Antioxidant Effects of Salicornia bigelovii Seed Extracts

  • The objective of this study was to compare antioxidant effects of different extracts of Salicornia bigelovii seed. DPPH radical scavenging activity, chelating activity of ferrous, superoxide anion radical-scavenging activity, hydroxyl radical scavenging activity, and contents of total polyphenol and flavonoid were measured for different extracts. Ethanol extract (10 mg/mL) was found to have the maximum DPPH radical scavenging and superoxide anion radical scavenging activities (65.26 and 62.36%, respectively). However, hot water extract (10 mg/mL) showed the highest hydroxyl radical scavenging and chelating activities (51.50 and 53.24%, respectively). Total phenolics and flavonoid contents in the ethanol extract were 522.8 and $480.35{\mu}g/mg$, respectively. They were 401.51 and $352.60{\mu}g/mg$, respectively, in the hot water extract. These results indicate that ethanol and hot water extracts of S. bigelovii seed have good potential to be used as a new source of antioxidants for functional foods, cosmetics, and pharmaceuticals.

민들레 물추출물의 항산화 및 자유라디칼 소거활성 (Antioxidative and Free Radical Scavenging Activity of Water Extract From Dandelion (Taruaxacum officinale))

  • 민들레 잎과 뿌리로부터 물추출물을 제조하여 항산화 활성 및 라디칼 소거활성을 분석하기 위하여 linoleic acid에 대한 과산화물 생성저해율, DPPH radical에 대한 소거활성, chemiluminescence를 이용한 hydroxyl radical에 대한 소거활성, EPR을 이용한 superoxide anion radical에 대한 소거활성및 hydrogen peroxide에 대한 소거활성을 측정하였다. 그 결과, 민들레 잎의 물추출물이 뿌리의 물추출물 보다 지방산에 대한 과산화물 생성 저해율이 높았고, DPPH radical, hydroxyl radical, superoxide anion radical 및 hydrogen peroxide에 대한 소거활성 역시 매우 높은 것으로 나타났다.

  • 현재 시판되고 있는 한약재 및 약용식물 118종에 대해서 80% 메탄올 추출물을 제조하였고 , 이들의 항산화 활성을 DPPH 와 superoxide anion radical 소거능 측정 방법을 이용하여 조사하였다. DPPH radical 소거 활성은 괴화 (Sophora japonica: 줄기부)의 메탄올 추출물에서 76.9%의 활성을 보였고 회수(Camptotheca acuminata Dence: 지상부)의 추출물에서는 50.9%의 활성을 보였으며 박하(Mentha arvensis: 지상부), 비파엽 (Eriobotryajaponica: 지상부), 소엽(Perilla frutescens: 줄기부), 애엽(Artemisia asiatica: 줄기부 ), 초과(Amomum. costatum: 지상부) 의 추출물과 산비장이(Serratula koreana: 잎), 살구나무(Prunus ansu: 잎)는 30% 이상의 활성을 보였다. Superoxide anion radical 소거능은 박하(Mentha arvensis), 비파엽(Eriobotrya jαponica), 초과(Amomum costatum), 희수(Camptotheca acuminata Dence)의 지상부에서 80% 이상의 활성을 보였으며 괴화(Sophora jαponica), 소엽(Perilla frutescens), 애엽(Artemisia asiatica), 순비기나무 (Vitex rotundifolia)의 줄기부 , 짚신나물(Agrimonia pilosa)의 잎, 창질경이(Plantago lanceolata) 지상부는 50% 이상의 활성을 나타내었다. 박하(Mentha arvensis) 와 비파엽(Eriobotrya jαponica), 초과(Amomum costatum), 희수(Camptotheca acuminata Dence)의 지상 부는 DPPH뿐만 아니라 superoxide anion radical 소거능에서도 높은 활성을 보였다.

