen tree radical, $H_2O_2$, was highly toxic on cultured skin fibroblasts derived from human, and also kaempferol of flavonoid showed the protection on $H_2O_2$-induced cytotoxicity.

Antioxidant and Free Radical Scavenging Activity of Different Fractions from Hawthorn Fruit

  • Hawthorn fruit is a conventional medicine used in treating cardiovascular diseases. Its therapeutic effects may relate to its antioxidant compounds. In this study, we evaluated the antioxidant activity of $CH_2Cl_2$, EtOAc, n-butanol and water fractions from 70% methanolic hawthorn fruit extract by total phenolic and flavonoid contents, total antioxidant activity, DPPH free radical scavenging activity, superoxide radical scavenging activity, reducing power assay, lipid peroxidation inhibitory activity and protective effect against hydroxyl-radical-induced DNA damage. Results showed that the EtOAc fraction contained significantly greater antioxidant activities than other fractions, which suggests that the potent EtOAc fraction should be used for further studies to identify the antioxidant compounds.

Neuroprotective and Free Radical Scavenging Activities of Phenolic Compounds from Hovenia dulcis

  • The EtOAc-soluble fraction from a methanolic extract of Hovenia dulcis Thunb. exhibited neuroprotective activity against glutamate-induced neurotoxicity in mouse hippocampal HT22 cells. The neuroprotective activity-guided isolation resulted in 8 phenolic compounds (1-8), such as vanillic acid (1), ferulic acid (2), 3,5-dihydroxystilbene (3), (+)-aromadendrin (4), methyl vanillate (5), (-)-catechin (6), 2,3,4-trihydrobenzoic acid (7), and (+)-afzelechin (8). Among these, compounds 6 and 8 had a neuroprotective effect on the glutamate-induced neurotoxicity in HT22 cells. Furthermore, compound 6 had a DPPH free radical scavenging effect with an $IC_{50}$ value of $57.7{\mu}M$, and a superoxide anion radical scavenging effect with an $IC_{50}$ value of $8.0{\mu}M$. Both compounds 6 and 8 had ABTS cation radical scavenging effects with $IC_{50}$ values of $7.8{\mu}M\;and\;23.7${\mu}M$, respectively. These results suggest that compounds 6 and 8 could be neuroprotectants owing to their free radical scavenging activities.
