Variation of Antioxidant and Anticancer Activities of Hull and Bran Extracts in Different Colored Rices (종피색이 다른 벼 품종의 영과 쌀겨 추출물의 항산화 및 항암활성 변이)

  • In order to confirm the biological activities of rice bran and hull, total phenolic content, antioxidant and anticancer activities were examined in three rice varieties that have different seed coat colors such as brown, black, and purple. The antioxidant activity was measured by free radical scavenging assay including DPPH, $ABTS^+$ and reducing power activity. The anticancer activity was evaluated by WST assay with human HepG2 cell lines. As the result, Huegjinju, black seed coat rice, showed higher of the total phenolics contents in both bran and hull, than purple Jeojinju and brown Ilpum. In the antioxidant activity, Heugjinju showed the highest among the bran extracts, but the activities of hull extracts were similar in three varieties. In the anticancer activity to HepG2 cell line, the hull extract was 20% higher than that of bran extract in the average of three varieties. The hull extracts of Ilpum and Jeokjinju showed similar anticancer activites about 92%, and that of Heugjinju showed the lowest activity of 30.4%. The anticancer activities of hull extracts in three varieties showed positive correlation with total phenolics contents, DPPH radical scavenging activities and reducing power activities. In conclusion, the rice bran and hull showed potent antioxidant and anticancer activities varied in three varieties that have different seed coat colors.

Physicochemical Properties and Biological Activities of Protaetia brevitarsis seulensis Larvae Fermented by Several Kinds of Micro-organisms (유용 미생물을 이용한 발효굼벵이 추출물의 이화학적 특성 및 생리활성효과)

  • In this study, the biological activities of aqueous, ethanol, and methanol extracts of larvae of the edible insect Protaetia brevitarsis seulensis, fermented using several kinds of microorganisms, were tested in in vitro experimental models. Six effective microorganisms were used for fermentation, namely Lactobacillus plantarum JBMI F3, Lactobacillus plantarum JBMI F5, Lactobacillus gasseri Ba9, Aspergillus kawachii KCCM 32819, Saccharomyces cerevisiae KACC 93023, and Bacillus subtilis KACC 91157. Biological activities (${\alpha},{\alpha}^{\prime}-diphenyl-{\beta}-picrylhydrazyl$ [DPPH] free radical scavenging activity, reducing power, and fibrinolytic activity), and biochemical properties (phenolic compounds and flavonoids) were examined in aqueous, ethanol, and methanol extracts from P. brevitarsis seulensis powder and fermented P. brevitarsis seulensis powder. The total phenolic compounds and flavonoid contents were highest in the aqueous extract of B. subtilis-fermented P. brevitarsis seulensis powder. DPPH radical scavenging activity and reducing power were stronger in the fermented group than the nonfermented group. Fibrinolytic activity were highest in the extract from B. subtilis-fermented P. brevitarsis seulensis powder. The ${\alpha}-amylase$ activity in starch was higher in the fermented group than the nonfermented group, but there was no significant difference. These results provide basic data to understand the biological activities of bioactive materials derived from fermented P. brevitarsis seulensis larvae for the development of functional foods.

Antioxidative Activity of Kalopanax pictus Shoot Extracted Using Different Extraction Methods (추출 방법이 개두릅 추출물의 항산화 활성에 미치는 영향)

  • The antioxidative activity of Kalopanax pictus shoots extracted using five different methods, stirrer extraction (SE), ultrasonification extraction (USE), vacuum extraction (VE), reflux extraction (RE), and reflux extraction after ultrasonification extraction (RUE), was evaluated to determine the most effective extraction method. RUE showed the highest yield of extract, and total polyphenol and flavonoid content. Additionally, the RUE extract showed higher antioxidative activity than that of the other extracts based on DPPH and ABTS free radical scavenging (90.85%, 99.83%), reducing power activity (OD 0.95, 700 nm), nitrite scavenging activity (55.46%), and ferrous ion chelating effect (45.12%).