Antiradical Capacities of Perilla, Sesame and Sunflower Oil

  • The aim of this study is to examine the radical scavenging activity of perilla and sesame oil that Koreans traditionally consume. For DPPH radical scavenging activity, oil and its hexane/70% methanol extracts (ME) are used and for superoxide and hydroxyl radical scavenging activities, ME are used. Unrefined perilla oil, sesame oil, and refined sunflower oil are used. The yields for ME of perilla, sesame and sunflower oil are 0.57, 0.61, and 0.30%, respectively, and the amounts of phenolic compounds in ME of corresponding oil are 18.77, 88.64 and $0.05\;{\mu}g$ tannic acid/mg, respectively. $IC_{50}$ for DPPH scavenging activity of perilla, sesame and sunflower oil are 2.12, 1.91, and 3.35 mg/mL, respectively and those for ME of corresponding oils are 0.42, 0.07, and 43.11 mg/mL, respectively. In DPPH assay, the solvent used for oil sample is iso-octane and that for ME is methanol. Superoxide anion scavenging activity of ME of perilla, sesame and sunflower oil tested at 1 mg/mL concentration are 21.10, 13.25, and 3.14%, respectively. Hydroxyl radical scavenging activities of those samples tested at 1 mg/mL concentration are 86.08, 93.30, and 93.17%, respectively. In summary, the refining process seems to remove the phenolic compound during oil processing. Antiradical substances in perilla and sesame oils responsible for scavenging DPPH radicals are present in the methanol fraction, while the antiradical substances in the sunflower oil are in the lipid fraction. DPPH scavenging activity of ME of sesame oil is significantly higher than that of perilla oil (p<0.05). However, superoxide anion scavenging capacity of ME of perilla oils was found to be greater than that of both sesame and sunflower oils (p<0.05).

Antioxidant activity of ethanol extracts from the root and bark of Ulmus davidiana

  • Antioxidant activities of Ulmus davidiana root and bark extracts were evaluated by various antioxidant tests, including DPPH radical-scavenging, nitric oxide-scavenging, superoxide anion radical-scavenging, and ABTS radical-scavenging assays, and superoxide dismutase (SOD)-like activity and reducing power analysis, along with the determination of total phenolic and flavonoid contents. Both the extracts showed strong antioxidant activities by these testing methods. Ulmus davidiana root extract possessed strong reducing power and nitric oxide-scavenging activity, and high scavenging activities against free radicals including the superoxide anion, and the ABTS and DPPH radicals, but a weaker scavenging activity of SOD. In contrast, the Ulmus davidiana bark extract exhibited a strong SOD-like activity, but all the other activities were weak. It was observed that the total phenolic and flavonoid contents of the Ulmus davidiana root extract were higher than those of the Ulmus davidiana bark extract.

가감형방지황탕 열수 추출물이 항산화 작용에 미치는 영향 (Antioxidant Property of the Gagam-Hyungbang-Gihwang-tang Using Biochemical Markers of Carcinogenesis)

  • Objectives : The verity extract of the Gagam-Hyungbang-Gihwang-tang (GHG) was assessed to determine the mechanisms of its antioxidant activity. Methods : The fellowing effects were measured : GHG exhibited a concentration-treatment; scavenging ${\alpha},\;{\alpha}-diphenyl-\beta-picrylhydrazyl$ (DPPH) radical, linoleic acid oxidation in a thiocyanate assay system, and superoxide anion, hydroxyl radical-induced DNA nicking. We investigated mRNA levels such as superoxide-dismutase. Results : The GHG extract showed dose-dependent free radical scavenging activity, including DPPH radicals, hydroxyl radicals, and superoxide anion, using different systems. The GHG was also found to be effective in protecting plasmid DNA against the strand breakage induced by hydroxyl radicals in Fenton's reaction mixture. Furthermore, SOD-1 mRNA expression levels increased in tat hepatoma H4IIE cells Conclusions : We expect that GHG will to helpful to the development of antioxidant activity treatments.

