Korean Red Ginseng enhances cardiac hemodynamics on doxorubicin-induced toxicity in rats

  • Background: Korean Red Ginseng (KRG) has been known to possess many ginsenosides. These ginsenosides are used for curing cardiovascular problems. The present study show the protective potential of KRG against doxorubicin (DOX)-induced myocardial dysfunction, by assessing electrocardiographic, hemodynamic, and biochemical parameters and histopathological findings. Methods: Animals were fed a standard chow and adjusted to their environment for 3 days before the experiments. Next, the rats were equally divided into five groups (n = 9, each group). The animals were administered with KRG (250 and 500 mg/kg) for 10 days and injected with DOX (20 mg/kg, subcutaneously, twice at a 24-h interval) on the 8th and 9th day. Electrocardiography and echocardiography were performed to study hemodynamics. Plasma levels of superoxide dismutase, catalase, glutathione peroxidase, and malondialdehyde were measured. In addition, the dose of troponin I and activity of myeloperoxidase in serum and cardiac tissue were analyzed, and the histopathological findings were evaluated using light microscopy. Results: Administration of KRG at a dose of 250 and 500 mg/kg recovered electrocardiographic changes, ejection fraction, fractional shortening, left ventricular systolic pressure, the maximal rate of change in left ventricle contraction (-dP/dtmax), and left ventricle relaxation (-dP/dtmax). In addition, KRG treatment significantly normalized the oxidative stress markers in plasma, dose dependently. In addition, the values of troponin I and myeloperoxidase were ameliorated by KRG treatment, dose dependently. And, KRG treatment showed better histopathological findings when compared with the DOX control group. Conclusion: These mean that KRG mitigates myocardial damage by modulating the hemodynamics, histopathological abnormality, and oxidative stress related to DOX-induced cardiomyopathy in rats. The results of the present study show protective effects of KRG on cardiac toxicity.

  • 대두(大豆)중에 존제하는 장내(腸內) 가스 발생인자인 raffinose와 stachyose의 제거에 이용될 수 있는 Aspergillus niger의 ${\alpha}$-galactosidase는 Czapeck-Dox 액체배지와 밀기울 배지에서 5일간 배양하였을 때 효소생성이 제일 높았다. Czapeck-Dox 액체배양에서 raffinose와 $NaNO_3$가 효소생산에 가장 효과적인 탄소원과 질소원으로 나타났으며 밀기울 배양에서는 질소원 또는 탄소원의 첨가효과가 없었다. 생성된 조효소(組酵素)용액의 작용 최적 pH는 4.0-5.0, 안정도 최적 pH는 3.5-6.5, 최적온도는 $40-50^{\circ}C$ 였다. 합성기질인 p-nitrophenyl-${\alpha}$-D-galactoside에 대한 Michaelis상수는 0.42mM, 최대반응속도는 152${\mu}moles$ substrate/min/kg (고체배지)이었다. $HgCl_2$는 강력한 비경쟁 저해제로 작용하였고 p-chloromercuribenzoate는 낮은 농도에서 경쟁 저해제로 작용하였다. 조효소 용액은 raffinose와 stachyose를 완전 분해하여 단당류 만을 생성하였다.

Combined Treatment with 2-Deoxy-D-Glucose and Doxorubicin Enhances the in Vitro Efficiency of Breast Cancer Radiotherapy

  • Doxorubicin (DOX) was introduced as an effective chemotherapeutic for a wide range of cancers but with some severe side effects especially on myocardia. 2-Deoxy-D-glucose (2DG) enhances the damage caused by chemotherapeutics and ionizing radiation (IR) selectively in cancer cells. We have studied the effects of $1{\mu}M$ DOX and $500{\mu}M$ 2DG on radiation induced cell death, apoptosis and also on the expression levels of p53 and PTEN genes in T47D and SKBR3 breast cancer cells irradiated with 100, 150 and 200 cGy x-rays. DOX and 2DG treatments resulted in altered radiation-induced expression levels of p53 and PTEN genes in T47D as well as SKBR3 cells. In addition, the combination along with IR decreased the viability of both cell lines. The radiobiological parameter (D0) of T47D cells treated with 2DG/DOX and IR was 140 cGy compared to 160 cGy obtained with IR alone. The same parameters for SKBR3 cell lines were calculated as 120 and 140 cGy, respectively. The sensitivity enhancement ratios (SERs) for the combined chemo-radiotherapy on T47D and SKBR3 cell lines were 1.14 and 1.16, respectively. According to the obtained results, the combination treatment may use as an effective targeted treatment of breast cancer either by reducing the single modality treatment side effects.

