Antioxidant Flavonoids from the Twigs of Stewartia koreana

  • In the course of screening for antioxidant compounds by measuring the radical scavenging effect on 1,1-diphenyl- 2-picrylhydrazyl (DPPH), a total extract of the twigs of Stewartia koreana (Theaceae) was found to show potent antioxidant activity. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of six phenolic compounds, ampelopsin (1), catechin (2), proanthocyanidin-A2 (3), fraxin (4), (2R, 3R)-taxifolin-3-${\beta}$-D-glucopyranoside (5), and (2S, 3S)-taxifolin-3-${\beta}$-D-glucopyranoside (6), as active principles. Their structures were elucidated by spectroscopic studies. Compounds 1-6 were isolated for the first time from this plant. Among them, three compounds 1-3 showed the significant antioxidative effects on DPPH, and riboflavin originated superoxide quenching activity. In riboflavin-nitrobluetetrazolium (NBT)-light system, compound 1 showed better superoxide quenching activity than vitamin C.

Biological Activities of Rosaceae Plants Extracts (장미과 식물 추출물의 생물학적 활성)

  • DPPH radical-generating system was used to evaluate the antioxidant properties of the Rosaceae. The inhibitory effects of ethanolic extracts from Rosaceae plants were investigated on melanin biosynthesis which is closely related to hyperpigmentation. Of the Rosaceae extracts, Prunus sargentii, Rubus coreanus, Chaenomeles sinensis, Photinia glabra and Pyrus pyrifolia showed a potent inhibition of tyrosinase, the enzyme which converts 3-(3,4-dihydroxyphenyl) alanine (dopa) to dopachrome in the melanin biosynthetic process. Furthermore, MMT assay was used to check the cytotoxicity of extracts on the human foreskin fibroblast cell line, Hs68. Among the Rosaceae, bark of Prunus sargentii, bark wood of Photinia glabra and all parts or Chaenomeles sinensis showed more than $50\%$ inhibition of mushroom tyrosinase activity at 100 ug/mL and more than $80\%$ of strong DPPH radical-scavenging activity at 10 ug/mL. In audition to, they had no cytotoxic activity on Hs68. These results suggest that these extracts might be except a controler in pigmentation.

Antioxidative Effects of Pine (Pinus denstifora) Needle Extracts. (솔잎 추출물의 항산화 효과)

  • Antioxidative activities of pine (Pinus denstifora) needle extracts were tested in vitro experimental models. The concentration of total polyphenolic compound of water extracts from pine needle was 1.61 %. In DPPH ($\alpha$, $\alpha'$-diphenyl-$\beta$-picrylhydrazyl) method, the electron donating activity of 0.1 % water extracts from pine needle was as high as BHT (0.05%, w/v). The antioxidative activity was measured by inhibition against lipid peroxidation of rat liver microsome, and this activity was shown in the following: 67.7% at 0.1% concentration >63.1% at 0.05% concentration > 28.2% at 0.01% concentration. In antioxidative activity determined by thiocyanate method against lipid peroxidation using linoleic acid, the antioxidative activities at all concentration of 0.01 %, 0.05% and 0.1 % were much higher than control during 7 days. In TBA method, the antioxidative activity was increased with increasing concentration until 6 days. These results support that water extracts from pine needle contain antioxidative compounds.

  • Four diarylheptanoids oregonin, hirsutanonol, 1,7-bis- (3,4-dihydroxyphenyl)-heptane-5-O-$\beta$-D-xylopyanoside and 1,7-bis-(3,4-dihydroxyphenyl)-heptane-5-0-$\beta$-D-glucopyranoside isolated from Alnus hirsuta were evaluated for their antioxidative effects with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation generation system mediated by addition of $H_2O$$_2$and/or Fe$^{2+}$ to rat liver homogenate. All these diarylheptanoids showed significant inhibitory activities against the DPPH radical and demonstrated strong activities in preventing the lipid peroxidation induced by $H_2O$$_2$.>.

The Antioxidant Activity of Cnidii Fructus and Torilis Fructus in Leydig cells (Leydig Cell의 항산화에 미치는 벌사상자와 사상자의 비교연구)

  • Objectives : The purpose of this study was to estimate the antioxidant activity of water extract of Cnidii Fructus (CF) and Torilis Fructus (TF) in Leydig cells. Methods : Free radical scavenging activity of CF and TF against 2,2-diphenyl-1-picrylhydrazyl (DPPH) was determined spectrophotometrically. We investigated the effect of CF and TF in Leydig cells by MTT assay. The protective effects of CF and TF against hydrogen peroxide-induced oxidative stress in Leydig cells. Superoxide dismutase (SOD), and catalase activity assays were performed in Leydig cells. Results : The results showed that CF scavenged DPPH radical in a dose-dependent manner by up to 81.2%, TF scavenged DPPH radical in a dose-dependent manner by up to 63.8%. CF showed cell viability as 121.0, 132.7, 126.6% in 5, 10, $100{\mu}g/ml$ concentrations. TF showed cell viability as 127.5, 111.8% in 5, $100{\mu}g/ml$ concentraions, respectively. The hydrogen peroxide-induced cytotoxicity of Leydig cells were protected to 86.3% by CF at concentration of $10{\mu}g/ml$ and protected to 83.5% by TF at concentration of $100{\mu}g/ml$. Both CF and TF at all concentrations, SOD activity was not significantly changed. Catalase activity was significantly increased at 10, $100{\mu}g/ml$ concentrations of CF, respectively. TF's catalase activity showed no significant difference from that of the control. Conclusions : These results suggest that CF, as an antioxidant, protects Leydig cells in hydrogen peroxide-induced oxidative stress. know that "Kwangjebikeup" played a role in settlement and spreading of foreign knowledge to civilians.