Changes in Antioxidant Activity, Total Phenolics and Vitamin C Content during Fruit ripening in Rubus occidentalis (블랙 라스베리의 과실 성숙에 따른 항산화 활성, 총페놀 함량 및 비타민 C 함량 변화)

  • In this study, changes in the overall antioxidant properties and chemical constituents including total phenolics and vitamin C of R. occidentalis fruit during ripening are studied. The antioxidant activity was measured by the free-radical scavenging activity(DPPH method) and reducing power(potassium ferricyanide method). Although, the weight and diameter of R. occidentalis fruit were increased with the progress of ripening, antioxidant activity and total phenolics were decrease during ripening. The highest free-radical scavenging activity(at $125{\mu}g/m{\ell}$) and reducing power(at $100{\mu}g/m{\ell}$) in fruit were 61.67% and 0.71, respectively. Total phenolic content and vitamin C content in fruit of 5 days after fruit set were $220.73{\mu}g/g$ and $540.45{\mu}g/g$, respectively. A linear correlation(r=0.9761) was shown between free-radical scavenging activity and total phenolic content.

Radical Scavenging Activities of Phellinus pini (상황버섯(Phellinus pini)의 라디칼 소거작용)

  • The concentration of phenolics in Phellinus pini (CY001) extracts, expressed as mg of GAEs per g of P. pini fractions, and the EtOAc fraction (436.5 mg GAEs/g) of P. pini had a higher phenolic content than other fractions. Several biochemical assays were used to screen antioxidant properties such as reducing power, 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity, NBT/XO superoxide system and inhibition of DCF/AAPH peroxyl radicals. Among the six mushroom extracts, the EtOAc fraction from P. pini (CY001) showed the most potent DPPH radical, superoxide radical, and peroxyl radical scavenging activities, with $IC_{50}$ values of $11.49\;{\mu}g/ml$, $8.32\;{\mu}g/ml$, and $1.91\;{\mu}g/ml$, respectively. The EtOAc fraction of P. pini (CY001) significantly inhibited enzymatic lipid peroxidation and effectively attenuated LPS-induced NO production of RAW 264.7 cells without cytotoxicity. We also found that the EtOAc fraction had a significant hepato-protectant effect on tacrine-induced cytotoxicity in HepG2 cells. These findings suggest that P. pini (CY001) may have potential as a natural antioxidant, which contains compound(s) with radical scavenging activity.

Nutritional composition and functionality of mixed cereals powder (혼합잡곡분말의 영양성 및 기능성)

  • Six types of mixed cereals powder were prepared using different mixing ratios of barley, proso millet, sorghum, and adlay, and their nutritional components and functionalities were analyzed. The mixed cereals powders contained high levels of crude protein. The free amino acid and mineral contents of Sample E were the highest among all the samples. Fructose, glucose, sucrose, and maltose were detected as the four major free sugars in all the mixed cereals powders, and Sample D had the highest amount of reducing sugar among all the samples. The total polyphenol and total flavonoid contents of Sample F were significantly higher than those of the other samples. Sample C showed the highest level of DPPH radical scavenging activity and reducing power. As for the ABTS radical scavenging activity, the $IC_{50}$ of Sample D was the lowest among all the samples, but did not significantly differ from that of Sample C. Therefore, we suggest 2:2:1:1 (in Sample D) as the best mixing ratio of barley, proso millet, sorghum, and adlay to produce a mixed cereals powder with excellent nutritional value and antioxidant activities.

A Study of Antioxidative and Antimicrobial Effects of Coffee Residue Extracts (커피 부산물의 항산화와 항균력에 관한 연구)

  • The purpose of this study is to analyze the possibility of a residual product of coffee (RC). RC oil extracted with n-hexane at $60({\pm}10)^{\circ}C$ for 24 hours. In this study, the cytotoxicity of RC oil was observed against B16F10 melanoma cells and RAW 264.7 macrophage cells by water solubletetrazolium salt-1 assay, and The RC oil measured by methods of DPPH radical scavenging and antimicrobial activities in Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans. As a result, the RC oil treatment-related cytotoxic effects appeared on B16F10 melanoma cells from 0.125 to $2{\mu}{\ell}/m{\ell}$ and RAW 264.7 macrophage cells from 0.125 to $0.5{\mu}{\ell}/m{\ell}$ concentrations in this study. RC oil is radical scavenging activity concentrations on dependent. The antimicrobial activity of RC oil ($150{\mu}l/{\ell}$) was determined by clear zone method. Straphylococcus epidermidis, Straphylococcus aureus, Escherichia coli, Candida albicans showed clear zone by each $11.3{\pm}0.4$, $12.{\pm}0.7$, $12.0{\pm}0.0$, $0.0{\pm}0.0mm$. It is suggested that RC oil have effects on the cytotoxicity, antioxidant and antimicrobial that could be applicable to cosmetics as a new material.

