해면으로부터 분리된 Bromotopsentin의 항산화활성 (Antioxidant Activities of Bromotopsentin from the Marine Sponge Spongosorites sp.)

  • Bromotopsentin (BSM) is a bisindole alkaloid compound, which is recognized as a metabolite of the marine sponge Spongosorites sp. In this study, the antioxidant activity of BSM was investigated. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, the trolox equivalent antioxidant capacity (TEAC) assay, the superoxide radical scavenging (NBT) assay, the lipid peroxidation and hydroxyl radical-induced DNA damage assays were carried out to evaluate the antioxidant activity of BSM. It was found that BSM had stronger scavenging activity on the stable free radical DPPH and superoxide radical than L-ascorbic acid with an $IC_{50}$ value of 62 and 64 ${\mu}M$, respectively. The TEAC value which indicated the total antioxidant capacity of BSM was about 0.8, which was also stronger than L-ascorbic acid. About 1.3 ${\mu}M$ of BSM induced 50% inhibition of lipid peroxidation. 60 nM of BSM exhibited a significant protective activity against DNA strand scission by hydroxyl radical on pBR322 DNA. Taken together, we suggest that BSM possesses strong antioxidant activity, and could be a valuable new addition to the list of anti-aging chemotherapeutic agents.

천연 식물자원에서 DPPH 라디칼 제거능과 Prolyl Endopeptidase 활성 저해능 탐색 (Investigation of DPPH Radical Scavenging and Pyolyl Endopeptidase Inhibitory Activities of Plant Extracts)

  • 현재 알츠하이머 질환의 주요한 치료제는 아세틸콜린 분해 저해제(acetylcholinesterase inhibitors)이나, tacrine의 경우 간독성 때문에 지금은 거의 사용되지 않고 있다. 이외에 알츠하이머 질환의 잠재적 치료로서 항산화제와 항아밀로이드 요법이 대두되어 치매의 예방 및 치료를 위한 무독성의 약물 개발을 위해서 많은 연구가 수행되고 있으나 아직까지 치매의 발병 기전이나 뚜렷한 효능을 가진 치료제에 대한 보고가 없는 실정이다. 따라서 본 연구에서는 새로운 자원에서 알츠하이머 질환의 치료제로서의 가능성을 탐색하고자 전초 추출물 35종과 부위별 추출물 36종의 식물 추출물 71종에서 DPPH 라디칼 소거능과 PEP 활성 저해율을 측정하였다. 항산화 활성을 살펴 본 결과, 식물 전초에서는 땃딸기(Fragaria yezoensis)에서 $90.4\%$의 라디칼 제거 효과를 보였고, 은행나무(Gingko biloba), 치자나무(Gardenia jasminoides for. grandiflora), 산철쭉(Rhododendron yedoensa var. poukhanense)등의 줄기, 털진달래(Rhododendron mucronulatum var. ciliatum), 치자나무(Gardenia jasminoides for. grandiflora)와 물개암나무(Corylus sieboldiana var. mandshurice)의 잎 부위에서 $88\%$ 이상의 높은 라디칼 제거 효과를, 산수유(Cornus officinalis) 열매 부위 추출물에서는 약 $95\%$ DPPH 라디칼 제거 효과를, 은행나무(Gi템 biloba), 우산고로쇠(Acerokamotoaum)등의 뿌리 부위에서 $85\%$ 이상의 DPPH 라디칼 제거 효과를 보였다. PEP활성의 저해율을 측정하여 비교한 결과, 식물 전초의 경우 딸기(Fragaria ananassa), 땃딸기(Fragaria yezoensis), 고추나물(Hyperircum erectum)에서 높은 저해율을 줄기 부위의 경우 다래(Actinidia arguta), 산철쭉 (Rhododendron yedoensa var. poukhanense)의 줄기 부위, 산철쭉(Rhododendron yedoensa var. poukhanense)과 생열귀나무(Rosa davurica), 삼나무(Cryptomeria japonica), 털진달래(Rhododendron mucronulatum var. ciliatum)의 및 부위에서 90$\%$ 이상의 높은 PEP 활성 저해율을, 산수유(Cornus officinalis)의 열매 추출물이 95.3$\%$의 저해율을, 우산고로쇠(Acer okamotoanum) 뿌리 추출물이 $83.3\%$의 저해율을 나타냈다. 식물 추출물 71종에서 땃딸기(Fragaria yezoensis) 전초, 산수유(Cornus officinalis) 열매 부위의 경우 DPPH 라디칼 소거능과 PEP 활성 저해율에서 모두 $90\%$ 이상의 활성을 보이는 등 DPPH 라디칼 소거능과 PEP 활성 저해율 사이에 유의수준 0.000에서 양의 상관관계가 있었다. 따라서 PEP 활성 저해 메커니즘에 대한 항산화 작용의 영향을 더욱 더 조사할 필요가 있을 것으로 판단되며, 산수유와 땃딸기와 같은 식물 자원은 항산화활성과 PEP 활성 저해능이 모두 높으므로 알츠하이머 질환의 치료 및 예방 물질로서의 개발 가능성이 큰 것으로 보인다.