Radical Scavenging Effect of Methanol Extracts from Seaweeds and Their Active Compounds (해조류 추출물 및 활성성분의 라디칼 소거능)

  • The radical scavenging activity of methanol extracts of seaweeds and their active compounds, alginic acid, fucoidan and phloroglucinol, were investigated under in vitro. Among methanol extracts of seaweeds (sea mustard, sea tangle, seaweed papulosa, fusiforme, sea lettuce, purple laver and chlorella), seaweed papulosa and sea tangle showed strong scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and hydroxyl radical (${\cdot}OH$). In addition, under in vitro, the scavenging activities on DPPH radical of alginic acid and fucoidan, which are active compounds of brown algae, and phloroglucinol, the active compound from Ecklonia species, were evaluated and compared. Fucoidan and phloroglucinol showed strong DPPH scavenging effect, in particular, phloroglucinol had strongest activity among the active compounds. On the other hand, alginic acid did not exert DPPH scavenging activity. From the present study, we could confirm the antioxidative activity of seaweeds and its active compounds.

The Experimental Study of Jeondo-san on Antioxidant Effects (전도산(顚倒散)의 항산화 효과에 관한 실험적 연구)

  • Objectives : This study was carried out to investigate the antioxidant effects of Jeondo-san(JDS). Methods : The antioxidant effects of JDS were measured by the scavenging for 2,2-Diphenyl-1-picrylhydrazyl(DPPH) radical, the formation of intracellular glutathione(GSH), the inhibition for reactive oxygen species(ROS). Results : 1. All concentrations of JDS showed antioxidant effect by decreasing the DPPH radicals. 2. All concentrations of JDS did not effect on the formation of intracellular GSH in HaCaT cell. 3. All Concentrations of JDS inhibited the production of ROS in the HaCaT cell stimulated with $H_2O_2$. Conclusion : The present date suggest that JDS has effects on the stage of inflammation.

Antioxidant Activity and Total Volatile Oil Content of Cassumunar Ginger (Zingiber montanum Roxb.) at Various Rhizome Ages

  • Cassumunar ginger (Zingiber montanum Roxb.) was grown in the experimental field at the Department of Horticulture, Kasetsart University, Thailand. The antioxidant activity and volatile oil content of rhizomes of varying age were measured. Antioxidant activity as determined using the DPPH (diphenyl-2-picrylhydrazyl) method differed significantly between samples of different ages. Antioxidant activity and rhizome age were positively correlated, with 22-month old rhizomes showing the highest radical scavenging activity (79.19%). Volatile oil was obtained by steam distillation of fresh rhizomes. The extraction yield of volatile oil was highest in l6-month old rhizomes (13.02 mL/kg). GC-FID data indicated the presence of three major compounds, sabinene, terpinen-4-ol and (E)-1-(3',4'-dimethylphenyl) butadiene (DMPBD), however none of the major components were correlated with the age of rhizome.

Antioxidant Activity of Substances Extracted by Alcohol from Chungkookjang Powder (분말청국장에서 알코올로 추출한 물질의 항상화능)

  • It is previously reported that Bacillus licheniformis B1 strain isolated from nature was successfully used for Chungkookjang fermentation. Antioxidant activity of its powder was determined in this study. Sephadex G-75 gel filtration chromatography was performed, far soluble fractions of the powder extracted by distilled water. The soluble fractions were separated into large and small fractions. The substance 1,1-diphenyl-2-picrylhydrazyl (DPPH) was used as an electron acceptor. Antioxidant activity was found in the small fractions. Five% solution of the Chungkookjang powder was the most effective in the extraction of antioxidant substances from the powder. It was proven in this study that strong antioxidant activity still remained in the Chungkookjang powder.

  • The antioxidant activity of Artemisia iwayomogi was determined by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of A. iwayomogi showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate > n-butanol > water > chloroform > n-hexane fraction. The ethyl acetate and n-butanol soluble fractions exhibiting strong antioxidant activity were further purified by repeated sitica get and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid was isolated as one of the active principles from the n-butanol fraction, together with the inactive components, 1octacosanol, scopoletin, scopolin, apigenin $7, 4^{I}$-di-O-methylether, luteolin $6, 3^{I}$-di-O-methylether (jaceosidin), apigenin methylether (genkwanin), 2, 4-dihydroxy-6-methoxyacetophenone $4-O-{\beta}-$D-glucopyranoside and quebrachitol. The antioxidant activity of chlorogenic acid was comparable to that of L-ascorbic acid, which is a well known antioxidant.