사과락(絲瓜絡)의 항산화효과 및 3T3-L1분화 시 Cytokine류에 미치는 영향 (Experimental Study on Antioxidative Effect of Luffae Fructus Retinervus and Their Effects on Cytokines to 3T3-L1 Cell Lines)

  • In recent year, We are concerned in anti-aging, disease-protection, long-life, many method are used in solving this problem. Recently, We heard that Luffae Fructus Retinervus(LFR) has effect of anti-aging, disease-protection, long-life. So I let made a experiment for this result. The purpose of this study is to; 1) the anti-oxidant effect of Luffae Fructus Retinervus(LFR) used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity, Superoxide anion radical scavenging activity, 2) cultivation 3T3-L 1 Preadipocytes and Protein chip used for ProteoPlexTM 16-Well Murine Cytokine Array Kit. We measured level of DPPH radical scavenging activity. And we experienced that the ability of DPPH radical's elimination was increased by rising concentration of LFR. When the concentration of LFR was 5 mg/ml, the ability of DPPH radical's elimination was Maximum. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was significant when concentration of LFR was from 1.25 mg/ml to 2.5 mg/ml. We measured level of Superoxide anion radical scavenging activity. And we founded that the ability of Superoxide anion radical's elimination was maximum when concentration of LFR was 0.3125 mg. When we inspected Antioxidative Effects with BSA, we experienced that ability of defense was increased by rising concentration of LFR. We known the immunity of LFR about 3T3-L1 Preadipocytes and gained the increase of Cytokines(IL-2, IL-4, GM-CSF) without IL-12p70, $INF-{\gamma}$, $TNF-{\alpha}$ So I guess that Luffae Fructus Retinervus(LFR) has effects of anti-aging, disease-protection, long-life, etc.

부위별 꽈리(Physalis alkekengi var. francheti) 추출물의 항산화효과 (Antioxidative Activities of Different Part Extracts of Physalis alkekengi var. francheti (Winter Cherry))

  • 본 실험에서는 꽈리를 열매, 꽃받침, 잎, 줄기, 뿌리 등 부위별로 구분하여 메탄올로 추출하고 추출 수율, 총페놀 함량, 총플라보노이드 함량, DPPH radical 소거능, 아질산염 소거능, superoxide anion radical 소거능, 환원력, 철 이온에 대한 킬레이트 효과 등을 측정하였다. 추출 수율은 열매가 가장 높았고 그 다음으로 꽃받침, 잎, 줄기, 뿌리 순이었다. 총페놀 및 총플라보노이드 함량은 잎 추출물이 가장 높았고 그 다음으로 꽃받침 추출물이었다. DPPH radical 소거능은 1 mg/mL 및 5 mg/mL의 농도에서 잎 추출물이 가장 높았고 꽃받침 추출물도 높은 활성을 나타내었다. 아질산염 소거능은 모든 부위에서 농도에 따라 유의적인 차이 없이 높게 나타났다. 꽈리 추출물의 superoxide anion radical 소거능은 꽃받침과 잎 추출물이 높은 활성을 나타내었다. 환원력은 잎 추출물에서 가장 높게 나타났고 철 이온에 대한 킬레이트 효과는 꽃받침, 줄기 및 뿌리 추출물이 높은 활성을 나타내었다. 이상의 결과를 종합하여 보면 꽈리의 잎 추출물에서 총 페놀 함량, 총플라보노이드 함량과 항산화 활성이 탁월하게 높게 나타났고 그 다음으로 꽃받침 추출물이 우수한 항산화 활성을 나타내어 향후 이를 이용한 천연 항산화제로 개발 가능성이 시사되었다.