Monascus 속 균주를 이용한 발효 홍국마 추출물의 생리학적 특성 (Physiological characteristics of Extracts from Monascus-Fermented Chinese Yam by Monascus sp. strain)

  • 본 연구는 발효 홍국마로부터 에탄올 추출물 농도에 따른 생리활성 효과를 조사하기 위하여 실시하였다. 발효 홍국마 에탄올 추출물로부터 색소, DPPH radical 소거활성, 환원력 및 monacolin K 함량을 조사하였다. 그 결과, 기질로서 장마를 Monascus sp. MK805 균주로 발효하였을 때 홍국색소는 29.6(황색), 15.1(오렌지색), 20.4(적색), DPPH radical 소거활성은 72.8%, 환원력은 1.74 및 monacolin K 함량은 480.6 mg/kg으로 각각 조사되었다. 이처럼 발효 홍국마는 항산화 및 항고지혈증 관련 기능성 식품 및 의약품 소재로의 이용 기능성이 높은 것으로 사료된다.

Quality Characteristics of Jelly Made from Fermented Red Ginseng Concentrate with Increased Ginsenoside Content by Enzyme Treatment

  • The purpose of this study is to investigate the physicochemical properties of jelly made from fermented red ginseng concentrate (FRGC) that can be easily absorbed and digested for the health promotion of the elderly. The pH of the jellies tended to decrease with increasing concentration of FRGC. Soluble solid content has significantly higher value when added more than 2%, and the water content of the sample was significantly lower when the FRGC was added 4%. As the amount of FRGC was increased, the total color difference increased, and the hardness of samples decreased significantly. On the other hand, the total ginsenoside contents of the FRGC was 45.50 mg/g. As the concentration of FRGC increased, the content of polyphenol and flavonoids increased. The increasing pattern of polyphenols and flavonoids showed a similar trend. As the content of FRGC increased, ABTS free radical scavenging activity significantly increased (p<0.05), and in the control, the minimum value (62.6 AEAC) and the 4% sample were highest (116.2 AEAC). DPPH radical scavenging activity was like that of ABTS radical scavenging activity. However, there was no significant difference in DPPH radical scavenging activity of 3% and 4% red ginseng jelly.

Antioxidant and Anti-diabetes Activities of Methanolic Extract and Fractions of Astragalus membranaceus Roots

  • The potential biological activities of methanol extract and 5 fractions (hexane, $CH_2Cl_2$, EtOAc, BuOH and water) from roots of Astragalus membranaceus were examined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, hydroxyl radical (${\cdot}OH$) scavenging activity, reducing power assays, lipid peroxidation inhibitory activity, $\alpha$-amylase and $\alpha$-glucosidase inhibition assays. The EtOAc fraction showed high DPPH free radical scavenging activity ($EC_{50}=170.34\;{\mu}g/mL$), hydroxyl radical scavenging activity ($EC_{50}=32.14\;{\mu}g/mL$), lipid peroxidation inhibitory activity ($EC_{50}=52.46\;{\mu}g/mL$) and a concentration dependence, with OD value ranging from 0.234 to 0.345 (0.1 to 0.5 mg/mL), for reducing power. The EtOAc fraction has the highest total phenolic content ($142.13\;Gal\;{\mu}g/mg$) and the $CH_2Cl_2$ fraction has the highest flavonoid content ($71.63\;Que\;{\mu}g/mg$). Meanwhile, hexane and EtOAc showed certain $\alpha$-amylase and $\alpha$-glucosidase inhibition activities. These results suggest that the methanol extract and fractions from Astragalus membranaceus root have significant antioxidant and anti-diabetes activities, which could be used as a potential source of pharmaceutical materials.

Natural Anthraquinone Derivatives from a Marine Mangrove Plant-Derived Endophytic Fungus Eurotium rubrum: Structural Elucidation and DPPH Radical Scavenging Activity

  • There is considerable interest in the isolation of potent radical scavenging compounds from natural resources to treat diseases involving oxidative stress. In this report, four new fungal metabolites including one new bisdihydroanthracenone derivative (1, eurorubrin), two new seco-anthraquinone derivatives [3, 2-O-methyl-9-dehydroxyeurotinone and 4, 2-O-methyl-4-O-(${\alpha}$-D-ribofuranosyl)-9-dehydroxyeurotinone], and one new anthraquinone glycoside [6,3-O-(${\alpha}$-D-ribofuranosyl)-questin], were isolated and identified from Eurotium rubrum, an endophytic fungal strain that was isolated from the inner tissue of the stem of the marine mangrove plant Hibiscus tiliaceus. In addition, three known compounds including asperflavin (2), 2-0-methyleurotinone (5), and questin (7) were also isolated and identified. Their structures were elucidated on the basis of spectroscopic analysis. All of the isolated compounds were evaluated for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity.