Characterization of the morphology and antioxidant content of shiitake cultivated in smart farm system (스마트 팜 시스템으로 재배된 표고의 외형평가 및 항산화능 활성)

  • In this study, the morphology and antioxidant content of shiitake mushrooms (Lentinula edodes) cultivated in smart farms and general farms have been compared. With regard to morphology, mushrooms produced in the smart farm system exhibited a slightly thicker and wider pileus and thicker and longer stipe than those in the general farm system. The stipe in the mushrooms from moderate-sized farms was harder, because the low relative humidity of cultivation rooms could induce mushroom tissue to harden. With regard to the antioxidant content, the free radical scavenging activity was evaluated by the DPPH assay. Among the various treatments, hot water extracts of freeze-dried shiitake produced from smart farms exhibited the highest DPPH value of 37.8%. In contrast, the lowest activity of 12.2% was observed in a 70% fermented alcohol extract of shiitake that was dried by hot air. The polyphenol content was higher in hot water extracts than in 70% fermented alcohol extracts. Additionally, the polyphenol content was higher in the freeze-dried samples than in hot-air dried ones. The smart-farm system was preferred over the general cropping system for cultivating shiitake mushrooms, because the antioxidant activity and polyphenol content of mushrooms from the smart-farm system was better; the functionality of this system was more improved than that of the general cropping system, and it enables mushrooms to be cultivated more efficiently. The antioxidant content is represented as the $mean{\pm}SD$ of three replicates. Different letters indicate significant differences among samples, i.e., p<0.05.

Antioxidant Properties of Various Microorganisms Isolated from Arctic Lichen Stereocaulon spp. (북극 지의류 Stereocaulon spp로부터 분리한 여러 미생물의 항산화 성질)

  • Lichens are symbiotic organisms composed of fungi, algae, or cyanobacteria which are able to survive in extreme environmental conditions ranging from deserts to polar areas. Some lichen symbionts produce a wide range of secondary metabolites that have many biological activities such as antibacterial, antifungal, antiviral, antitumor, antioxidant and anti-inflammatory etc. Among the symbionts of lichens, of the bacterial communities of lichen symbionts little is known. In this study, we isolated 4 microbial species from the Arctic lichen Stereocaulon spp. and evaluated their antioxidant properties using 1,1-diphenyl-2-picryl-hydrazyl assay as well as 2,2'-azino-bis(3-ethyl benzothiazoline-6-sulphonic acid) assay. Total phenolic contents and total flavonoid contents were also measured. A potent radical scavenging activity was detected in a number of the lichen extracts. Among the 4 species tested in this study, the ethyl acetate extract of Bosea vestrisii 36546(T) exhibited the strongest free radical scavenging activity, with an inhibition rate of 86.8% in DPPH and 75.2% in ABTS assays. Overall, these results suggest that lichen-bacteria could be a potential source of natural antioxidants.

Protective Effect of Rehmannia Radix Preparata Extract on the Cisplatin-induced Cytotoxicity of HEI-OC1 Cells via Scavenging of Free Radicals (숙지황 추출물이 Cisplatin으로 손상된 HEI-OC1 세포보호와 유리라디칼 소거능에 미치는 영향)

  • The steamed root of Rehmannia glutinosa has been used for treatment of inner ear diseases, such as tinnitus and hearing loss in traditional Oriental Medicine. In the present study, we investigated the effect of ethanol extract of steamed root R. glutinosa (SRG) on cisplatin cytotoxicity of HEI-OC1 auditory cells. In addition, to investigate the mechanism of SRG on cisplatin cytotoxicity, the effects of SRG on lipid peroxidation as well as scavenging activities against various free radicals were measured in cisplatin-treated cells. Treatment of SRG protected cells from cisplatin and reduced lipid peroxidation in a dose-dependent manner. Furthermore, SRG demonstrated significant scavenging activity against various free radicals, including superoxide radical, hydroxyl radical, hydrogen peroxide, and DPPH radical. These results indicate that SRG protects cisplatin-induced damages of HEI-OC1 cells through inhibition of lipid peroxidation and augmenting scavenging activities against free radials.