오계란 단백질 가수 분해물 제조 및 한외여과 분획물의 in vitro 항산화 활성 특성 (In vitro Antioxidant Activity of Ogae (Korean Native Black Fowl) Egg White Protein Hydrolysates Fractionated by Ultrafiltration)

  • 식물 및 동물성 단백질 유래 펩타이드 형태의 단백질 가수 분해물들은 항산화, 고혈압 완화, 면역조절, 진통완화 및 항균작용 등 생리활성이 있는 것으로 알려져 왔다. 본 연구는 연산오계란 단백질 가수 분해물을 Ultrafiltration를 이용하여 HDS(분획되지 않은 가수 분해물), 1 kDa, 5 kDa, 10 kDa, 50 kDa로 분획된 기능성 펩타이드의 DPPH radical scavenging activity, superoxide anion radical scavenging activity, hydroxyl radical scavenging activity 및 $Fe^{2+}$ chelation ability을 평가하였다. 그 결과 DPPH radical scavenging activity 최대값은 1 kDa(70.83 %), hydroxyl radical scavenging activity 최대값은 5 kDa (47.01 %), superoxide anion radical scavenging activity 최대값은 5 kDa(40.57 %), $Fe^{2+}$ chelation ability 최대값은 5 kDa(29.87 %)로 나타났다. Ultrafiltration를 이용하여 fractionation된 단백질 가수 분해물의 항산화 저해 능력 $IC_{50}$ 평가하였다. 그 결과 HDS의 최대값은 superoxide anion radical scavenging activity($IC_{50}$, 5.42 mg/ml)이고, 1 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 1.67 mg/ml)이고, 5 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 2.09 mg/ml)이고, 10 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 2.61 mg/ml)이고, 50 kDa의 최대값은 $Fe^{2+}$ chelation ability($IC_{50}$, 4.53 mg/ml)이다. 그러므로 본 연구 결과를 바탕으로 5 kDa를 이용하여 오계란 단백질에서 분획한 펩타이드는 항산화 기능성 식품소재로서 활용할 가치가 높을 것으로 기대한다.

복분자 추출액 급이 사양꿀과 일부 시판꿀의 항산화활성 비교 (Comparison of Some Antioxidative Activities of Feeding Honey from the Mixture of Extract of Rubus Coreanus Miquel and Sugar with Three Types of Honeys on the Market)

  • To make sure of the usefulness of extract of Rubus coreanus Miquel for producing functional feeding honey, we compared some antioxidative indicators of feeding honey using extract of Rubus coreanus Miquel with acacia honey, SueBee Clover honey(USA), feeding honey on the market. The water content of four honeys were 16.6~26.5%, pH were 3.18~3.70, and titratable acidity ranged 0.018~0.022%. The phenolic compound contents of SueBee Clover honey and feeding honey using extract of Rubus coreanus Miquel were 8.3 mg/100 g and 7.3 mg/100 g, respectively, and were significaltly higher thgheacacia honey and feeding honey on the market. The flavonoid contents per 100 g of honey ranged from 2.3 mg(acacia honey) to 15.0 mg(SueBee Clover honey). DPPH anion scavenging activity of four honeys were not high. 0.5~2.0 g/ml of feeding honey using extract of Rubus coreanus Miquel was 16~36% and showed a concentration-reliant figure. At the concentration of 0.25~0.75 mg/ml, the reducing power of four honeys increased concentration-dependently, and the power of 0.25 mg/ml of feeding honey using extract of Rubus coreanus Miquel was corresponding to thgt of $150{\mu}g$/ml of a vitamin C solution. ABTS radical scavenging activity of feeding honey using extract of Rubus coreanus Miquel was 43.3~68.4%; the highest activity amongst all samples. When plotting the dose-response curve, ABTS radical scavenging activity also increased as their concentration increased from 62.5 mg/ml to 500 mg/ml. The heat treatment reduced the phenolic compound contents of acacia honey and feeding honey using extract of Rubus coreanus DPPH anion scavenging activity of feeding honey on the market and feeding honey using extract of Rubus coreanus Miquel did not changed significaltly after heating for 20 min, and the three honey except SueBee Clover honey maintained the reducing power with the same treatment.