고삼의 인체 유방암세포에 미치는 효과 (Effects of Sophorae Radix on Human Breast Adenocarcinoma Cells)

  • Objective : The purpose of this study was to investigate the anti-cancer effects of Sophorae Radix and the effects of Doxorubicin (DOX) in human breast adenocarcinoma cells (MCF-7). Method : We used human breast adenocarcinoma cell line, MCF-7 cells. We examined cell death by MTT assay and caspase 3 assay with Sophorae Radix. To examine the inhibitory effects of Sophorae Radix, cell cycle analysis was done the MCF-7 cells after three days with Sophorae Radix. The reversibility of Sophorae Radix was examined on one day to five days treatment with 100 ${\mu}g/ml$ Sophorae Radix. Result : Sophorae Radix inhibited the growth of MCF-7 cells in a dose-dependent fashion. Also we showed that Sophorae Radix induced apoptosis in MCF-7 cells by MTT assay, caspase 3 assay and sub-G1 analysis. Sophorae Radix combined with DOX markedly inhibited the growth of MCF-7 cells compared to Sophorae Radix or DOX alone. After 3 days treatment of MCF-7 cells with Sophorae Radix, the fraction of cells in sub-G1 phase was much higher than that of the control group. Conclusion : Our findings provide insight into unraveling the effects of Sophorae Radix in human breast adenocarcinoma cells and developing therapeutic agents against breast cancer.

  • 본 연구는 Phellinus gilvus(PGE), Phellinus linteus (PLE) 그리고 Phellinus baumil(PBE)의 열수추출물에 대하여 SRB법과 MTT법으로 종양세포주(Sarcoma 180과 P388)를 이용하여 항암활성을 비교ㆍ평가하였다. 종양세포주는 PGE, PLE, PBE(7., 15, 30 ug/ml) 그리고 Doxorubicin(DOX)(0.001~10 mM)으로 처리되었다. 그 결과 DOX, PGE 그리고 PLE는 종양세포주에 대하여 농도의존적으로 억제하는 결과를 보였지만, PBEssm 30 ug/ml의 농도에서만 억제되는 항암활성을 보여주었다. 결론적으로 이 연구에서 사용된 PGE, PLE 및 PBE는 Sarcoma 180과 P388에 항암활성을 보여주었다. PLE는 sarcoma 180종양세포에 가장 효과가 뛰어났으나(p<0.05), SRB법에서는 PGE가 P388에 대해 가장 큰 항암활성을 나타내었다(p<0.05).

검정곰팡이의 분화(分化)에 따른 총탄수화물(總炭水化物) 및 트레하로즈량(量)의 변동(變勳)에 대한 연구(硏究) (Studies on Carbohydrate Accumulation in Aspergillus niger During the Differentiation)

  • 검정 공팡이의 분화에 따른 균체내의 총 탄수화물 및 trehalose의 함량변화를 연구하였다. 검정 곰팡이를 Czapeck-Dox액체배지로서 진탕배양과 정치배양법에 의해 배양하여 검정 곰팡이의 각 분화과정에서의 균체내의 총 탄수화물 및 trehalose함량을 Trevelyan및 Harison(1955)의 탄수화물분화 및 정량법에 의해 측정하였다. 검정 곰팡이가 포자를 형성할 때에 총 탄수화물의 양이 증가하였고 trehalose는 특히 포자형성 직전에 극적으로 증가하였다. 또한 배지내의 성분함량의 결핍에 따른 탄수화물량의 변화를 연구하기 위해 본 실험에 사용된 Czapeck-Dox 배지내의 질소원, 인산원, glucose 등의 함량의 제한시 검정 곰팡이 분화과정의 각 단체에서의 탄수화물 및 trehalose함량 변화를 실험하였다. 질소원의 함량을 제한할 때, 총 탄수화물과 trehalose양이 기본 배지에서 배양했을 때보다 증가하였다. 인산원 제한시도 총 탄수화물과 trehalose양이 증가하였으나, 질소원 제한 배지에서 배양했을 때 보다는 뚜렷하지 못했다. 굴루코즈 함량제한시는 총 탄수화물 및 trehalose양의 변동을 관찰할 수 없었다. 이들 총 탄수화물 및 trehalose의 함량변동은 검정 곰팡이의 분화와 관련성이 있는 것으로 해석된다.

Low-dose metronomic doxorubicin inhibits mobilization and differentiation of endothelial progenitor cells through REDD1-mediated VEGFR-2 downregulation

  • Low-dose metronomic chemotherapy has been introduced as a less toxic and effective strategy to inhibit tumor angiogenesis, but its anti-angiogenic mechanism on endothelial progenitor cells (EPCs) has not been fully elucidated. Here, we investigated the functional role of regulated in development and DNA damage response 1 (REDD1), an endogenous inhibitor of mTORC1, in low-dose doxorubicin (DOX)-mediated dysregulation of EPC functions. DOX treatment induced REDD1 expression in bone marrow mononuclear cells (BMMNCs) and subsequently reduced mTORC1-dependent translation of endothelial growth factor (VEGF) receptor (Vegfr)-2 mRNA, but not that of the mRNA transcripts for Vegfr-1, epidermal growth factor receptor, and insulin-like growth factor-1 receptor. This selective event was a risk factor for the inhibition of BMMNC differentiation into EPCs and their angiogenic responses to VEGF-A, but was not observed in Redd1-deficient BMMNCs. Low-dose metronomic DOX treatment reduced the mobilization of circulating EPCs in B16 melanoma-bearing wild-type but not Redd1-deficient mice. However, REDD1 overexpression inhibited the differentiation and mobilization of EPCs in both wild-type and Redd1-deficient mice. These data suggest that REDD1 is crucial for metronomic DOX-mediated EPC dysfunction through the translational repression of Vegfr-2 transcript, providing REDD1 as a potential therapeutic target for the inhibition of tumor angiogenesis and tumor progression.