오배자의 항산화 활성성분 및 자유라디칼 소거효과 (Constituents of Antioxidative Activity and Free Radical Scavenging Effect from Galla Rhois (Rhus javanica Linne))

  • This study was carried out to investigate the structure of antioxidative constituents and the free radical scavenging effect of the main ingredients from Galla Rhois(Rhus javanica Linne). Antioxidative activities of n-hexane, EtOAc and BuOH extracts of Galla Rhois were similar or even higher than that of natural (tocopherol) or synthetic antioxidant (BHA). It is suggested that major fractions for the antioxidative activity of Galla Rhois were the n-hexane, EtOAc and BuOH extract compartments. In the subsequently experiment, one active compound from n-hexane extract, three active compounds from EtOAc extract and one active compound from BuOH extract were isolated. Their chemical structures were identified as syringic acid, protocatechuic acid, gallic acid methylester, gallic acid and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ on the basis of the speculation of spectral data and chemical reaction. Among the compounds, protocatechuic acid, gallic acid methylester and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ showed most potent radical scavenging effect using DPPH method.

Multiresponse Optimization and Prediction of Antioxidant Properties of Aqueous Ginger Extract

  • The influence of extraction temperature, powder concentration, and extraction time on the antioxidant properties of aqueous ginger extract was investigated. The possibility of estimating the antioxidant properties of the extract from its absorbance and colour properties was also investigated. Results indicated that powder concentration was the most significant factor to consider in optimizing antioxidant extraction. However, temperature and time still influenced the 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity while extraction temperature influenced the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of the extract. Using the total phenol content, total flavonoid content, ABTS radical scavenging activity, and DPPH radical scavenging activity of the extract, the multiresponse optimization condition for extraction of antioxidant based on the experimental range studied is $96^{\circ}C$, 2.10 g/100 mL, and 90 min. The absorbance of the ginger extract at 610 nm could be exploited for rapid estimation of its total flavonoid and polyphenol with a $R^2$ of 0.713 and 0.753, respectively.

골질 보호 작용이 있는 수종 생약 추출액의 래디칼 소거능 및 DNA 보호 효과 (Radical Scavenging Activities and Protective Effects against Oxidative Damage to DNA of Extracts from Medicinal Plants with known Osteoprotective Effects)

  • Among aging diseases, the most basic problem is a decrease in bone mineral density. Patients with osteoporosis are steadily increasing in the world. This study was investigated to show the relationship between osteoporotic effects and oxidative damage. Water extracts of 15 medicinal plants and ethanol extracts of 14 medicinal plants with known anti-osteoporotic effects, were tested for their radical scavenging activity using DPPH, ABTS, SRSA and FRAP assay. Water extract of Cornus officinalis, Rubus coreanus and ethanol extract of Rubus coreanus, Viscium album var. coloratum, Cimicifuga heracleifolia showed about 15-20 mg/g of total phenolic contents. Water extract of Cornus officinalis, Rubus coreanus and Epimedium koreanum showed high radical scavenging activity. Ethanol extract of Drynaria fortunei, Cornus officinalis, Rubus coreanus, Gentiana scabra and Astragalus membranaceus showed high radical scavenging activity. Water extract of Drynaria fortunei, Cornus officinalis, Nelumbo nucifera, Epimedium koreanum, and Gentiana scabra showed very strong protective effect against oxidative damage to DNA. These results suggest the correlation between anti-osteoporotic effects and antioxidative effects.

Free Radical Scavenging and Antioxidant Activities of Water Extracts from Amannia multiflora, Amannia coccinea, Salix gracilistyla Inhabiting Along the Nakdong River (Republic of Korea)

  • Plant-derived antioxidants are used as a healthy diet and are known to inhibit various human diseases. In this study, we investigated free radical scavenging and antioxidant activity of extracts from three plants (Ammannia multiflora, Ammannia coccinea and Salix gracilistyla) with the most DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity from 196 plant extracts inhabiting along Nakdong River in Republic of Korea. The three extracts also have strong total antioxidant activity. Moreover, the extracts inhibited hydrogen peroxide (H2O2)-induced reactive oxygen species production and depolarized mitochondrial membrane potential in RAW264.7 macrophages. In zebrafish larvae, 2',7'-dichlorodihydrofluorescein diacetate (DCFDA) fluorescent intensity, induced by H2O2, was markedly reduced by the extracts of A. multiflora, A. coccinea and S. gracilistyla. Meanwhile, the extracts were upregulated Nrf2 and HO-1 expression, and an HO-1 inhibitor reversed the extract-induced oxidative responses both in vivo and in vitro. The data suggest that the extracts of A. multiflora, A. coccinea, and S. gracilistyla exert potential free radical scavenging and antioxidant capacities both in vivo and in vitro by activating the Nrf2/HO-1 signaling pathway.