Influence of Electrolyte Composition on Electrochemical Performance of Li-S Cells

  • The electrochemical performance of Li-S cells was investigated in various ternary electrolyte solutions composed of 1,2-dimethoxyethane (DME), tetra(ethylene glycol) dimethyl ether (TGM), and 1,3-dioxolane (DOX). The discharge capacity values and cycle data obtained at each composition were statistically treated with the Minitab program to obtain mixture contour plots, from which the optimal composition of the ternary solvent systems was predicted. The discharge capacities and capacity retention were quite dependent on the electrolyte composition. It was estimated from the contour plots of the capacity at 1.0 C that the discharge capacity sharply increased with a decrease in the TGM content. High capacities greater than 900 mAh/g at 1.0 C were expected for the electrolyte composition with a volume ratio of DME/TGM/DOX = 1/0/1. In contrast, it was predicted from the mixture contour plot of the capacity retention that the cycle performance would significantly increase with an increase in the DME content.

Anti-tumor Effect of 4-1BBL Modified Tumor Cells as Preventive and Therapeutic Vaccine

  • We have previously reported that genetically modified tumor cells with 4-1BBL have anti-cancer effects in a CT26 mouse colorectal tumor model. In this study, genetically modified tumor cells with 4-1BBL were evaluated for their potential as candidates for preventive and therapeutic cancer vaccine. To identify the effect of preventive and therapeutic vaccine of genetically modified tumor cells with 4-1BBL, tumor growth pattern of CT26-4-1BBL as a cancer vaccine was examined compared to CT26-beta-gal. In therapeutic vaccination, CT26-WT was inoculated into mice and then vaccinated mice with doxorubicin (Dox)-treated CT26-beta-gal and CT26-4-1BBL (single or three times). Triple vaccination with Dox-treated tumor cell inhibited tumor growth compared to single vaccination. Vaccination with CT26-4-1BBL showed an efficient tumor growth inhibition compared to vaccination with CT26-beta-gal. For preventive vaccination, Dox-treated CT26-beta-gal and CT26-4-1BBL was vaccinated into mice with three times and then administered mice with CT26-WT. Preventive vaccination with CT26-4-1BBL showed no tumor growth. Preventive vaccination with CT26-beta-gal also led to tumor-free mice. These results suggest that genetically modified tumor cells with 4-1BBL can be used as therapeutic or preventive cancer vaccine.

퍼지앤트랩-기체크로마토그래피(PT-GC)를 이용한 식품첨가물 중 1,4-디옥산 및 클로로히드린류 분석 (Analysis of 1,4-Dioxane and Chlorohydrins in Food Additives by Purge & Trap GC)

  • 폴리소르베이트류, 염화콜린, 주석산수소콜린, 변성전분 및 향신료 중 1,4-디옥산 및 클로로히드린류의 분석법을 개발하고 확립된 방법으로 잔류량을 조사한 결과는 다음과 같다. 분석에 사용된 트랩으로는 Vocarb 3000이 적합하였고 이때의 검출한계는 DOX 1.38$\mu\textrm{g}$, EPC 0.23$\mu\textrm{g}$, PCH 3.30$\mu\textrm{g}$, ECH 3.97$\mu\textrm{g}$, DCP 20.43$\mu\textrm{g}$이었다. 검체 온도증가와 무수황산나트륨 농도 증가에 따라 분석감도는 증가하였으나 6$0^{\circ}C$ 이상에서는 상대표준편차(RSD, %)가 6%를 초과하여 재현성이 떨어져 분석온도를 5$0^{\circ}C$로 설정하였다. 폴리소르베이트류에서의 회수율은 염을 첨가하지 않고 측정하였을 때 EPC(66%)를 제외하고는 90%이상을 나타내었다. 염화콜린 및 주석산수소콜린의 경우에는 대부분 회수율이 100% 이상이었으나 주석산수소콜린의 EPC는 20%로 낮게 나타났다. 히드록시프로필인산이전분, 히드록시프로필전분 및 찰옥수수전분에서는 검체를 가수분해한 후 회수율을 측정한 결과 검체종류별 큰 차이 없이 회수율이 90% 이상을 나타내었다. 향신료의 분석에서는 무수황산나트륨을 첨가하여 분석한 결과 회수율이 90%이상이었다. 구매한 폴리소르베이트류, 염화콜린, 주석산수소콜린, 향신료 중 1,4-디옥산 및 클로로히드린류를 분석한 결과 트윈 80에서 DOX가 2.5ppm 검출되었으나 이외의 검체에서는 모두 불검출되었고 직접 제조한 히드록시프로필전분에서도 모두 불검출되었다.
